방풍통성산(防風通聖散)이 아토피 피부염을 유발한 동물모델의 피부 손상에 미치는 영향 (The Effects of Bangpungtongsungsan Extract to the Skin Damage on Mice Model after Atopic Dermatitis Elicitation)

  • Objectives : Atopic dermatitis has a close relationship with damage of skin barrier function. To investigate the effects of Bangpungtongsungsan(BT) extract to the skin damage on mice model after atopic dermatitis elicitation, this study was done through forcing injury to mice's skin. Methods : The BALB/c mice were distributed into three groups: control(CON) group, atopic dermatitis(AD)-elicited group, Bangpungtongsungsan(BT)-treated group. AD-elicited and BT-treated group were caused AD according to the method of Christophers E., Mrowietz and Minehiro. The BT extract was administered for 48 hours to BT-treated group. We observed changes of external dermal formation, eosinophils in vasculature, lipid formation in stratum corneum, distribution of ceramide, distribution of capillary, $I{\kappa}B$ kinase(IKK) and induce nitric oxide synthase(iNOS) mRNA expression. We used the statistical methods of student t-test(p<0.05). Results : After dispensing BT extract into the AD-elicited group, the number of eosinophil as an atopic index in mice noticeably decreased and dermal injury decreased. Also the decrease of hyperplasia, degranulated mast cells, angiogenesis and substance P were shown. The lipid lamellae, lipid protect formation, were repaired and the distribution of ceramide which inhibit protein kinase C(PKC) activation increased, and the PKC caused inhibition of nuclear $factor(NF)-{\kappa}B$ activation. As a result of inhibition of $NF-{\kappa}B$ activation, iNOS production were inhibited and apoptotic cell were increased. Moreover the decrease of IKK and iNOS mRNA expression in BT-treated RAW 264.7 cell were noted. Conclusion : BT mitigated skin damage on mice model after atopic dermatitis elicitation through recovering skin barrier function and inhibiting nuclear $factor(NF)-{\kappa}B$ activation.

Efficacy of Hataedock Treatments for Maintenance and Formation of Lipid Barrier in Obese NC/Nga Mice with Dermatophagoides Farinae-Induced Atopic Dermatitis

  • Objectives: HTD treatment is a traditional preventive therapy for neonatal inflammatory diseases such as AD. The aim of this study was to investigate the efficacy of HTD treatments for the maintenance and formation of lipid barrier in Dermatophagoides farina-induced obese NC/Nga mice. Methods: 20 mg/kg of CRGR extracts as HTD treatments were orally administered to NC/Nga mice. To induce obesity, high fat diet was served. Dermatophagoides farina extracts was applied on the 4th-6th and 8th-10th weeks to induce AD-like skin lesions in NC/Nga mice. Changes of skin conditions in mice were observed by histochemistry and immunohistochemistry. Results: The results showed that HTD treatments effectively maintained and formed the lipid barrier. In the experimental groups, restorations of Lass2 expression and distributions of filaggrin, involucrin, loricrin, ASM, and LXR means that HTD treatments maintained and generated the lipid barrier. In the dermal papillae, HTD treatments reduced PKC production accompanied by epidermis damage. Furthermore, levels of IL-4, and STAT6 was low. HTD treatment may be effective for preventing inflammation induced by Th2-skewed condition by suppressing the main pathway of Th2 differentiation. Conclusions: HTD treatment alleviated the inflammatory damage in the skin tissues of the NC/Nga mice by maintaining the lipid barrier and suppressing Th2 differentiation.

Adenophorae Radix 뿌리 추출물에 의한 Keratinocyte의 분화 및 피부장벽 기능에 대한 연구 (Study on Keratinocyte Differentiation and Skin Barrier Function of Adeonphorae Radix Root Extracts)

  • Triterpenoid, saponin, 전분 등이 함유되어 있는 것으로 알려진 Adenophorae radix (A. radix)의 뿌리 추출물을 이용하여 각질형성세포의 분화와 피부장벽기능에 대한 연구를 수행하였다. A. radix의 뿌리 추출물은 CV-1 세포를 이용하여 $PPAR{\alpha}$ 발현을 살펴본 결과, Wy-14,643 $0.5-1.0{\mu}M$ 수준의 발현양을 나타내었다. 인체 각질형성 세포주(HaCaT)와 각질형성세포(nomal human keratinocyte)에 대한 각질형성능(cornified envelop formation, CE)은 대조군에 비해 통계적으로 유의한 증가를 보였다. HaCaT 세포에 A. radix의 뿌리 추출물 처리하였을 때, transglutaminase (TGase-1)의 유의적 증가를 보였다. A. radix의 뿌리 추출물을 함유한 간단한 화장품 제형을 약 2주간에 걸쳐 임상시험을 실시한 결과, TEWL의 유의적 감소와 수분량의 증가를 살펴볼 수 있었으며, 하박 내측에서 지질을 추출하여 세라마이드를 분석한 결과 통계적으로 유의한 증가를 관찰할 수 있었다. 이를 통하여 A. radix의 뿌리 추출물을 건조피부나 아토피 등의 피부질환과 관련된 질환의 예방 및 치료제로 사용될 수 있을 것이다.

Enhancement of skin barrier and hydration-related molecules by protopanaxatriol in human keratinocytes

  • Background: Protopanaxatriol (PPT) is a secondary intestinal metabolite of ginsenoside in ginseng. Although the effects of PPT have been reported in various diseases including cancer, diabetes and inflammatory diseases, the skin protective effects of PPT are poorly understood. Methods: HaCaT cells were treated with PPT in a dose-dependent manner. mRNA and protein levels which related to skin barrier and hydration were detected compared with retinol. Luciferase assay was performed to explore the relative signaling pathway. Western blot was conducted to confirm these pathways and excavated further signals. Results: PPT enhanced the expression of filaggrin (FLG), transglutaminase (TGM)-1, claudin, occludin and hyaluronic acid synthase (HAS) -1, -2 and -3. The mRNA expression levels of FLG, TGM-1, HAS-1 and HAS-2 were suppressed under NF-κB inhibition. PPT significantly augmented NF-κB-luc activity and upregulated Src/AKT/NF-κB signaling. In addition, PPT also increased phosphorylation of the mitogen-activated protein kinases (MAPKs) ERK, JNK and p38 and upstream MAPK activators (MEK and MKK). Furthermore, transcriptional activity of AP-1 and CREB, which are downstream signaling targets of MAPK, was enhanced by PPT. Conclusion: PPT improves skin barrier function and hydration through Src/AKT/NF-κB and MAPK signaling. Therefore, PPT may be a valuable component for cosmetics or treating skin disorders.

Improvement of Skin Photoaging by Polysaccharide Extract Derived from Tremella fuciformis (White Jelly Mushroom)

  • Chronic ultraviolet (UV) radiation causes photoaging, which represents skin damage, disrupts skin barrier function, and promotes wrinkle formation. We investigated that the polysaccharide extract of an edible basidiomycetous white jelly mushroom, Tremella fuciformis, (TF-Glucan®) exhibited statistically photoprotective activity by inhibiting matrix metalloproteases (MMPs) and increasing collagen synthesis, and an anti-inflammatory activity by inhibiting nitric oxide and pro-inflammatory cytokines at the concentrations of less than 1000 ㎍/ml, which is not cytotoxic (p < 0.05). Additionally, TF-Glucan® increased the expression of involucrin and filaggrin to prevent the disruption of UVB-induced barrier function (p < 0.05). TF-Glucan® was assessed as a safe material by the human primary skin irritation (1, 3, 5%), human repeated insult patch test (no sensitization at 5%), 3T3 NRU phototoxicity assay (no phototoxicity, PIF < 2, MPE < 0.1), eye irritation test test by BCOP (no category, IVIS ≤ 3) and local lymph node assay (negative at 10, 25, 50%) for identifying potential skin sensitizing. These results suggest that TF-Glucan® may be useful as an anti-photoaging ingredient for developing cosmeceuticals.

Effect of Skin Lipid Barrier Formation on Hataedock Treatment with Douchi

  • Objectives: Hataedock is indigenous treatment of Korean medicine that administers herbal extracts orally to newborn infants for remove the fetal heat. The purpose of this study was to evaluate skin lipid barrier formation effect of Hataedock treatment with Douchi. Methods: We measured the Western blot to observe the expression of protein such as involucrin and loricrin. Moreover, we observed immunohistochemical changes in NC/Nga mice. The 3-week-old NC/Nga mice were divided into 3 groups: the 3-week-old control group (3w-Ctrl), 5-week-old control group (5w-Ctrl), and the Hataedock-treated group (5w-FGT). Only the 5w-FGT group was treated with Douchi at the 3rd week. We identified the changes of the lipid skin barrier and protein differentiation through immunohistochemical changes of involucrin, loricrin, filaggrin and acid sphingomyelinase (ASM) in the stratum corneum. Results: The expression of involucrin and loricrin was increased in the Western blot that was treated with concentration of Douchi extracts. In 5w-FGT group, loricrin-positive reaction was increased by 54.0%, involucrin-positive reaction was increased by 84.0%, filaggrin-positive reaction was increased by 108.0% and ASM-positive reaction was increased by 91.0% in the stratum corneum. Conclusions: These results suggest that Hataedock treatment with Douchi promoted skin lipid barrier formation by promoting differentiation of keratinocytes.

각질형성세포에서 Fisetin의 피부장벽 기능 개선 및 항노화 효능 검증 (Roles of Fisetin on Skin Barrier Function and Anti-aging in Epidermal Keratinocyte)

  • 플라보노이드(flavonoid)는 식물 등의 대사체에서 유래한 폴리페놀 계열의 화합물이며, 다양한 인체생리작용을 조절할 수 있는 것으로 알려져 있다. 이중 3,3',3',7-tetrahydroxyflavone (fisetin)은 다양한 과일과 채소에서 발견되며, 최근 노쇠용해(senolytic) 활성을 통해 특정 조직의 기능을 회복시킨다는 것이 알려졌다. 본 연구에서는 인간 표피 각질세포를 대상으로 하여 fisetin의 피부장벽 유전자 발현 조절 및 항노화 효능을 분석하였다. Fisetin은 말단소립 역전사효소(telomerase)의 활성을 증가시켰으며, CDKN1B 유전자의 발현을 감소시켰다. 또한 피부장벽을 구성하는 주요 유전자인 KRT1, FLG, IVL, DSP의 발현을 증가시켰으며, 세라마이드 합성효소의 일종인 CerS3, CerS4 유전자의 발현을 증가시켰다. 이러한 결과는 fisetin의 효능이 노쇠용해에 국한되지 않고 인간 각질세포의 다양한 생리학적 조절에도 관여함을 보여준다. 따라서 fisetin은 화장품 및 의약품 등의 생리활성 조절물질로 활용될 수 있다고 사료된다.

The preparation of skin analogue composition having the liquid crystalline structure and its cosmetic applications

  • Recently, many cosmetic researchers have been focused on the development of high functional cosmetics including anti-wrinkle and whitening. In these studies, they couldn't afford to pay a deep attention to stable encapsulations for unstable materials and efficient drug deliveries for them. Particularly, in order to show a degree of instant effects as cosmetics, they can't also ignore moisturizing effect enough to satisfy customers just after applying and its maintenance by improving the function of skin barrier as well as above two effects. Therefore, skin analogue systems have attracted considerable attention in the view of structural and compositional similarity to intercellular membrane in stratum corneum. And, some models for skin analogue composition were developed to improve the function of skin barrier, stably encapsulate unstable materials such as retinol, vitamin B, C, E, etc., and control their skin penetration in order to show good effects as cosmetics. In this study, we suggest the new skin analogue model having the compositional similarity as well as conventional structural ones. Our skin analogue membrane(SAM) is mainly composed of ceramide/ cholesterol/phosphatidylcholin/fatty acids and its structural defects are compensated by including cholesterol amphiphile and controlling the ratio of ceramide/cholesterol. It was possible to confirm the formation of skin analogue membrane having highly-densed multilamella structure and compare them according to the change of each ratio with a polarized microscope, X-ray diffraction. More detaily, we observed their structures with a electron microscope(TEM). Finally, we dispersed them in excess of continuous water phase, observed the formation of maltese-cross liquid crystalline and measured the efficiency of drug deliveries and moisturizing effects.

Effect of Standardized Boesenbergia pandurata Extract and Its Active Compound Panduratin A on Skin Hydration and Barrier Function in Human Epidermal Keratinocytes

  • The skin plays a key role in protecting the body from the environment and from water loss. Cornified envelope (CE) and natural moisturizing factor (NMF) are considered as the primary regulators of skin hydration and barrier function. The CE prevents loss of water from the body and is formed by cross-linking of several proteins. Among these proteins, filaggrin is an important protein because NMF is produced by the degradation of filaggrin. Proteases, including matriptase and prostasin, stimulate the generation of filaggrin from profilaggrin and caspase-14 plays a role in the degradation of filaggrin. This study elucidated the effects of an ethanol extract of Boesenbergia pandurata (Roxb.) Schltr., known as fingerroot, and its active compound panduratin A on CE formation and filaggrin processing in HaCaT, human epidermal keratinocytes. B. pandurata extract (BPE) and panduratin A significantly stimulated not only CE formation but also the expression of CE proteins, such as loricrin, involucrin, and transglutaminase, which were associated with $PPAR{\alpha}$ expression. The mRNA and protein levels of filaggrin and filaggrin-related enzymes, such as matriptase, prostasin, and caspase-14 were also up-regulated by BPE and panduratin A treatment. These results suggest that BPE and panduratin A are potential nutraceuticals which can enhance skin hydration and barrier function based on their CE formation and filaggrin processing.
