Evaluation Method of Cosmetics for the Effect of Fine Dust Adhesion Prevention Using Floating Chamber (부유챔버를 이용한 화장품의 미세먼지 부착방지 효과 평가법)

  • Particulate matters (PM) are small particulate pollution that decrease the function of skin barrier, which causes inflammatory skin diseases and extrinsic aging. In this study, we evaluated the effect of preventing the adherence of PMs from several cosmetic products applied to human skin using iron oxide black. The PM floating chamber consists of skin exposure area, PM inlet, floating power device, and an outlet so that PM can be naturally attached to the skin while floating in the chamber. The change in skin brightness according to the floating concentration of alternative fine dust was checked to confirm the optimal floating concentration conditions. The intensity difference (before-after intensity, Δ) before and after adhesion of iron oxide black was proportional to the amount of PM adhered. The anti-adherence effect of iron oxide black on five cosmetic products were evaluated through 20 each subjects by comparing the amount of iron oxide black adhered on the control and treatment. The difference in brightness before and after the iron oxide black attached to the skin was calculated and compared with the control group(p < 0.05). When over 150 mg of iron oxide black was adhered on the skin, the interference of intensity was low and clearly showed the skin adhered pattern. According to the application of the five cosmetics, the intensity difference was significantly lower than the control group. This means that depending on the product, it prevented the attachment of iron oxide black. This study is a safe and useful method to confirm the prevention of PM skin adherence. In conclusion, cosmetics can prevent the adherence of PM on the skin according to the formulation or ingredients characteristics.

  • The skin barrier function is one of the vital functions. Horny layer and epidermis among various skin layers play an important role in protecting body from environmental insults. Stratum corneum is completely formed by differentiation processes from the basal layer and newly formed corneocytes push the older ones from skin surface. In this study we investigated changes of various parameters - capacitance, transepidermal water loss (TEWL), pH and amount of removed corneocytes - according to the stratum corneum's depth. We removed the stratum corneum using tape stripping technique. We found that uppermost stratum corneum play an important role in skin surface moisturization and lower stratum corneum in protecting transepidermal water loss. In addition, skin pH is more acidic than surface pH. In the stratum corneum, skin pH is mote acidic in the deeper layer. Furthermore amount of removed corneocyte is also decreased according to depth because of stronger cell-cell union strength. From these results we suggest that stratum corneum has different characteristics depending on their position.

Protective Effect of Cosmetics Containing Red Beet against Cigarette Smoke-induced Oxidative Damage in Human Skin (레드비트를 함유하는 화장품의 담배 연기에 의한 피부 지질 산화 방지 효과)

  • In cosmetics market, anti-pollution products recently come up with new solution for skin health. Environmental oxidation mechanisms are realized as bio-marker of atmospheric pollution upon skin by environmental pollutant such as ozone, UV rays, particulate matter (PM) as well as cigarette smoke. The exposure of cigarette smoke directly or indirectly causes the oxidation of the stratum corneum skin lipids, resulting in the conversion of squalene to squalene monohydroperoxide and/or generation of malondialdehyde (MDA) as a product of lipid peroxidation. The aim of this study is to see whether new cosmetics product containing red beet has anti-oxidation effect on skin exposed by cigarette smoke. So as to determine oxidative damage to human skin at biochemical level, each unit area of volar forearms was exposed to cigarette smoke through device (3.3 cm, diameter) for fifteen minutes, then measured MDA using standardized TBARS assay kit. Compared to negative control (untreated and unexposed area), the level of MDA was significantly increased at positive control (untreated and exposed area) more than 3.7 times, indicating the pollutant induced-oxidative damage on the skin barrier. Whereas, the pre-applied area with the cosmetics products containing red beet revealed a decrease of 25% compared with positive control. As reports, these data demonstrated that cigarette smoke induce peroxidation of stratum corneum skin lipids. Conclusively, we suggest that anti-pollution effect of the cosmetics product containing red beet is beneficial to prevent the oxidation of skin lipids by atmospheric pollution.

  • Objective: A case report on the improvement of athlete's foot using Ortho-Cellular Nutrition Therapy (OCNT). Methods: A Korean male in his 50s is suffering from athlete's foot for 10 years. Results: Keratinization of the soles and deformity of the toenails were alleviated after undergoing OCNT. Conclusion: Application of OCNT to patients with athlete's foot can help protect the skin barrier and improve symptoms.

Triterpenoid-Containing Liposome by Micelle-to-Vesicle Transition and Their Biological Activities

  • Ursolic acid (UA) and oleanolic acid (OA) are pentacyclic triterpenoids which are widely distributed in plants, and their derivatives are aglycones of many naturally occurring saponins. It is known that pentacyclic acids may possibly enhance the mechanical barrier functions of cell membranes in plants. Recently, it has been reported that OA and UA have interesting biological activities on skin, such as anti-inflammatory and anti-wrinkle activities. Since triterpenoids are extremely insoluble and their solubility problem limits skin-care application, OA and UA were encapsulated in liposomes via micelle-to-vesicle transition to overcome poorly soluble property and enhance biological efficacy. Optimal molar ratio of OA to lecithin was found to exist for producing liposomes of small hydrodynamic size and liposomal suspensions without recrystallized precipitation of OA. From electron micrograph and dynamic light scattering studies, reconstituted OA-containing liposomes without severe mechanical treatment showed small hydrodynamic size about 150 nm. Wide-angle X-ray diffraction coupled with dynamic light scattering revealed that optimal amount of OA in liposome was 25.4 mole %. In biological evaluation, OA-containing liposome significantly increased filaggrin and transglutaminase as markers of keratinocyte differentiation in epidermal layer of hairless mouse, whereas ursolic acid-containing liposome did not show noticeable increase of filaggrin and transglutaminase compared to empty liposome. It is concluded that nano-scaled liposomes containing triterpenoids were spontaneously prepared by vesicular transition from mixed micelle and liposomal triterpenoids can enhance skin absorption of triterpenoid and biological efficacy.

A Case of Acral Lick Dermatitis in a Dog with Multiple Lesions

  • A 7-year-old intact male Maltese dog was presented with firm, raised, erythematous, alopecic, and pruritic skin lesions on the right dorsal distal metatarsal region and the tail. The skin lesion started a year ago with a crust-like lesion on the right dorsal distal metatarsal region which gradually swelled as the patient continued to lick and chew without healing. Recently, similar lesions occurred on the tail. Based on the licking history and ruling out other diseases through skin examination, acral lick dermatitis (ALD) was diagnosed. Treatment included antidepressants, preventive antibiotics, topical corticosteroid, and wearing Elizabethan collar as a physical barrier. After three weeks, the lesion on the hind limb healed entirely. However, the tail lesion recurred as it was able to lick the tail again with a type of an E-collar. This case shows that it is essential to block the contact lesion and provide medical treatment until the lesion has been completely resolved for successful management of ALD.

Effects of Esculetin on TNF-α Induced MMP-1 Expression in Human Fibroblasts Hs68 (인간섬유아세포 Hs68에서 esculetin이 TNF-α로 유도된 MMP-1 발현에 미치는 효과)

  • The skin is an important barrier that protects the body from harmful environments such as UV rays. When the skin is repeatedly stimulated, such as UV rays, ROS and pro-inflammatory cytokines are overproduced. As a result, the proteins and nucleic acids that make up the skin are damaged, and aging occurs. Esculetin is known to have anti-inflammatory, antioxidant and UV-induced MMP-1 inhibitory effects. However, the inhibitory effect of MMP-1 on TNF-α-induced fibroblasts is not known. Therefore, in this study, the MMP-1 inhibitory effect of esculetin was confirmed in TNF-α-induced fibroblasts. As a result of confirming the cytotoxicity of esculetin in Hs68 cells by MTT assay, there was no significant toxicity up to 200 µM. As a result of real-time PCR and ELISA, it was confirmed that esculetin inhibited the expression of MMP-1. Esculetin did not inhibit MAPK (ERK, JNK, p38) phosphorylation, but inhibited phosphorylation of the mTOR-p70S6k signaling pathway. In addition, it was confirmed that the phosphorylation of the transcription factor NF-κB was inhibited. These results suggest that esculetin has potential as an anti-aging material.

Characteristics of Percutaneous Absorption for Three Kinds of Phthalate (Phthalate 3종에 대한 경피투과 특성 연구)

  • Objectives: Phthalates are used in a large variety of products including as coatings of pharmaceutical tablets, film formers, stabilizers, dispersants, emulsifying agents, and suspending agents. They have been the subject of great public concern in recent years. The extensive uses of this material have attracted attention and issues regarding its safety have been raised. Methods: In this study, three types of phthalate skin permeation were studied using matrixes such as ointments, creams and lotions in vitro. The absorption of phthalate diesters [Dimethyl phthalate (DMP), Di-n-propyl phthalate (DPP) and Di-n-pentyl phthalate (DNPP)] using film former has been measured in vitro through rat skin. Epidermal membranes were set up in Franz diffusion cells and their permeability to PBS measured in order to establish the integrity of the skin before the phthalates were applied to the epidermal surface. Results: Absorption rates for each phthalate ester were determined and permeability assessment made to quantify any irreversible alterations in barrier function due to contact with the esters. Types of phthalate in vitro experimental results quickly appeared in the following order DMP > DPP ${\geq}$ DNPP. Conclusions: In the experimental results, lotion> cream> ointment, and the permeation rate of lotion with a great amount of moisture was the fastest. Skin permeation rate is generally influenced by the chemical characteristics of a given chemical, such as molecular weight and lipophilicity. As the esters became more lipophilic and less hydrophilic, the rate of absorption decreased.

  • Long-term exposure to environmental pollutants can impair the human skin's barrier function and promote skin aging mechanisms. The visible consequences of these effects are dryness, wrinkles, black spots and worsening skin sensitivity. As awareness of the effects of environmental stressors on the skin has recently increased, consumers' demand for cosmetics that can provide anti-pollution effects is increasing. In this study, the possibility of anti-pollution cosmetic material was investigated by measuring the inhibitory effect of free radicals using a mixture of Nypa fruticans Wurmb (NF), Saussurea neoserrata (SN), Codium fragile (CF) and Enteromorpha compressa (EC), which has anti-inflammatory and antioxidant effects against irritation caused by dust. Combining the experimental results of a mixture of NF, SN, CF and EC by induction of fine dust stimulation, inhibition of inflammatory factors (PGE2) and intracellular free radicals (ROS) by inhibiting effects were significant, indicating the possibility of use as anti-pollution cosmetics. The most plants used as sources of anti-pollution cosmetic ingredients contain antioxidants as active substances. In our highly industrialized and chemically polluted world, it is not surprising that most plants used as sources of anti-pollution cosmetic ingredients contain antioxidants as active substances. Considering our results, it can be suggested that this anti-pollutant consisting of NF, SN, CF and EC may be a good ingredient for skincare products for the cosmetic industry due to their antioxidant properties, which may especially alter skin aging.
