Fructose 1.6-diphosphate Prevents Cyclooxygenase-2 and Matrix Metalloproteinases Expression by Inhibition of UVB-induced Signaling Cascades in HaCaT Keratinocytes (인체각질형성세포에서 Fructose 1,6-diphosphate의 자외선에 의해 유도되는 Cyclooxygenase-2 and Matrix Metalloproteinases의 발현억제기전)

  • UV radiation exerts various influences in the skin, including photoaging and inflammation (1). The MMPs (Matrix metalloproteinases), which are induced by UV irradiation, can degrade matrix proteins, and these results in a collagen deficiency in photodamaged skin that leads to skin wrinkling. It has been known that the production of PGE$_2$ stimulates MMPs expression, and inhibits procollagen (2). Thus, it is possible that the induction of MMPs and the inhibition of matrix protein synthesis by UV -induced PGE$_2$ may play some role in UV-induced collagen deficiency in photoaged skin. Fructose-1,6-diphosphate (FDP), a glycolytic metabolite, is reported to have cytoprotective effects against ischemia and postischemic reperfusion injury of brain and heart, presumably by augmenting anaerobic carbohydrate metabolism (3). And also, FDP significantly prevent skin aging by decreasing facial winkle compared with vehicle alone after 6 months of use. We studied the mechanism of anti-aging effect of FDP on UVB-irradiated HaCaT keratinocyte model. FDP has protective role in UVB injured keratinocyte by attenuating prostaglandin E$_2$ (PGE$_2$) production and COX-2 expression. And FDP also suppressed UVB-induced MMP-2 expression. Further, to delineate the inhibition of UVB-induced COX-2 and MMPs expression with cell signaling pathways, treatment of FDP to HaCaT keratinocytes resulted in marked inhibition of UVB-induced phosphorylation of ERK1/2, JNK. It also prevents UV induced NFB translocation, which are activated by cellular inflammatory signal. Our results indicate that FDP has protecting effects in UV-injured skin aging by decreasing UVB-induced COX-2 and MMPs expression, which are possibly through blocking UVB-induced signal cascades.

Antioxidant and Anti-wrinkling Effects of Aruncus dioicus var. kamtschaticus extract (눈개승마 추출물의 항산화 및 주름개선 효과)

  • In this study, the antioxidant and anti-wrinkling effects of extracts from Aruncus diocius var. kamtschaticus (ADV) were investigated. According to the results, the ethanol extract has better antioxidant and anti-wrinkling effects than the water extract. The amounts of total polyphenol and flavonoid compounds in the ethanol extract were 122 and 36 mg/g, respectively, while those in the water extract were 87 and 26 mg/g. The antioxidant activities of the ethanol and water extracts were 395 and 4,682 ${\mu}g/mL$ as the $RC_{50}$ values for the DPPH radical scavenging activity, and 227 and 366 ${\mu}g/mL$ for the $ABTS^+$ radical scavenging activity, respectively. The reducing power of the ethanol extract (1.58 at 2 mg/mL) was higher than that of the water extract (0.88 at 2 mg/mL). The astringent activities of the ethanol and water extracts were 91.27 and 16.35% at 10 mg/mL, respectively. Furthermore, the ADV ethanol extract treatment of the fibroblast cell after UV irradiation resulted in increased cell viability (10% at 100 ${\mu}g/mL$) and collagen biosynthesis (33% at 100 ${\mu}g/mL$), with a lowering in the MMP-1 expression level (16.8 % at 100 ${\mu}g/mL$). These results demonstrate that AVD provides a remarkable and significant tensor and anti-wrinkling effect on the skin, which could be of a great use in anti-aging skin care products.

Beneficial Effect of a Collagen Peptide Supplement on the Epidermal Skin Barrier (콜라겐 펩타이드의 피부 장벽 보호 효과)

  • Recent studies have revealed that collagen peptide (CP) plays a protective role in skin by improving the activity of antioxidants and acts as an inducer of skin regeneration by positive feedback. In this study, we focused on the beneficial effect of reinforcing the CP skin barrier. To evaluate the skin barrier, hairless mice were exposed to UVB irradiation and acetone-treatment, with or without oral administration of CP. The effects on skin appearance, trans-epidermal water loss, epidermal thickness, and cytokine content were measured using bioengineering and histochemical methods. In the CP treated group, the skin had better appearance and less damage than that of the control. Furthermore, in HaCaT cells, the amount of serinepalmytoyl transferase (SPT) mRNA increased by about 1.6-fold after treatment (CP, 100 mg/L), reflecting that CP can induce SPT expression and reinforce the recovery of skin barrier function. These results suggest that CP is not only an anti-wrinkling agent but also a potent candidate as an epidermal moisturizer.

The Level of UVB-induced DNA Damage and Chemoprevention Effect of Paeoniflorin in Normal Human Epidermal Kerationcytes

  • Ultraviolet (UV) radiation to mammalian skin is known to alter cellular function via generation of Reactive Oxygen Species (ROS), DNA damage and DNA lesions, such as pyrimidine dimmers and photoproducts, which could lead to DNA mutation if they are not repaired. In this study, we have investigated the reduction of DNA damage and of apoptosis with a particular attention to genetic effect of paeoniflorin in Normal Human Epidermal Keratinocytes (NHEK). After UVB irradiation from $10\;to\;500mJ/cm^{2}$ to NHEK, Mean Tail Moments (MTM) were increased with UVB dose increase. The greatest amount of strand breaks was induced at $500mJ/cm^{2}$ of UVB. Even at the lowest dose of UVB ($10mJ/cm^{2}$), change in MTM was detected (P<0.0001). Pretreated cell with 0.1% paeoniflorin maximally reduced the level of DNA damage to about 21.3%, compared to untreated cell. In the lower concentrations less than 0.01% of paeoniflorin, MTM had a small increase but paeoniflorin still had reductive effects of DNA damage. We measured the apoptosis suppression of paeoniflorin with annexin V flous staining kit. As we observed under the fluorescence microscopy to detect apoptosis in the irradiated cell, the fluorescence intensity was clearly increased in the untreated cell, but decreased in treated cells with paeoniflorin. These results suggest that paeoniflorin reduces the alteration of cell membranes and prevents DNA damage. Therefore, the use of paeoniflorin as a free radical scavenger to reduce the harmful effects of UV lights such as chronic skin damage, wrinkling and skin cancer can be useful to prevent the formation of photooxidants that result in radical damage.

  • DA-3711 is a novel anti-wrinkle agent containing growth factors derived from culture medium of artificial human skin. Photoprotective effect by DA-3711 against chronic UVB (ultraviolet B)-induced skin damage was investigated in hairless mice model. Methods: After hairless mice were irradiated to induce photodamage for 8 weeks with UVB, grouped mice were treated topically once a day with lotion base, DA-3711 (30% or l5%), Cylasphere retinol$^{\circed{R}}$ (2500 I.U.), NouriCel$^{\circed{R}}$ along with concomitant exposure to UVB for further 8 weeks. Then mice were sacrificed to assess photodamage-protective effect by replica analysis, biochemistry and histology. DA-3711 of 30% lotion significantly reduced UVB radiation-induced wrinkling, histological alterations and increased collagen contents. Whereas DA-3711 of l5% lotion and NouriCel$^{\circed{R}}$ treatment showed a partial protective effect on skin wrinkle, epidermal and dermal thickness, and collagen content, Cylasphere retinol$^{\circed{R}}$ showed no protective effects. These results demonstrate that topical application of DA-3711 can alleviate UVB-induced photodamage and potentially be used for reduction of UVB-induced photodamage.

Enhancement of Anti-wrinkle Activities of Abeliophyllum distichum Nakai through Low Temperature Extraction Process (미선나무의 저온 추출 공정에 의한 항주름 활성 증진)

  • Skin anti-wrinkle activities of the stems and leaves of Abeliophyllum distichum Nakai were evaluated by the extracts obtained from various extraction processes such as using hot water at $100^{\circ}C$, 70% ethanol at $85^{\circ}C$, and 70% ethanol with ultrasonication at $60^{\circ}C$ The ultrasonicated extract showed 95.62% of the highest cell viability in addition of $0.3mg/m{\ell}$ of the extracts into the normal human fibroblast cell, CCD-986sk. For antioxidant activities, the extracts using ultrasonicated extract showed the highest DPPH free radical scavenging as 80.27%, followed by 75.88% and 62.44% for the extracts using ethanol extract and water extract. The ultrasonicated extract also showed the highest elastase inhibition activity as 25.32%, compared to ethanol extract and water extract based method at 22.01% and 12.88%, respectively. MMP-1 production was most effectively decreased down to $2908.1pg/m{\ell}$ with ultrasonicated extract while $6640.8pg/m{\ell}$ with water extract and $3609.3pg/m{\ell}$ with ethanol extract, in addition of $0.3mg/m{\ell}$. Collagen production was increased up to $154.7ng/m{\ell}$ in addition of ultrasonicated extract, and followed by $121.4ng/m{\ell}$ and $31.2ng/m{\ell}$ for ethanol extract and water extract, respectively. These results indicate that the ethanol extract should have skin anti-wrinkling activities and can be improved by the ultrasonication process that high energy input elute more amounts of bioactive substances eluting more amounts of bioactive substances from the high energy input of ultrasonication.

Beneficial Effect of Collagen Peptide Supplement on Anti-aging Against Photodamage (콜라겐 펩타이드의 피부 광노화 예방 효과)

  • Recent research has revealed that hydrolyzed collagen peptides have beneficial effects in various diseases such as osteoarthritis and human rheumatoid arthritis and also play a protective role in skin by improving the activity of antioxidants. In this study, we investigated the effects of a novel mixture (AP-CPM01) containing collagen peptides and elastin peptides on photoaged hairless mice skin both in vivo and in vitro. To evaluate the effects of AP-CPM01 on UVBinduced skin wrinkle formation in vivo, the hairless mice were exposed to UVB irradiation and orally administered the AP-CPM01 at 333 mg/kg per day for 10 weeks. The effects on skin appearance and epidermal thickness were measured using bioengineering and histochemical methods. In addition, the influence of AP-CPM01 on collagen metabolism in human skin fibroblasts was also investigated. The skin of mice in the AP-CPM01 treated group had better appearance and less wrinkling than that of mice in the control group. In the human fibroblast cells, the amount of de novo procollagen synthesis was increased after AP-CPM01 treatment, reflecting that AP-CPM01 can induce de novo procollagen synthesis and reduce UVB-induced skin wrinkle formation. These results suggest that AP-CPM01 is a potent candidate for antiphotoaging functions.

Effects of Ehwa Makgeolli Containing Oriental Herbs on Skin Whitening and Wrinkles (한방 이화주의 미백 및 피부 주름 개선 효과)

  • Extrinsic skin aging is characterized by the loss of skin tone and resilience, irregular pigmentation, and deep wrinkles. The aim of this study was to investigate the effects of Ehwa Makgeolli containing oriental herbs (Glycyrrhiza uralensis Fisch., Lycium chinense MILL., Morus alba L., and Saururus chinensis Baill) on skin whitening and wrinkling in human skin cells. We prepared Makgeolli extracts (HEE) with 70% ethanol. HEE significantly inhibited in vitro mushroom tyrosinase activity and reduced the cellular and secreted melanin content of mouse melanoma melanocytes (B16F1 cells). HEE down-regulated the protein expression of tyrosinase related protein (TRP)-1/-2, a key player in melanogenesis. Treatment with HEE in human keratinoctyes (HaCaT cells) inhibited the proteolytic activities of matrix metalloproteinase (MMP)-2/-9 in a dose-dependent manner and dramatically reduced the expression of MMP-2/-9. In addition, HEE attenuated lipopolysaccharide (LPS)-induced nitric oxide production in murine macrophages (RAW264.7 cells). These results indicate that HEE may be a great cosmeceutical ingredient for its whitening, anti-wrinkle, and anti-inflammatory effects.

Optimal design of a lightweight composite sandwich plate used for airplane containers

  • Composite material-due to low density-causes weight savings, which results in lower fuel consumption of transport vehicles. The aim of the research was to change the existing base-plate of the aluminum airplane container with the composite sandwich plate in order to reduce the weight of the containers of cargo aircrafts. The newly constructed sandwich plate consists of aluminum honeycomb core and composite face-sheets. The face-sheets consist of glass or carbon or hybrid fiber layers. The orientations of the fibers in the face-sheets were 0°, 90° and ±45°. Multi-objective optimization method was elaborated for the newly constructed sandwich plates. Based on the design aim, the importance of the objective functions (weight and cost of sandwich plates) was the same (50%). During the optimization nine design constraints were considered: stiffness, deflection, facing stress, core shear stress, skin stress, plate buckling, shear crimping, skin wrinkling, intracell buckling. The design variables were core thickness and number of layers of the face-sheets. During the optimization both the Weighted Normalized Method of the Excel Solver and the Genetic Algorithm Solver of Matlab software were applied. The mechanical properties of composite face-sheets were calculated by Laminator software according to the Classical Lamination Plate Theory and Tsai-Hill failure criteria. The main added-value of the study is that the multi-objective optimization method was elaborated for the newly constructed sandwich structures. It was confirmed that the optimal new composite sandwich construction-due to weight savings and lower fuel consumption of cargo aircrafts - is more advantageous than conventional all-aluminum container.

Occurrence of Kiwifruit Vine Decline Syndrome and Its Prevention Using Rootstock Tolerant to Waterlogging (키위 쇠락증상 발생 및 습해 저항성 대목을 이용한 예방)

  • Kiwifruit industry has been threatened by the emergence of kiwifruit vine decline syndrome causing plant death within one or two years from symptom appearance. The main symptoms of this syndrome are root cortex breakdown, leaf necrosis, phylloptosis, fruit skin wrinkling, and twig wilting. Kiwifruit vine decline syndrome occurred on both Actinidia chinensis var. chinensis and A. chinensis var. deliciosa in mid-summer after rainy season. Kiwifruit vine decline syndrome was turned out to be severely occurred in wettable clay soils affected by waterlogging or poor aeration. No pathogens were directly correlated with the syndrome. Kiwifruit vine decline syndrome could be expected to be efficiently prevented controlled using Bounty 71 rootstock tolerant to water stress such as waterlogging.
