The dyeability and antimicrobial activity of Sophora Radix ethanol extracts - Characteristics of dyed silk - (고삼 에탄올 추출액의 염색성과 항균성 - 염색 견포를 중심으로 -)

  • The aim of study was to elucidate dyeability and antimicrobial and antifungal activity of silk fabrics dyed with Sophora Radix extracts according to different mordants. Dyes were extracted from Sophora Radix using ethanol. Then, silk fabrics were dyed with extracts two times by post-mordanting method in which the extract was 60%(owf), the mordant was 3%(owf), L.R was 1:20, the temperature was $60~60^\circ{C}$, the time of dyeing was 60min., and the time of mordanting was 60min. The dyeability was evaluated by surface color, K/S values and durability of dye. The skin microorganisms used in this study was S. sureus, B. subtilis, S. epidermidis, P. acnes, P. aeruginosa, E coli, A. niger, C. albicans and T. mentatrophytes. The results are as follows; 1. When mordants were added, K/S value of silk dyed was not improved much and surface color was 2.2Y to 8.8Y in H(hue) value which indicated greenish yellow to raddish yellow 2. The color fastness tests to light, perspiration, dry-cleaning, rubbing, and stain fabric washing show 4~5th degree which were valuated excellent. The color fastness to fade washing was improved to 3~4th degree by addition of $K_2CrO_7$ mordants. 3. Antibacterial activity of silk dyed using no-mordant as well as mordants was excellent on S. aureus, B. subtilis, S.epidermidis and P.acnes, but showed poor antibacterial activities on P.aeruginosa and E.coli such as gram negative baterials 4. Antifungal activity of silk dyed with ethanol extracts was good on A.niger, C.candida and T.mentagrophytes. Especially, on T. mentagrophytes there was no growth of fungus during 72 gous in silk dyed mordanting with $SnCl_2\cdot{2H}_2O$.

  • In this study, partition coefficients(Kw) of phenoxyethanol in oil-water phase according to type of oils was investigated. Partition coefficients(Kw) were experimented by classified oils that hydrocarbones, fatty alcohols, fatty acids, esters, triglycerides and silicones. It was found that partition coefficients(Kw) of hydrocarbons and silicones were low, but partition coefficients(Kw) of fatty acids, fatty alcohols, esters and triglycerides were high. It was known that the emulsions which were made of oils having low partition coefficients value had a good antimicrobial effects. Thus, the cosmetics stability of microorganisms and skin safety of phenoxyethanol could be improved by using the oils which have low partition coefficients value.

Long-term outcome of patients with p22phox-deficient chronic granulomatous disease on Jeju Island, Korea

  • Purpose: This study investigated the long-term clinical outcomes of patients with $p22^{phox}$-deficient chronic granulomatous disease (CGD) on Jeju Island and retrospectively evaluated the effects of interferon-gamma (IFN-${\gamma}$) prophylaxis. Methods: The medical records of 15 patients with CGD were retrospectively reviewed. The efficacy of IFN-${\gamma}$ prophylaxis was evaluated by comparing the frequency of severe infections before and after starting continuous prophylaxis with IFN-${\gamma}$. Results: At the time of the analysis, 14 patients were alive, with a median age of 14.3 years. The diagnosis of CGD was made at a median age of 2.4 years, and the median age at onset of severe infection was 0.3 years. Thirteen of the 15 patients had their first severe infection within the first year of life. The overall incidence of severe infection was 1.36 infections per patient-year; pneumonia, suppurative lymphadenitis, and skin and subcutaneous abscesses were the most common infections. Aspergillus species were the most frequently isolated microorganisms, present in 15.8% of isolates. IFN-${\gamma}$ did not significantly change the rate of severe infection. The survival rate for patients after 2 years of age was 93%; there was a prolonged survival plateau beyond the age of 2. Conclusion: Compared with cases of X-linked CGD reported in other studies, patients with CGD on Jeju Island did not show obviously different clinical manifestations, but they had a significantly higher survival rate. Further studies with a substantially longer period of observation, and with more patients under intensive surveillance are necessary to elucidate the prophylactic efficiency of IFN-${\gamma}$.

  • Bioconversion, the enzymatic process by microorganism on organic precursor to desired products. The natural extract is converted into a form that can be easily absorbed into the skin, while scaling up of to higher quantity is possible. Selection of naturally processed raw material rather than chemically processed is preferred. Therefore, fermentation was carried out by mixing Rubus coreanus Miquel, soybean, Zanthoxylum schinifolium as bioconverting materials, the possibility of inflammation, whitening material were checked. In this study, useful microorganisms were isolated from various salted fish, and 16S rDNA sequence was analyzed to confirm their genetic characteristics. Combining the three natural materials using bioconversion technology to study their activity before and after fermentation. To evaluate the antioxidant activity and the active ingredient content the DPPH radical scavenging activity and the total polyphenol content were examined. Raw 264.7 cells were used to evaluate MTT assay, NO and $TNF-{\alpha}$ production inhibitory activity. Also, to evaluate the whitening activity, tyrosinase inhibitory activity and melanin formation inhibitory activity were measured using B16F10 cells. In total 34 strains were obtained from various salted fish. The effective strains useful for the bioconversion process, showed that DPPH radical scavenging ability and polyphenol content were increased in the two kinds of microbial treatment groups compared to the untreated group. 16S rDNA sequencing analysis of the strains showed excellent in Pediococcus pentosaceus B1 in comparison. An increase of up-to 156% in anti-oxidative activity and 141% in polyphenol content was observed after bioconversion. In addition, after mixed fermentation the toxidty of Raw 264.7 and B16F10 cells tended to decrease and a significant increase was observed in anti-inflammatory activity as well. Also, tyrosinase activity and melanin significantly. synthesis decreased significantly.

A Study on Antimicrobial and Antioxidant Activity of Ethanol Extract of Rhus chinensis Mill (붉나무(Rhus chinensis Mill) 에탄올 추출물의 항균 및 항산화 활성에 대한 연구)

  • In this study, the Rhus chinensis Mill was divided into bark and inner bark. The antimicrobial activity and the antioxidative activity of the extracts were investigated by using the organic solvent fractions after the extraction and concentration with ethanol. This study showed the possibility of functional materials such as raw materials for cosmetics and food supplements. This study was the antimicrobial activity and antioxidant activity of five microorganisms (S. aureus, E. coli, B. subtilis, P. aeruginosa, and P. ovale). The following conclusions were obtained. First, the antimicrobial activity of B. subtilis was found to be high in the ethanol extract of. Second, DPPH scavenging activity was 86.4% free radical scavenging activity at 2.5 mg/mL bark part and 61.9% free radical scavenging activity at 2.5 mg/mL in inner Rhus chinensis Mill bark part. The ABTS scavenging activity was 79.2% free radical scavenging at 1 mg/mL bark fraction and 63% free radical scavenging activity at 1 mg/mL in inner bark, and bark showed higher antioxidant activity than inner bark. These results suggested that the antimicrobial activity and antioxidant activity of Rhus chinensis Mill extract can be used as a natural material. Specific and diverse physiological activity studies are expected in the future.

  • Leather (skin & hide) is a body organ, comprising 3 to 5% of the animals weight. The cross-section of a leather is composed of two major divisions: the epidermis or grain layer and the corium or split layer. The leather is naturally covered with bacteria and fungi, because it is a particularly rich source of a wide variety of microorganisms. Stains or coatings of different colours occur in patches or over large areas, depending on the type of mould spore infestation. We examined the antibiotic effect of leather after washing. Upon applying equal fungicide, antibiotic effects increased as follows: grain layer>middle layer>flesh layer. Antibiotic effect decreased with increasing frequency of washing. Decrease in antibiotic effect was lower in OITZ fungicide than in TCMTB and CMK fungicides. Sulfated fatliquor showed higher antibiotic effect than phosphated fatliquor.

Antioxidant and Antimicrobial Activities of Combined Extracts of Galla rhois, Achyranthes japonica Nakai, Terminalia chebula Retz and Glycyrrhiza uralensis (오배자, 우슬, 가자 및 감초를 포함하는 혼합추출물의 항산화 및 항균활성)

  • In this study, antioxidant and antimicrobial activities of ethanolic crude extract and its five different solvent subfractions (namely, ethyl acetate fraction, n-butanol fraction, chloroform fraction, n-hexane fraction and the aqueous fraction) from a mixture of four different medicinal herbs (Galla rhois, Achyranthes japonica Nakai, Terminalia chebula Retz and Glycyrrhiza uralensis) were investigated. Among all the tested mixture combination of four medicinal herbs, 5:3:1:1 ratio of Galla:Achyranthes : Terminalia : Glycyrrhiza had the best antimicrobial effects against four strains of microorganisms (Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa and Escherichia coli) and exhibited the highest DPPH radical-scavenging activity. Further sub-fractions with solvents were screened for antioxidant and antimicrobial activities. Antioxidant activity in order was ethyl acetate fraction > n-butanol fraction > chloroform fraction > nhexane fraction > aqueous fraction. The n-butanol extracted fraction showed the highest level of antimicrobial activity in com- parison to other fractions. In addition, all those fractions did not show any cytotoxicity against human skin cell CCD-986sk. These results suggest that 5:3:1:1 combination extracts of medicinal herbs (Galla : Achyranthes : Terminalia : Glycyrrhiza) may be potentially used as a safe natural antimicrobial preservative.

  • The aim of this study was to assess the cosmeceutical activies of four kinds of Korea herb medicine extracts using in cosmetics and related industries. The cosmeceutical activities of extracts were investigated by tyrosinase inhibition, astringent, anti-bacterial and MTT assay for cell viability. In the whitening effect, PA that the highest tyrosinase inhibition activity showed 56% at 10 ppm in ethanol extract. Also water and ethanol extract of RE showed 54%, 68% at 1,000 ppm, respectively, but LE and CA showed lower effect. Astringent effect of water and ethanol extract of PA appeared over 60% at 1,000ppm concentration but other extracts showed no astringent effect. In the anti-bacterial test, water and ethanol extract of PA showed no anti-bacterial effect against all microorganisms. But water and ethanol extract of RE showed anti-bacterial effect on Staphylococcus aureus, ethanol extract of showed on Staphylococcus aureus and Propionibacterium acnes, ethanol extract of CA showed on Candida albicans. The resUlt of stability test showed that the emulsion of containing PA were very stable at various temperature and sun-light test. Viscosity and pH of emulsion did not change. From the results of human patch test to assess the safety of cosmetics containing PA there was no negative reaction on skin was found.

  • The purpose of this study was to evaluate hot spring water (HSW) from Deokgu as a cosmetic ingredient in the preparation of a body lotion. The HSW was tested for its suitability as an aqueous-phase main component. Microbiological and chemical stability tests of the HSW were carried out. Microorganisms including E. coli were not detected or detected below the detection limits, and no harmful heavy metals were found. The cytotoxicity of the HSW was also considered, and its pH determined over a period of three months. Further, sensory characteristics were assessed for consumer acceptance by performing sensory tests on body lotions formulated using either Deokgu HSW or distilled water. Skin moisturization, irritation and tension reinforcement were found to be enhanced when using the HSW lotion rather than that formulated with distilled water. Taken together, the results of this study show that the use of HSW in cosmetic formulations contributes to the efficacy of these products.

  • Food irradiation is important not only in ensuring safety but also improving antioxidant activity of peaches. Our objective was to establish the best irradiation treatment for peaches by calculating dose distribution using Monte Carlo simulation. 3-D geometry and component densities of peaches, extracted from CT scan, were entered into MCNP to obtain simulated dose distribution. Radiation energies for electron beam were 1.35 MeV (low energy) and 10 MeV (high energy). Co (1.25 MeV) and the Husman irradiator, containing three sealed Cs source rods in an annular array, were used for gamma irradiation. At 1.35 MeV electron beam simulation, electrons penetrated well beyond the peach skin, enough for surface treatment for microorganisms and allergens. At 10 MeV electron beam simulation, for top-beam only treatment, doses at the core were the highest and for double beam treatment, the electron energy was absorbed by the entire sample. At Co source, the radiation doses were presented on the whole area. At Cs source, the dose uniformity ratios were 2.78 for one source and 1.48 for three ones at 120 degrees interval. Proper control of irradiation treatment is critical to establish confidence in the irradiation process.
