Gentisic acid is a skin-whitening agent which inhibits the tyrosinase activity, an essential enzyme in the process of biological synthesis of melanin. Since melanin is synthesized in melanocytes located between the viable epidermis and dermis layer, drug amount delivered into the epidermis/dermis layer can provide valuable information for the biological effect of skin-whitening agents. The purpose of this study was to prepare the gentisic acid patches with 2% dodecylamine as enhancer, and to observe the in vitro skin permeation and in vivo skin deposition of gentisic acid. Gentisic acid in DuroTak 87-2510 patch formulation permeated across hairless mouse skin at the rate of $40.79\;{\mu}g/cm^2/hr$. In vivo study showed that the gentisic acid amount in both the stratum corneum and the viable epidermis/dermis increased with the increase of application time. The amount of gentisic acid in the stratum corneum was higher than that in the epidermis/dermis layer, and was expected to provide a reservoir effect even after removing the patches. Thus, the patch formulation seems to be useful for the topical delivery of skin-whitening agent into the epidermis/dermis layer, the target site.
Psolarea corylifolia extract that contains bakuchiol is known to have anti-microbial, anti-inflammatory and anti-scarring effects. In this study, a vesicles such as liposome, niosome, and transfersome were produced to encapsulate P. corylifolia extract and measured their stability and physiochemical property. The skin permeation and partitioning of P. corylifolia extract in the vesicles were elucidated in nude mouse skin by using Franz diffusion cells after topical application for 24 h. After storage at 25, 40, $70^{\circ}C$, and light, the stability of bakuchiol incorporated into the vesicles was maintained for 30 days. The optimal concentration of P. corylifolia extract entrapped into the vesicles was found to be 5~10%. From the physicochemical studies, after storage at 4, 25, and $40^{\circ}C$, the viscosity and particle size of the vesicles remained in 30~80 cP and the nanosize range for 6 months, respectively. From the permeation experiments, niosome showed a higher amount of bakuchiol permeated through the mouse skin compared to liposome and transfersome after 24 h. From these results, niosome and transfersome could be a good bioavailability enhancement system (BAES) for P. corylifolia extract to improve the skin permeation and stability.
Antisense oligonucleotides seem to provide a promising new tool for the therapy. Choi et al. (1995) reported antisense phosphorothioate oligonucleotides (PS-ODN, 25 mer) complementary to TGF-.betha. mRNA designed for scar formation inhibitor to eliminate scars, which was caused by undesired collagen deposition due to overexpression of TGF-.betha., in wounded skin. PS-ODN were evaluated in vitro for skin penetration using normal and tape-stripped damaged rat skin. The in vitro skin transports were carried out with partially modified PS-ODN (6S) and fully modified PS-ODN (25S). The cumulative amount of PS-ODN (6S) penetrated through normal rat skin was $0.234{\pm}0.041{\mu}g/cm^2$ and that of tape-stripped damaged rat skin was $1.077{\pm}0.301{\mu}g/cm^2$ over 8 hrs. PS-ODN (25S) can not be found in receptor medium through normal skin due to high molecular weight (Mol.Wt.=8,000) and polyanionic charge. However, the cumulative amount of PS-ODN (25S) penetrated across damaged rat skin in PBS was $0.340{\pm}0.296{\mu}g/cm^2$ over 8 hrs. The absense of dermis raised the cumulative amount of PS-ODN (6S) penetrated through rat skin. And the fluxes of PS-ODN (6S) and PSODN (25S) at 8hrs across damaged rat skin were $134.63{\pm}37.67{\mu}g/cm^2$ h, and $42.50{\pm}36.95ng/cm^2$ h, respectively. While PS-ODN (25S) was stable in 10% heat inactivated fetal bovine serum (FBS) during 24 hrs, PS-ODN (6S) was less stable than PS-ODN (25S), but was markedly stable than unmodified phosphodiester. It is suggested that the cumulative amount of PS-ODN (6S) penetrated through damaged rat skin is larger than that of PS-ODN (25S) since the former is easier to degrade by nuclease than the latter and then is apt to penetrate into skin. Thus, PS-ODN represents a logical candidate for further evaluation due to the potential for delivery into the wounded skin.
During cosmetic treatments, SURFACTANTS penetrate into KERATINOUS TISSUES (hair, skin and nails). Whereas some of these surfactant molecules migrate to the vital tissues, a considerable fraction remains bound to the keratin. The extent of binding depends both on the nature of the head group and the length of the hydrophobic tail of the detergent molecules. In addition to entering the amorphous region of the keratin, some of the detergents also penetrate into the crystalline microfibrils and change their structures affecting their tensile properties. Owing to an uneven distribution of detergent molecules in the tissues, an anisotropy of the elastic moduli will occur, resulting in considerable internal stresses which, in rum, might lead to a deterioration of hair, skin and nails. The chemical behavior of keratins is also influenced by the presence of absorbed detergent in their structures. Depending on the detergents and the conditions, these effects can be either protective or detrimental. The deposition of detergent molecules into keratin can be enhanced or diminished by the inclusion of appropriate ingredients into the product formulae.

Omega-3 rich fish oils are extremely labile, thus requiring control of oxidation and off flavor development. A recently proposed emulsification method, layer-by-layer (LbL) deposition, was found to be a plausible method to enhance the characteristics of bioactive ingredients, especially lipids. The present work was designed to test the possibility of enhancing the uptake and utilization of omega-3 fatty acids present in fish oil. The bioavailability of nano-emulsified fish oil was monitored in terms of intestinal absorption as well as skin permeability by using the everted intestinal sac model and Franz cell model. The skin permeability and intestinal absorption characteristics was significantly improved by LbL emulsification with lecithin/chitosan/low methoxypectin. Multilayer encapsulation along with nano-emulsification can be a useful method to deliver biologically active lipids and related components, such as fish oil. The protective effect of this tool from lipid oxidation still needs to be verified.
꿀벌(Apis mellifera)의 독에 의해 야기되는 포유동물 피부의 조직병리학적 및 미세구조적 변화와 그 수복과정을 확인하기 위하여 실험용 생쥐의 피부에 직침법으로 꿀벌의 독을 주입한 후, 회복된 시점까지 일정시간 간격으로 조직의 표본을 제작하여 광학 및 전자현미경으로 관찰하였다. 독 주입 직후의 표본에서는 표피의 상피세포와 진피의 결합조직에서 현저한 염증반응이 유도되었고, 일부세포의 괴사가 관찰되었다. 고배율의 전자현미경상에서 교원섬유의 직경이 크게 증가되었으며, 면역 단백물질로 추정되는 전자밀도가 높은 grain의 침착이 확인되었다. 이러한 조직병리현상은 독 주입 후 12시간이 경과된 조직의 표본에서 서서히 회복되는 것으로 관찰되었다. 봉침 주위 피부조직의 조직학적 및 미세구조적 변화는 수 일간 지속되었으나, 병리학적 반응은 3일 이내에 거의 거의 소멸되는 것으로 관찰되었다. 또한 생쥐 피부에 대한 꿀벌 독의 병리반응은 다른 절지동물의 독에 비하여 비교적 경미한 것으로 확인되었다.
The present study was conducted with the aim to investigate the ameliorative effects of a new soybean product (cheonggukjang) fermented with Bacillus amyloliquefaciens SCGB1 (SFBA) in atopic dermatitis (AD) mouse model. Visual evaluation of AD induction in the mice indicated the remarkable control of SFBA in reducing the pathological severity of AD-like skin lesions reported as the SCORAD score of AD clinical symptoms. The results revealed that SFBA reduced dorsal skin and epidermal thickness to a similar extent with prednisolone. Further analysis revealed the dominance of SFBA in restraining mast cell infiltration in the dermis; immunoglobulin-E expression in serum; and TH2 IL-4 cytokine and itch-related IL-31 cytokine in the mice skin and serum. SFBA also suppressed scratching behaviours in mice induced by compound 48/80. Further histological findings also revealed the alleviation of collagen fiber deposition in dermal skin of the AD mice model. These actions of SFBA were examined to be mediated by its suppression of the phosphorylation activation of key signalling molecules such as $NF-{\kappa}B$ and MAPK responsible for the induction of cytokine production. Thus, SFBA can be considered as a promising functional food for managing clinical, histological and immunological spectra associated with AD.
본 연구는 사료내 색소원으로 paprika와 동물플랑크톤 혼합첨가가 흰줄납줄개의 carotenoid 축적 및 색택에 미치는 영향을 조사하였다. 대조사료(Con)를 제외한 모든 실험사료에 paprika 분말 5%를 첨가하고, paprika 분말과 동물플랑크톤의 상호작용을 알아보기 위하여 paprika 분말만 첨가한 실험사료(P5), P5사료에 water flea Moina macrocopa 3% 첨가사료(P5M3), Artemia nauplius (Artemia sp.) 3% 첨가사료(P5A3), rotifer Brachionus Plicatilis 3% 첨가사료(P5R3)로 총 5개의 실험사료를 제조한 후, 2반복으로 10주간 사육실험 하였다. 실험종료 후, 모든 실험구간에서 생존율의 차이는 없었으며, 총 carotenoids 함량은 동물플랑크톤이 첨가된 실험 구에서 높은 다소 높은 값을 보였다. 특히, P5A3 실험구에 가장 높은 값을 나타냈다. 흰줄납줄개의 색택은 paprika와 동물플랑크톤 혼합 첨가한 실험구가 대조구 및 P5 실험구에 비해 붉은색이 더 선명하였다. 이상의 결과를 고려할 때, 흰줄납줄개의 배합사료내 paprika와 동물플랑크톤의 혼합첨가는 붉은 색 개전에 도움이 되었고, 특히 Artemia nauplius 또는 rotifer 첨가가 좋을 것으로 판단된다.
Purpose: Heterotopic calcification is the abnormal deposition of calcium salts in tissues other than bone and enamel, and it occurs in the form of dystrophic calcification or metastatic calcification. This deposition can occur under many conditions, but in some rare cases, it may develop in burns and nonhealed scars. It is difficult to treat the combination of heterotopic calcification and ulceration in scar tissues by using conservative therapy and to determine the margin of excision in such cases. Our study proposes the use of intraoperative C-arm-guided mapping of lesions with heterotopic calcification, and adequate excision of ulcers in chronic scars where heterotopic calcification is also observed. Methods: This study included 2 patients and was conducted from January 2010 to July 2010. The first patient was a 63-year-old woman who presented with atypical calcium deposits and chronic ulceration in the lower one-third region of the right leg. The second patient was a 38-year-old man who presented with a nonhealing ulcer that had developed on the right leg 3 months earlier he had a history of 40% scalding burns on the entire body. Surgery is the most reliable method for treating heterotopic calcification therefore, both patients were treated using intraoperative C-arm-guided marginal mapping of heterotopic calcification, followed by release of contracture, and eventually split-thickness skin grafting. Results: Plain radiographs of the leg showed spotty radiopaque areas in the hard part of the scar well superficial to the underlying bones. Histopathological analysis revealed multiple foci of calcified deposits, increased fibrosis, and inflammation in the scar tissue. Surgery-related complications were not observed. Conclusion: C-arm guided excision of calcified scars and the release of contracture can cure nonhealing ulcers and may therefore prevent recalcification.
투명차폐재를 목적으로 Indium Tin Oxide (ITO) 투광성 박막을 제조하고 전자파 차폐특성에 대해 조사하였다. 박막은 RF magnetron co-sputtering 증착장비를 사용하여 제작하였다. RF 인가전력, Ar 및 $O_2$분압, 기판온도를 변화시키며 전기전도도와 투광성을 겸비한 박막의 조성과 구조에 관한 실험을 진행하였다. 최적의 증착조건은 $300^{\circ}C$의 기판온도, 20sccm의 아르곤 유량, 10sccm의 산소유량, 그리고 In과 Sn의 인가전력이 각각 50W와 30W일 경우였으며, 이때 얻어진 박막은 육안으로 분명할 정도의 투광성을 보였고 5.6$\times10^4$mho/m의 높은 전기전도도를 나타내었다. 이렇게 제조된 ITO 박막의 전자파 차폐효과를 차폐이론에 의해 분석하였다. 박막의 전기전도도, 두께, skin depth로부터 차폐기구(흡수손실, 반사손실, 다중반사 보정항)에 대해 고찰하였다. 계산된 차폐효과는 26dB의 값을 보여 투광성 차폐재로 ITO 박막의 사용 가능성을 제시할 수 있었다.
