지용성 비타민 C 유도체 및 갈릭산을 함유한 지질나노입자 제조 및 특성 (Preparation and Characterization of Lipid Nanoparticles Containing Fat-Soluble Vitamin C Derivatives and Gallic Acid)

  • 지질나노입자(lipid nanoparticles, LNPs)란, 세포 불투과성인 치료용 핵산, 단백질 및 펩타이드와 같은 생물학적 활성 화합물을 미세한 환경 변화에 의한 분해로부터 보호해 안정적이면서 효과적인 약물전달시스템이다. 본 연구에서는 지용성 비타민 C 유도체인 테트라헥실데실아스코르베이트(tetrahexyldecyl ascorbate, THDC)의 생체 내 반감기를 효과적으로 연장시키기 위해 산화방지제인 갈릭산(gallic acid, GA)을 함유하는 지질나노입자를 개발하였다. 마이크로플루이딕스칩(microfludics chip)을 통해 상온 및 상압 조건 하에서 작고 균일한 크기로 제작하였다. 기존 마이크로플루다이저(microfluidizer)를 통해 고압 및 고온 조건 하에 제작된 리포좀과 비교하였을 때, 마이크로플루이딕스칩(microfludics chip)을 통해 제작된 LNPs인 경우, 분산 및 온도에 따른 안정성이 우수하였으며, 지용성 비타민 C 유도체의 산화 안정성을 향상시켰을 뿐 아니라 피부 흡수율도 개선된 것을 확인하였다. 향후 연구에서는 이러한 결과를 더욱 입증하기 위해 피부 개선 효과를 연구하기 위한 생체 외 및 생체 내 평가에 중점을 둘 것이다.

  • The objective of this study is to prepare a stable delivery systems containing polyethoxylated retinamide(PERA) - derivatives of retinoic acid, effective anti-wrinkle and anti-acne agent. Cubic liquid crystalline phase gel (CLCPG) and cubosomes containing various concentrations of PERA were prepared to investigate the physicochemical properties. Furthermore, stability and transdermal absorption efficacy of the CLCPG containing PERA were investigated in comparison with oil-in-water (O/W) emulsions which are predominantly used as a topical formulation. CLCPG increase the stability of PERA in comparison with O/W emulsion. For tropical application, CLCPG containing PERA shows higher moisturizing effect than that of O/W emulsion. In skin permeation test, CLCPG shows higher PERA deposit on epidermis. With its specific physicochemical property caused by the glyceryl oleate, CLCPG itself could be used for stabilizer of various actives and applied as an effective delivery system for topical application. Cubosome, nano-sized dispersed CLCPG, is also expected to be applied in a various field of industry like food, cosmetics and pharmaceuticals.

Chelation of Thallium (III) in Rats Using Combined Deferasirox and Deferiprone Therapy

  • Thallium and its compounds are a class of highly toxic chemicals that cause wide-ranging symptoms such as gastrointestinal disturbances; polyneuritis; encephalopathy; tachycardia; skin eruptions; hepatic, renal, cardiac, and neurological toxicities; and have mutagenic and genotoxic effects. The present research aimed to evaluate the efficacy of the chelating agents deferasirox (DFX) and deferiprone (L1) in reducing serum and tissue thallium levels after the administration of thallium (III), according to two different dosing regimens, to several groups of Wistar rats for 60 days. It was hypothesized that the two chelators might be more efficient as a combined therapy than as monotherapies in removing thallium (III) from the rats' organs. The chelators were administered orally as either single or combined therapies for a period of 14 days. Serum and tissue thallium (III) and iron concentrations were determined by flame atomic absorption spectroscopy. Serum and tissue thallium (III) levels were significantly reduced by combined therapy with DFX and L1. Additionally, iron concentrations returned to normal levels and symptoms of toxicity decreased.

Autologous Fat Graft in the Reconstructed Breast: Fat Absorption Rate and Safety based on Sonographic Identification

  • Background Autologous fat graft has become a useful technique for correction of acquired contour deformity in reconstructed breasts. However, there remains controversial regarding the efficacy and safety of the practice for reconstructive breast surgery. Methods A retrospective review was performed on 102 patients who had secondary fat grafting after breast reconstruction. Fat harvest, refinement and injection were done by Coleman's technique. All patients were followed up postoperatively within 1 month and after 6 months including physical examination and ultrasonography. In 38 patients, the reabsorption rate was calculated by serial changes of thickness between skin and pectoral fascia in the ultrasonic finding. Locoregional recurrence rate was compared with control group of 449 patients who had breast reconstruction without fat graft in the same time period. Results Average 49.3 mL fat was injected into each breast. The most common location of fat graft was upper pole, followed by axilla, lower and medial breasts. During 28.7 months of average follow-up period, 2.9% of total patients had symptoms of palpable mass on fat graft side and ultrasonography identified fat necrosis and cyst formation in 17.6% of the patients. Calculated fat reabsorption rate was 32.9%. Locoregional recurrence was occurred in 1 patient (0.9%) and the rate was not different significantly with control group (2%). Conclusions Although further studies are required to provide surgeons with definitive guidelines for the implementation of fat grafting, we propose autologous fat graft is an efficient and safe technique for secondary breast reconstruction.

세포투과 펩티드를 이용한 주름개선 펩티드 GHKs의 피부흡수 증진 (Enhancement of Skin Permeation of Anti-wrinkle Peptide GHKs Using Cell Penetrating Peptides)

  • 본 연구에서는 경피흡수가 잘 안되는 주름개선 펩티드인 GHK, GHK-Cu, Pal-GHK에 대하여 세포투과 펩티드인 알르기닌 올리고머(tetra-D-arginine, R4)와 hexa-D-arginine, R6)를 첨가한 후 경피 투과도를 측정하여 그 결과를 다음 6가지 경우로 분석하였다. 첫번째로 주름개선 펩티드만 함유한 경우는 구리이온(Cu2+)과 팔미트산이 경피 투과율을 증진시키는 것을 알 수 있다. 두번째로, GHK에 알르기닌 올리고머(R4, R6)를 첨가한 경우는 알르기닌 올리고머(R4, R6)가 경피 투과율을 증가시켰으며, R4에서 더 좋은 경피 투과율 증가를 나타났다. 세번째로, GHK-Cu에 R4, R6를 첨가한 경우는 경피 투과율 증가가 나타났으며, R6 < R4 경피 투과율 순서로 나타났다. 네번째로 Pal-GHK에 R4, R6를 첨가한 경우에도 경피 투과율 증가가 나타났으며, R6 < R4 경피 투과율 순서로 나타났다. 다섯번 째로, R4를 GHK, GHK-Cu, Pal-GHK에 첨가한 경우에는 GHK+R4 < GHK-Cu+R4 < Pal-GHK+R4 순서로 경피 투과율 증가가 나타났다. 마지막으로 R6를 GHK, GHK-Cu, Pal-GHK에 첨가한 경우에는 GHK+R4 < GHK-Cu+R4 < Pal-GHK+R4 순서로 경피 투과율 증가가 나타났다. 이를 통하여 주름 개선 펩티드인 GHK, GHK-Cu, Pal-GHK의 피부 투과를 증가를 위한 최적의 조건을 제시하여 그 효능을 극대화할 수 있는 방안을 제시함으로써 주름 개선 기능성 화장품에서의 폭넓은 활용과 응용을 제안하고자 한다.
