Effect of Halophyte (Spartina anglica and Calystegia soldanella) Extracts on Skin Moisturizing and Barrier Function in HaCaT Cells (염생식물인 갯끈풀과 갯메꽃 추출물의 HaCaT 세포에서 피부 보습 및 피부 장벽 기능에 미치는 영향)

  • As aging progresses, reactive oxygen species (ROS) reduces skin moisturization and collapses skin barrier function. In this study, we evaluated the efficacy of skin moisturizing and skin barrier function enhancement by extracts from halophytes using HaCaT cells. Spartina anglica (S. anglica; SAE) and Calystegia soldanella (C. soldanella; CSE), a kind of halophytes, were collected from Dongmak beach in Incheon, and extracted with 70% ethanol. At the first, we evaluated the cytotoxicity of extracts in HaCaT cell using WST-8 Kit. As a result, the other experiment was conducted by setting the concentration at which the cell viability was 90% or more. SAE and CSE showed high radical scavenging activity through ABTS assay. Expression levels of genes related to skin moisturizing and skin barrier functions, were analyzed by real-time qPCR. As a result, it showed that the expression of aquaporin 3, hyaluronan synthase 2, and transglutaminase 1 was increased by SAE treatment but not changed by CSE. Activation of extracellular signal regulated kinase (ERK), c-Jun N-terminal kinase (JNK), and p38 mitogen activated protein kinase was induced by SAE. These results suggest that SAE can be used as functional materials for cosmetics for skin moisturizing and barrier function enhancement.

  • In this study, we investigated the skin safety, antioxidant, moisturizing, and anti-inflammatory effects of a composite extract of Prunus mume flower, Lonicera japonica flower, Chrysanthemum indicum flower, and Phyllostachys bambusoides (Royal TheraTea GuardTM, RTG), which has been used as an internal medicine in the royal medical treatment for preventing skin damage caused by external environmental factors. In a cytotoxicity test on HaCaT cells, RTG complex extract showed no change in cell viability at concentrations of 0.125, 0.25, 0.5, and 1%, and a skin irritation index of 0.00 in a human patch test, confirming that it is non-irritating to the skin. The antioxidant effects was confirmed by the presence of 497.83 ㎍ GAE/g of polyphenols and increased DPPH radical scavenging activity, with a significant increase in catalase activity in the stratum corneum, showing potential as an antioxidant that protects cells from oxidative stress. The anti-inflammatory effects was observed through reduced erythema in skin stimulated by tape stripping and treated with fine dust in keratinocytes. The moisturizing effects was shown by increased expression of hyaluronan synthase (HAS)2, 3 and keratin1 in keratinocytes treated with fine dust compared to the control group, as well as increased skin moisture content and decreased skin roughness. These results suggest RTG can be used as a cosmetic ingredient to prevent skin damage caused by external environmental factors.

A Study on the Design of Stearic Acid-Based Solid Lipid Nanoparticles for the Improvement of Artificial Skin Tissue Transmittance of Serine (Serine 의 인공피부조직 투과 개선을 위한 Stearic Acid 기반 고형지질나노입자의 설계 연구)

  • Stratum corneum known as a skin barrier, which maintains water in skin, is the outer layer of the skin. Natural moisturizing factors (NMF) are one of the constituents in stratum corneum and amino acids are the highest components among NMF. In this study, we designed stearic acid-based solid lipid nanoparticles (SLNs) for improved skin penetration of serine (Ser). Ser-capsulated SLN was manufactured by double-melting emulsification method. The mean particle size and zeta potential of SLNs were 256.30 ~ 416.93 nm and -17.60 ~ -35.27 mV, respectively. The higher the degree of hydrophobicity or hydrophilicity of emulsifiers, the smaller the particle size and the higher the stability and capsulation rate. In addition, skin penetration was conducted using SkinEthicTM RHE which is one of the reconstructed human epidermis models. The results of Ser penetration demonstrated that all SLNs enhanced than serine solution. The amount of enhanced Ser penetration from SLNs were approximately 4.1 ~ 6.2 times higher than that from Ser solution. Therefore, Ser-loaded SLN might be a promising drug delivery system for moisturizing formulation in cosmeceutical.

Study on the Relationship between Skin Dryness and Amino Acids in Stratum Corneum (아미노산 동시분석을 통한 피부보습능과 각질 중 아미노산 함량과의 상관관계 연구)

  • Natural moisturizing factors (NMFs) are hydrophilic and water-soluble components in stratum corneum of the skin. NMFs absorb water from outer environment and enhance the water-holding capacity of stratum corneum and thereby, prevent the dryness and increase flexibility and plasticity of skin. NMFs are mainly composed of amino acids and their metabolites that are produced from the degradation of filaggrin. Here we established a simultaneous quantification method for 22 amino acids in tape-stripped stratum corneum samples using UPLC-PDA. With this method, we analyzed amino acid contents from tape-stripped stratum corneum samples of forearm and forehead regions from 15 healthy volunteers. Amino acid contents of inner (or upper) region were higher than outer (or lower) region of stratum corneum. Amino acid contents of stratum corneum of forearm were higher by 1.5 fold than forehead region. Of note, total amino acid contents were significantly and inversely correlated with trans-epidermal water loss (TEWL), an index for skin barrier function. Especially, Ser, Glu, Gly, Ala and Thr were determined to positively affect skin mositurizing activities. In conclusion, we could demonstrate that amino acid contents of stratum corneum are closely linked with skin barrier and moisturizing function, providing an important and fundamental methodology for the study of amino acids in skin physiology.

Skin Barrier Strengthening and Moisturizing Effect of Essence Containing Coptis Japonica Extract (황련 추출물을 함유한 에센스의 피부장벽강화 및 보습 효과)

  • The purpose of this study is to verify the effect of essence containing Coptis Japonica extract on skin barrier strengthening and moisturizing. The essence containing the Coptis Japonica extract was used by 22 subjects for four weeks, and the amount of percutaneous water loss (g/m2h) and water (A.U.) in the designated upper limit were measured. As a result of measuring percutaneous water loss using a vapometer, it was found to be 11.08±2.55 after 4 weeks of use from 14.82±4.01 before use, and the significance probability decreased significantly by 25.27% to p<0.001. The amount of skin moisture measured by the Corneometer was found to be 46.73±7.45 after use from 31.73±8.09 before use, and the significance probability increased by 47.26% to p<0.001, and the subject's subjective efficacy evaluation through a questionnaire showed that all subjects were 100% satisfied without skin adverse reactions. As a result of this, it has been confirmed that Coptis Japonica essence is effective in strengthening skin barriers and moisturizing, which is expected to be a cornerstone for the development of cosmetics using various activities of Coptis Japonica extract in the future.

Skin Barrier-Enhancing Effects of Dermabiotics HDB with Regulation of Skin Microbiota

  • In the regulation of inflammatory responses and skin homeostasis, the skin and its microbiota are closely related. Studies have reported that lactic acid bacteria extracts can improve the skin condition and microbiota. In our previous study, we developed probiotic lysates, which are efficacious in improvement of human skin cells and the skin barrier. The skin-moisturizing effect of Dermabiotics HDB (HDB) prepared with Lactiplantibacillus plantarum, and the correlation between changes in the skin microbiota and moisture contents, were evaluated and analyzed in clinical trials. The clinical parameters on the cheeks of 21 female participants were measured using biophysical tools before and after (2 weeks) using HDB or control. The skin microbes were collected and identified using 16s rRNA gene sequencing. HDB significantly improved moisture intensity, transepidermal water loss (TEWL), and hot flush level on the cheek. The beta-diversity of the skin microbiota was different from that of the control in the unweighted UniFrac principal coordinate analysis after using HDB. The genus Lawsonella demonstrated a positive correlation with TEWL and a negative correlation with the moisture contents of the keratin layer, regardless of the use of HDB and control. Conversely, after HDB use, the genus Staphylococcus was increased and associated with a lower hot flush level, while the genera of the phylum Proteobacteria tended to decrease, which is associated with an improved skin condition. Overall, HDB showed clinically proven effects, including skin moisturization with regulation of the skin microbiota.

Effect of Galgeunhwanggeumhwangryeon-tang Extract on Skin Moisturizing, Antibacterial, and Tight Juntion Recovery in Atopic Dermatitis-induced Mice (갈근황금황련탕추출물의 아토피피부염 유발 백서에서의 피부 보습, 항균, 밀착연접 회복)

  • Objective The purpose of this study was to confirm the effects of Galgeunhwanggeumhwangryeon-tang (PSCG) extract on skin moisturizing, antibacterial, and tight junction recovery in atopic dermatitis-induced mice. Methods In this study, we used 4-week-old NC/Nga mice divided into four groups: control (Ctrl), lipid barrier elimination (LBE), dexamethasone (Dx) after lipid barrier elimination (DEX), and PSCG after lipid barrier elimination (PSC). Ten rats were assigned to each treatment group. Three days after drug administration following lipid barrier elimination, ceramide kinase, caspase 14, sodium hydrogen antiporter (NHE), cathelicidin, claudin, and toll-like receptor (TLR)-2 were observed to confirm the restoration of skin moisturizer production, antimicrobial barriers, and tight junctions in the skin barrier. Results Ceramide kinase and caspase 14 positive reaction were significantly higher in PSC than in LBE and DEX. Both NHE and cathelicidin showed higher positive reactions in PSC than in LBE and DEX. Claudin, and TLR-2 showed higher levels of positive staining in the PSC group than in the LBE and DEX groups. Conclusion It was confirmed that the PSCG extract can have the potential to restore the damaged skin barrier in atopic dermatitis.

Skin Regeneration and Whitening Effects of the Complex Extract of Dendrobii Caulis and Centella asiatica (석곡 및 병풀 복합 추출물의 피부 색소 침착 방지 및 피부재생 효과)

  • The objective of this study was to assess the effect of preventing pigmentation caused by external stimuli, promoting skin regeneration and whitening of the skin composition containing complex extract of Dendrobii Caulis and Centella asiatica (CE). We evaluated cell viability, tyrosinase and melanin inhibition activity, skin irritation test, and skin moisturizing and regenerative effects using CE. As a result of the tyrosinase inhibitory activity, 100 ㎍/mL CE (35.31%) showed higher value than kojic acid (21.32%). The results of melanin synthesis inhibition in B16F10 melanoma cells after treatment with α-melanocyte stimulating hormone showed a similar level of activity to that of arbutin, indicating an excellent whitening effect. In clinical test of the skin composition containing CE, we confirmed that CE is non-irritated in human skin primary irritation test as well as have a high skin moisturizing and regenerative effect. From these results, we suggested the CE not only prevents skin damage and pigmentation caused by external stimuli but has remarkable skin brightening activity and skin regeneration effect.

Antibaterial Activity and Skin Moisturizing Effect of Cedrela sinensis A. Juss Shoots Extracts (참죽나무 새순 추출물의 항균 활성과 피부 보습효과에 관한 연구)

  • In this study, we investigated the antibacterial activity and the skin moisturizing effect of cream contaning Cedrela sinensis A. Juss shoots extracts. MIC values of ethyl acetate fraction from Cedrela sinensis A. Juss shoots extracts on P. acnes and S. aureus were 0.13 %, 0.03% respectively and P. ovale and E. coli were 0.25 % equally. The results showed that the antibacterial activity of the ethyl acetate fraction on P. acnes and S. aureus were higher than the methyl paraben. The cream containing the ethyl acetate fraction from Cedrela sinensis A. Juss shoots extracts was formulated and its skin moisturizing effect was evaluated in a clinical trial. The skin hydration and transepidermal water loss (TEWL) were investigated after topical application of the cream. The skin hydrating effect of the cream containing Cedrela sinensis A. Juss shoots extract was 10 ~ 15 % higher than the placebo cream, and transepidermal water loss was decreased to 6.7 g/$m^2h$ in the subjects applied with cream containing the ethyl acetate fraction of Cedrela sinensis A. Juss shoots extracts. These results indicate that ethyl acetate fraction from Cedrela sinensis A. Juss shoots extracts could be applicable to new functional antibacterial cosmetics.

In Vitro Cytotoxicity, Skin Regeneration, Anti-wrinkle, Whitening and In Vivo Skin Moisturizing Effects of Oncheongeum (온청음 물 추출물의 세포독성, 피부재생, 주름개선, 미백 및 보습 효과)

  • Objectives: The objective of this study was to evaluate the effects of cytotoxicity, skin regeneration, anti-wrinkle, whitening and skin moisturizing of Oncheongeum (OCE).Methods: The cytotoxicity of OCE lyophilized aqueous extracts (yield=13.82%) was observed against human normal fibroblast cells and B16/F10 murine melanoma cells by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium Bromide (MTT) assay, and skin regeneration and anti-wrinkle effects were also evaluated through the assay of collagen type I synthesis compared to the transformation of the growth factor (TGF)-β1, hyaluronidase, collagenase and matrix metalloproteinase (MMP)-1 inhibitory assays compared to oleanolic acid (OA), and elastase inhibitory effects compared to phosphoramidon disodium salt (PP). In addition, OCE’s whitening effects were measured by a tyrosinase inhibitory assay and melanin formation test in B16/F10 murine melanoma cells compared to arbutin, and skin moisturizing effects were observed through a mouse skin water content test, respectively. Results: No OCE treatment-related cytotoxic effects appeared on human normal fibroblasts and B16/F10 murine melanoma cells. OCE concentration-dependently increased the collagen Type I synthesis on human normal fibroblast cells, and also effectively inhibited hyaluronidase, elastase, collagenase and MMP-1 activities. In addition, OCE inhibited melanin production of B16/F10 murine melanoma cells and activity of tyrosinase. And significant and dose-dependent increases of skin water content were detected in OCE-treated mouse skin compared to vehicle control skins. Conclusions: OCE showed favorable and sufficient effects in skin regeneration, anti-wrinkle, whitening and skin moisturizing in this experiment. But more detail mechanisms and studies on the skin protective efficiency of in vivo are needed with the screening of active biological compounds in individual OCE herbs.
