  • Silicagel colum chromatography of the ether extract of the fruits of Angelica decursiva $F_R$. et SAV. gave four crystalline products of coumarins and one steroid. The following furocoumarins and ${\beta}-sitosterol$ were identifed by UV, IR, NMR spectra and physicochemical tests: ${\beta}-sitosterol$, iso-imperatorin, imperatorin, bergapten, umbelliferone.

  • The phytochemical investigation of Halocharis hispida revealed the presence of 1-heptatriacontanol, ${\beta}-sitosterol$, ${\beta}-sitosterol-3-O-glucoside$, kaempferol, vitexin and isorhamnetin-3-O-galactoside in addition to vanillic, ferulic, isoferulic, syringic and caffeic acids. The different isolated compounds were identified by different physical, chemical, chromatographic and/or spectral methods.

The Inhibitory Effects of Lactose-${\beta}$-sitosterol on the Inflammatory Responses of HMC-1 Cells and EoL-1 Cells

  • ${\beta}$-sitosterol glucoside exists in a variety of plants and have anti-tumor, anti-microbial, and immunomodulatory activities. Mast cells and eosinophils play important roles in a variety of inflammatory diseases, specifically asthma and atopic dermatitis. In the present study, we used lactose-${\beta}$-sitosterol (L-BS) and investigated the effect of L-BS on inflammatory responses of the human mast cell line, HMC-1 and the human eosinophilic leukemia cell line, EoL-1. In HMC-1 cells, L-BS significantly inhibited cell migration in response to stem cell factor without cytotoxicity. However, the mRNA expression of CC chemokine receptors (CCRs), including CCR1-5, were not altered after L-BS treatment in HMC-1 cells. LPS-induced IL-4 production was also suppressed by L-BS in a dose-dependent manner. In EoL-1 cells, the concentration ranging from 0.1 ${\mu}M$ to 10 ${\mu}M$ of L-BS had no cytotoxicity and had no effect on mRNA expression of major protein-mediators derived from activated eosinophils. However, 100 ${\mu}M$ of L-BS induced the apoptosis of EoL-1 cells in a time-dependent manner. This finding indicates the possibility of L-BS as a potential therapeutic molecule in inflammatory diseases and may contribute to the need to improve current therapeutic drugs.

Quantitative and Classification Analyses of Lupenone and ${\beta}$-Sitosterol by GC-FID in Adenophora triphylla var. japonica Hara and Codonopsis lanceolata

  • A simple GC method with a FID detector was developed in order to determine two main compounds (${\beta}$-sitosterol and lupenone) for Adenophorae Radix. ${\beta}$-Sitosterol and lupenone were analyzed by the gradient thermal ramping method. Nitrogen was used as the carrier gas at 108 kPa. The flow rate of gas was 2.0 mL/min; $2{\mu}L$ of filtered sample was injected at a split ratio of 1 : 80. This method was fully validated with respect to linearity, precision, accuracy and robustness. Further, this GC-FID method was applied successfully in order to quantify two compounds in an Adenophorae Radix extract. The GC analytical method for classification analysis was performed by repeated analysis of 59 reference samples in order to differentiate between Adenophora triphylla var. japonica Hara and 14 Codonopsis lanceolata. The results indicate that the GC-FID method is suitable and reliable for the quality evaluation of Adenophorae Radix.

가을재배 찰옥수수의 등숙에 따른 지방산 조성 및 Phytosterol 함량 변화 (Changes in Fatty Acid Composition and Phytosterol Content During Ripening Period of the Autumn-Sown Waxy Corn)

  • 찰옥수수 가을재배시 등숙에 따른 종실의 지방산조성과 phytosterol의 함량변화를 검토하여 고품질 풋옥수수 생산을 위한 기초자료로 활용하고자 본 연구를 수행하여 얻어진 결과를 요약하면 다음과 같다. 1. 찰옥수수 출사후 일수가 경과할수록 종실의 조지방 함량은 지속적으로 증가 하였고, 흑진주찰은 일미찰보다 조지방 함량이 높았다. 2. 출사후 일수에 따른 지방산의 조성비는 뚜렷한 변화를 보였는데, palmitic acid 및 linoleic acid는 점차 감소되었으나 oleic acid는 증가되었고, 흑진주찰은 일미찰에 비해 불포화지방산의 조성비가 다소 높았다. 3. 출사후 일수가 경과할수록 총 phytosterol의 함량은 증가하여 출사후 33일에 함량의 최대치를 보였는데, 일미찰의 총 phytosterol 함량은 937.7 mg/100 g, 흑진주찰은 867.9 mg/100 g이었으나 품종간 통계적 함량의 차이는 없었다. 4. Phytosterol의 조성은 품종간 차이를 보였는데, 일미찰은 ${\beta}-sitosterol$ > stigmasterol > campesterol 순으로 함량이 높았으나, 흑진주찰은 ${\beta}-sitosterol$ > campesterol > stigmasterol 순으로 함량이 높았고, 성분별로 볼 때 campesterol 함량은 품종간 차이가 없었으나 stigmasterol 및 ${\beta}-sitosterol$은 흑진주찰이 일미찰에 비해 함량이 높았다. 5. Palmitic, linolieic, linolenic acid는 campesterol 및 ${\beta}-sitosterol$과 유의한 부(-)상관이 있었고, stearic acid는 stigmasterol과 정(+)상관이 인정되었다. Oleic acid는 campesterol 및 ${\beta}-sitosterol$과 정상관이 있었으나 stigmasterol과는 부상관이 인정되어 찰옥수수 등숙중 종실의 지방산 조성은 phytosterol의 함량의 변이에 영향을 미치는 것으로 판단되었다.

  • In order to find out the inhibitors of acetic acid fermentation in Korean ginseng (Panax Sin son C. A. Meyer), total aglycone, panaxadiol, panaxadiol, oleanolic acid and ${\beta}$ -sitosterol were added to the basal medium, respectively, and a surface culture was carried out at 30$^{\circ}C$. The results were as follows: 1 . Saponins lost their activity to inhibit the acetic acid fermentation by hydrolysis. 2 Panaxadiol inhibited slightly, and the degree of inhibition was about 1/300 of that of free saponins. 3. Panaxadiol and oleanolic acid inhibited silighly similar to total aglycone. 4. Acetic acid fermentation was stimulated at the early stage when ${\beta}$-sitosterol was added to the media below the level of 0.000815%. But the fermentation was inhibited when media contained it more than that media 5. An over-oxidation of acetic acid was observed when the media contained total aglycone. panaxadiol, panaxatriol, oleanolic acid and ${\beta}$-sitosterol, respectively, while the media which contained sucrose, ginseng extracts ginseng saponins was shown not to be over-oxidized.

Identification of Metabolites of Phytosterols in Rat Feces Using GC/MS

  • $\beta$-Sitosterol, campesterol and stigmasterol have been known to the phytosterols the most frequently found in plants. Metabolism of phytosterols was investigated using rat feces and liver microsomes. Feces were collected after phytosterols (a well characterized mixture of $\beta$-sitosterol 40%, campesterol 30% and dihydrobrasicasterol) were administered orally (0.5 ${g/kg$) to rats. Metabolites of phytosterols were identified using GC/MS. Three peaks were eluted at 12.47, 12.65, 12.87 min and had characteristic molecular ions m/z 428, 430, 432, respectively. Three fecal metabolites were identified as androstadienedione, androstenedione, and androstanedione. No metabolites could be detected in the rat liver microsomal reaction mixture. The results suggest that the metabolites of phytosterols in rat feces are formed by oxidation at 3- position, saturation at 5- and 6- position, and 17- side chain cleavage in the rat large intestine.

  • ${\beta}-Sitosteryl-3-O-{\beta}-D-glucopyranoside$ was isolated from the seeds of Plantago asiatica (Plantaginaceae). The assignments of $^{13}C$ NMR spectra of ${\beta}-sitosterol$ and ${\beta}-sitosteryl-3-O-{\beta}-D-glucopyranoside$ were made by comparing with $^{13}C$ NMR spectra of cholesterol and $cholesteryl-3-O-{\beta}-D-glucopyranoside$. Our data indicate that the revision of previous $^{13}C$ NMR spectral assignment is needed.

  • Five steroids and two norsesquiterpene glycosides were isolated from the methanol extract of H. japonicus. Their structures were determined by means of physio-chemical and spectral data to be friedelin (1), stigmast-5-en-3-${\beta}$-ol (${\beta}$-sitosterol) (2), 7-keto-${\beta}$-sitosterol (3), 6${\beta}$-hydroxy-4-stigmasten-3-one (4), 7${\alpha}$-hydroxy-${\beta}$-sitosterol (5), 3-hydroxy-4,4-dimethyl-4-butyrolactone (6), daucosterol (7), (6S, 9S)-roseoside (8), and (9S)-drummondol-9-O-${\beta}$-D-glucopyranoside (spinoside B) (9). The compounds 1, 3, 4, and 6 - 9 were first isolated from this plant source.
