In vitro effects of nitrofurantoin, an antimicrobial agent for acute and chronic urinary tract infection, on the lung microsomal lipid peroxidation and the generation of reactive oxygen radicals were investigated to elucidate the biochemical mechanisms of its in vivopulmonary toxicity. The interaction of nitrofurantoin with porcine lung microsome resulted in significant lipid peroxidation. In addition, nitrofurantoin stimulated the generation of reactive oxygen radicals, $O^{-}_{2}{\cdot},\;H_2O_2$ as well as a highly reactive secondary oxygen species, $OH{\cdot}$. The stimulation of lipid peroxidation was inhibited not only by superoxide dismutase and catalase, but also by hydroxyl radical scavengers, mannitol and thiourea. Neither singlet oxygen $({^1}O_{2})$ was detected during the incubation of microsome with nitrofurantoin, nor lipid peroxidation was inhibited by singlet oxygen scavengers. When incubated anaerobically under the nitrogen atmosphere, the ability of nitrofurantoin to stimulatle lipid peroxidation was abolished. It appears that NADPH-dependent metaboliam of nitrofurantoin in pulmonary microsome under aerobic condition is accompanied by the stimulation of lipid peroxidation through the mediation of reactive oxygen radicals, particularly hydroxyl radical. It is strongly suggested from these results that the stimulation of pulmonary microsomal lipid peroxidation by the reactive oxygen radical may be a in vivo mechanism of pulmonary toxicity caused by nitrofurantoin.

Photodegradation rate of flupyrazofos in aqueous solution were measured under various test conditions mainly following the guidelines of U.S. EPA and OECD. It was observed that the flupyrazofos was degraded by simple hydrolysis but not degraded by light in pure water. Using acetone as a known photosensitizer, the minimal concentration of acetone needed to photo-degrade the flupyrazofos in % and molar terms were 0.047% and 0.006, respectively. When treated with acetone, it was also found that the ringlet oxygen is a very effective photo oxidant in the degradation of flupyrazofos, but the effect of hydroxyl radical was not observed at the treatment level of hydroxyl radical, isopropylbenzene. In an actinometer experiment, quantum yield of flupyrazofos (0.4 ppm with 2% acetone) was $17.66{\times}10^{-5}$ and degradation rate and half-life were 0.038/hr and 18.2 hours, respectively.
Using solid inorganic peroxides (including Li2O2, Na2O2, SrO2 and BaO2) as starting materials, three reaction paths for singlet oxygen (1O2) production were developed and studied. Their 1O2 emission spectra in the near- IR region and visible region from these reaction paths were simultaneously recorded by a near-IR sensitive Optical Multichannel Analyzer and a visible sensitive Optical Spectrum Analyzer, respectively. The comparison of their 1O2 emission spectra indicated that: (1) in term of the efficiency for 1O2 production, the gasliquid- solid reaction path (in which Cl2 or HCl and H2O reacted with the solid inorganic peroxides suspension in CCl4) was prior to the gas-solid reaction path (in which Cl2 or HCl reacted with the solid inorganic peroxides suspension in CCl4), but was inferior to the gas-liquid reaction path (in which Cl2 or HCl reacted with the solid inorganic peroxides solution in H2O or D2O); (2) the alkali metal peroxides (such as Li2O2 and Na2O2) was prior to the alkaline earth metal peroxides (such as SrO2 and BaO2) as the solid reactants, and Cl2 was favorable than HCl as the gas reactant in efficiency for 1O2 production in these reaction paths.
The antioxidant potentials of ten kinds of medicinal plants used as natural dyeing agents were tested. Among the plant extracts and floral waters that were examined in this study, the antioxidant activities of the DPPH and ABTS radical scavengers increased with increasing amounts of the extracts. The hot-water extracts from Aphis chinensis, Caesalpinia sappan L., Rumex crispus L., and Areca catechu effectively inhibited the DPPH and ABTS radicals at concentration below 0.1 mg/mL. The floral water obtained through steam distillation from Areca catechu, Rubia akane Nakai, Coptis chinensis, and Phellodendron amurense showed relatively valid antioxidant activities. In particular, the natural colorants extracted from Rumex crispus L., Areca catechu, and Aphis chinensis effectively suppressed the photogenerated singlet oxygen induced by a photosensitizer in in-vitro assay systems. The concentrations ($IC_{50}$) of the hot-water Aphis chinensis extract required to exert a 50% reduction effect on DPPH, ABTS, and singlet oxygen were found to be 8.5, 8.0, and $210{\mu}g/mL$, respectively. The study results suggest that extracts derived from naturally occurring colorants as well as floral waters could be used as natural antioxidants in the food and health-care industries.
The photodegradation of TCBs in humic acid solutions at pH 7 was evaluated under laboratory sunlight simulated irradiation for 8 and 42 days. The irradiation to TCBs in solution in the presence of humic acid produced trichlorophenols, dichlorophenols in addition to the minor photodegraded products such as dichlorobutadiene, dichlorobenzaldehyde and pentachlorobiphenyls. The formation of photodegradation products indicate that the excited singlet oxygen and solvated electrons play a major contributors to the photodegradation process.

The effect of antioxidants on photochemical reaction of pefloxacin by UVB (290~320 nm) was investigated and the possible mechanism of phototoxicity on the skin was also studied. The photo-degradation of pefloxacin by UVB was suppressed by cysteine, reduced glutathione and ascorbic acid, but was promoted by ${\alpha}-tocopherol$. Squalene, accounts for more than 10% of skin surface lipids, was peroxidized by pefloxacin through both radical and singlet oxygen mechanism.
Direct photoisomerizations of benzalpyrrolinones yield the corresponding E-isomers via a singlet state, since no effect of oxygen on the reaction rates was observed. In photodynamic therapy (PDT), oxygen plays important role. Because of singlet oxygen which is produced by activated photosensitizer after laser irradiation of specific wavelength. The aim of this study is to find how oxygen concentration affects anticancer effect in PDT. Groups were divided into PDT with oxygen applied group and only PDT applied group. PDT with oxygen applied group supplied oxygen for 15 minute before laser irradiation. In vitro, CT-26 cell was incubated with various concentration of photofrin $(50.0{\sim}0.05{\mu}g/ml)$ and was irradiated with 632nm diode laser 6hr after application of photofrin. The cell viability of two groups was assessed by MTT assay. In vivo, CT-26 cell line was transplanted into the subcutaneous tissue of BALB/c mouse. The anticancer effect of two groups was measured by tumor volume change. In vitro study, the cell viability was significantly decreased at $1.56{\sim}3.13{\mu}g/ml$ in PDT with oxygen applied group. In vivo study, the PDT with oxygen applied group significantly higher reduction rate of tumor volume 7 days after PDT compared to PDT only group. The high oxygen concentration might enhance the anticancer effect of the photodynamic therapy.
