Study on the Single Oral Dose Toxicity of High Quality Rice Varieties (국내육성 벼 주요 품종의 Ethanol 추출물 단회 경구투여 안전성 연구)

  • The present study was carried out to investigate the potential acute toxicity of ethanol extracts of the aleurone layer of Oryza sativa cvs. Obongbyeo, Ilpumbyeo, and Aranghyangchalbyeo by a single oral dose in ICR mice. The test article was orally administered once by gavage to male and female mice at dose levels of 0, 2.5, 5.0, and 10.0 g/kg body weight (n=10 for male and female mice for each dose). We examined numbers of deaths, general signs, weight measurement and biochemical analysis for sexes and doses of mice control and experimental groups. All mice were alive during the experimental period so can not yield death rate and $LD_{50}$. Any significant clinical symptom was not observed in all treated groups. No significant body weight changes in treatment groups in comparison with those of control groups was observed at any dose levels in experimental groups. Plasma glucose levels were valued both control and treated groups and there were no significant differences between groups. The activities of ALT and AST were not increase in all sample treated groups when compared with the control groups. The results suggest that the toxicity of Oryza sativa cvs. Obongbyeo, Ilpumbyeo, and Aranghyangchalbyeo are low and its $LD_{50}$ is over 10.0 g/kg body weight in both male and female mice.

  • Burkholderia pyrrocinia CAB08106-4 has an anti-bacterial effect on Garlic White Rot caused by Sclereotium cepivorum and Sclereotium sp.. It is an environmentally friendly microbial product that prevents and controls a variety of phytopathogens involving Garlic White Rot caused by Sclereotium cepivorum and Sclereotium sp.. The aim of this study was to assess and to compare the pathogenicity of Burkholderia pyrrocinia CAB08106-4 by single exposure of rats through several routes such as oral, intranasal and intravenous. For the acute toxicity / pathogenicity study, the animals were sacrificed on days 3, 7, 14 and 21, and macroscopically observed their organs to examine the numbers of internally-retained pesticidal microbes. Clinical examinations were performed daily during administration period, and body weight gain was evaluated. In the study, no clinical sign, weight gain and mortality were observed in relation to the administration of test article. The significant changes of internal/external microbes by test article were not detected. The pathological findings in relation to the administration of the test article in the necropsy were not observed. It could be concluded that the microorganism was not toxic or pathogenic in rats via oral, intranasal and intravenous route.

Primary dermal irritation study of oregano oil in rabbits (토끼에서 oregano oil의 일차 피부 자극시험)

  • The present study was carried out to investigate the skin irritation potential of oregano oil in rabbits. A volume of 0.5 ml of test article was applied to intact and abraded skins, respectively, for 24 h in 6 healthy male New Zealand White rabbits. Parameters measured during 72 h observation period were mortality, clinical signs, body weight changes, and local irritation. Treatment-related toxic symptoms, as evidenced by anorexia and decreased locomotor activity, were observed in all rabbits tested. Two rabbits out of the 6 total died on day 2 after the application of test article due to treatment-related toxicity. The mean body weight of test animals decreased progressively during the 72 h observation period. All animals showed very slight edema at 24 h and then recovered at 72 h. These animals also showed very slight to severe erythema and/or slight eschar formation at both 24 h and 72 h. The eschar formation induced by oregano oil was not recovered at the end of additional 11-day recovery period. Based on these results, it was concluded that a single dermal application of the undiluted oregano oil caused an increased incidence of clinical signs and death and a decrease in the body weight and showed a primary irritation index score of 2.79 indicating moderate irritation.

DNA Damage Effect of Botanical Insecticides Using Chinese Hamster Lung Cells

  • BACKGROUND: Botanical insecticides, especially Azadirachta Indica extract (AIE) and Sophorae radix extract (SRE) are widely used in Agriculture field. In our previous studies on genotoxicity test of AIE and SRE samples, a suspicious clastogenic properties was shown. Herein, we investigated the DNA damage effect of these botanical insecticide samples through the in vitro comet assay. METHODS AND RESULTS: Chinese hamster lung (CHL) fibroblast cell line was used, and methyl methanesulphonate was as positive control. Respective two samples of AIE and SRE were evaluated using Single Cell Gel Electrophoresis (Comet) assay and measured as the Olive tail moment (OTM). Results from this study indicated that all tested AIE and SRE samples did not show DNA damage in comet assay using CHL cells, compared with control. CONCLUSION: AIE and SRE samples used in this study were not cause genetic toxicity and are suitable for use as organic materials.

The Effect of Glutathione on High Dose Cisplatin-Induced Cellular Toxicity in Non-small Cell Lung Cancer Cell Lines (비소세포폐암 세포주에서 고용량 Cisplatin 세포독성에 대한 Glutathione의 효과)

  • Background : This study was designed to examine how glutathione, one of the nucleophilic sulfur compounds, effects the cisplatin cellular toxicity in the non-small cell lung cancer cell lines and normal lung epithelial cell line. Materials and Methods : Three cultured cell lines, the lung adenocarcinoma cell(NCL-H23), the lung squamous carcinoma cell(SK-MES-1) and the normal lung epithelial cell(L-132) line were exposed to various concentrations of cisplatin with or without glutathione. The relative viability was estimated as a means of measuring the cisplatin cellular toxicity using the MTT method. Results : In NCI-23, the response to cisplatin was sensitive but glutathione markedly increased the relative survival of the tumor cells by removing the antitumor effect of cisplatin. In both SK-MES-1 and L-132, the responses to cisplatin were less sensitive, and the chemoprotective effect of glutathione compared to and equal cisplatin dose was significantly higher in L-132 than in SK-MES-1(p<0.05). Conclusion : The protective effectes of of glutathione on cisplatin-induced cellular toxicity is more significant in normal lung epithelial cells than in squamous carcinoma cells.

Noninvasive Monitoring of Bleomycin-induced Lung Injury in Rats Using Pulmonary Function Test

  • The single intratracheal instillation (ITI) of bleomycin (BLM) is a widely used method for inducing experimental pulmonary fibrosis in rat model. In the present study, pulmonary function tests (PFTs) of tidal volume ($V_T$), minute volume ($V_M$), and respiratory frequency ($F_R$) have been applied to study their possibility as a tool to monitor the progress of BLM-induced lung injury in rat model. Rats were treated with a single ITI of BLM (2.5 mg/kg) or saline (control). Animals were euthanized at 3, 7, 14, 21, and 28 days post-ITI. Lung toxicity effects were evaluated by inflammatory cell count, lactate dehydrogenase (LDH) activity in the bronchoalveolar lavage fluid (BALF), and light microscopic examination of lung injury. The PFT parameters were measured immediately before the animals were sacrificed. BLM treatment induced significant cellular changes in BALF-increase in number of total cells, neutrophils, and lymphocytes along with sustained increase in number of macrophages compared to the controls at days 3, 7, and 14. BALF LDH level was significantly increased compared to that in the controls up to day 14. On day 3, infiltration of neutrophils was observed in the alveolar spaces. These changes developed into marked peribronchiolar and interstitial infiltration by inflammatory cells, and extensive thickening of the interalveolar septa on day 7. At 14, 21, and 28 days, mild peribronchiolar fibrosis was observed along with inflammatory cell infiltration. The results of PFT show significant consistencies compared to the results of other toxicity tests. These data demonstrate that the most suitable time point for assessing lung fibrosis in this model is 14 days post-ITI of BLM based on the observation of fibrosis at 14, 21, and 28 days. Further, the progress of lung injury can be traced by monitoring the PFT parameters of $F_R$, $V_T$, and $V_M$.

  • Developing effective tools for predicting absorption, distribution, metabolism, excretion properties and toxicity (ADME/T) of new chemical entities in the early stage of drug design is one of the most important tasks in drug discovery and development today. As one of these attempts, support vector machines (SVM) has recently been exploited for the prediction of ADME/T related properties. However, two problems in SVM modeling, i.e. feature selection and parameters setting, are still far from solved. The two problems have been shown to be crucial to the efficiency and accuracy of SVM classification. In particular, the feature selection and optimal SVM parameters setting influence each other, which indicates that they should be dealt with simultaneously. In this account, we present an integrated practical solution, in which genetic-based algorithm (GA) is used for feature selection and grid search (GS) method for parameters optimization. hERG ion-channel inhibitor classification models of ADME/T related properties has been built for assessing and testing the proposed GA-GS-SVM. We generated 6 different models that are 3 different single models and 3 different ensemble models using training set - 1891 compounds and validated with external test set - 175 compounds. We compared single model with ensemble model to solve data imbalance problems. It was able to improve accuracy of prediction to use ensemble model.

Acute Toxicity of Leuconostoc Citreum GR1 Isolated from Kimchi in Mice (김치로부터 분리한 Leuconostoc citreum GR1의 마우스에 대한 급성독성)

  • The present study was carried out to investigate the in vivo single-dose acute toxicity of Leuconostoc citreum GR1 (Leuc. citreum GR1), a lactic acid bacteria isolated from kimchi, in ICR male and female mice. The test article was orally administered once to both sexes of mice. The mortalities, clinical findings, autopsy findings, and body weight changes were monitored daily for two weeks. The male and female mice were gavaged with Leuc. kimchi GR1 of four doses (625, 1,250. 2,500 and 5,000 mg/kg). The oral $LD_{50}$ of Leuc. citreum GR1 was considered higher than 5,000 mg/kg. No significant changes in the general conditions, body weights, clinical signs and presence of gross lesions were observed in both sexes of mice to whom Leuc. citreum GR1 was administered orally. The results indicated that the 5,000 mg/kg dose of Leuc. citreum GR1 showed no adverse effect.

Toxicity of Eun-bi San Pharmacopuncture Injection inthe Muscle of Rats

  • Objectives: This study was carried out to analyze the single-dose toxicity of Eun-Bi San pharmacopuncture injected into the muscle of Sprague-Dawley (SD) rats. Methods: All experiments were performed at Biotoxtech, an institution certified to conduct non-clinical studies under the Good Laboratory Practice (GLP) regulations. Six week old SD rats reared by ORIENTBIO were chosen for this pilot study. The reason SD rats were chosen is that they have been widely used in safety tests in the field of medicine, so the results can be easily compared with many other databases. The Eun-Bi San pharmacopuncture was made in a clean room at the Korean Pharmacopuncture Institute (KPI, K-GMP). The constituents of the Eun-Bi San pharmacopuncture are Angelicae gigantis radix, Strychni semen and Glycyrrhizae radix. These were extracted at low temperature and low pressure in an aseptic room at the KPI. Doses of Eun-Bi San pharmacopuncture, 0.25, 0.5 and 1.0 mL, were administered to the experimental groups, and a dose of normal saline solution, 1.0 mL, was administered to the control group. This study was performed under the approval of the Institutional Animal Ethics Committee of Biotoxtech Co., Ltd. Results: No deaths or abnormalities occurred in any of the four groups. No significant changes in weight, hematological parameters or clinical chemistry between the control group and the experimental groups were observed. To determine if abnormalities existed in any organs and tissues, we used microscopy to examine representative histological sections of each specified organ; the results showed no significant differences in any of the organs or tissues. Conclusion: The results of this study suggest that treatment with Eun-Bi San pharmacopuncture is relatively safe and that its clinical use may be beneficial. Further evaluations and studies on this subject will be needed to provide more concrete evidence in support of these conclusions.

Utilization of Potato Starch Processing Wastes to Produce Animal Feed with High Lysine Content

  • This work aims to utilize wastes from the potato starch industry to produce single-cell protein (SCP) with high lysine content as animal feed. In this work, S-(2-aminoethyl)-L-cysteine hydrochloride-resistant Bacillus pumilus E1 was used to produce SCP with high lysine content, whereas Aspergillus niger was used to degrade cellulose biomass and Candida utilis was used to improve the smell and palatability of the feed. An orthogonal design was used to optimize the process of fermentation for maximal lysine content. The optimum fermentation conditions were as follows: temperature of 40℃, substrate concentration of 3%, and natural pH of about 7.0. For unsterilized potato starch wastes, the microbial communities in the fermentation process were determined by terminal restriction fragment length polymorphism analysis of bacterial 16S rRNA genes. Results showed that the dominant population was Bacillus sp. The protein quality as well as the amino acid profile of the final product was found to be significantly higher compared with the untreated waste product at day 0. Additionally, acute toxicity test showed that the SCP product was non-toxic, indicating that it can be used for commercial processing.
