[$Na^+-Ca^{2+}$ Exchange Curtails $Ca^{2+}$before Its Diffusion to Global $Ca^{2+}{_i}$ in the Rat Ventricular Myocyte

  • In the heart, $Na^{+}-Ca^{2+}$ exchange (NCX) is the major $Ca^{2+}$ extrusion mechanism. NCX has been considered as a relaxation mechanism, as it reduces global $[Ca^{2+}]_i$ raised during activation. However, if NCX locates in the close proximity to the ryanodine receptor, then NCX would curtail $Ca^{2+}$ before its diffusion to global $Ca^{2+}_i$ This will result in a global $[Ca^{2+}]_i$ decrease especially during its ascending phase rather than descending phase. Therefore, NCX would decrease the myocardial contractility rather than inducing relaxation in the heart. This possibility was examined in this study by comparing NCX-induced extrusion of $Ca^{2+}$ after its release from SR in the presence and absence of global $Ca^{2+}_i$ transient in the isolated single rat ventricular myocytes by using patch-clamp technique in a whole-cell configuration. Global $Ca^{2+}_i$ transient was controlled by an internal dialysis with different concentrations of BAPTA added in the pipette. During stimulation with a ramp pulse from +100 mV to -100 mV for 200 ms, global $Ca^{2+}_i$ transient was suppressed only mildly, and completely at 1 mmol/L, and 10 mmol/L BAPTA, respectively. In these situations, ryanodine-sensitive inward NCX current was compared using $100{\mu}mol/L$ ryanodine, $Na^+$ depletion, 5 mmol/L $NaCl_2$ and $1{\mu}mol/L$ nifedipine. Surprisingly, the result showed that the ryanodine-sensitive inward NCX current was well preserved after 10 mmol/L BAPTA to 91 % of that obtained after 1 mmol/L BAPTA. From this result, it is concluded that most of the NCX-induced $Ca^{2+}$ extrusion occurs before the $Ca^{2+}$ diffuses to global $Ca^{2+})i$ in the rat ventricular myocyte.

  • In this paper, we examined noise figure and the 3rd order intercept point in sensitivity single tone desensitization, and response attenuation by intermodulation spurious of receiver and also analyzed them through simulation based on J-STD-0.18, which is the minimum specification of PCS mobile station. The result of simulation was as follows ; In sensitivity specification, the total noise figure was 5.99dB and the 3rd order intercept point was -33.979dB. Ins ingle tone desensitization and intermodulation spurious response attenuation specification, the total noise figure was 6.375dB and the 3rd order intercept point was -26.99dBm. When there is an interference tone, the nosie figure and the 3rd order intercept point have been increasedly 0.367dB respectively, compared wit that of no interference tone. Because the amount of increase in the 3rd order intercept point was highly raised more than that of noise figure, mixer, closely related to interference tone should be specified in high 3rd order intercept point. At a frequency spacing of 1.25MHz and 2.05MHz from center frequency in IF SAW filter, attenuation should be more than 45dB.

  • To clarify characteristics of yolk protein of abalone, yolk protein was purified from the ovarian egg extracts of mature female Haliotis discus hannai by a gel chromatography of sepharose CL-4B. From the results of immuno-electrophoresis and Ouchterlony's diffusion test to male and female sera and ovarian egg extracts using antibodies raised against mature female and male sera and male sera and ovarian egg extrascts, it was identified that the mature female serum had female specific serum protein and its antigenecity shared with ovarian egg extracts. A single type of yolk protein was purified from ovarian egg extracts, and it was composed of two subunits. Their molecular weights were estimated to be approximately 166 KDa and 113 KDa by SDSPAGE. The antiserum against yolk proteins cross-reacted with a mature female specific serum protein and extracts of hepatopancreas of vitellogeing females, but did not reacted with extracts of hepatopancreas of mature male.

  • Today, wireless LANs are widely deployed in various places such as corporate office conference rooms, industrial warehouses, Internet-ready classrooms, etc. However, new concerns have been raised regarding suity. Currently, both virtual private network(VPN) and WEP are used together as a strong authentication mechanism. While security is increased by using VPN and WEP together, unnecessary redundancy occurs causing power consumption increase and authentication speed decrease in the authentication process. In this paper a new synchronization protocol for authentication is proposed which allows simple authentication, minimal power consumption at the mobile station, and high utilization of authentication stream. This is achieved by using one bit per a frame authentication, while main authentication process including synchronization is handled by access points. Computer simulation reveals that the proposed scheme significantly improves the authentication efficiency in terms of the number of authenticated frames and authentication speed compared with an earlier protocol employing a similar authentication approach.

  • A photovoltaic (PV) system generates electricity by installing a solar energy array; therefore, the photovoltaic system can be easily exposed to external factors, which include environmental factors such as temperature, humidity, and radiation. These factors-as well as shading, in particular-lead to power degradation. When there is an output loss in the solar cell of a PV module package, the output loss is partly controlled by the bypass diode. As solar cells become highly efficient, the characteristics of series resistance and parallel resistance improve, and the characteristics of reverse voltage change. A bypass diode is connected in parallel to the string that is connected in series to the PV module. Ideally, the bypass diode operates when the voltage is -0.6[V] around. This study examines the bypass diode operating time for different types of crystalline solar cells. It compares the reverse voltage characteristics between the single solar cell and polycrystalline solar cell. Special modules were produced for the experiment. The shading rate of the solar cell in the specially made solar energy module was raised by 5% each time to confirm that the bypass diode was operating. The operation of the bypass diode is affected not only by the reverse voltage but also by the forward bias. This tendency was verified as the number of strings increased.

  • Nowadays, dental zirconia is widely used as a framework material for a fixed dental prosthesis as well as a single restoration. However, clinical studies have reported high incidence of veneer chipping of zirconia-ceramic restorations compared to that of metal-ceramics. Several factors were raised as the possible causes of veneer ceramic chipping, however, it is still in debate. Recently, residual stresses in the veneer ceramics after cooling process gathers attention as one possible cause of chipping and many studies reported that the rate of cooling significantly influenced the types and the amount of residual stress. The purpose of current review was to briefly describe the effect of cooling rate on the residual stress in zirconia-ceramics. It was also described that the different behavior of residual stress between zirconia-ceramics and metal-ceramics following different cooling rate.

A Study on Optimal Combustion Conditions with a Design and Manufacture of the Long-Stroke Slow Speed 4 Cycle Diesel Engine (장-행정 저속 4 사이클 디젤기관의 제작 및 최적 연소조건에 관한 연구)

  • Recently, fuel prices have been continually raised in diesel engine. Such a change in the fuel price influences enormously the development trend of marine diesel engines for slow speed, In other words, the focus was shifted from large diameter and high speed to low fuel consumption. Accordingly, more efforts are being made for engine manufacturing and development to develop highly efficient engines. In this study. a single cylinder 4 stroke cycle DI slow speed diesel engine was designed and manufactured, a 4 stroke cycle was configured and basic performances were evaluated. The results are as follows. The optimal fuel injection timing had the lowest value when specific fuel consumption was in BTDC 8~$10^{\circ}$, a little more delayed compared to high speed diesel engines. Cycle variation of engines showed about 5% difference at full loads. This is a significantly small value compared to the cycle variation in which stable operation is possible, showing the high stability of engine operation is good. The torque and brake thermal efficiency of engine increased with an increase of engine 250-450 rpm. but fuel consumption ratio increased from the 450 rpm zone and thermal efficiency abruptly decreased. Mechanical efficiency was maximally 70% at a 400 rpm that was lower than normal engines according to the increase of mechanical frictional loss for cross head part. The purpose of this study was to get more practical engines by comparing the above results with those of slow speed 2 stroke cycle diesel engines.

Effects of Culture Methods on the Growth Rates and Fatty Acid Profiles of Euglena gracilis (배양방법에 따른 Euglena gracilis의 성장 및 지방산 조성)

  • The quality and quantity of live food sources strongly influence the success of fish production in farming operations. Thus, critical studies of live forage species are a crucial element for progress in fish aquaculture. The fat content of food is an especially important determinant of growth in marine fish. Omega-3 highly unsaturated fatty acids (HUFA) are essential components of diet that determine the nutritional value of larval fish. Euglena is a protist that has potential as a forage species. These single-celled organisms have plant and animal characteristics they are motile, elliptical in shape and 15–500 μm in diameter. Their nutritional content is excellent, but most studies have focused on cells raised in autotrophic culture. We therefore examined differences in the lipid and fatty acid contents, and the growth of Euglena cells grown under autotrophic, heterotrophic, and mixotrophic conditions. Biomass production reached 15.03 g/L, 12.28 g/L, and 3.66 g/L under mixotrophy, heterotrophy, and autotrophy, respectively. The proportional n-3 HUFA content differed among culture methods: 10.04%, 5.80% and 10.01% in mixotrophic, heterotrophic and autotrophic cultures, respectively. Mixotrophy was to be the best form of cultivation for improving the growth and nutritional content of Euglena.

  • Larval excretory-secretory products of Anisakis simplex are known to cause allergic reactions in humans. A cDNA library of A. simplex 3rd-stage larvae (L3) was immunoscreened with polyclonal rabbit serum raised against A. simplex L3 excretory-secretory products to identify an antigen that elicits the immune response. One cDNA clone, designated as ${\alpha}$-methylacyl CoA racemase (Amacr) contained a 1,412 bp cDNA transcript with a single open reading frame that encoded 418 amino acids. A. simplex Amacr showed a high degree of homology compared to Amacr orthologs from other species. Amacr mRNA was highly and constitutively expressed regardless of temperature (10-$40^{\circ}C$) and time (24-48 hr). Immunohistochemical analysis revealed that Amacr was expressed mainly in the ventriculus of A. simplex larvae. The Amacr protein produced in large quantities from the ventriculus is probably responsible for many functions in the development and growth of A. simplex larvae.

Assessment of Tissue Perfusion Following Conventional Liposuction of Perforator-Based Abdominal Flaps

  • Background The effect of liposuction on the perforators of the lower abdominal wall has been investigated in several studies. There are controversial results in the literature that have primarily demonstrated the number and patency of the perforators. The aim of this study was to determine the effect of liposuction on the perfusion of perforator-based abdominal flaps using a combined laser-Doppler spectrophotometer (O2C, Oxygen to See, LEA Medizintechnik). Methods Nine female patients undergoing classical abdominoplasty were included in the study. Perforators and the perfusion zones of the deep inferior epigastric artery flap were marked on the patient's abdominal wall. Flap perfusion was quantitatively assessed by measuring blood flow, velocity, capillary oxygen saturation, and relative amount of hemoglobin for each zone preoperatively, after tumescent solution infiltration, following elevation of the flap on a single perforator, and after deep and superficial liposuction, respectively. Results The measurements taken after elevation of the flap were not significantly different than measurements taken after the liposuction procedures. Conclusions The liposuction procedure does not significantly alter the perfusion of perforator-based abdominal flaps in the early period. The abdominal tissue discarded in a classic abdominoplasty operation can be raised as a perforator flap and has been demonstrated to be a unique model for clinical research.
