The Status of Distribution for Native Freshwater Turtles in Korea, with Remarks on Taxonomic Position

  • Korean freshwater turtles are divided into native turtles, Chinemys reevesii in Geoemydidae, Pelodiscus sinensis in Trionychidae, and imported turtles, Trachemys scripta elegans, T. s. scripta, Pseudemys rubriventris and Ocadia sinensis. In this study, we determined the distribution of Korean freshwater turtles based on a field study and literature study, and considered the taxonomic position of unidentified native turtles. The study was carried out between May 2010 and November 2011 during the day time, and the capturing tool used to collect turtles consisted of net and metal frame. C. reevesii and P. sinensis were found in 16 and 11 places, respectively. C. reevesii, which inhabits the Korean peninsula, is a taxonomically controversial species, but it is presumed that the turtle belongs to Mauremys. Moreover recent taxonomic studies of the soft-shell turtle have shown that the Korean native species is P. maackii, and P. sinensis was imported from abroad and has been in the natural habitats of Korea since 1970s. The exterior shape and skeletal form of P. maackii and P. sinensis are nearly similar. However, the skin color and yellow spots on the shell could be used to distinguish the two species.

The Study on Depigmentation Effects of Aloe, Camellia sinensis and Mel (알로에(蘆회), 녹차(綠茶), 꿀(蜂蜜)의 미백효과에 관한 연구)

  • Objectives : This study was performed to investigate the depigmentation effects of Aloe, Camellia sinensis and Mel. Methods : Inhibition of tyrosinase activity, melanin production & melanoma cell viability in cultured B16 melanoma cells, UV screen and cytoprotective effects on PC12 cells injured by hydrogen peroxide were measured. Results : Aloe has some inhibitory effects on tyrosinase activity, on the other hand Camellia sinensis and Mel do not have. They did not show any inhibitory effects on melanin production in melanoma cells and cytoprotective effects on PC12 cells injured by hydrogen peroxide. Aloe and Camellia sinensis have some inhibitory effects on UV screen. Conclusions : This study shows that Aloe and Camellia sinensis which were generally used for external application have some depigmentation effects. Following this, We should use them for whitening agents and the depigmentation effects of the other natural subjects which were generally used for external application should be examined.

The Energy Flow and Mineral Cycles In a Zoysia japonica and a Miscanthus sinensis Ecosystem on Mt. Kwanak 4. The Cycles of Phosphorus (관악산의 잔디와 억새 생태계에 있어서 에너지의 흐름과 무기물의 순환 4.인의 순환)

  • In this investigation, the accumulation mineralization and annual cycle of organic P has been studied in grassland ecosysterns of a Z japonica grassland and a M sinensis grassland on Mt. Kwanak. The basic models of the accumulation and mineralization for ash components of a grass-litter have been presented as the equations (1), (2), and (3). The equations (7)~(10) for organic P are derived from these basic concepts. There was a highty significant relationship between organic matter and organic P. The estimates between organic matter and organic P correlated very high significance. The parameter factors k or k' of mineralization of organic P for the Z. japonica and M sinensis asslands were k=0.412 or k'=0.292 and k=0.224 or k'=0.183, respectively. The time required for a cycle to be completed from organic P to inorganic P of 50, 95 and 99 % are 3.9, 16.7 and 27.8 years in the Z. japonica grassland and 4.1, 17.7 and 29.4 years in the M sinensis grassland. The annual P cycle formulae for mineralization were based on the equations (5), (11) and (12). Annual yields of mineralization for organic P in the steady state grasslands of Z. japonica and M sinensis were 0.407 and 0.504g /$m^2$, respectively.

Growth Environment Characteristics and Decline in Mt. Seunghak's Miscanthus sinensis Community (승학산 참억새군락의 생육환경 특성 및 쇠퇴에 관한 연구)

  • Mt. Seunghak's Miscanthus sinensis community is not only a landscape resource in terms of cultural services within the Ecosystem Services but also a site that is visited by many mountaineers in autumn. As the current Miscanthus sinensis community has been experiencing a rapid decline due to Korean forest succession characteristics, ongoing artificial management is thought to be needed for landscape resource use. The purpose of this study was to determine growth environment characteristics and the cause of the rapid decline of the Miscanthus sinensis community in Mt. Seunghak, which is located inside a large city with a large scale and outstanding accessibility. As the Miscanthus sinensis community is the representative early vegetation that appears temporarily in dry, barren soil, the Miscanthus sinensis community in Korean forest succession tends to be unsustainable. As the current soil on Mt. Seunghak is inappropriately fertile for the Miscanthus sinensis community, other wetland woody plant communities are anticipated to succeed it. If Miscanthus sinensis community maintenance is needed for Miscanthus sinensis landscape scenery, various alternatives apart from overall Miscanthus sinensis community maintenance should be determined for cost-effective management. For example, while many byways toward the inside of the Miscanthus sinensis community have affected the Miscanthus sinensis community growth environment, the installation of wooden fences and ropes has been a control in approach. As a result of this positive effect, many byways toward the inside of Miscanthus sinensis community have been restored naturally. Through viewable range analysis, as good scenery sites on the observatory have a good viewable range on the main trail as well, if these scenery sites are intensively managed, effective Miscanthus sinensis ccommunity management will be done despite maintenance budget cutbacks. This study is expected to be used as a basic material regarding the alternatives for a sustained Miscanthus sinensis community and the possibility of cultivating other growth in poor soils of fallow fields and unused land.

Reduction of Mitochondrial Electron Transferase in Rat Bile duct Fibroblast by Clonorchis sinensis Infection (간흡충(Clonorchis sinensis)감염에 의한 흰쥐 담관 섬유모세포 미토콘드리아 전자전달효소의 감소)

  • Fibroblasts are the most common cells in connective tissue and are responsible for the synthesis of extracellular matrix components. The fibrosis associated with chronic inflammation and injury may contribute to cholangiocarcinoma pathogenesis, particularly through an increase in extracellular matrix components, which participate in the regulation of bile duct differentiation during development. Mitochondria produce ATP through oxidative metabolism to provide energy to the cell under physiological conditions. Also, mitochondrial dysfunction and oxidative stress have been implicated in cellular senescence and aging. Alternations in mitochondrial structure and function are early events of programmed cell death or apoptosis and mitochondria appear to be a central regulator of apoptosis in most somatic cell. Clonorchis sinensis, one of the most important parasite of the human bile duct in East Asia, arouses epithelial hyperplasia and ductal fibrosis. Isolated fibroblast from the bile ducts of rats infected by C. sinensis showed increase of cytoplasmic process. In addition, decrease of cellular proliferation was observed in fibroblasts which was isolated from normal rat bile duct and then cultured in media containing C. sinensis excretory-secretory product. However, the effects of C. sinensis infection on the mitochondrial enzyme distribution is not clearly reported yet. Therefore, we investigated the structural change of C. sinensis infected bile duct and mitochondrial enzyme distribution of the cultured fibroblast isolated from the C. sinensis infected rat bile duct. As a result, C. sinensis infected SD rat bile ducts showed the features of chronic clonorchiasis, such as ductal connective and epithelial tissue dilatation, or ductal fibrosis. In addition, fibroblast in ductal connective tissue was damaged by physical effect of fibrotic tissue and chemical stimulation. Immunohistochemically detected mitochondrial electron transferase (ATPase, COXII, Porin) was decreased in C. sinensis infected rat bile duct and cultured fibroblast from infected rat bile duct. It can be hypothesized that the reason why number of electron transferase decrease in fibroblast isolated from the rat bile duct infected with C. sinensis is because dysfunction of electron transport system is occurred mitochondrial dysfunction, increase of ROS (reactive oxygen species) and apoptosis after chemical damage on the cell caused by C. sinensis infection. Overall, C. sinensis infection induces fibrotic change of ductal connective tissue, mutation of cellular metabolism in fibroblast and mitochondrial dysfunction. Consequently, ductal fibrosis inhibits fibroblast proliferation and decreases mitochondrial electron transferase on fibroblast cytoplasm. It was assumed that the structure of bile duct could not normalized and ductal fibrosis was maintained for a long period of time according to fibroblast metamorphosis and death induced by mitochondrial dysfunction.

Quality Characteristic and Antioxidant Activities Maejakgwa Added Cedrela sinensis Powder (참죽 분말을 첨가한 매작과의 품질 특성 및 항산화 활성)

  • Cedrela sinensis is a Korean traditional wild herb that has a special taste and aroma. This study is conducted to investigate the effect of Cedrela sinensis powder on the antioxidant activity and the quality characteristics of Maejakgwa (Korean traditional cookie). Maejakgwa was prepared with different amounts of Cedrela sinensis powder (in ratios of 0, 5, 10, 15 and 20% to the flour quantity). The antioxidant activity was estimated by DPPH free radical scavenging activity and the total phenol content in Cedrela sinensis powder and Maejakgwa. For analyzing the quality characteristics, the bulk density and pH of the dough, moisture contents, volume, color, texture profile analysis, and sensory evaluations were measured. The bulk density, moisture contents, volume, total phenol contents and DPPH free radical scavenging activity of Maejakgwa significantly increased with increasing Cedrela sinensis powder (P<0.001), while the pH of the dough, L values and b values of Maejakgwa significantly decreased with increasing Cedrela sinensis powder content (P<0.001). The consumer acceptability score for 10% Cedrela sinensis Maejakgwa ranked significantly higher (P<0.001) than those of the other groups in overall preference, flavor, taste, crispiness and color. From these results, we suggest that Cedrela sinensis is a good ingredient for increasing the consumer acceptability and functionality of Maejakgwa.

A rare duodichogamous flowering system in monoecious Toona sinensis (Meliaceae)

  • Background: Duodichogamy is the rarest form of dichogamy in angiosperms, which is characterized by flowering in the sequence of male${\rightarrow}$female${\rightarrow}$male. Disentangling factors promoting duodichogamy require the discovery of more duodichogamous species in angiosperms. However, extremely limited information on duodichogamous species makes it difficult to make general conclusions. Given the inflorescence morphology and flowering characteristics, the Meliaceae family is highly likely to contain duodichogamous species. Methods: We selected 48 individuals from 20 populations in Korea and investigated their flower morphology, arrangement of flowers by sexual condition within inflorescences, and flowering phases and duration of male and female flowers of Toona sinensis (Meliaceae) for 5 years (2011-2015) to determine if the species shows duodichogamous flowering. Results: Toona sinensis belonging to Meliaceae possessed functionally unisexual flowers with rudimentary parts of the opposite sex. The floral organs in male were larger than those in female, except for ovary length and width. In dichasium, male flowers were observed on primary or lateral branches, whereas female flowers were borne only on lateral branches. Overall, individuals from six different populations flowered in the male${\rightarrow}$female${\rightarrow}$male sequence, thereby male is blooming far longer than female flowers at the level of individual trees (male vs. female = 17-20 days vs. 2-4 days). Conclusions: This is the first study to report a duodichogamously flowering species, T. sinensis, within Meliaceae. Several flowering characteristics observed from T. sinensis may be important clues used to discover additional duodichogamous Meliaceae species. Short flowering period and relatively small number of female flowers, which is analogous to reduced ovule numbers observed in other duodichogamous species, may intensify male-male competition in T. sinensis. This study contributed to narrowing down potential candidates of duodichogamy based on their geographic distributions and flowering time.

Sex Ratios of Juveniles Hatched by Constant Temperatures Artificial Incubation From Eggs Collected in the Natural Nest of Soft-Shelled Turtle, Trionyx sinensis Strauch (1862) (한국산 자라, Trionyx sinensis (1862)의 자연산란둥지에서 수거한 난들의 항온인공부화에 의해 출생한 새끼자라들의 성비)

  • Recently, in many reptiles (14 genera of turtles in five families), common characteristics of incubation temperatures are known to determine the sexes of hatchlings in many species of turtles, including the map turtles, painted turtles and snapping turtles, emys turtle, etc. According to many researcher's reports, in general, incubation at $25^{\circ}C$ (cooler temperatures) produces all or mostly males, however, incubation at $31^{\circ}C$ (higher temperatures) or higher produces all or mostly females. Exceptionally, even cooler temperature ($20^{\circ}C$) produce females, they produced all or mostly females. Accordingly, it is well-known that incubation temperature is the sex determining agent in these turtles. However, this paper presents study of the sex ratio and nest ecology in natural spawning nest: Observations on hatching sex ratios of eggs collected from natural nests of T. sinensis are similar to a previous report of the same genus Trionyx in the soft-shelled turtles. However, this genus (or species) showed some different phenomena to other kinds of turtles such as various kinds turtles mentioned above. After collection of naturally spawned eggs (17 eggs of T. sisnensis) on the natural nests, a laboratory experiment by the constant incubation temperatures was conducted with natural fluctuating soil temperatures in the natural nest with the soft-shelled turtle, T. sinensis. And also laboratory experiments were conducted using constant incubation temperatures of $25^{\circ}C$ (cooler temp.) and $30^{\circ}C$ (higher temp.) with the turtle, T. sinensis. Exceptionally, it was confirmed that the first and second incubation temperatures can't control sex-determination in the freshwater soft-shelled turtle, T. sinensis. The sex ratio approximated 1:1 (${\chi}^2=0.06$, P>0.05 (the Ist experiment). And the sex ratio approximated 1:1 independently of incubation temperature (${\chi}^2=0.33$, P>0.05 (the 2nd experiment). Consequently, temperature has no effect on sex determination in the genus Trionyx in a soft-shelled turtle.

Egg Production of Clonorchis sinensis in Different Strains of Inbred Mice (근교계 마우스에서 간흡충 기생기간과 산란력의 변동)

  • In order to compare the intraspecific variation in host-parasite relationship of Clonorchis sinensis, six strains of inbred mice, ICR, DDY, GPC, BALB/c, nude and DS, were infected orally with 20 metacercariae of C. sinensis. The biologic incubation period of C. sinensis was the shortest in DDY mice, 21.2 days in average, followed by GPC 21.4, BALB/c and DS 23.2, ICR and nude 23.4 days, respectively. The fertile period of the cuke was also the longest in the DDY strain, 164 days on average, followed by GPC 132, BALB/c 97, nude 37, DS 32 and ICR 28 days. The egg-laying capacity of the cuke in DDY and GPC was relatively high and stable compared with the other four strains of mice. It was found that there are intraspecific variations in biologic incubation period, fertile period, and fecundity of C. sinensis. The DDY mouse is likely to be the most suitable experimental animal among the six strains of the mice tested. Key words: Mouse strain, Clonorchis sinensis, egg-laying capacity.

High Prevalence of Clonorchis sinensis and Other Zoonotic Trematode Metacercariae in Fish from a Local Market in Yen Bai Province, Northern Vietnam

  • A small survey was performed to investigate the recent infection status of Clonorchis sinensis and other zoonotic trematode metacercariae in freshwater fish from a local market of Yen Bai city, Yen Bai province, northern Vietnam. A total of 118 fish in 7 species were examined by the artificial digestion method on March 2016. The metacercariae of 4 species of zoonotic trematodes, i.e., C. sinensis, Haplorchis pumilio, Haplorchis taichui, and Centrocestus formosanus, were detected. The metacercariae of C. sinensis were found in 62 (69.7%) out of 89 fish (5 species), and their intensity of infection was very high, 81.2 per fish infected. Prevalences of 3 intestinal flukes, H. pumilio, H. taichui and C. formosanus, were 75.0%, 47.6%, and 31.7% in positive fish species, respectively, with the metacercarial intensities of 15.5, 10.3, and 2.2 per fish infected. From the above results, it has been confirmed that various species of freshwater fish continue to play the role of the infection source of C. sinensis and other zoonotic trematodes in Yen Bai city, Yen Bai province, northern Vietnam. It is of particular note that the prevalence and intensity of C. sinensis metacercariae are much higher than those reported in previous studies in fish in northern Vietnam.
