Bacterial Traits Involved in Colonization of Arabidopsis thaliana Roots by Bacillus amyloliquefaciens FZB42

  • Colonization studies previously performed with a green-fluorescent-protein, GFP, labeled derivative of Bacillus amyloliquefaciens FZB42 revealed that the bacterium behaved different in colonizing surfaces of plant roots of different species (Fan et al., 2012). In order to extend these studies and to elucidate which genes are crucial for root colonization, we applied targeted mutant strains to Arabidopsis seedlings. The fates of root colonization in mutant strains impaired in synthesis of alternative sigma factors, non-ribosomal synthesis of lipopeptides and polyketides, biofilm formation, swarming motility, and plant growth promoting activity were analyzed by confocal laser scanning microscopy. Whilst the wild-type strain heavily colonized surfaces of root tips and lateral roots, the mutant strains were impaired in their ability to colonize root tips and most of them were unable to colonize lateral roots. Ability to colonize plant roots is not only dependent on the ability to form biofilms or swarming motility. Six mutants, deficient in abrB-, sigH-, sigD-, nrfA-, yusV and RBAM017410, but not affected in biofilm formation, displayed significantly reduced root colonization. The nrfA- and yusV-mutant strains colonized border cells and, partly, root surfaces but did not colonize root tips or lateral roots.

  • 차세대 정지영상 압축방식인 JPEG2000의 엔트로피 코더는 컨텍스트 추출부(BPC)와 산술부호화기(AC)로 구성되는데 본 논문에서는 효율적인 컨텍스트 추출부 설계에 새로운 알고리즘을 제안하였고, 설계하였다. BPC(Bit Plane Coding)는 context 기반의 부호화기를 사용하였고, 현재의 SigStage register의 값과 상위 비트 플랜의 column 값을 가공한 데이터와 현재의 column 값을 이용하여 코딩패스를 미리 예측하는 기법을 사용하여, 각 bit plane에서 사용되어지는 상태 정보 레지스터와 이 상태 정보 레지스터를 접속하는 Access time을 줄일 수 있다. 본 논문에서 제안된 방법으로 설계된 Bit Plane Coding은 Verilog HDL 모델링후 Xilinx FPGA technology를 이용하여 합성한 후 동작을 검증하였다.

가스하이드레이트 구조 변형을 통한 메탄 저장에 관한 연구 (Study of Methane Storage through Structure Transition of Gas Hydrate)

  • Structure H formation experiments were conducted in a semi-batch stirred vessel using methane as the small guest substance and neohexane(NH), tert-butylmethylether(TBME) and methylcyclohexane(MCH) as the large molecule guest substance (LMGS). The results indicate that the rates of gas uptake and induction times are generally dependent on the magnitude of the driving force. When tert-butyl methyl ether is used as the LMGS rapid hydrate formation, much smaller induct ion time and rapid decomposition can be achieved. Liquid-liquid equilibrium (LLE) data for the above LMGS with water have been measured under atmospheric pressure at 275.5, 283.15K, and 298.15K. Effects of Asymmetric Airway Inertance on Mean Lung Volume During High Frequency Ventilation(HFV)

  • A possible asymmetry in airway inertance was modeled based on previously reported radiographic visualization data of the airway wall fluctuation in intact dogs. Effects of asymmetric Inertance on mean lung volume during high frequency ventilation (HFV) were evaluated in terms of mean inertive pressure drop across the airways. It was found that a negligible inertlve pressure drop was expected ($<1{\;}cmH_20$) in normal subjects, while a sig- nificant rise in mean alveolar pressure compared to mean mouth pressure by approximately $3{\;}cmH_20$ was resulted for about 40% airway fluctuation representing bronchoconstriction state by Histamine induction. These results demonstrate that asymmetric Inertance could lead patients with airway diseases to a significant lung hyperinflation (LHI), and bronchodilation treatment is recommended prior to applying HFV to prevent those patients from a possible barotrauma.

  • 니클로사미드 일수화물의 결정구조는 흑연으로 단색화된 Mo-Ka 방사선을 사용하는 자동 CAD4 회절 기로 측정된 1976개의 독립반점을 이용하여 결정되었다. 결정계는 단사정계이며, 공간군은 P211c이고, 293k에서 단위세포 상수는 a=11.331(3), b=6.964 (2), c=7.347(4)A, p=98.20(3)°, Z=4이다. 구조는 ggg Parity군에 속하는 반불변수를 이용하는 직접법으로 해석하였으며, 완전행렬 최소자승법으로 정밀화하여 최종 신뢰도 R=0.046인 모형을 구하였다. 분자 모양은 ab평면상에 평행하게 놓이는 평면성 구조를 나타낸다.

  • Dry grinding of a mixture of CaCO$_3$ and Ca(H$_2$PO$_4$)$_2$.$H_2O$ was conducted using a planetary ball mill in order to investigate solid state reaction for a synthesis of hydroxyapatite(Ca$_{10}$(PO$_4$)$_{6}$(OH)$_2$, HAp) through mechanochemical treatment method. The raw materials, which are composed of waste oyster and calcium biphosphate Ca(H$_2$PO$_4$)$_2$.$H_2O$, were mixed and then treated mechanochemically. The synthesis of hydroxyapatite(Ca$_{10}$(PO$_4$)$_{6}$(OH)$_2$, HAp) from the mixture was almost completed by about 60 minute grinding. The formation of HAp monophase in the ground mixture was characterized through X-ray diffraction (XRD) analysis. Moreover, the formation of HAp monophase depending on the grinding time was improved by increasing the grinding time.ime.ime.

한국내 고추나물의 하이퍼리신 유전자(HyH)의 탐색과 유럽의 서양고추나물과 비교 (Detect of Hypericin (HyH) gene in Hypericum erectum in Korea and Comparison of H. perforatum in Europe)

  • 하이퍼리신(Hypericin, HyH)은 예로부터 성 요한의 풀(St. John's Wort)로 널리 알려져 있는 서양고추나물(Hypericum perforatum)에서 추출되는 약리성분이다. 서양고추나물은 국내에서는 자생하지 않으나 같은 속의 고추나물(H. erectum) 등이 이속에 속하며 우리나라에 자생한다 . 고추나물을 조직배양으로 RNA를 추출하고 cDNA를 합성한 후 Hyp 유전자를 추출하여 서열화한 결과, 전체 크기는 732 bp로 나타났으며 서양고추나물의 HyH-1과 거의 99.8% 서열 일치를 나타내었다. 이 서열의 152개 아미노산 역시 서양고추나물의 항산화효소와 98% 일치하였으며 자작나무와 능금속 식물에서 유발되는 알레르기 유발유전자와 약 60% 일치를 나타내었다. 본 연구 결과 우울증치료제로 사용되는 하이퍼리신 추출에 우리나라 자생종인 고추나물이 이용될 수 있을 것으로 사료된다.

상압건조 나노다공성 실리카 에어로젤에 대한 용매의 영향 (Influence of solvent on the nano porous silica aerogels prepared by ambient drying process)

  • Nano porous, transparent silica aerogels monoliths were prepared under ambient drying (1 atm, $270^{\circ}C$) condition by the combination of sol-gel process and surface modification with subsequent heat treatment. Three kinds of solvent, n-hexane, n-heptane and xylene, were selected in the point view of low surface tension and vapor pressure in order to restrain a formation of cracks during drying. Crack-free silica aerogels with over 93 % of porosity and below $0.14g/cm^3$ of density were obtained by solvent exchange and surface modification under atmosphere condition. Optimum solvent was confirmed n-heptane among these solvents through estimation of FT-IR, TGA, BET and SEM. Modified silica aerogel exhibited a higher porosity and pore size compare to unmodified aerogels. Hydrophobicity was also controled by C-H and H-OH bonding state in the gel structure and heat treatment over $400^{\circ}C$ effects to the hydrophobicity due to oxidation of C-H radicals.

만성 Babesia 감염견에서의 Diminazene Aceturate의 반복투여에 따른 혈액학 및 혈액화학적 변화 (Hematological and Serum Chemical Findings following Repeated Medication of Diminazene Aceturate in Canine Babesiosis)

  • Effects of repeated administration of diminazene aceturate (Hoechst Veterin r GmbH. Ger- many) that has been introduced as effective compound against Babesiosis and trppanosomiasis were investigated in dogs experimentally infacted with Babesia gibsoni. Adull mongrel dogs of both sexes were inoculated will nonpreserved infected blood and then maintained chronic anemia. A single dose of diminazene aceturate of 7mg/kg b.w. was administrated intramnuscularIy on day 7.1 arid ,7. Clin- ical and hcmatological findings following inoculation and medication were observed and 7enlm bio chemical analysis also was monitored. Parasitemia was detected between 3 and 6 days after inoculation. The rate of parasitized erythrocytes,1 in peripheral blood reached the peak on the 13th day and was maintained the percentage of 0.1 to 1.0 until the medication of diminazene aceturate. RBC was significantly (p<0.01) decreased on the 3rd day and then kept on decreasing. The lowest value was observed on the 16th day. WBC remained generally within normal ranges. PCV revea1ed the sig-nificant (p<0.01) decrease within the range of 24-27% and platelet was significantly (p<0.05) decreased during the period. Senum chemical values (ALT, AST. total bilirubin. LDH BUN, area- tinine, total protein. albumin and glucose) were within normal ranges during the experimental period. Serum CPK values were significantly (p<0.01) increased on the 3rd day. There was no clinically, sig-nificant difference in a single dose of diminazene aceturate of 7 mg/kg b.w. But the administration of diminazene aceturate of 14 mg/kg b.w. revealed vomiting and anorexia and one dog died in 30 hours after administration. The administration of 14mg/kg b.w. resulted in vomiting, salivation, actor- exia, tremor of head and involuntary movement and one dog died in 27 hours after administration. WBC, RBC, PCV and Platelet values were no significant difference and hematological findings revealed persistent anemia and thrombocytopenia during chronic anemia after inoculation. AST activity its was significantly (p<0.01) increased 11\\`from 3 days after medication and AST activity was on the same trend. Serum CPK activity revealed significant (p<0.01) increase within 6 hors)\\`s after every administration and decreased in 48 howl·s after administration.

알칼리성 Cellulase를 생산하는 호알칼리성 Bacillus sp. HSH-810의 분리 및 효소 특성 (Isolation and Characterization of an Alkaline Cellulase Produced by Alkalophilic Bacillus sp. HSH-810)

  • 일반 토양과 부엽토, 퇴비로부터 alkaline cellulase 생성이 우수한 균주를 분리한 후 형태적, 배양학적 및 생화학적 동정을 실시한 결과 호알칼리성 Bacillus sp. HSH-810 균주인 것으로 판명되었다. 분리균 Bacillus sp. HSH-810의 생육과 효소 활성은 $30^{\circ}C$와 pH 10.0에서 가장 높은 것으로 나타났다. 본 균주는 배지 중에 탄소원과 질소원, 무기염으로 1.0%의 CMC와 0.5%의 peptone, 0.02%의 $CaC1_2$, $CoC1_2$를 사용하였을 경우 최대의 alkaline cellulase생산성을 나타내었다. 효소의 최적 활성 pH와 온도는 각각 10.5와 $50^{\circ}C$였다. 이 효소는 pH 6.0-13.0과 $50^{\circ}C$의 온도에서 매우 안정하였다. 효소액에 계면활성제를 첨가하여 효소 활성을 측정한 결과, 효소액은 sodium-$\alpha$-olefin sulfonate (AOS)와 sodium dodecyl sulfonate (SDS), Tween 20, Tween 80에 대하여 안정성 이 높은 특징을 보였으나 0.1%의 linear alkyl-benzene sulfonate (LAS)를 첨가한 경우에는 효소 활성이 심하게 저해되었다.
