Pedigree Indexing of Imported American Brahman Breeder Cattle in the Philippines

  • Pedigree data were used to predict the performance of 1,166 imported Americas Brahman breeder cattle, composed of 104 bulls and 1,062 heifers with an average age of 24.4 months (range of 17 to 40 months). A total of 13 full sib, 10 maternal half sib, and 228 paternal half sib groups were identified, each with average group size of 2.08, 2.00, and 4.49, respectively. Only 758 (64.9% of total) imported cattle were found to have at least one ancestor with expected progeny differences (EPDs) reported in the Spring 1995 Brahman Sire Summary. Moderate average accuracy values of .71, .69, .52, and .52 for birth weight, weaning weight, yearling weight, and maternal milk, respectively, were noted for EPDs of the ancestors. Prediction equations were derived by multiple regression analysis of available EPDs of sire, paternal grand sire, and maternal grand sire. Based on pedigree indexes that involve various combinations of available ancestral information, the average predicted EPDs (lbs) for imported cattle were $1.76{\pm}0.54$, $14.93{\pm}4.86$, $25.10{\pm}9.50$, and $5.86{\pm}2.08$ for birth weight, weaning weight, yearling weight, and maternal milk, respectively. Significant correlations (p < .05) were also found between sire and son EPDs (+.27) for yearling weight; and between sire and paternal grand sire EPDs for birth weight (+.34), weaning weight (+.51), yearling weight (+.49), and maternal milk (+.55).

국내산 돼지고기의 원산지 검증을 위한 SNP Marker Set 개발 (Development of SNP Markers for Domestic Pork Traceability)

  • 본 연구는 돼지고기 원산지 식별에 활용될 수 있는 최적의 SNP marker set을 개발 및 확립하기 위해 수행되었다. 선발된 51개의 SNP marker들의 효율성, 다형성 및 독립성 검증을 실시하였으며 51개의 SNP marker set은 MassARRAY method에 의해서 Multiplex-PCR panel 4개로 디자인 되었으며, 농장별, 생산조합별, 모돈별, 웅돈별로 효과적으로 고유 유전자형 지문분석이 가능하게 제작되었고, 다른형태의 SNP 유전자형 분석 플랫폼에 적용될 수 있는 적절한 마커 갯수이다. 또한 51개의 SNP marker set을 적용하여 모의 실험 및 친자감별확율을 계산하였을 때 무작위 교배 집단(PI), 반형매 교배집단($PI_{half-sib}$)과 전형매 교배집단($PI_{sibs}$)을 통해 모의 실험을 한 결과 $5.63{\times}10^{-33}$, $4.35{\times}10^{-15}$ 그리고 $1.32{\times}10^{-15}$로 분석되었으며 친자확인률에서도 모두 100%에 가까운 확률값을 나타내었다. 따라서 본 연구에서 개발된 SNP marker set을 이용하여 돈육제품의 원산지를 추적에 이용한다면 개별돼지의 고유한 DNA 지문 정보를 생성할 뿐만 아니라, 이를 통하여 모돈과 웅돈을 식별하여 농장원산지를 확인이 가능 할 수 있을 것으로 사료된다. 따라서 국내산 돼지의 생산에서부터 돈육제품으로의 소비까지 이력추적이 가능한 도구로 제공 될 것이다.

테다소나무 7-1037 클론의 단일 반형매 풍매가계 6년생 생장에 대한 QTL mapping과 QTL 대립유전자 치환의 평균효과 (QTL Mapping for 6-Year-Old Growths of a Single Open-Pollinated Half-Sib Family of a Selected Clone 7-1037 in Loblolly Pine(Pinus taeda) and Average Effect of QTL Allele Substitution)

  • 테다소나무 7-1037 클론에서 얻은 반형매 풍매 차대의 반수체 DNA에 대해 AFLP 표지자 분석을 수행하여 유전연관지도를 작성하고, 6년생 때의 수고 및 흉고직경 생장에 대한 QTL mapping을 수행하였다. 121개 AFLP 표지자로 전체 연관거리 1,869 cM, marker간 평균거리 18.5 cM의 20개 framework map을 작성하였다. Composite interval mapping 방법에 의해 수고 생장의 전체 표현형 변이의 5.9%를 설명할 수 있는 l개의 QTL과 흉고직경 생장 변이의 3.9~5.6%를 설명할 수 있는 3개의 QTL을 동정하였으며, QTL 간의 상호작용 효과는 없었다. 수고 생장에 대한 QTL의 유전적 효과는 39.6cm이었고, 흉고직경 생장에서는 7.20~9.41 mm인 것으로 추정되었다. 상기 QTL들과 가장 가깝게 연관되어 있는 표지자를 이용하여 대립유전자 치환의 평균효과(average effect of gene substitution)를 산출한 결과, 수고생장에서는 44.3 cm, 흉고직경 생장에서는 8.38~11.81 mm이었다. 테다소나무의 생장에 대한 가계내 개체유전력을 0.2 이하로 가정한다면, 본 연구에서 확인된 QTL은 7-1037 클론의 반형매 풍매 차대가 보유한 상가적 유전분산의 26.8%를 설명할 수 있어 표현형에 의한 개체선발보다 선발효율에서 5배나 높은 것으로 추정되었다. 본 연구에서 제시된 반형매 풍매 차대를 이용한 QTL mapping 분석은 채종원을 기반으로 하는 선발육종 사업에서 필요한 breeding value 등의 정보를 제공한다는 측면에서 인공교배 가계를 이용한 기타의 QTL 분석에 비해 보다 현실적이고 적용성이 높은 방법론이라 생각된다.

  • In this paper, we perform a simulation study of genetic model-free age-of-onset methods in linkage tests which has been proposed by Zhu et al. (1997). They performe. Haseman-Elston regression on a set of bipolar pedigree data using each of three dependent variables: a binary trait indicating disease concordance or discordance, a binary trait adjusted for age-of-onset, and the residuals from a survival analysis. We compare the powers of the proposed test statistics for various situations. Simulations that we have carried out show that the gains in power are observed when the residuals from a survival analysis are used in linkage tests.

Comparison of Mating Systems in Populations of Gleditsia japonica var. koraiensis

  • The mating systems of two groups of natural populations of Gleditsia japonica var. koraiensis in Korea were determined using allozyme analysis. The result suggests that G. japonica var. koraiensis is predominantly outcrossing. The tm values of eight populations in Korea varied from 0.667 (Mdh-1) to 0.938 (ldh-1), giving an average 0.820. Population and individual outcrossing estimates were associated with flowering tree density or degree of spatial isolation. The reason for relatively low outcrossing rates of some populations could be attributed to reduction of effective population sizes of sib for the medicine, small population size, and isolation of flowering mature trees. The heterozygote excesses were observed in some natural populations, whereas other populations exhibited varying degrees of inbreeding and heterozygotes deficit. Thus, selection against homozygotes operated in the progeny populations throughout the life cycle.

Characterization of Phosphoinositide-3-kinase, Class 3 (PIK3C3) Gene and Association Tests with Quantitative Traits in Pigs

  • This study deals with the characterization of porcine PIK3C3 and association tests with quantitative traits. PIK3C3 belongs to the class 3 PI3Ks that participate in the regulation of hepatic glucose output, glycogen synthase, and antilipolysis in typical insulin target cells such as those in the such as liver, muscle system, and fat. On the analysis of full-length mRNA sequence, the length of the PIK3C3 CDS was recorded as 2,664 bps. As well, nucleotide and amino acid identities between human and pig subjects were 92% and 99%, respectively. Five SNPs were detected over 5 exons. We performed genotyping by using a SNP C2604T on exon24 for 145 F$_2$ animals (from a cross between Korean native boars and Landrace sows) by PCR-RFLP analysis with Hpy8I used to investigate the relationship between growth and fat depot traits. In the total association analysis, which doesn' consider transmission disequilibrium, the SNP showed a significant effect (p<0.05) on body weight and carcass fat at 30 weeks of age as well as a highly significant effect (p<0.01) on back fat. In an additional sib-pair analysis, C allele still showed positive and significant effects (p<0.05) on back fat thickness and carcass fat. Moreover, the effects of C allele on the means of within-family components for carcass fat and back fat were estimated as 2.76 kg and 5.07 mm, respectively. As a result, the SNP of porcine PIK3C3 discovered in this study could be utilized as a possible genetic marker for the selection of pigs that possess low levels of back fat and carcass fat at the slaughter weight.

Microsatellite marker를 활용한 칡소의 유전적 다양성 및 유연관계 분석 (Studies on Genetic Diversity and Phylogenetic relationships of Chikso (Korea Native Brindle Cattle) Using the Microsatellite Marker)

  • 본 연구는 microsatellite marker를 이용하여 국내에서 사육되고 있는 칡소 9지역 간의 유전적 거리 분석 및 계통 지도 작성 등의 계통유전학적 분석을 실시하였다. 11종의 MS 마커를 이용하여 대립유전자의 수(No. of allele)를 확인한 결과 8에서 24개로 확인되었으며, 기대이형접합율(expected heterozygosity, Hexp)은 0.672에서 0.834 범위 안에 나타났으며, 관측이형접합율(observed heterozygosity, Hobs)은 0.687에서 0.886, 다형성정보지수(Polymorphism information content, PIC)은 0.638에서 0.876로 확인되었다. 무작위 교배집단(Random)으로 가정하였을 경우 동일개체 출현빈도는 11개의 marker를 사용하였을 때, 5.24×10−19 빈도로 출현하는 것을 확인 할 수 있었으며, 반형매 교배집단(Half-sib)과 전형매 교배집단(sib)으로 가정했을 경우에는 2.63×10−06, 2.63×10−06으로 각각 확인되었다. 이러한 결과는 칡소의 개체식별 및 친지확인 marker로 11종의 MS marker가 충분히 활용 가능할 것으로 사료된다. Phylogenetic tree (Neighbor-Joining tree), Principle Component Analysis (PCA) 그리고 Factorial Component Analysis (FCA) 분석을 통해 9 지역의 칡소 집단 간의 유연관계를 확인하였다. 이러한 결과는 칡소 품종을 중요한 가축유전자원으로써 인식하고 국내 타 품종과의 유전적 차별화와 순수성 보존과 능력을 개량하는데 있어 기초 자료로 활용 가능할 것으로 사료된다.

  • Perpendicular guideway mechanism has a different behavior with horizontal guideway mechanism due to the slider weight. So, to decrease its weight effect, counter balances such as weight type and hydraulic cylinder type are used. But it can also make another motion behavior by weight rate of slider and counter balance, its connected position. Therefore, it is necessary to find design parameters and analyze their effect. This paper dealt with the optimal design of perpendicular guideway mechanism. For analysis, the theoretic model as same as real machine tool and sib plate to adjust the clearance was used. Rotational angle and displacement of slider, pressure distribution, friction distribution were calculated. Stick slip, intermittent motion of slider according to friction change was adapted to the perpendicular guideway mechanism.

Seismic evaluation of isolated skewed bridges using fragility function methodology

  • A methodology, based on fragility functions, is proposed to evaluate the seismic performance of seismic isolated $45^{\circ}$ skewed concrete bridge: 1) twelve types of seismic isolation devices are considered based on two different design parameters 2) fragility functions of a three-span bridge with and without seismic isolation devices are analytically evaluated based on 3D nonlinear incremental dynamic analyses which seismic input consists of 20 selected ground motions. The optimum combinations of isolation device design parameters are identified comparing, for different limit states, the performance of 1) the Seismic Isolated Bridges (SIB) and 2) Not Seismic Isolated Bridge (NSIB) designed according to the AASHTO standards.

Genetic diversity of Halla horses using microsatellite markers

  • Background: Currently about 26,000 horses are breeding in Korea and 57.2% (14,776 horses) of them are breeding in Jeju island. According to the statistics published in 2010, the horses breeding in Jeju island are subdivided into Jeju horse (6.1%), Thoroughbred (18.8%) and Halla horse (75.1%). Halla horses are defined as a crossbreed between Jeju and Thoroughbred horses and are used for horse racing, horse riding and horse meat production. However, little research has been conducted on Halla horses because of the perception of crossbreed and people's weighted interest toward Jeju horses. Method: Using 17 Microsatellite (MS) Markers recommended by International Society for Animal Genetics (ISAG), genomic DNAs were extracted from the hair roots of 3,880 Halla horses breeding in Korea and genetic diversity was identified by genotyping after PCR was performed. Results and conclusion: In average, 10.41 alleles (from 6 alleles in HTG7 to 17 alleles in ASB17) were identified after the analysis using 17 MS Markers. The mean value of $H_{obs}$ was 0.749 with a range from 0.612(HMS1) to 0. 857(ASB2). Also, it was found that $H_{\exp}$ and PIC values were lowest in HMS1 (0.607 and 0.548, respectively), and highest in LEX3(0.859 and 0.843, respectively), and the mean value of $H_{\exp}$ was 0.760 and that of PIC was 0.728. 17 MS markers used in this studies were considered as appropriate markers for the polymorphism analysis of Halla horses. The frequency for the appearance of identical individuals was $5.90{\times}10^{-20}$ when assumed as random mating population and when assumed as half-sib and full-sib population, frequencies were $4.08{\times}10^{-15}$ and $3.56{\times}10^{-8}$, respectively. Based on these results, the 17 MS markers can be used adequately for the Individual Identification and Parentage Verification of Halla horses. Remarkably, allele M and Q of ASB23 marker, G of HMS2 marker, H and L of HTG6 marker, L of HTG7 marker, E of LEX3 marker were the specific alleles unique to Halla horses.
