Presently, we report a simple, reproducible and high frequency plant regeneration in Hibiscus syriacus L. using axillary buds. H. syriacus was regenerated from axillary buds directly or through a callus phase. Regenerated shoots were directly induced from young and fresh axillary buds cultured on Murashige and Skoog medium (MS) supplemented with 0.01 mg/L of the growth regulator thidiazuron (TDZ) after 2 weeks of culture. Directly induced shoots were transferred to hormone-free MS medium and root development was observed after 6 weeks. On the other hand, old and stale axillary buds were regenerated to shoots via callus induction on MS medium containing 0.01–2 mg/L TDZ after 4 weeks. A TDZ concentration of 0.01 mg/L was most effective in callus formation. Green callus was transferred to MS medium containing 0.01 mg/L α-naphthalene acetic acid (NAA) and 0.5 mg/L benzylaminopurine (BA). After 4 weeks, callus had developed into multiple shoots. Plantlets were formed from 10 week cultures of single shoots on hormone-free MS medium. Regenerated plantlets were cultured on MS medium for one month and then transferred to pots containing garden soil. Potted plants were acclimatized for one month and grown to maturity under greenhouse conditions. The present study has shown that various concentrations of plant growth regulator can be effective for in vitro plant regeneration of H. syriacus. The direct and indirect regeneration protocol presented here will be useful for understanding the manipulation and propagation of H. syriacus.

Lily is an important cut-flower in Korea and world as well due to it's a variety of flower colors and various sizes of flowers. To develop elite lily cultivars, conventional breeding techniques have been used so far. However, an introduction of tissue culture system in mass propagation of bulbs and regeneration of shoots with a high efficiency is prerequisite at this moment. Especially, growth of bulbs and shoots as well as reduction of browning is critical factor to proliferate bulbs with shoots of superior lines or cultivar in lily. For this purpose, we tried to test whether ascorbic acid, citric acid and silver nitrate in medium to facilitate growth of bulbs and shoots as well as reduction of browning of bulb scales in lily. As a result, ascorbic acid and silver nitrate showed no significant differences compared to control plants in the growth of bulbs and shoots. When bulb scales were treated with $150mg{\cdot}l^{-1}$ of citric acid, formations of shoots and bulbs showed best result. While, bulb scale treated with $100mg{\cdot}l^{-1}$ of citric acid showed the growth of shoot and root as well as increasing of fresh weight compared to other treatments. Regarding the reduction of browning, $150mg{\cdot}l^{-1}$ of ascorbic acid showed the best result with the less than 2%. Although more experiments with several commercial varieties are needed in the future to establish mass propagation of bulbs of lily, results obtained in this study are supposed to provide the basic knowledge and contribution in tissue culture system of lily.

This study was conducted to examine differences in shoot regeneration among chrysanthemum cultivars. Leaf explants of chrysanthemum cultivars 'Sulhwa', 'Puma', 'Geummokseo' and 'Sulpoong' were used. Explants cultured on the medium for 2 weeks formed calli at the cut surfaces. Shoots regenerated on MS basal medium supplemented with various concentration combinations of NAA and BAP. Explants were cultured under cool-white fluorescent lamps with a light intensity of $40\mu{Mm}^{-2}$.$s^{-1}$ for 16 $hday^{-1}$, at $25^{\circ}c$ and 70-80% relative humidity. 'Geummokseo' and 'Sulpoong' were the most responsive cultivars in shoot regeneration. Most effective medium for 'Sulhwa' and 'Puma' was MS basal medium supplemented with 10.0 $\mu{M}$ NAA and 5.0 $\mu{M}$ BAP and for 'Geummokseo' MS supplemented with 10.0$\mu{M}$ NAA and 20.0$\mu{M}$ BAP. Regeneration of multiple shoots was observed on MS basal medium supplemented with 1.0$\mu{M}$ or 10.0 $\mu{M}$ NAA and 5.0$\mu{M}$ BAP. High frequency regeneration of adventitious shoots from leaf explants and efficient induction of root from these regenerated shoots were obtained.

The response of dissolved oxygen (DO) concentrations caused significant change in root number, root length, coleoptile length, shoot length and leaf age of seedlings. The genotypic difference in the effect of DO also highly significant (P<0.01) for all of the seedling traits. The number and length of root were extremely inhibited at the condition of $0.39\pm0.09$ DO concentration. While the coleoptile elongated markedly in the lowest DO concentrations, the shoot did not develop. The root growth was improved slightly at the $1.39\pm0.27mg L^{-1}$, however, there were no difference among genotypes at these two low DO concentrations. As the DO concentration become higher, the growth of root and shoot was improved remarkably. Root number, root length and shoot length was significantly different between $20\;and\;30^{\circ}C$ in DO rich and normal conditions, the development of those traits were apparently accelerated in high water temperature, however those traits of seedlings in DO deficiency were not different between the two temperatures except for shoot length. On the other hand the coleoptile length was not affected by the stagnant water temperature; it was stimulated by the low DO concentration. The competition of DO was greater as the seedling density was increased in the stagnant water, therefore the seedlings grown under high density have long and white coleoptiles, and the growth of roots and shoots was retarded severely.

As the zygotic embryo of North American ginseng (Panax quinquefolius L.) matured during stratification over 203 days it grew from 0.75 to 5.2 mm. Embryo excision and culturing on media containing different concentrations of two growth regulators, gibberellic acid ($GA_3$, 1 to 10 ${\mu}M$) and benzyladenine (BA, 1 to 5 ${\mu}M$), during stratification, showed that shoot and root number and the shoot, root and cotyledon length increased with increased stratification time. Gibberellic acid was the more effective growth regulator for increasing shoot and root number and shoot, root and cotyledon lengths. Immature embryos (stratified for up to 63 days) needed growth regulators for further development. Cultures on $GA_3$ at the last culture date (stratified for 203 days) when embryos were mature, produced multiple shoots but there was no effect of $GA_3$ concentration. Benzyladenine inhibited shoot and root growth regardless of embryo stratification. Growth regulators had little effect on cotyledon length of mature embryos. Embryos cultured on $GA_3$ combined with BA were green on all culture dates whereas greening in the control and BA treatments increased with culture date. The BA treatments induced 100% swelling of the embryos on the final culture date while in the control and $GA_3$ treatments there was no swelling. There was little or no curling in the control and BA treatments and a linear decrease in curling with culture date in the $GA_3$ and $GA_3$ + BA treatments.

Tamarix chinenis blooms twice a you and its flowers, branches and leaves make the adjustment of tree shape. Propagation methods and growth characteristics of T. chinensis were studied in order to ascertain its potential use as one of vegetation resources for coast forestation and landscaping. The study results indicated that 1 or 2 you old hard wood cuttings showed higher rooting ratio than greenwood or semi hard wood cuttings. One to one mixture between vermiculite and pearlite appeared to be the best for bed soil, and sea sand and silt(loess) mixture was the next. Sea sand and granitic soil followed after. In terms of seasonal differences, spring cuttings showed the best rooting ratio, root number, and root length. Fall cuttings followed after spring cutting, and summer cuttings showed worst results regarding rooting ratio, root number, and root length. The best rooting promotion effects of growth regulators were observed with sea sand bed soils. There was no significant difference among growth regulators in terms of rooting and shoot growth. Low concentration below 100 ppm of growth regulators was enough for rooting promotion effect. In general, the number and mean length of roots and shoots were showed the excellent records in the sites with high rooting ratio. The study result strongly showed that T. chinensis can be considered as a suitable tree for coast forestation and landscaping because of its easy cutting propagation and rapid growth on saline lands.
The effect of nickel (Ni) on growth and some tolerance strategies with regard to heavy metal tolerance mechanism was investigated in radish (Raphanus sativus) seedlings. The protective effect of histidine on nickel stress conditions was also monitored. The seedling growth decreased with an increase in metal concentrations. The inhibitory effect was more pronounced in the root elongation than in the shoot elongation. Increasing Ni supply showed a progressive increase of Ni concentrations in the roots and shoots. Ni content was higher in the shoots than in the roots. In the presence of nickel, radish exhibited an antioxidative defense mechanism, as evidenced by the elevated malondialdehyde(MDA), showing that nickel is an efficient inducer of lipid peroxidation. Exposure of radish to elevated concentrations of nickel was accompanied by an increase in the proline content. Supplemental histidine in the presence of Ni ameliorated metal-induced growth inhibition and lipid peroxidation. Combinations of Ni and histidine resulted in a significant decline in proline content compared with Ni stress alone, indicating that histidine may provide protection against the adverse effect of Ni stress. From the results it is suggested that histidine is an efficient chelator by complexing metal ion within the plant and may playa role in nickel tolerance implicated in metal detoxification.
Morphological characteristic during formation of tobacco crown gall tumor and genetic tumor, and their differential response to growth regulator were investigated in in vitro culture. Crown gall tumor was induced from tumor tissue transformed by infecting Agrobacterium tumefaciens C58. Genetic tumor was induced from tumor tissue which was induced spontaneously from reciprocal interspecific hybrids between Nicotiana glauca (2n=24) and Nicotiana langsdorffii (2n=18). Morphological characteristic of crown gall tumor, genetic tumor, and teratoma shoot was very similar, and they were actively proliferated on hormone-free medium. Typical tumor callus and teratoma shoot formed from crown gall tumor on the hormone-free medium. On the contrary, tumor callus derived from genetic tumor formed as a crown gall tumor callus on the medium supplemented with 0.5 mg/L of 2,4-D, and lots of teratoma shoots without any root formed on the hormone-free medium. Root development from the teratoma shoots was hardly obtained on the medium with IAA, GA and active carbon. However, teratoma shoots with roots, as normal shoots, were initiated occasionally on the hormone-free medium. These shoots also formed new genetic tumor on the stem, which leaves formed lots of teratoma shoot on the hormone-free medium in in vitro culture.
Experiments were conducted to investigate the optimum season for mulberry graftages harvesting before the period of natural leaf fall in autumn. Harvesting season was examined at 10 days intervals from September 30 to November 20, 1985 and 1986, at nine sites in eight provinces. The results were : 1. Diameter of graftages increased until October 30. 2. Early harvest of graftages decreased survived rate, number of shoots, and total shoot length the next spring. The October 30 harvest yielded the best growth the following spring. 3. Harvests after October 20 allowed for good survial of mulberry shoots and sprouting the following spring. 4. Total N and P in plants increased with late harvests, K decreased, and Ca content was not affected by harvest date. 5. Total N, P, and Ca were higher in stem cortex than root bark, while K appeared to be equally distributed between root and shoot bark.
Combinations of plant growth regulators, darkness treatments, and the order of expanding leaves for explants were evaluated for optimizing in vitro shoot regeneration rate of 'Whangkeumbae' pear. In a MS medium, supplemented with $8.88{\mu}M$ 6-benzylaminopurine (BA) and $0.49{\mu}M$ indole-3-butyric acid (IBA), green foci were observed on the surface of the callus 8 days after culture initiation. Some adventitious buds were later induced from those green foci, resulting in the appearance of normal shoots. In a medium containing $22.20{\mu}M\;BA$, the surface of the callus became compact and greenish, and many adventitious buds were formed over the entire area of the callus surface. When comparing BA concentration via histological observation, the section which had been treated with $22.20{\mu}M\;BA$ exhibited closer cell aggregation than those with $8.88{\mu}M\;BA$. The darkness treatment enhanced the formation of adventitious shoots for up to 3 weeks. The youngest two expanding leaves, proximal to the shoot apex, were proved to be the most regenerative, and yielded the highest shoot number per regenerating leaf. A fourth strength MS medium, which was supplemented with $0.54{\mu}M\;NAA$, yielded good quality plantlets, with regard to root number and root length.
