Optimization of Explosive Compounds (TNT and RDX) Biodegradation by Indigenous Microorganisms Activated by External Carbon Source (외부탄소원으로 활성화된 토착미생물에 의한 화약물질(TNT and RDX) 분해 최적화)

  • Contamination of explosive compounds in the soils of military shooting range may pose risks to human and ecosystems. As shooting ranges are located at remote places, active remediation processes with hardwares and equipments are less practical to implement than natural solutions such as bioremediaton. In this study, a series of experiments was conducted to select a suitable carbon source and to optimize dosing rate for the enhanced bioremediation of explosive compounds in surface soils and sediments of shooting ranges with indigenous microorganisms activated by external carbon source. Treatability study using slurry phase reactors showed that the presence of indigenous microbial community capable of explosive compounds degradation in the shooting range soils, and starch was a more effective carbon source than glucose and acetic acid in the removal of TNT. However, at higher starch/soil ratio, i.e., 2.0, the acute toxicity of the liquid phase increased possibly due to transformation products of TNT. RDX degradation by indigenous microorganisms was also stimulated by the addition of starch but the acute toxicity of the liquid phase decreased with the increase of starch/soil ratio. Taken together, the optimum range of starch/soil ratio for the degradation of explosive compounds without significant increase in acute toxicity was found to be 0.2 of starch/soil.

  • In this study, we analyzed the angle of shooting based on the footage of shooting training of basketball student athletes taken by the basketball team leaders. In the shooting video, the shot height of the player and the trajectory of the ball were drawn through a solid line, and the player was shown to guide the player to adjust the angle of projection to a situation where the success rate was higher. Through this study, the video shows that efficient guidance can be made available to basketball players.

Effects of osmoticum treatments and shooting chances on the improvement of particle gun-mediated transformation in Phalaenopsis (유전자총을 이용한 팔레놉시스 형질전환 효율향상에 삼투압 조절제 및 발사횟수차이가 미치는 영향)

  • This study was carried out to develop an efficient transformation protocol via particle bombardment with PLBs (protocorm-like bodies) in Phalaenopsis. To achieve this aim, osmoticum treatment and an increasing shooting chances in particle bombardment process were applied for this study. In addition, pCAMBIA3301: ORE7 vector which contains a herbicide-resistance bar gene as a selectable marker and ORE7 gene as a gene of interests were employed. With regard to the increasing chances of shooting in particle bombardment, double shooting was the best results with 1.5 ~ 2.5 times higher than those of a single or triple shooting treatment in the productioon of PPT (D-L-phosphinothricin)-resistant PLBs. However, regeneration rate of shoots in double shooting was not high as a single shooting. Further, double shooting showed 35 ~ 40% higher than that of a single shooting in the frequency of browning. Regarding effects of different osmotic treatments, combination of 0.2 M sorbitol with 0.2 M mannitol showed the best results in transformation efficiency, regeneration of transformants and reduction of browning. Putative transgenic Phalaenopsis plants were analyzed by PCR analysis and confirmed the presence of bar and ORE 7 gene. Also, real-time PCR was conducted by using 21 transgenic plants and showed only 4 plants had one copy of transgene; whereas, the other 17 plants had more than 2 copies of transgene. Transgenic phalaenopsis plants produced in this study were transferred to pots and flowered normally without morphological variations in flower and leaf.

  • Effects of cutting time and scions section on shooting and plantlets growth characteristics were investigated to increase plantlets products by root cuttings in lacquer tree. Shoot induction from scion section was developed lately as it planted in earlier season. However, shooting rates were over 84% or more in cut planting on 15, 25 March and 5 April than sinee 15 April. Shoots were developed evenly as late cut planting. Generally, plant height, branch number, leaf number, stem diameter, and leaf area per plantlets was excellent in cut planting on 25 March and 5 April. Shooting rate from the root scion varied greatly by root sectionings. The rate of the 10 cm-long-scion was reached 84%, whereas only 54% in no trimmed root scion after 10 weeks of cutting. Regenerated plantlets from root scion without cut were slightly good in shape, but their height, number of branch per plantlets, and stem diameter were not significant.

  • Effects of growth regulators, scion length and scion diameter on shooting and seedling characteristics were investigated to increase seedling products by root cuttings of lacquer tree. Shoots developed and emerged above-ground from the root scion at 3∼5 weeks after cut plating, most of that was a shoot but some of that were 2∼3 shoots. Otherwise, non-shooting was the reason that cortical layer was decayed. Soaking in zeatin was the most effective on shooting of root cuttings and high concentration was effective. Also soaking on high contents of BA or kinetin was effective on shooting but was lower totally than soaking on zeatin. Seedling characteristics among 3 growth regulators and 3 concentrations were not any difference in statistical significant, but were move excellent than control. As root scion was tall, shoots were developed at early. 15cm length scion was the most excellent to use seedlings but 3cm length scion was wickedness. 10cm scion, however, was highest to product of seedlings than any others. 0.3∼0.5 cm length scion was showed 76% shooting rate at 10 weeks after cut planting, but seedling characteristics were not any difference among the kind of 3 diameters.

Development of Floating-Islands with a Sod Mat by Shooting and Rooting from Shoot Nodes of Common Reed (갈대 줄기의 마디부 발근을 이용한 뗏장 식물섬 개발)

  • The community of common reed (Phragmites australis) is expanded by the development of new shoots from nodes of rhizomes and old shoots in natural environments. We developed the useful technique to construct reed mats of floating islands developed from shoot nodes of reeds for the purpose of the rapid stabilization of vegetation and the application of adapted plants with their specific environmental conditions. The vegetation development was compared in the floating islands planted with the different reed samples. The reed samples were consisted of four types : long shoot in the length of 100 cm (AG I), short shoot of 15 cm (AG II), long rhizome of 100 cm (BG I) and short rhizome of 15 cm (BG II). Shooting started earlier in BG I and BG II than in AG I and AG II. But the rate of shooting was higher in AG I and AG II than in BG I and BG II. After four weeks, AG I and AG II were higher than BG I and BG II in the biomass and density of newly developed shoots. These results showed that the construction of sod mats by planting only reed shoots was a useful technique to develop vegetation on the floating islands.

Distribution and Behavior of Mixed Contaminants, Explosives and Heavy Metals, at a Small Scale Military Shooting Range (국내 소규모 군사격장 복합오염물질(화약물질 및 중금속)의 분포 및 거동)

  • A phase II site investigation and feasibility study was conducted at a military mortar shooting range near the demilitarized zone (Kyunggi, South Korea) to assess the extent of contaminants migration to the nearby Imjin river in which a flood control dam is under construction. The results showed that silty-clay soils around target areas were co-contaminated with heavy metals (Cd, Cu, and Pb) and explosives (HMX, RDX, and TNT). The total amount of contaminant was estimated to be 497.1 kg-RDX, 20.6 kg-HMX, 1.4 kg-TNT, 35.2 kg-Cd, 4,331 kg-Cu, and 5,115 kg-Pb, respectively. Both heavy metals and explosives were almost equally distributed on each soil particle size fraction. Neither subsurface soil samples nor ground water samples showed signs of contamination above the environmental criteria. The major migration route of contaminants was soil particles in surface run-off during rain at which a mass discharge rate of 30.0 mg-RDX/hour was observed.

Efficiency of Closed Cutting Propagation Affected by Closed Periods, Leaflet Number and Photoperiod in Rose (Rosa hybrida) (밀폐 기간, 소엽수 및 광주기에 따른 장미의 밀폐삽목 번식 효율)

  • When it comes to single node leafy stem cuttings of rose (Rosa hybrida), environmental management such as air temperature, relative humidity, and light affect productivity. In order to investigate the effect of air temperature and relative humidity on the cutting success rate and rooted cuttings quality, a transparent airtight box was used to implement a closed system. We have also tried to find out the most effective photoperiod and the number of leaflets in closed system using artificial light (white LED, 104.0 µmol·m-2·s-1 photosynthetic photon flux density). The first experiment was conducted for a total of 6 weeks under 4 airtight period conditions. The number of roots and longest root length decreased as the airtight period increased. But there were no significant differences in the survival rate, shooting rate, and rooting rate according to airtight periods. In the second experiment the results indicated that survival and shooting rate were significantly affected by the photoperiod (0/24, 2/22, 4/20, 8/16, and 16/8 h), the number of leaflets (0, 2, and 4 leaflets) of the cuttings and their interaction. The survival rate was the highest in the 16-h day length and 4 leaflets. By considering survival rate and shooting rate with energy efficiency, the 8-h day length and 2 or 4 leaflets were judged to be the most effective.

Effects of Cutting Media in Root Cuttings of Lacquer Tree (Rhus verniciflua Stokes) (옻나무 근삽시 삽상배지의 효과)

  • Effects of cutting media on shooting and seedling characteristics were investigated to determine productivity of seedling by root cuttings of lacquer tree. Shoots developed at 3 weeks after cut planting on PV (peat moss : Vermiculite : 1 : 1), PVS (peat moss : Vermiculite : Sand : 1 : 1 : 2) and PVSC (peat moss : Vermiculite : Sand : Clay loam : 1 : 1 : 2 : 2) media, and shooting rate was the highest in PVSC medium at 10 weeks after cut planting among the treatments. Shooting numbers per scion on PH and PVC media were 2.2 but those on HSC medium was 1.3, which was not significantly different among media. PVSC medium was suitable to use the root cuttings of lacquer tree as it meets the qualifications requirements. Seedlings on PVSC medium were excellent to transplant in field as tree height, branch number, leaf number, stem diameter and leaf area were good.

  • The grinding process is very complex and relates many parameters to control the process. As this reason, a theoretical analysis and a quantitative estimation of the grinding process has not been well established. In this study, the in-process monitoring system was suggested by applying the neural network for monitoring and shooting the malfunction of cylindrical plunge grinding process. This system used the power signals from the electric power meter. This neural network was composed of processing elements [4-(5-5)-3] with 4 identified power parameters. Because sensitivity is blunted some minute vibration components, the simulation result of this system has appeared about 10% erroneous recognition in the uncertain pattern and the average success rate of the trouble recognition was about 90%. Consequently, the developed system, which applied to the power signals, can be recognize enough to monitor the grinding process as in-process.
