Effects of Medium Components and Growth Regulators on Callus Development and Shoot Regeneration from Shoot Explants of Black Locust (Robinia pseudoacacia)

  • Effects of growth regulators and medium components were tested for shoot multiplication and callus growth from shoot explants of black locust. During shoot multiplication, callus growth at the cut end of shoot explants proceeded shoot development. The basal callus growth seemed to be a function of both mineral components and cytokinin supplemented in the medium. Maximum callus growth was induced by 0.5${\mu}$M BAP and the callus growth decreased as the level of BAP increased. Positive correlations were found between basal callus growth, and shoot multiplication and growth. Shoot multiplication was greatest on BSM medium (black locust shoot culture medium) supplemented with 1 $\mu$M BAP. With medium containing high nitrogen content, both shoot multiplication and growth were significantly enhanced. A new BRM medium was the most effective for rooting of black locust among three rooting media tested.

Effects of Plant Growth Regulators on in vitro Propagation of Echinosophora koreensis Nakai

  • To establish in vitro nodal culture conditions of Echinosophora koreensis Nakai, one of rare and endangered species famous for beautiful flowers in the Korean Peninsula, the influence of plant growth regulators (PGRs) on shooting and rooting from in vitro shoots was investigated. In shoot multiplication, addition of 6-benzylaminopurine (BA) to the half-strength Driver and Kuniyuki's media in the range of 2.22 to 8.88 ${\mu}M $induced 2.5 to 2.7 shoots per axillary bud; and addition of 2.27 ${\mu}M $ thidiazuron (TDZ) produced 3.2 shoots, during 4 weeks of culture, while zeatin and isopentenyl adenine (2ip) were not effective on shoot multiplication as observed from several combination treatments of BA with other PGRs. Shoots established were smaller than 2 cm in length, in most of the treatments. while in BA 8.88 ${\mu}M $ treatment more than 30% of shoots were longer than 2 cm and shorter than 4 cm. In rooting, naphthalene acetic acid (NAA) from 5.37 to 21.48 ${\mu}M $ showed the rooting rate from 40.0 to 62.5%. Indole butyric acid (IBA) addition had little effect on rooting (<10%), although some roots in IBA-containing media were longer than those in NAA. Micropropagation from axillary buds of nodular explants was applicable and promising to multiplication and conservation of Echinosophora koreensis Nakai.

정아배양에 의한 산돌배나무의 기내번식 (Micropropagation Using Shoot Tip Culture of Pyrus ussuriensis Maximowicz)

  • 산돌배나무의 기내번식 기술을 개발하고자 유묘의 정아를 재료로하여 기내 식물체 증식에 미치는 생장조절제의 효과를 시험하였다. 정아를 MS 배지에 BA 단독처리 혹은 BA+NAA 혼용처리 결과 대부분 줄기는 1-2개로 유도 되었으며 처리에 따른 차이는 나타나지 않았다. 줄기의 생장과정에서 정아괴저가 흔히 관찰되었으며 다경이 유도되는 줄기에서는 과수화 현상이 나타났고 줄기의 기부에는 캘러스가 형성되었다. 줄기의 증식 과정에서 BA+NAA 처리구에서 약 20%의 자발적인 발근이 이루어 졌다. 다경으로부터 얻어진 줄기는 gelrite 경화배지에서 기내발근이 저조하였으며, 기외삽목시 100 mg/L IBA 처리로 50%까지 발근되었다. 발근묘는 순화 후 정상적으로 생장하였다.

  • 복숭아 품종 백미조생, 대구보, 유명의 기내 신초 증식 및 발근에 미치는 생장조절제의 종류 및 농도의 영향을 알아보기 위하여 일련의 실험을 실시하였다. 기내 신초 대량증식에 효과적인 cytokinin은 2.0 mg/L BA처리였으며, BA와 auxin과의 혼용처리 결과 2.0mg/L BA와 1.0mg/L IAA혼용처리가 백미조생 품종의 신초 증식률에 가장 효과적이었다. 배양된 신초의 기내발근에 효과적인 auxin은 IAA였으며,그 농도는 백미조생에서는 5.0mg/L, 대구보에서는 3.0mg/L였다.

정단배양에 의한 Delphinium cv. Princess Caroline의 대량번식 (Micropropagation of Delphinium cv. Princess Caroline through Shoot Tip Culture)

  • Delphinium cv. Princess Caroline을 대량번식하기 위하여 신초를 MS 배지에 사이토킨닌과 오옥신이 단용 또는 혼용으로 첨가된 배지에서 배양하였다. 사이토킨닌 단용처리에서 kinetin이나 2iP 첨가배지보다는 BA 첨가배지에서 신초 증식율이 높았다. 농도는 BA 1.0-5.0 mg/L가 첨가된 배지에서 신초수가 5개 이상으로 증식이 양호하였다. BA와 IAA를 혼용처리한 배지에서는 BA 1.0-3.0 mg/L와 IAA 0.1-0.5 mg/L 첨가배지에서 신초수가 5개 이상으로 높은 신초증식을 보여주었다. 그러나 BA와 IAA를 혼용첨가 하였을 때, 생체중은 약간 증가하였으나 저농도의 오옥신 첨가에 의한 신초증식 효과는 나타나지 않았다. IBA 또는 NAA가 농도별로 첨가된 배지에서 신초는 거의 발근되지 않았으며, 활성탄소가 첨가된 배지에서도 전혀 발근이 되지 않았다. 기내에서 생산된 신초의 기부에 Rootone을 뭍혀 기외발근을 한 결과, 펄라이트 1과 버미큘라이트 1의 배합토에서는 발근율이 68%, 뿌리수 약 4개, 뿌리길이 2.2cm로 발근이 양호하였다.

Practical Factors Controlling in vitro Multiplication and Rooting in Empetrum nigrum var. japonicum, an Endangered Woody Species

  • The plant Empetrum nigrum, valued in the traditional system of medicine, is well known for its antibacterial, antifungal, and antioxidant properties. In the present work, the effect of removal of shoot apical meristem (SAM) on shoot proliferation was studied. It was observed that removal of SAM promoted shoot proliferation whereas intact tip resulted in higher survival percentage. Further, the effect of different concentrations of BA on above was also studied. During root formation the effect of light quality after treatment with IBA was investigated. For rooting, continuous red light without IBA resulted in maximum rooting percentage. The above factors when taken into consideration during micropropagation of this endangered plant can result in healthier plantlets. The results show that the species could be successfully conserved by in vitro propagation system.

티크의 기내 줄기 생장 및 발근에 미치는 LED (light-emitting diode) 효과 (Effect of light-emitting diode (LED) on in vitro shoot growth and rooting in teak (Tectona grandis L.))

  • 본 연구는 티크의 기내 생장 및 발근에 미치는 light-emitting diode (LED) 효과를 구명하기 위해 수행되었다. 티크의 정아 절편체의 줄기 신장은 LED 혼합광(BR, 50% 청색광 + 50% 적색광) 하에서 DKW 배지 배양 시 3.2 cm로 가장 좋았고, 줄기유도는 광원 및 배지(MS 및 DKW) 효과가 그리 크지 않은 것으로 나타났다. BA 농도 처리 시 최대 줄기 유도는 BR 광원 및 1.0 mg/L 처리구에서 2.4개, 최대 줄기 신장은 BR 및 0.5 mg/L 처리구에서 4.94 cm로 가장 효과적이었다. 발근에 영향하는 IBA 농도 효과는 정아 절편체를 BR 및 0.5 mg/L 처리구에서 배양 시 최대 발근율(93.8%)과 최대 뿌리 길이 신장(1.3 cm)을 보였고, 뿌리 수는 0.2 mg/L (3.1개/절편) 처리구에서 가장 좋은 것으로 나타났다. 발근 식물체는 상토로 이식 후 순화 과정을 거쳐 정상적은 생장을 보였다.

  • 미선나무의 기내번식을 위하여 MS배지에 세 가지 싸이토키닌 (zeatin, kinetin 및 BA)을 세 가지 농도로 처리하여 액아 배양을 실시하였다. 줄기유도는 BA가 가장 효과적이었고, 생장은 zeatin이 좋았다. Kinetin은 2.0 및 5.0 mg/L 처리에서 줄기발생 효과가 있었으나 그 효과는 zeatin, BA에 비해 저조했다. 500 lux정도의 약광 하에서 배양은 싸이토키닌 처리에 관계없이 줄기의 생장을 촉진하였다. 발근은 IBA가 첨가된 1/2 GD배지에서 주효했다. 발근된 개체는 인공배양토에서 100% 활착되었고, 정상 생장이 가능했다. 이상의 결과에서 미선나무의 기내 번식이 가능함을 보여 주었다.

In vitro Conservation of Coleus forskohlii- an Endangered Medicinal Plant

  • Protocols for in vitro conservation was developed for Coleus forskohlii. Plants maintained both in field served as explant source. Shoot tips and single node cuttings were used to optimize protocols for in vitro multiplication. MS basal medium supplemented with $0.54\;{\mu}M$ naphthalene acetic acid (NAA) and $8.87\;{\mu}M$ benzy-ladenine (BA) induced multiple shoots in shoot tips and nodes. Shoot multiplication was amplified with a gradual decrease of BA concentration, leading to its final omission after 4 months. Concomitant rooting on multiplication media enabled successful establishment extra vitrum. For in vitro conservation studies, experiments were carried out with 2-3 week maintained in vitro plants under standard and reduced culture conditions (SCC, RCC). In vitro plants could be successfully conserved in full strength MS medium (FMS) under SCC for 6 months without subculture with full potential to regenerate, producing viable shoots and nodes. The root production remained unaffected due to conservation, showing high rooting activity in mannitol and low temperature treatments. Preset low temperature (15 and $10^{\circ}C$) and reduction in media constituents does not appear to favour conservation, although the former accomplished conservation levels equal to (FMS) under SCC.

Micropropagation of Echinosophora koreensis Nakai, a Korean Endemic Species in Danger, Using Axillary Buds

  • To establish in vitro axillary bud culture conditions of Echinosophora koreensis Nakai, one of Korean endemic endangered species famous for beautiful flowers, we tested the influence of plant growth regulators (PGRs) in shooting and rooting stage from in vitro plants. In shoot multiplication, addition of 6-benzylaminopurine (BA) to the media induced 2.5 to 3 shoots per bud during 4 weeks of culture. And media including 0.5 mg L-1 thidiazuron (TDZ) produced 3 to 4 shoots per bud. However, zeatin and isopentenyl adenine (2-ip) were not successful to increase shoot number, and the combination treatments of BA with other PGRs were also not effective. Shoots were smaller than 2 cm in length, in most of the treatments. In rooting, naphthalene acetic acid (NAA) treatments in the range of 0.5 to 4.0 mg L-1 appeared to increase rooting rate by 10% to 60% approximately when compared with the control but roots developed with callus clusters. Indole butyric acid (IBA) addition had little effect on rooting (below 10%), while some roots were longer than in NAA treatments and some shoots were longer on high IBA concentrations (4.0 to 8.0 mg L-1). It is suggested that micropropagation is a highly applicable and promising to multiplication and conservation of rare and endangered endemic species.
