Cloning, Purification, and Structural Characterization by 1D 1H-NMR of the PDZ domain of the Shank3 protein (Shank3 PDZ 도메인의 동정, 정제 및 1차 NMR 구조분석)

  • We wished to create a set of small molecular weight PDZ domain ligands that may be used in functional studies on the proteins AF6, PSD-95 and Shank. As a starting point, the Shank3 PDZ domain was cloned, purified, and characterized the structure of Shank3 PDZ domain by 1D $^1H-NMR$. The chemical shift dispersion of the proton signals indicates that the purified Shank3 PDZ protein is very pure and globally well folded. Currently, we are working on improving the yield of the protein production for complete NMR structural analysis of the Shank3 PDZ domain.

Disease Resistance of Race Differential Varieties and Improved Varieties(Nicotiana tabacum L.) on Black Shank (Phytophthora parasitica var. nicotianae) Infested Field (담배역병균(P. parasitica)에 오염된 포장에서의 Race 판별품종 및 육성품종(N. tabacum L.)의 발병반응)

  • This study was conducted to identify useful germ plasm sources of resistance to black shank, Phytophthora. parasitica, in Nicotiana tabacum and to characterize the interaction between host plant resistance to black shank and pathogen variability. The six internationally represen-tative set of black shank resistant and susceptible tobacco genotypes and four Korean lines were tested for black shank on the field infested with P. parasitica at Chonju Experiment Station, Korea Ginseng & Tobacco Research Institute during the 1992~1997 growing season. Almost 100% of the plants of the most susceptible genotype, Ky 14, did not survive. Complete survival of (MS Ky 14 x L8)F$_1$, which is resistant to reee 0 and ausceptible to reace 1, suggests that Race 0 is the only race of the P. parasitica in this bield. Coker 371 Gold and Beinhart 1000-1 having excellent resistance to black shank could be recommended as the parental cultivars for improving black shank resistance in Korea. Data of the diseased plants showed that KB 101 and KB 110 were medium high resistant and KB 103 and KB 108 being low resistant to black shank.

Estiniation of Genetic Parameters for Body Weight, Shank Length, and Shank Width in Korean Pheaaant (한국꿩의 체중과 정강이 길이 및 정강이 두께에 대한 유전모수 추정에 관한 연구)

  • This study was carried out to estimate the heritability and genetic correlation for body weight, shank length and shank width at various wk of age in growing Korean pheasant. All the measurements were done from one day to 20 wk of age with 4 wk interval. The heritability estimates for body weight were in the range of 0.54~0.59 for male and of 0.49~0.81 for female from 4 to 20 wk of age. The heritability estimates of male and female were in the range of 0.38~0. 82 and 0.41~0.67 for shank length, and of 0.48~0.88 and 0.49~0.71 for shank width, respectively. Genetic and phenotypic correlation coefficients between the measurements at different ages in each trait of body weight, shank length and shank width were medium to high positive values. Because the estimates of heritabilities and genetic correlations were high, the genetic gain for body weight is expected by either a direct selection or an early selection based on the record of early growing stage.

Effects of the Water Extract of Beef Shank Bones on the Physical and Sensory Characteristics of Cooked Rice for JeonJu Bibimbap (사골 추출물이 전주비빔밥용 밥의 물리적 및 관능적 특성에 미치는 영향)

  • The effects of the water extract of beef shank bones on the physical and sensory characteristics of cooked rice for JeonJu Bibimbap were investigated. Five kinds of cooked rice were prepared with 0%(control), 20%, 40%, 60%, and 100% of the beef shank bones extract. An increase in the beef shank bones extract concentration was associated with a lower moisture content of the cooked rice. The color(L, b value) of the cooked rice prepared with the beef shank bones extract became gradually darker during storage at $60^{\circ}C$. In regards to the textural properties of the cooked rice, the hardness increased with the addition of the beef shank bones extract and chewiness was changed in a similar pattern to that of hardness. However, the adhesiveness significantly decreased. In the sensory evaluation, the cooked rice prepared with 20 and 40% of the beef shank bones extract had the best sensory quality for JeonJu Bibimbap.

  • An experimental and numerical approach has been carried out for to see the noise characteristics of a circular diamond saw. The noise level measurement result for the several kinds of shanks shows us that the noise level reduction in high frequency range could be an effective way to control the noise problem. Sandwich type shank whose center part is laminated with a low Young‘s modulus material greatly reduces the noise level. The noise level for the shank where several curve shaped slits are provided in the circumferential direction is reduced considerably too. The response spectrum analysis by the FEM shows us to be an alternative for predicting the noise characteristics of the shank.

A Study on the Static Stiffness in the Main Spindle Taper of Machine Tool (공작기계 주축 테이퍼 결합부 정강성에 관한 연구)

  • This paper presents the experimental study of the static stiffness for the BT shank(7/24 long taper) and the HSK tool shank(1/10 short taper) in the main spindle taper of machine tool. The static stif71ess test was performed under different experimental conditions. It is turned out that the effective axial drawing force is larger than 6kN in the 7/24 test tool shank and BkN in the 1/10 test tool shank. As a test result, considering that the actual drawing force of the machining center is about 1300k2f and axal drawing force 12kN is equivalent amount as a 1220kgf, it is turned out that 1/10 test tool shank superior to 7/24 test tool shank in the static stiffness.

  • Black shank(Phytophthora parasitira roar. nicotianae) resistant burley tobacco (Nicotiana tabacum L.) germplasms, KB 104 and KB 106, were developed by Korea Ginseng & Tobacco Research Institute. KB 104 was developed from the single cross of Burley 21$\times$Newton 77, using a modified pedigree method. KB 104 was highly resistant to black shank, and its agronomic characteristics and chemical contents were comparable to those of Burley 21, and value per 10a was slightly higher than Burley 21, KB 106 is a maternally derived doubled haploid made by N. africana method from the single cross of Burley 21$\times$ Va 509. KB 106 was also highly resistant to black shank, had two more harvestable leaves per plant and flowered three days later than Burley 21 did. Total alkaloid and nicotine contents of KB 106 were significantly lower than those of Burley 21. But its nornicotine content was higher than Burley 21 5. A Study on the Static Stiffness in the Main Spindle Taper of Machin Tool (공작기계 주축 테이퍼 결합부 정강성에 관한 연구)

  • This paper presents the experimental study of the static stiffness for the BT Shank(7/24 Long Taper) and the HSK Tool Shank(1/10 Short taper). The static stiffness test was performed under different experimental conditions. The results obtained are as follows ; As known in the analysis results of the Load-Deflection diagram of the 7/24 Test tool shank, it is turned out that the diagram is a linear characteristics without regard to axial drawing force and according as the axial drawing force get to the 6kN, the static stiffness of the shank increase linearly. Thus the effective axial drawing force which maintains the static stiffness of the Main spindle taper of Machine Tool is larger than 6kN. It is found that the Load-Deflection diagram with 6kN of drawing force in the 1/10 Test tool shank is characterized by non-linear. But according as the axial drawing force is increasing by the 8kN, the diagram is characterized by linear. And increasing amount of deflection is about 60%. Therefore commendable axial drawing force is larger than 8kN. As a result, considering that the actual drawing force of the Machining Center is about 1300kgf and axial drawing force 12kN is equivalent amount as a 1220kgf, it is turned out that 1/10 Test tool shank superior to 7/24 Test tool shank in the static stiffness.

  • This study carried out to know whether corn shank can be used as the crude fiber source for the larval growth or not, The results obtained from this study are summarized as follows: 1. It seems that there are no bad effects of the artificial diet containing corn shank on feeding of the newly hatched larva. 2. Author did not find any bad effects of the artificial diet containing corn shank on larval growth and cocoon qualifies. 3. In view of these results, it seems that the corn shank is suitable for use as the crude fiber source for the silkworm artificial diet.

Breeding of Burley Tobacco KB 110 Resistant to PVY and Black Shank and its Agromomic Characteristics (감자바이러스 Y 및 역병 저항성 연초 버어리종 신품종 KB 110의 육성 및 농경적 특성)

  • The vein-necrosis strain or potato virus Y (PVY-Vff) and black shank (Phytophlhora parasitica roar. nicotianae) causes severe damage on burley tobacco(Wicotiana tabacum L.) in Korea, A new burley tobacco resistance to PVY and black shank, KB 110, was developed by Korea Ginseng and Tobacco Research Institute. It was developed from the cross of Burley 21 with TC 591 in 1990, and was backrossed to Burley 21 in the following season. TC 591 has resistance to PVY and moderate resistance to race 0 of black shank, but it is susceptible to tobacco mosaic vim (TMV). KB 110 was evaluated for its resistance to PVY, TMV and black shank in the greenhouse and at fields for preliminary and performance trials. KB 110 which has secreting glandular trichomes was resistant to PVY-VN, TW and black shank. It had an erect growth habit and two more leaves per plant than that of Burley 21, and matures two to three days later. It yielded approximately 3 percent more cured leaf than the standard cultivar Burley 21, but other plant characteristics were very similar to those of Burley 21. It had acceptable standards for chemical and physical characteristics of lured leaf on regional farm test in 1995-1997. KB 110 produced average yields of good quality tobaccos and was appeared to be resistant to PVY inwhere occurrence of the virus are severe chronic at burley growing area.
