Purge and Trap Headspace 법에 의한 솔잎(Pinus densiflora S.)의 휘발성 성분 (Volatile Components of Pine Needle(Pinus densiflora S.) by Purge and Trap Headspace)

  • 솔잎(Pinus densiflora S.)의 휘발성 성분을 추출시간과 온도를 달리하면서 headspace(purge & trap)-GC/MS로 분석하여 총61개의 휘발성 성분을 확인하였다. 확인된 성분을 관능기별로 분류하여 보면 hydrocarbons 35종, alcohols 16종, carbonyls 4종, ethers 3종, esters 2종, acid 1종으로 나타났다. 솔잎에서 확인된 주요 휘발성 성분으로는 $\alpha$-pinene(1.5~15.7%), $\beta$-pinene(0.4~5.4%), $\beta$-myrcene(13.2~15.6%), $\beta$-phellandrene (12.0~16.0%), cis-3-hexenol(4.0~18.3%), $\beta$-caryophyllene (3.6~6.8%) 등이 확인되었다. 또한 headspace(purge & trap)법에 의해 솔잎의 휘발성 성분 분석시 최적 조건을 검토하기 위해 실온에서 2시간과 20시간동안 추출한 것과 6$0^{\circ}C$에서 2시간과 20시간동안 각각 추출했을 때 실온에서 2시간 추출시 가장 많은 56개의 성분이 확인되었고, 실온에서 2시간 추출시 가장 적은 45개의 성분이 확인되었다. 시험조건에 따른 휘발성분 분석에서 비점이 낮은monoterpenes화합물들은 추출시 초기에 주로 추출되어 지고 alcohols류와 비점이 높은 sesquiterpenes 화합물들은 추출시간을 길게 했을 때 더 많은 성분이 추출된다는 것을 알 수 있었다. 4가지 추출조건을 종합해 볼 때 고비점 휘발성 성분 분석에는 6$0^{\circ}C$에서 20시간 추출이 가장 양호하나, 전반적으로 실온에서 20시간 추출이 가장 적합한 조건인 것으로 나타났다.

Anti-oxidative, Acetylcholinesterase Inhibitory Activities and Acute Toxicity Study of Nepeta sibirica L.

  • Nepeta sibirica L. or Siberian catmint is a medicinal plant species used in Mongolian traditional medicine for curing human different disorders and veterinary practices. The previous study of the whole plant concentrated on the determination of its essential oil composition and reported that the major ones are sesquiterpenes, including nepetalactone. The aim of this study was to reveal a new biological activity of the above-ground parts of N. sibirica L. and compare the activity of different extracts correlating with the content of biologically active compounds and evaluate their toxicity. For this purpose, anti-oxidative and acetylcholinesterase inhibitory activities of the above-ground parts of N. sibirica L. aqueous and ethanol (EtOH) (40%, 70%) extracts were assayed spectrophotometrically. The aqueous extract showed positive anti-oxidative activity by both tested DPPH and FRAP assays with IC50 134.24 ± 1.42 mg/mL and FRAP value 1385.15 ± 8.12 µmol/L at 200 ㎍/mL, in contrast to 40% and 70% EtOH extracts. The 70% EtOH extract presented the highest acetylcholinesterase inhibitory activity (IC50 77.29 ± 0.38 mg/mL) followed by 40% EtOH extract (176.72 ± 0.35 mg/mL) and aqueous extract (275.41 ± 0.23 mg/mL). Total phenolics were found to be gallic acid equivalent, % 3.74 ± 0.05 (70% EtOH), 3.94 ± 0.04 (40% EtOH), and 3.79 ± 0.16 (aqueous), whereas the total flavonoids as a rutin equivalent, % as 2.01 ± 0.12, 1.44 ± 0.17 and 1.99 ± 0.02, each. The aqueous extract showed the best anti-oxidative and lowest activity against the acetylcholinesterase; however, the 70% EtOH extract showed the opposite effects than that of the aqueous. No mortality incidence was visible at various doses, indicating that the oral median lethal dose of aqueous and 70% EtOH extracts were considered greater than 5000 mg/kg. N. sibirica L. belongs to the non-toxic category of the OECD 423 classification.

침향(沈香)의 국내외 연구동향과 본초학에 대한 문헌고찰 (Agarwood's Domestic and International Research Trends and Literature Review of Herbal Medicine)

  • Objectives : To investigate the modern research achievements of agarwood and its association with the efficacy of herbal medicine based on the in vivo and in vitro activities of volatile compounds detected in agarwood. Methods : Databases such as PubMed and ScienceOn were searched for medicinal in vivo and in vitro activity studies on agarwood. They were categorized into "medicine and pharmacy" and "others not related to medicine and pharmacy," and the studies on medicine and pharmacy were organized according to active efficacy. The efficacy and virtue of agarwood as identified in the book (or herbal medicine/herbology) corresponded to modern medical terms and diseases in reference to the . Separately, the Gas Chromatography & Mass spectrometer (GC-MS) analysis results of agarwood from a total of 5 production areas of Aquilaria crassna from Vietnam, Indonesia, Malaysia, Myanmar, and Cambodia as identified in previous studies were consulted to search for research papers on the medicinal activity of the 13 compounds of the aromatic sesquiterpene family detected in agarwood, and they were categorized according to topic. Results : There were 123 studies on the medicinal activity of agarwood. Overall, 46 studies on single extracts of agarwood reported activities such as improving mental health, including anti-anxiety and stability, and antiallergic, antioxidant, antibacterial, and digestive system protective effects. In total, 59 papers on the activities of single compounds isolated from agarwood reported anti-inflammatory and mental health benefits. Separately, 13 compounds detected in agarwood, such as α-agarofuran and β-caryophyllene, were reported by 110 studies to have anticancer, stabilizing, antibacterial, and anti-inflammatory activities. There were also papers on the cultivation methods and resin formation conditions of agarwood trees unrelated to the efficacy of herbal medicine. Conclusion : Among the pharmacological papers, a total of 57 papers corresponded to the effects of agarwood in traditional herbal medicine, including 33 papers related to agarwood extracts and 24 papers analyzing 8 types of sesquiterpenes, such as β-caryophyllene and cedrol, from previous studies. Based on the research findings of each paper, it was possible to correlate the effects of agarwood in traditional herbal medicine with the achievements of modern pharmacological research. In addition, further research is anticipated in new areas related to traditional herbal medicine, including the improvement of mental health such as anti-depression, as well as activities related to anticancer, antioxidant, and hair growth.

오미자(Schisandra chinensis) 열매의 과육과 씨에서 정유 및 리그난 성분 비교 (Comparison of essential oils and lignans compositions from pulp and seed of Omija (Schisandra chinensis) berry)

  • 오미자 (Schisandra chinensis Baillon)는 다양한 생리활성을 지니고 있어 오래전부터 약용 및 식용으로 이용되어왔다. 식품 분야 측면에서 오미자는 가공과정에서 씨(seed)가 부산물로 얻어지고 있으나 이에 대한 활용도는 낮은 편이다. 따라서 본 연구에서는 오미자 씨의 활용성 증대하기 위해 오미자를 부위별(과육과 씨 부위)로 구분하여 일반성분, 정유성분 및 주요 리그난 성분들의 함량을 비교 조사하였다. 일반성분으로서 건조 오미자(whole berry)의 조지방 함량은 27.97%이었고, 과육(21.84%)보다 씨(38.16%)에서 함량이 높았고, 조단백질 함량도 씨 부위(18.09%)가 과육(3.53%)에 비해 약 5.1배 높았다. 건조한 오미자 열매 전체의 정유함량은 1.33%이었고 과육(0.91%)보다 씨 (2.65%)에서 약 2.9배 정도 높았다. 오미자의 정유에서 동정된 62종의 성분들 중에서 부위에 관계없이 가장 많이 검출된 성분은 α-ylangene이었고, 과육에서 α-ylangene의 함량은 195.08 ㎍/g이었으나 씨에서는 6,687.76 ㎍/g이었다. 그 다음으로 cuparene이 과육에서 77.46 ㎍/g, 씨에서 2,098.72 ㎍/g, alloaromadendrene은 과육에서 106.20 ㎍/g, 씨에서 708.96 ㎍/g이 검출되어 씨에서 각각 27.1배 및 6.7배 많이 함유되어 있었다. 특히 여러 가지 약리활성을 지니고 있는 것으로 밝혀진 nootkatone의 경우 과육에서는 60.83 ㎍/g이 검출된 반면 씨에서는 과육보다 약 27배 많은 1,645.66 ㎍/g이 검출되었다. 리그난 성분인 schizandrin, schizandrol B, schizandrin A 및 γ-schizandrin도 과육보다는 씨에 5-18배 많이 함유되어 있어 식품 원료로서 활용잠재력을 가지고 있었다.
