Effects of Seomaeyakssuk (Artemisia argyi H.) Vinegar on Lipid Metabolism in Rats Fed a High-Fat and High-Cholesterol Diet (섬애약쑥 식초가 고지방-고콜레스테롤 급이 흰쥐의 체내 지질대사에 미치는 영향)

  • The effects of orally administered Seomaeyakssuk (Artemisia argyi H.) vinegar on lipid metabolism in Sprague-Dawley rats fed a high-fat and high-cholesterol (HFC) diet were analyzed. The experimental animals were divided into five groups: a normal diet group (normal, N), HFC diet group (control, C), HFC diet with lovastatin at 20 mg/kg body weight (B.W.) group (positive control, PC), HFC diet with malt vinegar group (TM), and HFC diet with Seomaeyaksuk vinegar group (TS) (2 mL/kg B.W.). After 4 weeks of feeding rats the experimental diet, contents of serum total lipids and total cholesterol levels of TM and TS groups were significantly lower than those of the PC group. Triglyceride contents of the TM and TS groups were not significantly different from those of the PC group but significantly lower than those of the C group. Content of serum high density lipoprotein-cholesterol was significantly lower than that of the N group but higher than that of the C group. Low density lipoprotein-cholesterol content of serum was 190.68 mg/dL in the TS group, which was the lowest except for the N group. Aspartate transaminase and albumin transaminase activities as a measurement of liver damage index were not significantly different between the TM, TS, and C groups. Serum thiobarbituric acid reactive substance content of the TS group was reduced to a similar level as the N group but was lower than that of the C group in the liver and significantly higher than that of the N group. Antioxidant activity of the TS group was 55.69% in serum, which was a similar to that of the N group, and was 52.39% in the liver, which was not significantly different than that of the C group. From these results, we conclude that Seomaeyakssuk vinegar improves serum lipid content as a result of the complex action of vinegar, an active ingredient of Seomaeyakssuk and a product of the fermentation process.

Changes of the blood chemistry, lipid and protein components in blood and liver tissue of the rat after oral combined administration of caffeine, iron and vitamin E (Caffeine, 철분 및 vitamin E 혼합투여시 rat의 혈액과 간조직내에서 혈액화학성분과 지질 및 단백질 구성성분의 변화)

  • This study was conducted to identify the effects of caffeine or combinations of caffeine and iron or vitamin E on the lipid and protein components or blood chemistry levels of the serum as well as the total homogenate, mitochondrial and microsomal fraction of the rat(Sprague-Dawley, female) liver. Chronic test were conducted to determine those effects. The chronic test was conducted by dividing rats into 5 groups according to the type of drugs and dosages administrated as follows; the control(group A), and group B was given 25mg/kg caffeine orally once daily for 30 days, group C was given 50mg/kg caffeine orally once daily for 30 days, group D was given 25mg/kg caffeine and orally ferric chloride once daily for 30 days and group E was given 25mg/kg caffeine and 25mg/kg vitamin E once daily for 30 days. The concentrations of glucose, urea nitrogen, uric acid, creatinine, total protein, albumin, A/G ratio, triglyceride, total cholesterol, HDL-cholesterol, free fatty acid, phospholipid as well as the activities of alanine aminotransferase(ALT), aspartate aminotransferase(AST) and alkaline phosphatase(ALP) were measured in the serum of each experimental groups. The concentrations of the carbonyl group and malondiaidehyde(MDA) and the patterns of the SDS-PAGE(Sodium Dodecyl Sulfate - Polyacrylamide Gel Electrophoresis) and fatty acid compositions in free fatty acids and phospholipids were analyzed to determine the oxidative damages and metabolic changes on the lipid and protein components in the serum, and total homogenate, mitochondrial and microsomal fractions of the rat liver. The results obtained from this study were summarized as follows; 1. Body weights of groups B, C, D and E were significantly decreased(p < 0.01) in comparison with that of the control in the chronic test. 2. The concentrations of serum glucose in groups B(124.5mg/dl), C(130.1mg/dl), D(122.1mg/dl), E(119.3mg/dl) were significantly higher(p < 0.01) in comparison to that of the control(101.5mg/dl). But, there were no significant differences in the concentrations of urea nitrogen, uric acid, creatinine, total protein, albumin and A/G ratio in comparison to that of the control. 3. The concentrations of total cholesterol and HDL-cholesterol in serum of groups B(69.6, 53.4mg/dl), C(73.0, 56.3mg/dl), D(68.9, 51.1mg/dl) and E(68.2, 51.3mg/dl) were significantly higher(p < 0.01) in comparison to that of the control(52.6, 38.8mg/dl). On the other hand, the concentrations of triglyceride in serum of groups B(45.0mg/dl), C(40.4mg/dl), D(33.8mg/dl) and E(47.2mg/dl) were significantly lower(p < 0.01) in comparison to that of the control(66.2mg/dl). There were no significant differences in the activities of ALT, AST and ALP in comparison to that of the control. 4. The concentrations of free fatty acid and phospholipid in serum of groups B(45.7, 154.4mg/dl), C(50.0, 167.2mg/dl), D(52.5, 148.4mg/dl) and E(41.1, 159.2mg/dl) were higher(p < 0.01) in comparison to that of the control(35.2, 125.3mg/dl). And the concentrations of the carbonyl group and malondialdehyde in serum of group D(1.82, 0.52nM/mg protein) were significantly higher(p < 0.01) in comparison to the control(1.53nM/mg protein). 5. The concentrations of carbonyl group in total homogenate, mitochondrial and microsomal fraction of group D(1.45, 0.94, 1.67nM/mg protein) were significantly higher (p < 0.01) in comparison to the control(1.16, 0.66, 1.27nM/mg protein). And the concentrations of malondialdehyde in the total homogenate, mitochondrial and microsomal fraction of group D(6.70, 6.10, 1.36nM/mg protein) were significantly higher(p < 0.01) in comparison to the control(5.17, 3.64, 0.68nM/mg protein). 6. As the analytical results of the fatty acid compositions of free fatty acid in serum, the proportions of stearic acid and arachidonic acid of groups B(16.52, 12.62%), C(17.52, 15.18%), D(19.73, 13.47%) and E(17.62, 13.28%) were significantly higher(p < 0.01) in comparison to the control(14.75, 7.88%), but the proportions of oleic acid and linoleic acid of groups B(12.97, 32.59%), C(10.88, 31.23%), D(12.37, 30.66%) and E(11.95, 32.41%) were significantly lower(p < 0.01) in comparison to the control(16.44, 35.12%). Otherwise, as the results of the fatty acid compositions of phospholipid in serum, the proportions of stearic acid and arachidonic acid of groups B(39.37, 16.39%), C(40.63, 17.83%), D(42.73, 15.39%) and E(39.16, 15.70%) were significantly higher(p < 0.01) in comparison to the control(37.74, 14.24%), but the proportions of oleic acid and linoleic acid of groups B(4.03, 14.38%), C(3.54, 12.38%), D(4.52, 11.68%) and E(4.29, 13.64%) were significantly lower(p < 0.01) in comparison to the control(5.53, 16.14%). 7. As the analytical results of the fatty acid compositions of free fatty acid in total homogenate, mitochondrial and microsomal fraction of liver, the proportions of oleic acid of groups B(7.8**, 8.73**, 6.88%) and C(6.89**, 7.75**, 6.58%) were lower(**:p < 0.01) in comparison to the control(8.67, 10.08, 7.81%), but the proportions of arachidonic acid of group C(22.62, 19.79, 23.71%) were significantly higher(p < 0.01) in comparison to the control(20.93, 18.47, 22.24%). And the proportions of palmitic acid of group D(25.95**, 26.16, 26.34**%) were significantly higher(**:p < 0.01) in comparison to the control(24.43, 25.42, 23.34%). In addition, the proportions of linoleic acid of group D(23.43, 25.02, 23.95%) were also significantly higher(p < 0.01) in comparison to the control(22.17, 23.75, 21.26%). The proportions of stearic acid of group D(19.87, 19.76**%) in mitochondrial and microsomal fraction were lower(**:p < 0.01) in comparison to the control(21.01, 24.18%), and the proportions of stearic acid of group E(16.71*, 19.65**%) in mitochondrial and microsomal fraction were significantly lower(**:p < 0.01, *:p < 0.05) in comparison to the control(21.01, 24.18%), and the proportions of linoleic acid of group E(25.04, 29.20, 26.48%) in total homogenate, mitochondria and microsome were significantly higher(p < 0.01) in comparison to the control(22.17, 23.75, 21.26%). 8. As the results of the fatty acid compositions of phospholipid in total homogenate, mitochondrial and microsomal fraction of liver, the proportions of palmitic acid of group D(17.58**, 18.78*, 18.23%**) were significantly higher(**:p < 0.01, *:p < 0.05) in comparison to the control(16.28, 17.22, 16.38%), and the proportions of stearic acid of group D(36.41, 37.23, 39.53%) were also significantly higher(p < 0.01) in comparison to the control(34.18, 34.16, 36.04%). But the proportions of oleic acid(3.41*, 3.11**, 3.12**%) and linoleic acid (18.03**, 15.79**, 14.74**%) of group D were significantly lower(**:p < 0.01, *:p < 0.05) in comparison to the control(oleic : 3.63, 3.72, 3.79%, linoleic : 20.03, 18.71, 18.48%). 9. In order to determine the oxidative damages to the protein in serum, mitochondrial and microsomal fraction of the rat liver, the patterns of the SDS-PAGE were identified, but the results of SDS-PAGE were not significantly different between the control and experimental groups.

Hypoglycemic and Antioxidant Effect of Dietary Hamcho Powder in Streptozotocin-induced Diabetic Rats (Streptozotocin-유발당뇨쥐에서 함초첨가 식이의 항당뇨 및 항산화 효과)

  • Male Sprague-Bawler rats were blocked into four groups which were normal rats fed control diet (NC) diabetic rats fed control diet (DC), normal rats fed Hamcho powder diet (NH), and diabetic rats fed Hamcho powder diet (DH). Diabetes was induced by single injection of streptozotocin (60 mg/kg B.W. i.p.). The animals were fed ad libitum for 5 weeks. Malondialdehyde (MDA), glucose 6-phosphtase (Gspase), glutathione S-transferase (GST) glutathione Peroxidase (GPx), and glutathione reductase (GR) activities were measured in the homogenates of liver and kidney, and total lipid, total cholesterol, triglyceride, and HDL-cholesterol concentrations in the blood serum. Food and water intakes were markedly higher in diabetic groups than those of normal groups and were not significantly decreased by Hamcho powder supplementation, But, FER (Feed efficiency ratio) of DH Brood was higher than that of U group. Total cholesterol level of DH group was decreased in the second and third week, and the weekly change of blood sugar was also decreased in the 5th week. Dietary Hamcho intake showed 41.2% of hypoglycemic effect in diabetics rats. Levels of total lipid and triglycerides of DH group were lower than those of DC group. Hepatic GR activity of DH group was higher than those of other groups. However, renal GR activity was lower than those of other groups. Hepatic G6Pase activity was significantly high in DH group and reduced by Hamcho powder supplementation. GST was reduced by Hancho diet in diabetic rats. In conclusion Hamcho supplementation decreased serum lipid and glucose concentration in STZ-induced diabetic rats and this effects of Hamcho might exert antidiabetic effect of Hamcho powder diet.

Difference in Serum Iron, Cardiac, and Biochemical Indices between Alcoholic and Non-alcoholic Fatty Liver (알코올성 지방간과 비알코올성 지방간 성인에 있어 혈청 철 표지자, 심장 표지자, 생화학적 표지자의 차이)

  • Although alcohol drinking may cause fatty liver to induce hepatocytic injury, other factors lead to it. We designed this study to investigate the differences in serum iron, cardiac, and biochemical indices in men with fatty liver and the difference between alcohol drinkers (Alcohol group) and non-drinkers (Non-alcohol group). The alcohol group had higher body indices than the non-alcohol group. Systolic and diastolic blood pressure (SBP and DBP), and right and left intraocular pressure in the alcohol group were higher than those in the non-alcohol group. Hemoglobin, hematocrit, mean corpuscular hemoglobin, mean corpuscular hemoglobin concentration, and monocyte counts were higher in the alcohol group than in the non-alcohol group. Alanine aminotransferase, ${\gamma}$-glutamyltranspeptidase, total cholesterol, triglyceride, low-density lipoprotein cholesterol, glucose, creatinine, uric acid, iron, total iron binding capacity, and ferritin levels in the alcohol group were greater than those in the non-alcohol group. The present data reveals that alcohol-induced fatty liver has more elevated level of iron indices than in non-alcohol fatty liver as well as biochemical and cardiac indices, indicating that alcohol- induced fatty liver may cause possibility of adult diseases including cardiovascular disease and metabolic syndrome.

The Antihyperlipidemic Effect of Lion's Mane Mushroom (Hericium erinaceus) in Hyperlipidemic Rats Induced by High Fat and Cholesterol Diet (고지방과 콜레스테롤 식이로 유도된 고지혈증 흰쥐에서 노루궁뎅이버섯의 항고지혈증 효과)

  • This study was conducted to investigate the dietary supplementation of fruiting body of Hericium erinaceus (HE) mushroom on lipid profiles of serum and histological changes of the liver in rats with high fat and cholesterol diet. Five-week old female Sprague-Dawley albino rats were divided into three groups of 8 rats each: The normal control diet (NC group), high fat and cholesterol diet (HFC group), and HFC diet supplemented with 5% fruiting powder of Hericium erinaceus (HFC+HE group). In the HFC+HE group, serum total cholesterol, low density lipoprotein, and triglyceride concentrations were significantly reduced compared with the NC group. Body weight gain of those in the HFC+HE group were lower than those in the HFC group; whereas HFC+HE had no effect on the levels of plasma albumin, creatinine, blood urea nitrogen, uric acid, glucose, and total protein. The enzyme activities related to the liver function, such as aspartate aminotransferase (AST), alanine aminotransferase (ALT), and, alkaline phosphatase (ALP), were lower in the NC group than in the HFC group, but without significance. Feeding the mushroom increased the excretion of total lipid and cholesterol. A histopathological analysis showed that the those in the HFC group developed hepatic steatosis, whereas those in the HFC+HE group developed small fat droplet. In conclusion, these results suggest that 5% HE supplementation to HFC diet provided health benefits by acting on lowering atherogenic lipid profile in rats with high fat and cholesterol diet.

Hepatoprotective and Anti-fatigue Effects of Lactic Acid Bacteria (Lactobacillus acidophilus, Bifidobacterium bifidum and Streptococcus thermophilus)

  • This study was carried out to investigate the effect of LAB (Lactic acid bacteria: Lactobacillus acidophilus, Bifidobacterium bifidum and Streptococcus thermophilus) on detoxication of damaged liver in carbon tetrachloride ($CCl_4$) and ethanol (25%)-treated rats. Rats had been daily (twice a day) pre-treated with saline (0.5 ml/kg: untreated group), $CCl_4$ (0.5 ml/kg: other groups) for 6 days. At seventh day, after treating rat with $CCl_4$ and then, mixture of LAB ($10^{11}$/0.5 ml: LAB group), saline (0.5 ml/kg: untreated group, $CCl_4$ group), and biphenyl dimethyl dicarboxylate (DDB) (50 mg/kg: DDB group) were treated orally with $CCl_4$ for 8 days. Ethanol is treated as the same manner instead of $CCl_4$. To investigate the hepatoprotective effect, rats treated with $CCl_4$ and ethanol were analyzed with serum GOT and GPT level. The GOT and GPT levels of LAB group was lower than the level of $CCl_4$ and DDB group. Especially, compared with data of $CCl_4$ group, GPT activity showed statistically significant result in the significance level of p < 0.05. The LAB group treated with ethanol also showed lower level of GOT and GPT than the other control groups treated with ethanol. The triglyceride level of serum decreased more in a group treated special materials (DDB and LAB group) than ethanol group. As well, the effect of LAB on the antifatigue has been investigated. The animals (10/group) were divided into 4 groups (untreated group, Carrier group, Red-ginseng group, LAB group). Each group was given carrier (0.9 mg/0.2 ml), red ginseng extract (200 mg/kg), and mixture of LAB ($10^{11}$/0.2 ml). Special materials were given for three weeks. After finishing treating through oral, horizontal wire test, rotarod test, and forced swimming test were performed. The time of resistance to fatigue of the group, fed with mixture of LAB, was longer than the time when mice treated with red-ginseng that the effect was already revealed. The result of this study revealed that LAB could decrease hepatocelluar injury compared with rats treated orally with $CCl_4$ and ethanol, and could also decrease fatigue.

Nutritional Status and Health Habit of College Students by Body Mass Index (수도권 남녀대학신입생의 체질량지수에 따른 건강행동과 영양상태)

  • It is important for young adults to establish good dietary behavior for healthy life. The prevalence of obesity in college students has increased gradually. The obesity is one of the most prevalent health problems in S. Korea. Moreover, obesity is closely related with the disease such as cardiovascular disease, diabetes mellitus. The purpose of this study was to evaluate health practice, serum components and nutrient intake of the college students according to Body Mass Index (BMI) level. 400 male and female freshmen students in 4 year university located near to Seoul metropolitan area were surveyed respectively through the health check-up procedure for college entrance in February. Survey was carried out for them by questionnaires, including a 24-hour dietary recall and anthropometric measurements. Blood samples were obtained while fasting. Nutrient intake was expressed by DDS(Dietary Diversity Score by 5 food groups) and DVS(Dietary Variety Score). Nutrition level was analyzed by Can-Pro (Korea Nutrition Association). And for the quantity intake, percentage ratio against daily requirement(by KDRIs) and MAR(Mean Adequacy Ratio) were calculated. Underweight group($BMI<18.5kg/m^2$), normal weight group($18.5) and overweight group($BMI<23.0 kg/m^2$) were 18.5%, 61.2% and 19.3% respectively. Nutrient intake levels and food intake status were not significantly different among three groups. The mean nutrient adequacy ratio (MAR) was not significantly different by BMI status. Nutrient intakes of calcium, iron, and riboflavin were especially low for all the students. Overweight group showed high blood pressure(120.9/79.3 mmHg) compared with the underweight group (116.4/ 75.8mmHg) but the difference was not significant. The serum triglyceride level in overweight group (94,0mg%) was higher than that in normal (70.9mg%) or underweight group(70.8mg%)(p<0.05). HDL-cholesterol level in overweight (43.8 mg%) group was not significantly different from that in underweight group (45.9mg%). BMI was positively correlated with blood pressure, hemoglobin, and the intake of total fat and cholesterol. But it was negatively correlated with HDL-cholesterol level. With these results overweight group is closely related with the cardiovascular disease risks, such as high blood pressure and high triglycerides and cholesterol. Thus, a health intervention program including weight control is required for young adults.

Associations between dietary patterns and hypertension among Korean adults: the Korean National Health and Nutrition Examination Survey (2008-2010)

  • The objective of this study is to identify the dietary patterns associated with the risk of hypertensions among Korean adults using data from the Korean National Health and Nutrition Examination Survey (KNHANES, 2008-2010). This study analyzes data from 11,883 subjects who participated in the health and nutrition survey, aging from 20 to 64 years. We performed factor analysis based on the weekly mean intake frequencies of 36 food groups to identify major dietary patterns. We identified three major dietary patterns in both sexes, namely "traditional", "western" and "dairy and carbohydrate" patterns. Participants in the highest quartile of western pattern scores had significantly higher blood pressure, serum total cholesterol, and triglyceride levels than those in the lowest quartile. Although not statistically significant, a trend (P for trend = 0.0732) toward a positive association between the western dietary pattern and hypertension risk was observed after adjustments for age, sex, education, income, body mass index (BMI), smoking, physical activity, and energy intake. The dairy and carbohydrate pattern was inversely related with BMI and blood pressures and positively associated with serum high-density lipoprotein (HDL)-cholesterol. After adjusting the age, sex, education, income, BMI, smoking, physical activity and energy intake, the dairy and carbohydrate pattern showed inverse associations with hypertension prevalence (OR = 0.64, 95% CI = 0.55-0.75; P for trend < 0.0001). Intakes of fiber, sodium, and antioxidant vitamins were significantly higher in the top quartile for the traditional pattern than in the lowest quartile for the traditional pattern (P for trend < 0.0001). Intakes of fiber (P for trend < 0.0001), calcium (P for trend < 0.0001), retinol (P for trend = 0.0164), vitamin $B_1$ (P for trend = 0.001), vitamin $B_2$ (P for trend < 0.0001), niacin (P for trend = 0.0025), and vitamin C (P for trend < 0.0001) were significantly increased across quartiles for the dairy and carbohydrate pattern whereas sodium (P for trend < 0.0001) intake was decreased for this pattern. In conclusion, the dairy and carbohydrate pattern may be associated with a reduced risk of hypertension whereas the western pattern may be associated with an increased risk of hypertension among Korean adults.

  • The experiment was conducted to investigate the effects of dihydropyridine on laying performance and fat metabolism of laying hens. Five hundred and forty laying hens, 40 weeks old, were randomly allotted to three groups, each of which included four replicates of 45 hens. The groups were given a basal corn-soybean meal diet supplemented with 0, 150 mg/kg and 300 mg/kg dihydropyridine. Results showed that compared with the control group (0 mg/kg dihydropyridine), supplements of 150 and 300 mg/kg dihydropyridine increased egg production rate by 9.39% (p<0.01) and 12.97% (p<0.01), increased mean egg weight by 3% (p>0.05) and 4.8% (p>0.05), and improved feed efficiency by 9.54% (p<0.05) and 7.25% (p<0.05), respectively; The addition of 150 and 300 mg/kg dihydropyridine decreased percentage of abdominal fat by 35.4% (p<0.05) and 46.9% (p<0.05), decreased liver fat content by 32.4% (p<0.05) and 10.5% (p<0.05), increased HSL activity of abdominal fat by 39.64% (p<0.05) and 48.48% (p<0.05), increased HSL activity of liver by 9.4% (p>0.05) and 47.34% (p<0.05) and increased the content of cAMP in adenohypophysis by 14.67% (p<0.05) and 10.91% (p<0.05), respectively; The inclusion of 150 mg/kg dihydropyridine increased liver superoxide dismutase activity by 69.61% (p<0.05), and increased hepatic apoB concentration by 53.96% (p<0.05); The supplementation of 150 or 300 mg/kg dihydropyridine decreased malondialdehyde concentration of hepatic mitochondria by 30.90% (p<0.01) and 10.39% (p<0.05), respectively; Supplemented dihydropyridine had no significant effects on TG, Ch HDL-C and VLDL-C concentrations in serum; addition of 150 or 300 mg/kg dihydropyridine increased T3 levels in serum by 15.34% (p<0.05) and 11.88% (p<0.05) and decreased insulin concentration by 40.44% (p<0.05) and 54.37% (p<0.05), respectively. The results demonstrated that adding dihydropyridine had the tendency of improving very low density lipoprotein receptor (VLDLR) content in the ovary. It was concluded that dihydropyridine could improve laying performance and regulate the fat metabolism of laying hens and that 150 mg/kg dihydropyridine is the optimum dose for laying birds in practical conditions.

Effects of Various Sodium Fluoride (NaF) Administration on Blood Glucose, Lipid Composition and A/G Ratio in Rats (NaF 함량을 달리한 투여가 흰쥐의 지질 및 혈당농도와 A/G 비에 미치는 영향)

  • The aim of the study was to investigate the influences of sodium fluoride on the body weight, lipid metabolism functional improvement, blood glucose level, protein component in the sera of rats which fed experimental diets for 5 weeks. Body weight gain of rats was higher in the NF25 group, NF30 group and NF50 group showed a significant difference from the basal diet group (BD group). The changes of the liver and the heart of each argan weights were observed compared to the BD group tends to appear lower in the oral NaF groups (p<0.05). Organ weight of each group of the others showed no significant difference (p<0.05). Serum concentrations of total cholesterol, low-density lipoprotein (LDL)-cholesterol, free cholesterol, triglyceride (TG), phospholipid (PL) and blood glucose were lower in BD group than basal diet plus sodium fluoride groups. Conversely depending on the concentration of sodium fluoride for the high-density lipoprotein (HDL)-cholesterol level, it showed lower results. Serum total protein was observed low showed a significant difference in the NF50 group, there was no significant difference in the other groups (p<0.05). globulin and albumin concentration levels also showed the lowest results in the NF50 group.
