  • Four out of 15 dogs were successfully induced diabetes mellitus with intravenous iqiection of 30 mg of streptozotgin and 50 mg of alloxan monohydrate per kilogram body weight and maintained more than 9 weeks without iqiection of insulin or oral hypoglycemic sgent Histopathologicallyi these four dogs have typical diabetic lesions such as degeneration and vacuolation of pancreatic islet cells, and fatty change of liver at necropsy in the end of study. Serum glucose level increased dramatically at 24 hours post-injection but serum fructosamine level increased gradually and reached plateau at 31-41 days post-injection of streptozotocin and alloxan. Serum fructosamine concert%lion correlated very well with serum glucose concentration of preceding 4-7 weeks in experimentally induced diabetic dogs. Our data suggest that serum fructosamine reflects mean glucose concentration of preceding 4-7 weeks in experimentally induced diabetic dogs.

The Effects of Dangguibohyultang and Ssangbohwan the Muscle Recovery in Rats (당귀보혈탕(當歸補血湯) 및 쌍보환(雙補丸)이 백서(白鼠)의 근육피로(筋肉疲勞) 회부(恢復)에 미치는 영향(影響))

  • An attempt was made to study the effects of dangguibohyultang and Ssangbohwan on the muscle recovery from exhaustion after severe exercise. Before rats were given severe swimming exercise, Dangguibohyultang and Ssangbohwan were admisterd. Then, the blood lactate level, the serum activities of LDH, CPK and the serum levels of FFA and glucose were measured. The results were as follows; 1. The level of the blood lactate were significantly decreased in the group of Dangguibohyultang. 2. The activities of serum LDH were significantly decreased in the groups of Dangguibohyultang and Ssangbohwan. 3. The activities of serum CPK were significantly decreased in the groups of Dangguibohyultang and Ssangbohwan. 4. The levels of serum FFA were significantly decreased in the groups of Dangguibohyultang. 5. The levels of serum glucose were significantly increased in the groups of Dangguibohyultang and Ssangbohwan. According to the above results, it is concluded that Dangguibohyultang was more effective on the muscle recovery than Ssangbohwan in rats.

  • Carcinoembryonic Antigen is one of most frequently checked tumor markers in cancer management. We performed statistical analysis with serum CEA data of 2626 persons who received regular health examination and were thought to be free of active disease to determine the cutoff values of serum CEA level in normal Korean adults and to study the factors influencing serum CEA levels in normal subjects. 1) The cutoff values of serum CEA in normal Korean adults in general were 9.28ng/ml for men, 5.90 ng/ml for women. 2) Serum CEA level was influenced by age, present smoking history, sex, and abnormal findings in chest X ray. 3) Serum CEA level had no correlation with the history of amount of alcohol consumption or obesity. 4) Cutoff values of serum CEA in normal Korean adults were tabulated according to age, sex, and smoking history. Serum CEA level was influenced by age, sex, present smoking history and abnormal findings in chest X ray and cutoff values of serum CEA were tabulated according to age, sex, and smoking history.

Serum and CSF Mineral Profile of Himalayan Yak (Bas grunniens) in their Natural Habitat

  • Mineral profile of serum and cerebrospinal fluid (CSF) of 15 Himalayan Yak (adult female, n=8; adult male n=4 and young male,n=3) was studied in their natural habitat at an altitude of 3300 meters above mean sea level at Sangla in north western Himalayas. The macro and micro minerals estimated in serum and CSF were; Sodium, Potassium, Calcium and Magnesium and Zinc, Copper and Iron respectively. The values recorded among different Yak groups did not significantly differ from each other except serum iron and haemoglobin which were significantly higher (p<0.05 ) in young male Yaks compared to the adult male and female Yaks. An observation of great significance was considerably higher potassium and lower sodium level in Yaks compared to other bovine species of plains. The serum potassium values in some adult female Yaks were recorded as high as 10.4 mEq/l and the values varied between 6.6 to 9.8 mEq/l in young male Yaks. The serum Sodium values and Na:K ratios in Yak serum ranged between 117.5 to 122.6 mEq/l and 13.7 to 15.3 respectively. The possible relationship of high serum Potassium value with hypoxic conditions and hostile mountain environment has been discussed.

Clinical Study for Low Dose & Short-Term Therapy of Biphenyl Dimethyl Dicarboxylate(DDB) in the Chronic Hepatitis. Patients with Elevated Serum Aspartate Aminotransferase and Alanine Aminotransferase Levels (Biphenyl Dimethyl Dicarboxylate의 저용량 단기 투여가 만성 간염환자의 상승된 Aspartate Aminotransferase와 Alanine Aminotransferase의 저하 효과에 관한 임상적 연구)

  • Biphenyl Dimethyl dicarboxylate(DDB) has been regarded as a safe, effective drug for decreasing serum aminotransferase levels from elevated serum aminotransferase levels, which cause acute or chronic hepatitis and chronic liver diseases. This study was designed to low dose(22.5mg/day) & short-term therapy effectiveness for 4 weeks of DDB in 30 chronic hepatitis patients with elevated serum aminotransferases. The following results were observed. 1. Serum alanine aminotransferase(ALT) levels significantly decresed from 173. $97\pm130.62(U/L)$ of pretreatment level to $32.23\pm19.22(U/L)$ after treatment for 4 weeks(p<0.00l) and normalized patients by $73\%$ 2. Serum aspartate (AST) aminotransferase levels significantly decreased from $94.90\pm49.17(U/L)$ of pretreatment level to $45.30\pm23.25(U/L)4 after treatment(p0<0.01). 3. However, no significant effects in the serum AST & ALT changes by which cause hepatitis and hepatitis duration (p>0.05). 4. No significant adverse effects were observed except for mild epigastric discomfort in one patient during DDB treatment It is suggested that DDB small dosage administration can result effectively decreasing serum aminotransferase levels from chronic hepatitis patients with elevated serum aminotransferase levels.

Regular Exercise-training Affects Serum Lipid and Carnitine Profiles in Some College Students

  • This study investigated the effect of regular exercise on serum lipid profiles and carnitine levels in college students. Daily nutrient intake, anthropometry , serum lipid, and carnitine profiles in serum and urine were evaluated prior to beginning the study and after 35 days of treadmill running for 30 minutes per day. The results obtained were summarized as follows : 1) Concentrations of total lipid and Triglyceride in serum were decreased by regular exercise in female subjects but unaffected in males. 2) Serum LDL-cholesterol was significantly decreased, but total cholesterol and HDL-cholesterol in serum were not affected in both male and female subjects. 3) nonesterified carnitinem, acid-insoluble acylcarnitine, and total carnitine levels in serum were not affected, but acid-soluble acylcarnitine level was increased by regular exercise in both subjects. 4) Urinary excretionof the acid-soluble acylcarnitine level was increased by regular exercise -training. These results suggest that regular exercise -training has different effects on serum lipid oxidation via carnitine metabolism in this condition.

Dietary Zinc Analysis and Changes of Zinc Nutriture with Zinc Supplementation in Korean Adults (한국성인의 아연 섭취실태 및 아연보충에 의한 아연 영양상태 변화)

  • The purpose of this was to analyse zinc intakes and effect of Zn(30mgZnSO4/day) supplementation on plasma zinc level, serum HDL-cholesterol and serum Alkaline Phosphatase (AP) activity of Korean adults. The men consumed 8.52($\pm$2.08)mg of zinc, and the women consumed 6.4($\pm$2.62)mg of zinc. Although protein intakes of subjects were lower than normal values. The first source of zinc was cereal and grain group, the second was meat, fish, egg and soybean group. Two food groups supplied about 80% of zinc. After two weeks of zinc supplementation, the zinc concentration in plasma was significantly increased. The highest plasma zinc level was 78.80ug/dl(men), 76.04ug/dl(women) at 2 weeks after zinc supplementation(p<0.05). Serum DHL-cholesterol was significantly decreased by zinc supplementation. The lowest serum HDL-cholesterol level was 39.29mg/dl(men), 44.84mg/dl(women) at 4 weeks after zinc supplementation(p<0.01). Serum AP activity was significantly increased by zinc supplementation. The highest AP activity was 86.40units/L(man), 67.93units/L(women) at 2 weeks after zinc supplementation(p<0.05). It seems that the supplementation of 30mg ZnSO4/day can be beneficial for improving zinc nutriture. However it can be negative factor on coronary heart disease because serum HDL-cholesterol was significantly decreased(p<0.01)

  • Mammalian cell cultures have been used extensively to produce proteins for therapeutic agent because of their ability to perform post-translational modification including glycosylation. To produce recombinant protein, many factors and parameter are considered such as media composition, host cell type, and culture process. In this study, recombinant human erythropoietin (rhEPO) producing cell line was established by using glutamine synthetase system. To reduce serum concentration in media, we compared direct adaptation with step adaptation. Cell growth was faster in step adaptation. In low-level serum media, there were insufficient glucose for cell growth. Thus, we added glucose in low-level serum media from 2 g/L to 4.5 g/L. Titer of rhEPO was higher than other conditions at 4.5 g/L of glucose. Additionally, N-methyl-D-aspartate (NMDA), 13-cis-retinal, and pluronic F-68 (PF-68) were added to enhance productivity in CHO cell cultures. In conclusion, we applied CHO cell producing rhEPO to low-level of serum in media using step-adaptation. Also, we confirmed positive effect of NMDA, 13-cis-retinal, and PF-68.

EFFECTS OF SUPPLEMENTAL CHROMIUM PICOLINATE ON GROWTH PERFORMANCE, CARCASS COMPOSITION AND SERUM TRAITS OF BROILERS FED DIETS VARYING IN PROTEIN AND LYSINE

  • Arbor Acres broiler chickens (N=288) with an average initial weight of 59.4 g were fed diets varying in protein and lysine (80, 100, 120% of NRC; 100, 120% of NRC, 1984) in order to investigate the effects of supplemental chromium picolinate on growth performance, nutrient utilizability, carcass composition, serum traits and in vitro protein synthesis. Six replicates of eight chicks were grouped into one treatment Six chicks were sacrificed from each treatment for carcass analysis, and six additional chicks were chosen and dissected for in vitro culture of liver tissue. Body weight gain, feed intake, feed conversion, mortality, carcass composition and serum glucose, HDL/cholesterol ratio, serum triglyceride and serum nonesterified fatty acid appeared to be affected by either the level of dietary crude protein or lysine when supplemented with 200 ppb chromium picolinate (p < 0.05). Retained and secreted proteins in liver acinar cell cultured in vitro were not affected by dietary lysine level but affected by dietary protein level when added with 200 ppb chromium picolinate.

Influence of fat levels and types of dietary fiber on lipid metabolism of rats (식이중 지방 수준과 Fiber 종류가 흰쥐의 지방대사에 미치는 영향)

  • The influence of fat level and dietary fiber on serum and liver lipid concentration of the rat was investigated. Groups of rats were fed respectively 3%, 5%, 20% fat containing and free fiber diet until 5th week. From 5th week to 9th week high fat diet groups of rates were devided high fat diet group, high fat pectin supplemented diet group and high fat celulose supplemented diet group. Pectin and cellulose at 10% was supplemented to high fat diets. After rats were fed for 9 weeks, all rats were sacrificed to collect the liver and blood samples by heart puncture. Serum and liver lipids were determined on all rats and compared the histochemical lipid staining method with the biochemical analysis of serum and liver lipids. A high fat diet caused increased level of the total lipids, total cholesterol and free cholesterol contents in the liver and the serum. Rats fed pectin reduced body weight, werum lipid and liver lipid but cellulose had no effect to reduce serum and liver lipid concentration. These results indicate that dietary fat level led to changes in the lipid metabolism of rats and that pectin was more effective in lowering serum and liver lipid than cellulose.
