청폐사간탕(淸肺瀉肝湯)이 Streptozotocin으로 유발(誘發)된 흰쥐의 실험적(實驗的) 당뇨(糖尿)에 미치는 영향(影響) (Effects of Chungpyesagantang on the Diabetic Rats induced by Streptozotocin)

  • In order to study the effects of Chungpyesagantang(淸肺瀉肝湯) on the diabetic rats induced by streptozotocin, during 15 days rats were administered Chungpyesagantang extract after streptozotocin injection (50mg/kg) (sample group). On 2nd, 9th, and 15th day, I investigated the levels of body weight, serum glucose, serum total cholestrol, serum triglyceride. The last day, I killed rats and investigated hepatic lipid peroxidase. The results were obtained as follows: 1. Body weight of the diabetic rats induced by streptozotocin and Chungpyesagantang, as compared with the control group on the 15th day, increased effectively. (p〈0.05 respectively) 2. Glucose levels in serum of the diabetic rats induced by streptozotoc in and Chungpyesagantang, as compared with the control group on the 9th and 15th day, decreased effectively.(p〈0.05 respectively) 3. Total cholesterol levels in serum of the diabetic rats induced by streptozotocin and Chungpyesagantang, as compared with the control group on the 9th and 15th day, decreased effectively.(p〈0.05 respectively ) 4. Triglyceride levels in serum of the diabetic rats induced by streptozotocin and Chungpyesagantang, as compared with the control group on the 9th and 15th day, decreased effectively.(p〈0.05 respectively) 5. On hepatic lipid peroxidethe of the diabetic rats induced by streptozotocin and Chungpyesagantang, as compared with the control group on the 9th and 15th day, decreased effectively.(p〈0.01 respectively)

택사(澤瀉) 시간별 투여(投與)가 Streptozotocin으로 유발된 고혈당 백서(白鼠)의 실험적(實驗的) 당뇨(糖尿)에 미치는 영향(影響) (Effects of Different Lengths of Treatment with Rhizoma Alismatis on Diabetic Mellitus of Streptozotocin-Induced Hyperglycemic Rats)

  • Objectives : The objective of the study was to observe the effects of different treatment lengths of Rhizoma Alismatis on diabetes mellitus. Methods : Rats were divided into three groups: normal(non-treated group), control (group administered saline for 4, 11, and 18 days), and RA (group administered 2.45mg/200g Rhizoma Alismatis for 4, 11, and 18 days). The experimental results were derived from the measurement of the levels of glucose, ALP, AST, ALT, creatinine and BUN in the serum from the rats on days 4, 11, and 18. Results : The glucose level in the serum significantly decreased on the 18th day. The ALP level in the serum did not show statistical significance. The GOT and GPT levels in the serum slightly decreased on the 4th, 11th, and 18th days, but did not show statistical significance. The creatinine level in the serum was unchanged for all of them. The BUN level in the serum did not show statistical significance. Conclusions : According to the above results, it is concluded that Rhizoma Alismatis has a therapeutic effect on diabetes mellitus with a longer period ofintake. As to influence upon the liver and kidney, there was no damage orside effects.

db/db 마우스에서 수풍순기환의 혈당강하 활성 및 기전연구 (Blood Glucose Lowering Activity and Mechanism of Supungsungihyan (SPSGH) in db/db Mouse)

  • Antidiabetic activity and mechanism of Supungsungihyan(SPSGH) were examined in db/db mice, which is a spontaneously hyperglycemic, hyperinsulinemic and obese animal model. SPSGH and acarbose were administered orally for 4 weeks. Fasting and non-fasting serum glucose, glycated hemoglobin and trig-lyceride of SPSGH treated group were all reduced when compared with those of db/db control group. At 12th week after birth, SPSGH increased an insulin secretion although statistic significance was not seen. Total activities of sucrose, maltase and lactase in SPSGH treated group were not significantly different from those in db/db control. On the other hand, sucrase and maltase activities in acarbose treated groups were increased. Effect of SPSGH on mRNA expression of glucose transporter(GLUT-4) was also examined by RT-PCR and in vitro transcription with co-amplification of rat $\beta$-actin gene as an internal standard. Muscular GLUT-4 mRNA expression in SPSGH treated group was increased significantly. These results may suggest that SPSGH lowered blood glucose ascribing to upregulation of muscular GLUT-4 mRNA expression.

불포화도가 다른 유지가 단식과 정상식이 흰쥐의 체중 및 간대사에 미치는 영향 (Effect of Various Fat and Oil Types on Weight Gain and Liver Metabolism in Fasting or Normal Diet Rats)

  • 단식 또는 기초사료를 투여한 성장기 rat 에 우지 또는 들깨유를 매일 일정량 투여하여 2일 간격으로 체중변화와 장기중량, 혈액의 생화학적검사 간조직검사를하여 다음의 결론을 얻었다. 완전 단식rat군(F)과 단식중 우지 (FT)와 들깨유(FP)를 투여한 rat군은 1일 평균 -6.2~-4.0%인 체중감소를 보이고 기초식이군 (B) 또는 기초식이에 우지 (BT)나 들깨유(BP)를 투여한 rat군은 1일 2.3~4.0%의 체중증가를 보였다. F군은 4일만에 모두 아사하였고, FT와 FP군은 6~10일 사이에 모두 아사하였다. 단식에 의한 영향으로 아사직전 이들 rat는 간중량(3.6~4.0g) serum cholesterol(70~73.2mg/㎗), serum triglyceride (6.5~7..5mg/㎗) blood glucose (76-91mg/㎗)의 현저한 감소와 GPT (117.5-186.5u), blood urea nitrogen (38~65mg/㎗)을 대폭 증가하는 특징을 가졌다. 유지시료 불포화도에 따른 영향에서 FP군은 FT군에 비해 약 2일 정도 생존일수의 감축과 serum cholesterol, serum triglyceride, blood urea nitrogen 감소를 보이나 GPT, blood glucose를 약간 증가하였다. BP군은 BT군에 비해 serum cholesterol, triglyceride를 상당히 감소하고 blood urea nitrogen의 근소한 감소와 GPT는 대폭 증가하였다. Blood glucose는 거의 차이가 없었다. 간조직 검사 결과 F군은 아사직전 간실질내 염증 세포 침윤을 보이고 FT, FP군은 유지 시료의 불포화도 차이에 관계없이 아사직전 일부 rat가 염증세포 침윤, 간실질의 염증, 간세포의 손상된 모양이 관찰되었다. 따라서 어떤 목적으로 단식 또는 유지만을 공급하는 무단백질 제한식이를 할 때 극심한 체중감소를 보이고 혈중 cholesterol과 trig1yceride는 크게 낮아지며 간을 포함한 모든 조직의 붕괴의 조짐으로 blood urea nitrogen이 크게 증가되고 그에 따른 간기능저하로 GPT도 크게 상승하고 rat의 경우 4~6일만에 사망하였다. 단식중이거나 혹은 정상식이내 고도 불포화 유지 (들깨유)의 섭취는 포화지방 (우지)에비해 혈중 cholesterol 과 triglyceride를 감소시키는 효과는 있지만 GPT가 유의적으로 증가됨을 볼때 장기간 고도 불포화 유지를 다량 섭취함은 간기능을 저하 시킬 수 있음이 나타났다.

Effect of Feeding Ammoniated Wheat Straw Treated with Hydrochloric Acid on Blood Biochemical Profile in Growing Male Buffalo (Bubalus bubalis) Calves

  • An experiment was conducted to study the effect of feeding ammoniated wheat straw treated with HCl on blood biochemical profiles in growing male buffalo (Bubalus bubalis) calves. Twenty-four growing male buffalo calves (one year of age, 88.54${\pm}$3.81 kg average body weight) were divided into three groups in a completely randomized design on the basis of their body weight. Animals in all the three groups were fed on concentrate mixture. In addition, they were offered wheat straw, ammoniated wheat straw (4% urea at 50% moisture level) and HCI treated ammoniated wheat straw (4% urea at 50% moisture level and HCl added to trap 30% of the NH3 evolved) in groups I, II and III, respectively for a period of 180 days, as per Kearl (1982) for body weight gain of 500 g/d. In all diets, concentrate:roughage ratio was fixed at 50:50 and were made isonitrogenous by adjusting CP levels of conc. mixtures. Blood was collected from jugular vein of each buffalo calf at the beginning and subsequently at two months interval of experimental feeding. Due to urea-ammoniation, the CP content of wheat straw increased from 2.90 to 6.96% and addition of HCl along with urea further increased the CP content to 10.09%. In all the three groups, the mean values of plasma glucose (mg %) and serum globulin (g %), showed a decreasing trend, while the mean value of serum TP (g %), serum A:G ratio, serum urea (mg %), serum creatinine (mg %), serum ALP (KA units), SGOT (units/ml.), SGPT (units/ml), serum T$_3$ and T$_4$ (ng/ml) showed an increasing trend with the advancement of feeding period. The cumulative period mean values of serum TP (6.15 to 6.20 g %), serum albumin (3.07 to 3.18, g %), serum globulin (2.98 to 3.09, g %), serum A:G ratio (1.03 to 1.10), serum ALP (23.15 to 23.63, KA units), serum T$_3$ (1.20 to 1.23 ng/ml) and serum T$_4$ (21.33 to 21.88 ng/ml) were comparable among the groups. The cumulative period mean plasma glucose (mg %) in group III (57.28) was similar to groups I (55.31) and II (59.41), however, the cumulative period mean plasma glucose in group II was significantly (p<0.01) higher than group I. The cumulative period mean serum urea (mg %) in group III (47.34) was significantly (p<0.001) higher than group I (38.38) and II (42.24), which were statistically alike. However, the cumulative period mean serum creatinine values (mg %) in groups II (1.43) and III (1.52) were similar and were significantly (p<0.01) higher than group I (1.24). The cumulative period mean SGOT (units/ml) in groups I, II and III was 91.71, 96.04 and 96.64, respectively. Similarly the cumulative period mean SGPT (units/ml) was 19.00, 19.93 and 20.01 in groups I, II and III, respectively. The cumulative period mean values of SGOT (p<0.05) and SGPT (p<0.001) in groups II and III were similar and were significantly higher than group I. The cumulative period mean serum T$_3$ and T$_4$ values in groups I (1.21 and 21.81), II (1.23 and 21.42) and III (1.20 and 21.33) were comparable. From the present study it may be concluded that feeding of AWS treated with and without HCI to growing male buffalo calves for 180 days had no significant adverse effect on blood biochemical profile.

비만 소아의 혈중 아디포넥틴과 인슐린저항성에 관한 연구 (Serum Adiponectin Levels and Insulin Resistance in Obese Children)

  • 목 적 : 식생활과 생활양식의 서구화로 인하여 우리나라도 비만의 유병률이 급격히 증가하는 추세이며 특히 소아비만은 이미 소아기에 합병증 발생의 위험과 더불어 성인비만으로 이행될 가능성이 크기에 이에 대한 관심이 날로 높아지고 있는 추세이다. 아디포넥틴은 최근 발견된 지방세포에서 분비되는 adipocytokine으로 비만에서 아디포넥틴의 감소가 인슐린저항성과 동맥경화증의 발병과 관련이 있는 것으로 알려져 있다. 본 연구는 비만아와 정상 체중아의 혈중 아디포넥틴 농도의 차이와 체질량지 수(BMI) 및 인슐린저항성과의 상관관계를 알아보고자 하였다. 방 법 : 2003년 1월부터 2003년 6월까지 BMI가 95 백분위 이상인 8-15세 사이의 비만아 82명(남아 54명, 여아 28명)과 대조군으로 정상 체중아 31명(남아 21, 여아10), 총 113명을 대상으로 신체계측을 하고 공복 후 채혈하여 혈중 아디포넥틴, 인슐린, 혈당 농도를 측정하였다. 혈중 아디포넥틴 농도를 비만여부와 사춘기 시기별로 비교하였으며 아디포넥틴과 체질량지수, 인슐린, HOMA 등과의 상관관계를 알아보았다. 결 과 : 1) 비만아의 평균 혈중 아디포넥틴 농도는 19.7 mg/mL, 정상 체중아는 27.6 mg/mL로 비만군이 유의하게 낮았으며(P<0.01), 전체 대상아 중 남아와 여아의 성별에 따른 차이는 없었다. 2) 사춘기 전보다 사춘기 시기의 소아에서 아디포넥틴의 농도가 의미있게 낮았다(P<0.01). 3) 비만아 중 27명을 대상으로 시행한 경구 당부하 검사에서 공복시에 비해 식후 2시간 후 인슐린 농도는 유의하게 높았으며($17.0{\mu}IU/mL$ vs $35.8{\mu}IU/mL$, P<0.01), 아디포넥틴 농도는 반대로 유의하게 낮았다(25.8 mg/mL vs 19.8 mg/mL, P<0.01). 4) 혈중 아디포넥틴과 BMI(r=-0.39, P<0.01), 인슐린(r=-0.28, P<0.01)은 각각 음의 상관관계를 보였으며, 혈중 지질치중 저밀도 지질단백(LDL) 콜레스테롤(r=-0.20, P<0.05)과도 음의 상관관계를 보였으나 총 콜레스테롤과 고밀도 지질단백(HDL) 콜레스테롤과, 중성지방과는 유의한 상관관계가 없었다. 결 론 : 혈중 아디포넥틴 농도는 비만아에서 정상 체중아보다 유의하게 낮았으며, 아디포넥틴은 BMI 및 혈중 인슐린과 음의 상관관계가 있었다. 혈중 아디포넥틴이 소아 비만아에서 인슐린 저항성의 예측인자로 유용하게 사용될 수 있을 것으로 생각된다.

The associations between serum leptin, adiponectin and intercellular adhesion molecule-1 in hypercholesterolemic patients

  • We examined the associations between adiponectin or leptin and serum ICAM-1 levels in seventy-six hypercholesterolemic patients (mean age 59 yrs, 25 males and 51 females, LDL-cholesterol>=130mg/dL at screening). Blood lipid profiles and HOMA-IR derived from fasting glucose and insulin concentrations were determined. Serum levels of adiponectin, leptin and ICAM-1 were analyzed using ELISA The results showed that serum levels of leptin were positively associated with serum levels of ICAM-1 independent of age, sex and BMI (r=0.392, p<0.001). Serum levels of adiponectin were negatively associated with serum levels of ICAM-1 independent of age, sex and BMI (r=-0.343, p<0.005). Stepwise multiple linear regression analysis showed that serum leptin was an independent factor to be associated with serum ICAM-1 levels after adjusting for age, sex, BMI, alcohol intake, smoking status, blood lipids such as total cholesterol, triglyceride, HDL cholesterol and LDL cholesterol and HOMA-IR (p<0.001). With respect to adiponectin, its association with serum ICAM-1 was attenuated but still significant when further adjustments were made for age, sex, BMI, alcohol intake, smoking status, blood lipids such as total cholesterol, triglyceride, HDL cholesterol and LDL cholesterol and HOMA-IR (p<0.005). In conclusion, this study suggests that adiponectin and leptin are associated with endothelial derived inflammation.

비만형 동물모델에서 복합 한약 추출물 CAPA의 비만 및 당대사 개선 효능 (The Mixed Herbal Extract, CAPA, Prevents Obesity and Glucose Intolerance in Obese Mice)

  • Objectives: This study investigated the effects of the mixed herbal extract from Cinnamomum cassia Blume, Atractylodes macrocephala Koidzumi, Pueraria lobata Benth, and Aconitum carmichaeli Debx (CAPA) on obesity and glucose tolerance in obese mice. Methods: Animals were divided in 6 groups, normal diet, high fat diet (HFD), HFD with CAPA 100 mg/kg (CAPA 100), CAPA 300 mg/kg (CAPA 300), and metformin 200 mg/kg or lorcaserin 10 mg/kg as positive controls, and treated for 16 weeks. Body weight, food intake, fasting blood glucose, and body temperature were checked every week and then organs, blood serums were collected after treatment. The oral glucose tolerance test was also carried out after treatment. Results: Compared to HFD, CAPA extract treated mice showed significant decreases in body weight, adipose tissue weight, lipid accumulations in liver and serum lipid levels without a reduction of food intake. And fasting glucose and glucose tolerance were all improved in the CAPA treated mice. Conclusions: Our results suggest that CAPA extract can prevent diet induced obesity and glucose intolerance without a reduction of energy intake in obese mice.

Streptozotocin 에 의해 유발된 당뇨병 쥐의 혈당과 당뇨병성 신증에 한약복합처방이 미치는 효과 (Effects of Herb-Combined Remedy(HCR) on Blood Glucose Levels and Diabetic Nephropathy in STZ-induced Diabetic Rats)

  • Diabetes mellitus is a worldwide epidemic. Global projections suggest that most nations will have a doubling of the incidence of Diabetes mellitus(DM) within 20years. The Herb-Combined Remedy(HCR) of DM was known as one of anti-hyperglycaemic agents. But it is unclear whether HCR can be used to treat patients with Diabetic nephropathy or not. The present study was designed to investigate the anti-hyperglycaemic and preventive effects from diabetic nephropathy of HCR in STZ-induced diabetic rats. Treatment with HCR regulated blood glucose levels, but did not affect body weights. In addition, HCR was able to decrease blood glucose levels after 120min. Serum BUN levels were not effective but, serum Creatinine levels were lowered by treatment with HCR or MF. In histopathological observation of renal tissue, both HCR and MF groups showed decreased tendency of inflammation and renal injury. In conclusion, these results demonstrate that HCR is effective to treat patients with DM and also has preventive activity on diabetic nephropathy.

Adiponectin Level in Non-Pregnant Women, Pregnant Women without Diabetes and Pregnant Women with Diabetes

  • Gestational diabetes mellitus (GDM) is a common complication during pregnancy and one of the main causes of adverse fetal-maternal outcomes. However, the pathogenesis of GDM has not been clearly stated. Adiponectin, an adipose tissue-derived plasma protein, is involved in regulation of insulin resistance and glucose hemostasis, and thus is a key modulator of insulin action and glucose metabolism. In this study, we investigated to compare serum adiponectin levels in pregnant women with diabetes, pregnant women who are without diabetes, and non-pregnant women, and to evaluate relationship between serum adiponectin. levels and metabolic parameters. Forty-one pregnant women with diabetes, fifty-nine pregnant women without diabetes and forty non-pregnancy women were recruited. Adiponectin levels were significantly lower in pregnant women with diabetes when compared to non-pregnant women and pregnant women without diabetes. Pregnant women without diabetes at second trimester had lower adiponectin levels compared to non-pregnant women. Adiponectin was negatively correlated with BMI, fasting insulin, HOMA-IR, total cholesterol, and triglyceride. In conclusion, this study confirmed that the decreased level of adiponectin precedes the onset of abnormal glucose level during pregnancy and also normal pregnant women had lower adiponectin levels compared to non-pregnant women. This knowledge may help to identify strategies for lowering the occurrence of GDM in women who are at high risk of developing the disorder.
