The protective effect of AI-1367 (Alnus japonica extract) on liver injury was investigated. Primary rat hepatocyte intoxication was induced by tert-butyl hydroperoxide (tBH), carbon tetrachloride ($CCl_4$), or D-glactosamine (D-GalN). Liver injury was induced by $CCl_4$, D-GalN or MCD (methionine choline deficient)-diet in mouse. The cellular leakage of lactate dehyrogenase and cell viability followed by the treatment of hepatotoxicants were significantly improved by AI-1367 treatment at a concentration range of 5~50 ${\mu}g/ml$ for tBH, 5~50 ${\mu}g/ml$ for D-GalN, and 5~100 ${\mu}g/ml$ for $CCl_4$, respectively. Treatment with AI-1367 (20, 10, 5 mg/kg, p.o.) on liver injury induced by subcutaneous injection of $CCl_4$ or D-GalN reduced significantly the levels of aspartate transaminase and alanine transaminase in serum. Histological observations revealed that fatty acid changes, hepatocyte necrosis and inflammatory cell infiltration in $CCl_4$ (D-GalN)-induced liver injury was improved by administration of AI-1367. AI-1367 treatment (10, 5, 2.5 mg/kg, p.o.) also significantly recovered the body weight change and serum levels of aspartate transaminase, alanine transaminase and triglyceride in liver injury induced by MCD diet. From these results, AI-1367 shows protective effects against tBH, $CCl_4$, D-GalN, or MCD diet-induced hepatotoxicity in vitro or in vivo.

In this study, the effects of Gamgung-tang gamibang on the hyperthyroidism induced by the intraperitoneal injection of 3,5,3-triiodothyronine was examined by the measurement of physical changes, body weight, the volume of food intake and rectal temperature, and heart weight, heart beat, blood pressure with contrast to propranolol, one of beta-blocking agents. the obtained results were as follows. The Gamgung-tang gamibang extract showed to inhibit the decrease of body weight and rectal temperature, and decrease the food intake, so the inhibitory effects of Gamgung-tang gamibang extract on the experimental hyperthyroidism were exhibited. The Gamgung-tang gamibang extract showed the inhibitory effects on the circulatory functions changed and enhanced by the experimental induced hyperthyroidism, the action of Gamgung-tang gamibang extract was less effective than the propranolol of D-CONT group. The Gamgung-tang gamibang extract showed significant effects to inhibit the concentration of serum thyroid houmone, more effective than the propranolol, beta-blocking agents. The Gamgung-tang gamibang extract showed the effective inhibitory reaction on the biochemical changes in serum, cholesterol, ketone bodies, free fatty acid, glucose in hyperthyroid rats induced by 3,5,3-triiodothyronine.

In this study, we investigated the dose-dependent effects of mixtures of Atractylodes macrocephala (AM) and Amomum villosum (AV) water extracts in a ratio of 3:1 on high fat diet (HFD)-induced obesity model. Oral administration of various concentrations with mixtures of AM and AV extracts in a ratio of 3:1 for 6 weeks inhibited HFD-induced increases of body, liver and epididymal fat weights in a dose-dependent fashions. Those effects may be mediated by decreased expressions of lipogenesis-related genes such as acetyl coA carboxylase (ACC) and fatty acid synthase (FAS) in liver. Also, increase of insulin and decrease of adiponectin in serum by HFD supply were inhibited by three different dosages of mixtures of AM and AV extracts in a ratio of 3:1. HFD supply induced increases of serum total cholesterol, triglyceride and LDL cholesterol. However, hyperlipidemia was significantly decreased in dose-dependent manners by treatment with mixtures of AM and AV extracts. Based on the results of the present study, hypolipidemic and anti-obesity effects by mixtures of AM and AV extracts were found in HFD-induced obesity model. Further clinical investigation is needed to develop anti-obesity therapeutic or preventive agents by using mixtures of AM and AV extracts.
We studied the effects of 2 different dosages of high-molecular-weight poly-${\gamma}$-glutamic acid (hm ${\gamma}$-PGA) derived from Bacillus subtilis chungkookjang on lipid metabolism in a high-fructose diet-induced hypertriglyceridemic animal model. For 4 weeks, rats were fed either AIN-93 diet (normal control, NC; n = 10) or modified AIN-93 diet in which cornstarch was substituted with 63% fructose (n = 30) to induce hypertriglyceridemia. After 4 weeks, the hypertriglyceridemic rats were treated with daily oral doses of 0 mg (hypertriglyceridemic control, HC), 2.5 mg (hypertriglyceridemic, low hm ${\gamma}$-PGA, HL), or 5 $mg{\cdot}kg{\cdot}bw^{-1}{\cdot}d^{-1}$ (hypertriglyceridemic, high hm ${\gamma}$-PGA, HH) hm ${\gamma}$-PGA for 4 weeks. The HL and HH groups exhibited significantly lower levels of serum triglyceride, total cholesterol, LDL cholesterol, and free fatty acids than the HC group. The administration of hm ${\gamma}$-PGA reduced serum ALT and AST levels. The activities of lipogenic enzymes such as hepatic malic enzyme and glucose-6-phosphate dehydrogenase as well as glucose-6-phosphate dehydrogenase mRNA expression were significantly decreased by hm ${\gamma}$-PGA administration (p < 0.05). These results indicate that hm ${\gamma}$-PGA has an anti-hypertriglyceridemic effect in high-fructose diet-induced hypertriglyceridemic rats.

X-linked adrenoleukodystrophy (ALD) is a uncommon metabolic disorder which derived by peroxismal ${\beta}$-oxidation and elevation of serum very long chain fatty acid (VLCFA). VLCFA is mainly accumulated in the myelin of the central nervous system and adrenal cortex, by which the expressed symptoms of this disease are mainly neurologic and endocrinologic (such as adrenal insufficiency). The mutations in the ABCD1 gene causes X-linked ALD, nevertheless its phenotypes and genotypes are poorly coordinated. We report the case of a 12-year-old boy with X-linked ALD who developed vomiting, fatigue and poor oral intake. Severe dehydration and hyponatremia were found in initial physical examination and laboratory test, but his motor/sensory nerve function and mental status were completely normal. We diagnosed ALD with diffuse high-intensity signal in both parietotemporal cerebellar white matter in brain MRI and elevated serum VLCFA. Later, we confirmed a novel c.1635-1G>A (IVS6-1G>A) mutations of the ABCD1 gene. With the discrepancy between its phenotypes and genotypes, various phenotypes could be seen in X-ALD patient. Careful examination and further studies for these patients will be needed.

Etiological studies of diabetes and its complications showed that oxidative stress might play a major role. Therefore, many efforts have been tried to regulate free oxygen radicals for treating diabetes and its complications. Cheonghyul-san has been known to be effective for the antidiabetic, antihyperlipidemic and antiobesitic prescription, and composed of four crude herbs. In male ob/ob mouse with severe obesity, hyperinsulinemia, hypergiycemia, hyperlipidemia, the acting mechanisms of Cheonghyul-san were examined. Mice were grouped and treated for 5 weeks as follows. Both the lean (C57/BL6J black mice) and diabetic (ob/ob mice) control groups received standard chow. The experimental groups were fed with a diet of chow supplemented with 7.5, 15 and 30 mg Cheonghyul-san per 1 kg of body weight for 14 days. The effects of Cheonghyul-san extract on the ob/ob mice were observed by measuring the serum levels of glucose, insulin, lipid components, and the kidney levels of reactive oxygen species (ROS), MDA+HAE, GSH and also the enzyme activities involved in polyol pathway. Cheonghyul-san lowered the levels of serum glucose and insulin in a dose dependent manner. Total cholesterol, triglyceride and free fatty acid levels were decreased, while the HDL-cholesterol level was increased, in Cheonghyul-san treated groups. Renal aldose reductase and sorbitol dehydrogenase activities were increased in the ob/ob mice, whereas those were inhibited in the Cheonghyul-san-administered groups. Cheonghyul-san inhibited the generation of ROS in the kidney. Finally, MDA+HAE level was increased and the GSH level was decreased in the ob/ob mice, whereas those were improved in the Cheonghyul-san-administered groups. The results suggested that Cheonghyul-san exerted the antidiabetic and antihyperlipidemic activities by regulating the activities of polyol pathway enzymes, scavenging ROS, regulating the MDA+HAE and GSH levels in the ob/ob mice.
Objective : This experimental study was designed to investigate the effects of Mahwangbokhapbang2 (every abbreviation from now on MHBHB2) on obesity care where it is prescribed for high fat diet-induced mice. Also designed to find out effects of MHBHB2 on controlling th obesity clinically) Method : In order to investigate th obese inhibitory effects of MHBHB2, C57BL/6 mice were induced by high fat diet C57BL/6 mice were divided into four group(normal, high fat diet, high fat diet with reductil, high fat diet with MHBHB2 extract) and fed for 15weeks. Result : 1. Mahwangbokhapbang2(MHBHB2) decreased significantly the body weig-ht. 2. MHBHB2 decreased significantly the weight of adipocyte. 3. MHBHB2 decreased significantly the blood level of serum ALT, AST. 4. MHBHB2 decreased significantly the blood level of serum total cholester-l, LDL- cholesterol, friglyceride and NEFA(free fatty acid). 5. MHBHB2 decreased significantly the blood level of HDL-cholesterol. 6. MHBHB2 500mg/kg decreased significantly the blood level of Leptin. Conclusion : Based on these results, it is prove that MHBHB2 is effective on obesity care and has obese-inhibitory effects in obese-mouse induced by high fat diet. So it is espected that the clinical application of MHBHB2 can help the treatment of obesity.
The effect of buckwheat dietary fiber (BDF) as hypolipidemic and antioxidant agent were determined in C57BL/6 mice fed a high fat diet (HFD) with different doses of 500 (low, BDF-L) or 1,000 (high, BDF-H) mg/kg of body weight, compared with the HFD-diet control group (HFD). The negative control group (ND) was fed the basal diet. Body weights in the BDF-L and BDF-H groups were significantly decreased as compared to those in the HFD group (p<0.05). BDF also improved the lipid profile in a dose-dependent manner; serum lipid profiles and levels of insulin, glucose, and free fatty acid were significantly decreased in the BDF-L and BDF-H groups, whereas HDL-C and adiponectin significantly increased as compared to the HFD group (p<0.05). Meanwhile, BDF lowered serum malondialdehyde (MDA) in comparison with the HFD group (p<0.05). The results demonstrate that the intake of BDF might prevent obesity and its related metabolic disorders by inducing dyslipidemia and oxidative stress.
We determined the risk factors for displacement of the abomasum (DA), and the relationships between DA and postpartum disorders, milk yield, and reproductive performance in dairy cows. Initially, we identified the risk factors for DA using data regarding cow health and calving season from 2,208 lactations. Then, we compared the incidence of postpartum disorders, culling, death, and reproductive performance between cows with DA and their control herdmates (each n = 57). In addition, serum metabolites concentrations and milk yield were compared between cows with DA and controls (each n = 33). Ketosis (odds ratio [OR] = 9.27, p < 0.0001) and twin calves (p = 0.06) increased the risk of DA. Cows with a parity of three had a higher risk (OR = 5.23, p < 0.01) of DA than primiparous cows. Serum total cholesterol concentration was lower but non-esterified fatty acid, ${\beta}-hydroxybutyrate$, and alanine aminotransferase concentrations were higher after calving in cows with DA than in controls (p < 0.05). The removal rate from the herd by 2 months after calving was higher (p < 0.05) but milk yield 1 and 2 months after calving (p < 0.01) and the rate of first insemination by 150 days postpartum were lower (hazard ratio = 0.49, p < 0.05) in cows with DA than controls. In conclusion, higher parity, twin calves, and ketosis are risk factors for DA in dairy cows, which is associated with a higher removal rate from the herd, lower milk yield, a longer calving to first insemination interval, and unfavorable levels of metabolites related to energy and liver function.
Purpose: Obesity is a serious public health issue for the modern society and is considered a chronic health hazard. There are many surgical and pharmacological approaches to treat obesity. However, various potentially hazardous side effects remain the biggest challenge. Therefore, diets based on foods derived from natural products have gained increasing attention compared to anti-obesity drugs. Recently, research on edible insects as a food source has been a topic of considerable interest in the scientific communities. This study examined the anti-obesity effects of ingesting an edible insect by feeding a high-fat diet (HFD)-induced obese mouse models with a diet containing Tenebrio molitor larvae powder (TMLP). Methods: Six-week-old female C57BL/6J mice were divided into 4 groups according to treatment: 100% normal diet (ND), 100% HFD (HFD), HFD 99% + TMLP 1% (TMLP), and HFD 97% + TMLP 3% (TMLP 3%). TMLP was added to the HFD for 6 weeks for the latter two groups. Results: Compared to the HFD group, mice in the TMLP group showed weight loss, and micro-computed tomographic imaging revealed that the volume of the adipose tissue in the abdominal area also showed significant reduction. After an autopsy, the fat weight was found to be significantly reduced in the TMLP group compared to the HFD group. In addition, the degree of fat cell deposition in the liver tissue and the size of the adipocytes significantly decreased in the TMLP group. Reverse transcription polymerase chain reaction analysis for the mRNA expression of adipogenesis-related genes namely CCAAT-enhancer-binding proteins (C/EBP-β, C/EBP-δ), and fatty acid-binding protein 4 (FABP4) showed that the expression levels of these genes were significantly reduced in the TMLP group compared to the HFD group. Serum leptin level also decreased significantly in the TMLP group in the comparison with the HFD group. In addition, total cholesterol, triglyceride, and glucose levels in mouse serum also decreased in the TMLP group. Conclusion: Taken together, our results showed that TMLP effectively inhibited adipocyte growth and reduced body weight in obese mice.
