Effects of Green Tea Catechin on Liver 3-Hydroxy-3-Methylglutaryl Co Enzyme A Reductase Activity and Serum Lipid Levels in Streptozotocin-induced Diabetic Rats (Streptozotocin 유발 당뇨쥐 간조직에서의 3-Hydroxy-3-Methylglutaryl Co Enzyme A Reductase 활성과 혈중지질수준에 미치는 녹차 Catechin의 영향)

  • The purpose of this study was to investigate the effects of green tea catechin on lipid metabolism in streptozotocin(STZ)-induced diabetic rats. Male Sprague-Dawley rats weighing 150$\pm$10gm were randomly assigned to one normal and three STZ-induced diabetic groups. Diabetic animals were fed catechin free diet(DM-0C group), 0.5% catechin diet(DM-0.5C group) and 1% catechin diet(DM-1C group). Diabetes was experimentally induced by intravenous injection of 55mg/kg body wt of STZ in citrate buffer(pH 4.3) after feeding of three experimental diets for 4 weeks. Animals were sacrificed at the 6th day of diabetic states. Levels of blood glucose were three fold higher in all three STZ-induced diabetic groups than that of the normal group. The levels of plasma insulin were markedly lower in three STZ-induced diabetic groups than that of the normal group. The levels of plasma cortisol were increased in DM-0C group compared with that of the normal group. Triglyceride, total-cholesterol and LDL-cholesterol levels in serum were increased in DM-0C groups compared with the normal group but were not significantly different between catechin diet groups and normal group. Serum HDL-cholesterol levels were reduced in DM-0C and DM-0.5C groups by 38% and 25%, respectively and had similar tendency in the DM-1C group compared with that of control group. Atherogenic index have shown same pattern as the result of total cholesterol. Activities of 3-hydroxy-3-methylglutaryl Co enzyme A(HMG-CoA) reductase were higher in DM-0C groups than those of the normal group but were not significantly different between catechin diet groups and the normal group. It is concluded that dietary catechins can modulate lipid levels of serum and liver HMG-CoA reductase activity in diabetic rats.

  • Objective : We analyzed the clinical and endocrinological results of the transsphenoidal microsurgery for ACTH secreting pituitary adenomas. Marerials and Methods : From October 1995 to August 2000, 18 patients underwent transsphenoidal microsurgery for Cushing's disease. We analyzed the surgical results of 17 patients, one patient who was previously operated from other hospital was excluded. Age of the patients were 18 to 61 years old(mean 37.7), male to female ratio was 1 : 3.3, and follow-up period was 3 to 50 months(mean 20.3). The selection of candidates for transsphenoidal exploration was based on endocrinologic criteria. Magnetic resonance imaging was the preferred radiologic test. Selective inferior petrosal sinus sampling of adrenocorticotropic hormone futher refined the diagnosis when endocrinologic and radiologic procedures were not definitive. Results : Results of the preoperative endocrinological test were : level of serum ACTH 29.4 to $225{\mu}g/dL$(mean $93.88{\mu}g/dL$) ; serum cortisol 11.9 to $47.5{\mu}g/dL$(mean $27.49{\mu}g/dL$) ; 24-hour urine free cortisol 235 to $1019{\mu}g/day$(mean $571.0{\mu}g/day$). Inferior petrosal sinus sampling for ACTH was performed in 11 patients and all were confirmed by Cushing's disease and we could predict the laterality of the tumor in 9 of 11 patients. We performed transsphenoidal selective adenomectomy in 5 patients, adenomectomy and subtotal hypophysectomy in 2 patients, adenomectomy and partial hypophysectomy in 9 patients, and in the remaining one patient, hemihypophysectomy followed by total hypophysectomy due to remission failure. Fifteen of 17 patients(88.2%) showed endocrinological remission. Glucocorticoid replacement therapy was performed in all the patients who showed remission for 1 to 24 months(mean 5.9 months), and 6 patients received steroid over 6 months. Conclusion : We conclude that the direct demonstration of a tumor in the pituitary gland by MRI is the most important and definitive diagnostic tool and the location of a mass should be confirmed with increased level of ACTH by the inferior petrosal sinus sampling. Transsphenoidal microsurgery is effective treatment modality for Cushing's disease and the immediate postoperative evaluation of the surgical resection of the tumor is very important. The patients should show hypocortisolism, decreased, subnormal serum ACTH and cortisol levels and 24-hours urine free cortisol. We performed 18 transsphenoidal microsurgery for Cushing's disease in 17 patients and 15 patients(88.2%) showed endocrinological remission.

  • The present study was carried out to study the serum concentration of peptide and steroid hormones in puerperal sow. Eight crossbred sows were used for collection of blood samples from day 20 prepartum to day 20 postpartum. FSH, LH, prolactin, estradiol-$17{\beta}$, progesterone and cortisol were assayed by radioimmunoassay methods. The mean serum FSH did not vary during the puerperal period and ranged from $8.1{\pm}1.8mIU/ml$ to $9.0{\pm}2.3mIU/ml$. LH concentrations increased from $2.6{\pm}0.3mIU/ml$ at day 20 prepartum to $3.9{\pm}1.1mIU/ml$ at the time of parturition, reached $3.2{\pm}0.9mIU/ml$ by day+2 and remained quite constant therafter. Prolactin reached a peak mean level of $68.5{\pm}9.5ng/ml$ at day 0. Estradiol-$17{\beta}$ increased from $205.0{\pm}29.5pg/ml$ at day 6 prepartum to $425.0{\pm}35.0pg/ml$ at the time of parturition. Progesterone remained fairly constant ($18.4{\pm}1.6$ to $20.2{\pm}2.1ng/ml$) from 20 to 6 days before parturition, began to decline on day-2, reached $0.9{\pm}0.3ng/ml$ by day+2 and remained quite constant thereafter. Cortisol reached a peak level of $86.5{\pm}10.5ng/ml$ at the day 0.

Effect of Horticultural Therapy on the Stress and Serum Cortisol of Demented Elders (원예치료가 치매노인의 스트레스 및 혈중 코티졸 호르몬에 미치는 영향)

  • This study was conducted in order to examine the effects of horticultural therapy on stress in demented elders, and for this purpose, we analyzed stress-related physiological changes in demented elders under the care of an elderly facility who had not taken medication and consented to blood tests. The horticultural therapy program included only flower decoration, which was preferred by the participants, aiming ultimately to reduce stress. First, a one-to-one session was given for forming rapport and the horticultural therapist had many conversations about plants with the elders. Furthermore, the participants were induced to be absorbed into the horticultural activities, though a short while, so that they might forget troubles. Evaluation was made by measuring subjective stress and the blood cortisol hormone concentration before and after the horticultural therapy and examining differences between them. Subjective stress increased from 12.88 to 17.88 in the control group, but decreased significantly from 13.88 before the therapy to 6.38 after in the experimental group (p = 0.007). When the blood cortisol hormone level was measured as a physiological indicator of stress, it decreased significantly from $11.33{\mu}g{\cdot}L^{-1}$ before the horticultural therapy to $9.85{\mu}g{\cdot}L^{-1}$ after (p = 0.037). These results suggest that horticultural therapy reduces the blood cortisol hormone level and is effective in easing stress.

  • Background: Acute and chronic airway inflammation are important in the pathogenesis of bronchial asthma. Corticosteroids have proved to be very effective in the management of asthma. Although the mechanism by which they produce this effect is still debated, suppression of the inflammatory response is thought to be the most likely. Although inhaled steroids are known to be safe and have less side effects than oral steroids, the extent which inhaled steroids have beneficial and the detrimental effects in the treatment of asthma has remained open to question. Budesonide is a recently developed corticosteroid for inhalation treatment with a strong local effect combined with rapid inactivation in the systemic circulation. We set out to look in more detail at the time course of change in bronchial reactivity, clinical symptoms and the effects on the adrenal function during 6 weeks of treatment with budesonide (800 ug per day). Methods: Clinical symptoms, pulmonary function test, histamine $PC_{20}$, serum ACTH and cortisol (8 AM and 4 PM) were measured in 23 allergic asthmatic patients before and after 6 weeks of treatment with budesonide. Results: 1) Pulmonary function test; PEFR, FEV1 and FVC after 6 weeks of treatment with budesonide were higher than those before treatment. 2) Clinical symptoms; Clinical symptoms were significantly improved after 3 weeks and 6 weeks of treatment with budesonide. 3) Histamine provocation; Histamine $PC_{20}$ after 6 weeks of treatment with budesonide was significantly higher than that before treatment. 4) Adrenal function; 6 weeks of budesonide therapy did not significantly affect the level of serum ACTH and cortisol. Conclusion: From these results, it is concluded that budesonide therapy improved the clinical symptoms, pulmonary function and bronchial hyperreactivity after 3 weeks of treatment and the improvement after 6 weeks of treatment was higher than that after 3 weeks of treatment. During 6 weeks of treatment with budesonide, the inhibitory effect on the adrenal function was not obvious.

Change of Concentration of Hormones and Metabolic Materials in Serum by Age in Hanwoo (한우 혈청에서 호르몬 및 대사물질 농도들의 연령에 따른 변화에 관한 연구)

  • This study was carried out to investigate the change of blood compositions by age in Hanwoo, and a total of 866 of Hanwoo, which consisted with 638 of steer and 228 of bulls, were used to measure serum concentrations. A multiple regression equation was estimated with collection age and blood composition as independent and dependent variables, respectively. Complicated regression equations for blood compositions in steer and bulls were IGF-I(cubic), calcium (linear), and IP(linear). Linear and cubic equations were fitted to testosterone in steer and creatinine in bulls, respectively. A cubic equation in steer and linear equation in bulls were fitted to HDLC. Equations of quadratic in steer and cubic in bulls were fitted to concentration of triglyceride, globulin, and A/G ratio. BUN was fitted by equations of cubic in steer and quadratic in bulls. TP and albumin were fitted by equations of quadratic in steer and linear in bulls. A cubic regression equation did not explain the change of cortisol by age in steer and bulls. A cubic regression equation did explain the change of glucose by age in steer, but not in bulls. Higher R-square values (R-SQUARE>0.1) were estimated to IGF-1, albumin, creatinine, Inorganic phosphorous(IP) and HDLC in steer, and testosterone, IGF-I, TP, albumin, glucose, creatinine, IP, and HDLC in bulls for the fitted regression equations of blood compositions. Therefore, IGF-I, albumin, creatinine, IP, and HDLC were regarded as comparatively large variation by age in steer and bulls.

Physiological Response of the Cultured Olive Flounder (Paralichthys olivaceus) to the Sharp changes of Water Temperature (급격한 수온변화에 따른 양식 넙치(Paralichthys olivaceus)의 생리적 반응)

  • The effects of sparp changes of water temperature (WT) on the stree response and physiological change of the cultured olive flounder (Paralichthys olivaceus) were examined by manipulating WT (3 patterns) in a running seawater culture system. In the first group (Exp. I), the WT was decreased from 18$^{\circ}C$ to 11$^{\circ}C$ within 6 hours and increased back to the original WT quickly. WT was decreased from 2$0^{\circ}C$ to 11$^{\circ}C$ within 5 hours and main-tained at 11$^{\circ}C$ for 10 hours. and then increased to 2$0^{\circ}C$ in the second group (Exp. II). In the third group(Exp. III) WT was decreased to 11$^{\circ}C$ within 5 hours (type A) or 10 hrs. (type B). In Exp. I and III, the level of serum cortisol was increased from 2.5$\pm$0.3 ng/ml and 2.6$\pm$0.9 ng/ml to 13.6$\pm$3.0 ng/ml and 12.4$\pm$3.2ng/ml, respectively, with WT decrease. However, no consistent tendency in the change of serum glucose level was shown according to WT decrease. In Exp. III, the glucose level of fish in type A was decreased until 5 hours and increased at 7 hours, then decreased until 12 hours where as the glucose level in type B was decreased until 5 hours and stayed at the level of 15.7 mg/dl. The serum osmolality was reduced with WT decrease and the response of serum electrolytes in this experiment conflicted, and a tendency in total protein, AST and ALT was not found following WT decrease. In conclusion, olive flounder responded to the stress caused by WT decrease and acclimated to this condition when the lower temperature was maintained. But there was no stress response in the blood of olive flounder when WT was increased. On the other hand, the degree of stress response in olive flounder was various according to the range and gradient of WT change.

Effect of Chromium Picolinate on Growth Performance, Carcass Characteristics, Serum Metabolites and Metabolism of Lipid in Pigs

  • The study was conducted to evaluate the effects of chromium picolinate (CrP) on growth, carcass characteristics and serum metabolites in growing-finishing pigs. A total of 96 Landrace$\times$Yorkshire$\times$Duroc hybrid pigs, initial live weight about $38.12{\pm}00kg$, were randomly assigned to 2 groups (16 pigs per pen, 3 pens per group), each group had 48 pigs with an equal number of barrows and gilts. The pigs were fed the diet with or without $200{\mu}g/kg$ Cr from CrP. The results indicated that the addition of $200{\mu}g/kg$ CrP increased ADG by 3.58% and decreased feed conversion rate (FCR) by 3.00% compared to the control group. Pigs fed CrP had 7.58% (p<0.05) higher carcass lean percentage, 15.55% (p<0.05) larger longissimus muscle area (LMA) and 10.90% (p<0.05) lower back fat thickness, 15.17% (p<0.05) lower carcass fat percentage. In addition, the IGF-I level in serum was elevated by 79.20% (p<0.05), the Insulin and cortisol level decreased by 27.35% (p<0.05) and 34.58% (p<0.05) respectively with supplementation of CrP. Analysis of subcutaneous fat (10th rib) showed that the activity of hormone sensitive lipase (HSL) increased by 79.58% (p<0.05) and the activities of isocitrate dehydrogenase (ISD) and malate dehydrogenase (MDH) decreased significantly by 15.06% (p<0.05) and 54.53% (p<0.05) respectively in the $200{\mu}g/kg$ CrP group. The concentration of RNA, RNA/DNA in LMA increased by 31.89% (p<0.05) and 5.41% (p<0.05) respectively with the addition of CrP. These results suggest that CrP reduced fat deposits by decreasing lipogenic enzyme activities and increasing HSL activity and may have promoted muscle anabolic metabolism through elevated IGF-I levels.

Metabolic Signatures of Adrenal Steroids in Preeclamptic Serum and Placenta Using Weighting Factor-Dependent Acquisitions

  • Although translational research is referred to clinical chemistry measures, correct weighting factors for linear and quadratic calibration curves with least-squares regression algorithm have not been carefully considered in bioanalytical assays yet. The objective of this study was to identify steroidogenic roles in preeclampsia and verify accuracy of quantitative results by comparing two different linear regression models with weighting factor of 1 and 1/x2. A liquid chromatography-mass spectrometry (LC-MS)-based adrenal steroid assay was conducted to reveal metabolic signatures of preeclampsia in both serum and placenta samples obtained 15 preeclamptic patients and 17 age-matched control pregnant women (33.9 ± 4.2 vs. 32.8 ± 5.6 yr, respectively) at 34~36 gestational weeks. Percent biases in the unweighted model (wi = 1) were inversely proportional to concentrations (-739.4 ~ 852.9%) while those of weighted regression (wi = 1/x2) were < 18% for all variables. The optimized LC-MS combined with the weighted linear regression resulted in significantly increased maternal serum levels of pregnenolone, 21-deoxycortisol, and tetrahydrocortisone (P < 0.05 for all) in preeclampsia. Serum metabolic ratio of (tetrahydrocortisol + allo-tetrahydrocortisol) / tetrahydrocortisone indicating 11β-hydroxysteroid dehydrogenase type 2 was decreased (P < 0.005) in patients. In placenta, local concentrations of androstenedione were changed while its metabolic ratio to 17α-hydroxyprogesterone responsible for 17,20-lyase activity was significantly decreased in patients (P = 0.002). The current bioanalytical LC-MS assay with corrected weighting factor of 1/x2 may provide reliable and accurate quantitative outcomes, suggesting altered steroidogenesis in preeclampsia patients at late gestational weeks in the third trimester.

Effects of Insulin on Acetyl-CoA Carboxylase in Bovine Mammary Secretory Cells

  • Objectives of this study were to determine effects of insulin on acetyl-CoA carboxylase (ACC) activity and correlate this activity with relative amounts of ACC in MAC-T cells. MAC-T cells were grown in Medium 199 supplemented with fetal bovine serum (5%), cortisol ($1{\mu}g/ml$), and insulin ($1{\mu}g/ml$). At confuluence, the cells were transferred to $100mm^2$ culture dishes coated with the extracelluar matrix. After 10 h of incubation, the media were replaced with media without fetal bovine serum and the concentration of insulin was lowered to 5 ng/ml. After 24 h, the media were changed to contain the varying concentrations of insulin and incubations continued for 48 h. The addition of insulin resulted in increases in the specific activity of ACC. The maximal effects of insulin on the ACC activity occurred at concentrations of insulin, 1,000 ng/ml. In contrast, the relative change in lactate dehydrogenase (LDH) activity in response to increasing insulin concentration was minimal as compared to the effects of insulin on ACC. Transblot and enhanced chemiluminescence (ECL) analysis indicated that the increase in ACC activity in MAC-T cells caused by insulin were due to actual increases in amounts of enzyme.
