시험스트레스와 혈청 Cortisol 및 Prolactin치 간의 관계 (The Relationship of Examination Stress with Serum Cortisol and Prolactin Levels)

  • 본 연구의 목적은 시험과 같은 심리적 스트레스와 혈청 cortisol 및 prolactin과의 관계를 알아보고, 주관적인 스트레스지각 및 정신병리가 혈청 cortisol 및 prolactin치 간의 상관성을 알아보는데 있다. 본 연구에는 의과대학 남학생 20명이 참여하였다. 이들을 대상으로 시험기간과 수업기간 중에 각각 global assessment of recent stress(GARS)척도와 SCL-90R척도를 사용하여 스트레스지각 및 정신병리를 평가하였다. 혈청 cortisol 및 prolactin치는 방사성 면역측정법에 의해 측정되었다. 혈청 cortisol치는 수업기간보다 시험기간 중에 유의하게 더 높았으나 혈청 prolactin치는 두 군 간에 유의한 차이를 보이지 않았다. 한편 시험기간 및 수업기간 중 스트레스지각점수는 혈청 cortisol치 및 prolactin치와 각각 유의한 상관성을 보이지 않았다. 그러나 정신병리에 있어서는 시험기간 중 대인예민척도점수가 혈청 cortisol치와 유의한 음 상관성을 보였다. 이상의 결과는 시험과 같은 심리적 스트레스가 prolactin보다 cortisol과 더 밀접하게 관련될 가능성이 높음을 시사한다. 그리고 대인예민과 같은 특정 정신병리는 낮은 혈청 cortisol치와 관련될 수 있음을 시사한다.

Tryptophan 및 Tyrosine 보강 식이가 Stress로 인한 혈청 Cortisol, Glucose 및 Free Fatty Acid의 농도 변화에 미치는 영향 (The Effects of Tryptophan and Tyrosine-Enriched Diet on the Serum Cortisol, Glucose and Free Fatty Acid Levels of Stressed Rats)

  • Sprague-Dawley 숫쥐에게 3.6%의 tryptophan 또는 tyrosine 보강 식이를 4일간 먹여 스트레스로 인한 혈청 cortisol, glucose, free gatty acid(ffa)수준 변화를 조사하였다. Trp-보강 식이를 먹은 쥐들은 혈청 및 뇌의 trp농도가 유의적으로(p<0.05) 증가하였고 cortisol과 ffa 수준은 정상 식이군과 같았으나 glucose는 유의적으로 낮았다. Trp- 보강 식이군이 stress를 받았을 때 혈청 cortisol 수준은 유의적으로 증가하였으나 정상식이군의 stress 반응 정도에는 미치치 못하는 유의적으로 낮은 증가 추세를 보였다. 또한 trp-보강 식이군의 혈청 glucose와 ffa 수준은 정상식이군과는 달리 스트레스로 인한 증가 현상이 나타나지 않았다. 이로서 Trp-보강 식이는 스트레스로 인한 대사적 교란(metabolic disturbance)를 방지하고, 뇌하수체-시상하부-부신 피질계를 통한 호르몬 반응의 강도를 줄이는 효과를 나타내었다. 한편 try-보강 식이를 먹은 쥐들은 혈청 try함량이 증가하였고 유의적이지는 않으나 전반적으로 낮은 cortisol 수준을 보였다. 이들의 giucose와 ffa농도는 정상 식이군과 비교해 차이가 없었다. Try-보강 식이군이 스트레스를 받으면 혈청 cortisol 수준에는 유의적인 변화가 없는 반면 glucose와 ffa함량은 정상 식이군과 동일하게 증가하였다. 즉 try보강 식이는 스트레스로 인한 대사적 교란을 방지할 수는 없었으나 시상하부-뇌하수체-부신 피질게를 통해 이루어지는 hormone 반응을 말소시킬 수 있는 것으로 보여진다.

위탁견에서 스트레스에 의한 혈중 Cortisol의 농도 변화 (Changes in Serum Cortisol Concentration Due to Boarding Stress in Dogs)

  • Among the dogs boarding at an animal hospital, 10 dogs each were selected as a free boarding group and a kennel boarding group. Each group was further divided into 5 puppies and 5 mature dogs to examine the changes in their serum cortisol concentrations during boarding (5 days). We collected blood at day 8 and then analyzed the blood corpuscles, ran a biochemical serum test, and an additional urine test before boarding to check their health status. After collecting the dog's blood at 9:00 am, 3:00 pm, and 9:00 pm, we analyzed the cortisol concentration by the radioimmunoassay (RIA) method. The daily serum cortisol concentration at 9:00 am, 3:00 pm, and 9:00 pm, had a circadian rhythm during both the free and kennel boarding periods in both puppies and adults. Furthermore, the average daily serum cortisol concentration was significantly increased during the 1st, 2nd, and 3rd day after free boarding in the puppy group (P < 0.01). In adults from the free boarding group, the serum cortisol concentration dramatically increased on the 1st day (P < 0.01), as well as on the 2nd and 3rd day (P < 0.05). Interestingly, the average daily serum cortisol concentration was significantly increased on the 1st day after kennel boarding as well as during the entire period of kennel boarding in the puppy group (P < 0.05). In the adult kennel boarding group, serum cortisol concentration was significantly increased during the whole kennel boarding period (P < 0.01). An interesting result was that circadian rhythmicity in the sum of the daily serum cortisol concentrations was present in the free boarding group, but not in the kennel boarding group in both puppies and adults. In summary, cortisol was released depending on the degree of stress in free and kennel boarded dogs. Taken together, these results suggest that cortisol, a stress hormone, should be maintained at physiological concentrations in a circadian rhythm when the animals are hospitalized.

ELISA에 의한 국내 재래종 산양의 정상상태에서의 혈중 cortisol 농도 (Serum cortisol concentrations on the normal condition in Korean native goats by ELISA)

  • This study was performed to examine the charges of serum cortisol concentrations on the normal conditions in Korean native goats by enzyme-linked immunosorbent assay(ELISA). The Korean native goats were 4 to 30 months old and weighted 6.0 to 28kg. The goats were allowed ad libitum access to water and food. To minimize the stress with handing and blood sampling, animals were adapted for 2 weeks before the experiment. After adaptation, intravenouse catheter was inserted left intact a jugular vein of 6 goats and blood samples were done 2~3 days later. Experimental animals were divided into 2 groups which was non-catheterized and catheterized group in order to examine the effect for changes of serum concentration and circadian rhythm of cortisol. The results were obtained as follows ; The sensitivity of serum cortisol concentration was $20pg/m{\ell}$. The intra-assay and inter-assay coefficient of variation were below 2.5% and 5%, respectively. Serum concentration of cortisol was more higher in noo-catheterized group$(32.84{\pm}16.78ng/m{\ell})$ than catheterized group$(23.20{\pm}10.29ng/m{\ell})$. The difference according to months old in serum concentration of cortisol was the more higber on 4~6 than 7~12 and lowest over 12 months old. A circadian rhythm in the serum concentration of cortisol in Korean native goats was not found significantly with 2 hours sampling intervals.

The Effect of Occupational Noise Exposure on Serum Cortisol Concentration of Night-shift Industrial Workers: A Field Study

  • Background: In both developed and developing countries, noise is regarded as the most common occupational hazard in various industries. The present study aimed to examine the effect of sound pressure level (SPL) on serum cortisol concentration in three different times during the night shift. Methods: This case-control study was conducted among 75 workers of an industrial and mining firm in 2017. The participants were assigned to one of the three groups (one control and two case groups), with an equal number of workers (25 participants) in each group. Following the ISO 9612 standard, dosimetry was adopted to evaluate equivalent SPL using a TES-1345 dosimeter. The influence of SPL on serum cortisol concentration was measured during the night shift. The serum cortisol concentration was measured using a radioimmunoassay (RIA) test in the laboratory. Repeated measure analysis of variance and linear mixed models were used with ${\alpha}=0.05$. Results: The results indicated a downward trend in the serum cortisol concentration of the three groups during the night shift. Both SPL and exposure time significantly affected cortisol concentration (p < 0.0001, p < 0.0001). Conversely, age and body mass index had no significant influence on cortisol concentration (p = 0.360, p = 0.62). Conclusion: Based on the obtained results, increasing SPL will lead to enhancement of serum cortisol concentration. Given that cortisol concentration varies while workers are exposed to different SPLs, this hormone can be used as a biomarker to study the effect of noise-induced stress.

스트레스 유형에 따른 고양이 혈중 Cortisol농도의 변화 (Change of Blood Cortisol Concentration in Domestic Cats by Various Stress Types)

  • The present study was performed to evaluate circadian rhythm of blood nortisol concentration and change of blood cortisol concentration by various stress type in domestic cats. The ELISA kit for cortisol measurement was evaluated on the effective assay range, day to day variation, within-run variation and accuracy. The results were 1-1,000 ng/ ml, 0.7-5.9%, 0.9-4.5% and 98%, respectively. The circadian rhythm of serum cortisol concentration is presented in cats. Change of serum cortisol concentration was also examined with several stressors, stimulation with hamster as a prey, beep electronic sounds, vehicle transportation and unfamiliar environment. Unfamiliar environment showed most remarkable change in serum cortisol level.

  • Background: High cortisol levels are frequently observed in patients with severe infections are of prognostic value in sepsis. The aim of this study was to evaluate the clinical usefulness of serum cortisol in assessment for the severity of community-acquired pneumonia (CAP). Methods: This study analyzed the results of 52 CAP subjects admitted in Changwon Fatima Hospital between July 2008 to May 2010. Total serum cortisol, infection markers such as C-reactive protein (CRP), procalcitonin (PCT) and CURB (Confusion, Uremia, Respiratory rate, Blood pressure)-65 were examined retrospectively. Results: In clinically unstable subjects on admission day 4, baseline serum cortisol, CURB-65, and CRP were elevated significantly compared to those of stable subjects. Area under curve (AUC) of cortisol, CRP, and CURB-65 from ROC curves were 0.847, 0.783, and 0.724 respectively. In the subjects with serum cortisol ${\geq}22.82{\mu}g/dL$, CRP, PCT, CURB-65 score, and mortality were significantly elevated. Conclusion: These findings suggest that measurement of serum cortisol in early stage may provide helpful information in the assessment of CAP severity.

Effect of Suckling Systems on Serum Oxytocin and Cortisol Concentrations and Behavior to a Novel Object in Beef Calves

  • We investigated differences between effects of natural- and bucket-suckling methods on basal serum oxytocin (OT) and cortisol concentrations, and the effect of OT concentration on affiliative and investigative behavior of calves to a novel object. Ten Japanese Black calves, balanced with birth order, were allocated evenly to natural-suckling (NS) and bucket suckling (BS) groups. Blood samples were collected at the ages of 1 and 2 months (1 week after weaning) calves, and serum OT and cortisol concentrations were measured using enzyme-linked immunosorbent assay and enzymeimmunoassay tests, respectively. Each calf at the age of 2 months (2 weeks after weaning) was released into an open-field with a calf decoy, and its investigative and affiliative behaviors were recorded for 20 minutes. In 1-month-old calves, the basal serum OT concentration ($25.5{\pm}4.9$ [mean${\pm}$standard deviation, pg/mL]) of NS was significantly higher than that of BS ($16.9{\pm}6.7$) (p<0.05), whereas the basal cortisol concentration ($5.8{\pm}2.5$ [mean${\pm}$standard deviation, ng/mL]) of NS was significantly lower than that in BS ($10.0{\pm}2.8$) (p<0.05). Additionally, a negative correlation was noted between serum OT and cortisol concentrations in 1-month-old calves (p = 0.06). Further, the higher serum OT concentration the calves had at 1 month old, the more investigative the calves were at 2 months old but not affiliative in the open-field with a calf decoy. Thus, we concluded that the natural suckling method from a dam elevates the basal serum OT concentration in calves, and high serum OT concentrations induce investigative behavior and attenuate cortisol concentrations.

지역사회 거주 성인의 지각된 스트레스, 혈중 코티졸 수준 및 텔로미어 길이의 관련성 (Associations of Perceived Stress Level, Serum Cortisol Level, and Telomere Length of Community-dwelling Adults in Korea)

  • Purpose: To investigate associations of perceived stress level, serum cortisol level, and telomere length of community-dwelling adults in Korea. Methods: Data of a total of 135 community-dwelling adults aged over 40 years living in D metropolitan city from December 2020 to March 2021 were collected. Perceived stress level over the past month were measured using the Perceived Stress Score. Serum cortisol level was analyzed using a chemiluminescent microparticle immunoassay. Telomere length was determined using quantitative real-time polymerase chain reaction. The statistical package SPSS 23.0 was used to perform Chi-square test, independent t-test, and Pearson's correlation coefficient analysis. Results: There was no association between perceived stress and serum cortisol level (r = .07, p= .402). Serum cortisol level was not significantly associated with telomere length either (r = -.15, p= .081). However, the higher the perceived stress level, the shorter the telomere length (r= -.29, p= .001). Conclusion: These results suggest that perceived stress might induce physiological stress, which might partially affect gene biology. Further longitudinal research is needed to investigate the effect of perceived stress on telomere length. Intervention for relieving stress should be included in stabilizing the genetic environment of adults.

단기 속박스트레스가 마우스 혈청 Cortisol, Dehydroepiandrosterone Sulphate 농도에 미치는 영향 (Effects of Short-term Immobilization Stress on the Mouse Serum Concentrations of Cortisol and Dehydroepiandrosterone Sulphate)

  • 단기 속박스트레스가 혈청 cortisol과 DHEAS 농도에 미치는 영향을 알아보고자 단기 속박스트레스 (IS)를 받은 BALB / c 숫컷 생쥐의 혈청 cortisol과 DHEAS 농도를 방사선면역측정 (RIA)을 이용하여 분석하였다. 그 결과 cortisol의 경우 30분 IS를 받은 그룹 (Ⅰ-30N)에서 대조군에 비해 유의적인 증가를 보였으며 (p<0.01), 120분 부과 (Ⅰ-120N) 시에도 대조군에 비해 유의적인 증가를 보였다. 가시오갈피 (SG) 처리군에서도 30분 IS를 받은 그룹 (Ⅰ-30T)에서 대조군에 비해 유의적 증가를 보였고 (p<0.01), 120분 부과(Ⅰ-120T) 시에도 대조군에 비해 유의적으로 증가하는 경향을 나타내었다. SG 처리군과 비처리군 모두에서 cortisol의 계속적인 증가를 관찰할 수 있었다. 30분 부과 그룹에서 SG 비처리군 (Ⅰ-30N)에 비해 SG 처리군 (Ⅰ-30T)에서 cortisol 농도의 유의적인 감소를 나타내었고, 120분 부과 그룹에서도 SG 비처리군 (Ⅰ-120 N)에 비해 SG 처리군 (Ⅰ-120T)에서 cortisol 농도의 유의적인 감소를 나타내었다. 이에 반해, DHEAS의 경우 30분 속박 (Ⅰ-30N)시 대조군에 비해 유의성 없는 감소 경향을 보였으나, 120분 속박 (Ⅰ-120N)시 대조군에 비해 유의적인 감소를 나타내었다 (p<0.01). SG 처리군에서도 30분 속박 (Ⅰ-30T) 시 대조군에 비해 유의성 없는 감소 경향을 보였으나, 120분 속박 (Ⅰ-120T)시 대조군에 비해 유의적인 감소를 나타내었다 (p<0.01). 그러나 30분, 120분 부과그룹 모두에서 SG 처리에 대한 유의적 차이는 없었다. 따라서 본 연구를 통하여 단기 속박이라는 국한된 스트레스에도 불구하고 DHEAS가 지속적으로 감소됨으로써 단기 속박스트레스가 혈청 cortisol과 DHEAS에 영향을 미치고 있음을 알 수 있었으며, 이를 통하여 DHEAS가 스트레스 관련 호르몬임을 확인할 수 있었다.
