Effect of Persimmon Vinegar on Serum Lipid Profile in Rats with High Cholesterol Diet (감식초가 고콜레스테롤 식이를 한 흰쥐의 혈중 지질 성분에 미치는 영향)

  • This study was performed to evaluate the effect of persimmon vinegar on serum lipid components in the hypercholesterolemic Sprague-Dawley male rats. Forty rats were divided into four groups and fed diets containing l% cholesterol for 4 weeks. Experimental diets added to distilled water, 1.5%, 3%, and 7% persimmon vinegar(PV) as drinking water. Concentration of total cholesterol, LDL and HDL cholesterol, triglyceride in serum and activity of AST, ALT in serum were assayed. Average food intake, weight gain and FER of experimental rats were not significantly different by contents of persimmon vinegar. Concentration of total cholesterol and LDL cholesterol in serum were lowest in 7% persimmon vinegar group. However concentration of HDL cholesterol in serum was highest in 3% persimmon vinegar group. Concentration of triglyceride in serum was significantly lower in persimmon vinegar group than control group. Activities of AST and ALT in serum were lowest in 7% persimmon vinegar group. The results in this study suggest that persimmon vinegar is effective in lowering serum triglyceride level.

  • Casein or soybean protein was subjected to there action with caffeic acidtyrosinase system at 30-35$\circ$C, pH 6.8 with aeration for 5hr. The resulting brown proteins were washed with acetone until the washings were on longer colored. However, modified protein still retained a light brown. The effects of the modified proteins and brown compounds on male Wistar strain rats were studied by pair-feeding of a cholesterol-free diet for 14days. Significant decrease in protein digestibility for the rats fed with the modified proteins were observed. Weight gain and protein digestibility were not influenced by feeding brown compounds, but the feeding of brown compound from casein caused an enlargement of caecum. The concentrations of serum cholesterol and triglyceride in the rats fed with modified proteins and brown compounds were mostly unchanged against the rats fed with untreated proteins. These results suggest that the decrease in protein digestibility induced by enzymic browning-reaction did not cause the decrease in concentration of serum cholesterol.

Influences of Clotrimazole on the Blood Cholesterol and HDL-Cholesterol level in Rats (흰쥐의 혈중 콜레스테롤 및 HDL-콜레스테롤 함량에 미치는 Clotrimazole의 영향에 관한 연구)

  • Influences of clotrimazole on the blood cholesterol and HDL-cholesterol level were studied in rats. Rats were provided food and water ad libitum and clotrimazole and methylcellulose were gavaged for 6 days. Clotrimazole was suspended in 1% methylcellulose solution as and administered at concentration 20mg/Kg, 40mg/Kg, 60mg/Kg. Body weight gain and liver weight/body weight ratio, serum cholesterol level, serum HDL-cholesterol level, serum triglyceride level, the activity of cytochrome p450 and erythromycin demethylase were determined at 6th day. Clotrimazole decreased the body weight gain a little as compared with control group and did not show any influence on liver weight/body weight ratio. Clotrimazole increased the serum HDL-cholesterol and serum triglyceride level significantly. Clotrimazole increased the microsomal cytochrome P450 significantly and increased the erythromycin demethylase (cytochrome P450 IIIA) significantly too. It might be conclued that clotrimazole showed a little influence on body weight and increased the serum lipid, especially HDL-cholesterol level. It also increased microsomal cytochrome P450 IIIA significantly. It might be concluded that clotrimazole showed a corelative influence between HDL-cholesterol and cytochrome P450 IIIA. In these results clotrimazole can be used as an anti-atherosclerotic agent by increasing the HDL-cholesterol but it is necessary that cloreimazole will show any adverse or side action on body or not.

  • Effects of amino acid composition and average hydrophobicity of soybean peptides on serum cholesterol in rats were investigated. Soybean protein(ISP), casein(SCN), their peptic hydrolyzates fractionated by acid precipitations(SHT, SH8, SH6, SH4, CHT), and amino acid mixtures of the same composition as the proteins(SAA, CAA) were prepared to feed to rats. The amino acid composition of the peptides was analyzed by HPLC and the concentration of serum cholesterol in the rats was measured. By data analysis, it was found that there was no relationship between ratio of Lys/Arg or molar ratio of hydrophobic amino acids and serum cholesterol level. And also there was no relationship between the concentration and average hydrophobicity calculated by the method of Tanford, Manavalan, for Meirovitch, only except by the method of Krigbaum(r=-0.736); the higher the average hydrophobicity of Krigbaum was, the lower the concentration of serum cholestrol became.

Effects of Feeding the Mixture of Linseed and Sunflower Seed Oil on the Lipid Components of Serum in Dietary Hyperlipidemic Rats (아마인유와 해바라기 종자유의 혼합급이가 식이성 고지혈증 흰쥐의 혈청 지질 성분에 미치는 영향)

  • This study was carried out to investigate the effect of the feeding mixture of linseed oil, rich in n-3 PUFA and the sunflower seed oil, rich in n-6 PUFA on the lipid metabolism in the dietary hyperlipidemic rats. After male Sprague-Dawley rats were induced hyperlipidemia by feeding the diet containing lard, butter and cholesterol for 3 weeks, then they were fed with the diet containing lard 3.0 % and butter 12.0% for control, the mixture in different proportion of both linseed oil and sunflower seed oil and antihyperlipidemic drugs for 2 weeks. Analysis of the lipid component of the serum showed following results. Concentration of total cholesterol in serum was significantly lower in the other groups except group 2 (lard 3.0 % + olive oil 12.0%) than in the control group , especially in groups 3 (cholestyramine 2.0%) and 5 (lard 3.0% + linseed oil 12.0%). HDL-C and the ratio of HDL-C to total cholesterol concentration were higher in groups 1, 3, 5 and 9 (lard 3.0% + sunflower seed oil 12.0%) , while the atherosclerotic index was low in groups3, 5. Concentrations of free cholesterol and choesteryl ester in serum were significantly lower in the other groups except group 2 than in the control group, especially lower in grop 5. Concentration of LDL was significantly higher in group 2 while it was remarkably lower in groups 3 and 5, while those of triglyceride in groups in 3 and 4 (lipraroid) were significantly higher than in the control groups. From the data on concentration of total choelsterol , HDL-cholesterol, LDL, phospholipid and triglyceride in serum, the results suggested that the feeding mixed with 3.0% lard and 12.0% linseed oil or 2.0% cholestyramine were most effective for the improvement of the serum lipids.

Effects of Caffeine Supplementation on Lipid Concentrations of Serum and Liver in Ovariectomized Rats (난소 절제 흰쥐에서 카페인 첨가가 혈액과 간 지질 농도에 미치는 영향)

  • The purpose of this study was to examine the impact of caffeine on lipid concentrations in ovariectomized rats. Forty-four rats (75${\pm}$5 g) were randomly divided into ovariectomy(ovx) and sham groups, each of which were randomly divided into two subgroups that were fed the control diet or a caffeine-supplemented diet (0.03%/diet). The experimental groups were classified into 1) sham-control, 2) sham-caffeine, 3) ovx-control and 4) ovx-caffeine groups. All rats had been fed on experimental diet and deionized water ad libitum for 6 weeks. Triglyceride concentration and total cholesterol were measured in serum and liver. Serum HDL-cholesterol was measured. Serum LDL-cholesterol and atherogenic index were calculated. Weight gain and food efficiency ratio were higher in ovx groups than in sham groups (p<0.05). Caffeine intake did not affect weight gain and food efficiency ratio. Mean food intake was significantly increased in ovx-caffeine group than other three groups. There were no significant differences in serum triglyceride, total cholesterol, HDL-cholesterol, LDL-cholesterol and atherogenic index between the control diet group and caffeine diet group. In addition, hepatic triglyceride and cholesterol level was unaffected by caffeine intake or ovariectomy. Therefore, we suggest that caffeine (0.03%/diet) intake may not have adverse effects on serum lipid level in ovariectomized rats.

Effect of Surface Hydrophobicity of Soybean Peptides on the Concentration of Serum Cholesterol and Fecal Steroid Excretion in Rats (대두 펩타이드의 표면소수도가 흰쥐의 혈청 콜레스테롤 농도 및 분변 스테로이드의 배설량에 미치는 영향)

  • Effect of surface hydrophobicity of soybean peptides on serum cholesterol in rats was investigated. Soybean protein(ISP), casein(CNP), and their peptic hydrolyzates fractionated by acid precipitations (SHT, SH8, SH6, SH4, CHT, CH6, CH5, CH4) were fed to rats and the concentration of serum cholesterol and the fecal steroid excretion were measured. And surface hydrophobicities of the peptide fractions were measured by determining by the ANS flourescence intensity and SDS binding capacity. It was found that the higher the surface hydrophobicity of peptides was, the more the fecal steroids excreted(r=0.801) and the lower the concentration of serum cholesterol became(r=-0.868). However, there was no relationship between SDS surface hydrophobicity and fecal steroids or serum cholesterol. ANS surface hydrophobicity of soybean protein was increased by enzymatic hydrolysis. These results suggest that high surface hydrophobicity of peptides formed during digestion is responsible for the hypocholestrolemic effect of soybean protein through the hydrophobic interaction between the peptides and bile salts in rats.

Effects of Green Tea Consumption on Serum Lipid Profiles (녹차 섭취가 혈중 지질 성상에 미치는 효과)

  • In the present study, we investigated the effects of chronic green tea consumption on the serum lipid profiles of 92 subjects (mean age: 59.5 $\pm$ 8.9, men: 41, women:51) living in a green tea cultivation village. The anthropometric and biochemical blood indices were measured, and nutritional intakes were assessed using 24-h recall method. Also, we administered a questionnaire regarding green tea consumption. The subjects were divided into three groups based on the period of green tea consumption (0, < 10 year, and $\geq$ 10 year). There were no significant differences in age, anthropometric indices (Wt, Ht, BMI, Body fat, LBM), and nutritional intakes among the groups. Serum LDL-cholesterol concentration was significantly lower in the $\geq$ 10 year group compared with the other groups (p < 0.05). Also, the LDL/HDL-cholesterol ratio was significantly lower in the $\geq$ 10 year group compared with the other groups (p < 0.05). The frequency of subjects with a normal range of serum LDL-cholesterol concentration (< 200 mg/dl) was significantly higher in the $\geq$ 10 year group compared with the other groups (p < 0.05). These results imply positive effects of chronic green tea consumption on blood lipid profiles, which requires confirmation in further studies.

Analysis of Serum Lipids and Lipoprotein Electrophoresis Fractions in Hypertensive Patients

  • We analyzed the concentration of serum total cholesterol (TC), high density lipoprotein cholesterol (HDLC), low density lipoprotein cholesterol (LDLC), triglyceride (TG) and the serum lipoprotein electrophoresis fractions in hypertensive patients (hypertension group, n=182). The average concentration of lipids and lipoprotein fractions in the hypertension group was compared to that of the normal group. The average concentrations of serum TC, LDLC and TG in the hypertension group were significantly higher than those of the normal group (3.3%, 11% and 70%, respectively) (P<0.05). But HDLC was nonsignificantly lower (2%) (P<0.05). In the hypertension group, the percentages of patients who had an abnormally high level of total cholesterol, HDLC, LDLC and TG were 12.1%, 8.2%, 24.2% and 44.5%, respectively. The average of ${\beta}$-lipoprotein (27.5%) and pre ${\beta}$-lipoprotein (12.6%) were significantly higher, whereas ${\alpha}$-lipoprotein (15.2%) was significantly lower in the hypertension group than in the normal group (P<0.05). The percentages of patients who had an abnormally high level of very low density lipoprotein (pre ${\beta}$-LP), LDLC (${\beta}$-LP) and chylomicron were 31.3%, 17.0% and 12.6%, respectively. This study suggests that an increasing of total cholesterol, LDLC and TG, pre ${\beta}$-LP, ${\beta}$-LP and chylomicron are associated with a risk factor for hypertension.

Lecithin : Cholesterol Acyltransferase Activities in Rats Fed Cow량s Milk with Different Levels of Cholesterol (식이 콜레스테롤의 섭취량에 따라 우유가 흰쥐의 Lecithin: Cholesterol Acyltransferase 활성에 미치는 영향)

  • The effect of milk in low and high cholesterol diets were investigated on serum cholesterol esterification. Weanling male Sprague-Dawley rats were divided into low(0.01% w/w) and high (1.01% w/w) cholesterol-diets groups. Bothlow and high cholesterol groups were consisted of three groups : control , LM(low heat milk), and HM (high heat milk) groups. After feeding these experimental diets for six weeks, serum cholesterol (free cholesterol and cholestryl ester) concentration and activity of lecithin.: cholesterol acyltransferase (LCAT) were measured, and serum lipoprotein profile was examined using gel column chromatography. According to the result, activity of LCAT was elevated independently by intakes of high cholesterol and milk , which resulted in the increase of daily turnover of serum cholesteryl ester. However, the turnover of HDL-cholesteryl ester increased only by milk. LCAT activity was moderately correlated with levels of total-and HDL-free choelsterol. It is concluded from the present study that milk had the cholesterol-lowering effect which partly appears to be mediated through facilitated reverse cholesterol transport.
