양식어류 고음과 잔사의 단백질 품질평가 (Protein Nutritional Qualities of Fish Meat Extracts and Their Residues)

  • To confirm the food quality of conventionally processed fish extracts, protein quality of boiled crucian carp(Carassius carassius) and bastard halibut(Paralichthys olivaceus) extracts and their residues were evaluated. For the both fish extracts, some of the essential amino acids were lowered significantly but two times more proline and glycine were detected in extracts than those in raw fish meats. Boiling(100oC, 5 hours) caused 1.8(crucian carp)~2.4(bastard halibut) times more total free amino acid contents in fish extracts as compared to those in original fish meats. Taurine, glutamic acid, proline, lysine, and ammonia were the predominant free amino acids released in fish extracts. In vitro digestibility of boiled fish extracts were lower at a level of 4~6% than those of raw fish meats. Fish extraction residue had a higher in vitro digestibility and had a 60% lower level of TI than that of original fish meats. 18(bastard halibut)~ 24%(crucian carp) of available lysine was reduced in boiled fish extracts but a remarkable variation was not noted between extracts and residues. PERs and NPRs of fish extracts were significantly lower than those of casein, while those values of extraction residue were slightly higher as compared to those of control(ANRC casein). In vivo apparent digestibility exhibited a similar trend to in vitro digestibility. Hematological properties in serum of rat fed with fish extracts and residue were not changed significantly but the serum cholesterol concentration were reduced in rats fed fish extraction residue comparing with those of control. These results suggest that body weight loss due to fish extracts may not affect physiological changes.

목향생화탕(木香生化湯)과 목향생화탕가녹이(木香生化湯加鹿茸)이 산후회복(産後回復) 및 유즙분비(乳汁分泌)에 미치는 영향(影響) (Effects of Mokhyangsaenghwa-tang and Mokhyangsaenghwa-tang plus Cervi Pantotrichum Cornu on Postpartum Recovery and Lactation)

  • Objective : This study was conducted to investigate the effect of Mokhyangsaenghwa-tang (MS) and Mokhyangsaenghwa-tang plus Cervi Pantotrichum Cornu (MS-C) on postpartum recovery and lactation. Materials and Methods : We used 18-week pregnant Spraque Dawley rats and administered the decoctions of MS and MS-C to rats once a day for 4 days or 8 days. Then we observed changes in the body weight of pup rats and complete blood cell count, liver function test, renal function test, mammary gland tissue, level of serum prolactin, ${\beta}-casein$ and WAP of postpartum rats. Result : A significant increase in body weight was observed in MS-C treated pup rats compared with in MS treated group. The levels of WBC and platelet from MS group and MS-C group were decreased compared with the control group. The levels of RBC, hemoglobin and hematocrit from MS group and MS-C group showed statistically significant increases compared with the control group. The levels of protein, albumin from MS group and MS-C group were increased compared with the control group. The levels of BUN, creatinine from MS group and MS-C group did not show statistically significant changes compared with the control group. The mammary gland tissues from MS group and MS-C group showed increased angiogenesis. The levels of serum prolactin from MS group and MS-C group were increased compared with the control group. The expression of ${\beta}-casein$ and WAP genes from postpartum rats treated with MS and MS-C was increased. Conclusion : This study shows that MS and MS-C improved postpartum recovery and lactation in rats.

영양실험을 위한십이지장 궤양 흰쥐 실험 모델 개발과 그 영양생리적특성 (Development of Experimental Animal Model with Duodenal Ulcer for Nutritional Study and its Nutritional Characteristics)

  • This study aimed to develop an experimental animal model of duodenal ulcer for application to nutrition, and to verify the nutritional characteristics. The 7 different doses and 8 different injections of cysteamine and 2 kinds of diets with different casein levels(10%, 20%) were tested to examine the incidence and the process of duodenal ulcer in female Sprague-Dawley rats. The result showed that the duodenal ulcer was induced when rats fed 10% casein diet(for 9 days) were injected 6 times with 13mg/100g BW cysteamine. The first injection was conducted after 24 hours of fasting, followed by the 2nd shot after 3 hours. The following shots were repeated every 3 day(4th and 7th days). Duodenal ulcer was observed 3 days after the last injection by optical microscope and the naked eye. The characteristics of nitrogen bioavailability of duodenal ulcer model were as followings : 1) gastric emptying rate was faster, 2) trypsin activity at duodenum was higher, 3) total protein concentration at serum and nitrogen retention rate were lower than the control. 단백질 급원의 변화와 열량제한 및 식이회복이 흰쥐의 성장발달에 미치는 영향 (Effects Protein Quality, Energy Restriction and Subsequent Rehabilitation on Growth in Young Rats)

  • Effects of dietary protein qualify, energy restriction, and subsequent rehabilitation were studied in the weanling rats. Rats were devided into the six experimental diet groups. Rats in AC (casein- containing diet ), AS (soyprotein-containing diet), and AG(gluten-containing diet) group consumed their diets ad libitum, and rats in PC, PS, PG groups were pair-fed in 60% of the of amounts eaten by corresponding rats in the AC, AS, AG groups, respectively. After 3 weeks of nutritional restriction, all rats were switched to the AC diet for 14 days. At the end of restriction period, body weight, weight gain, food intake, and FER were higher in casein- fed rats than other groups of rats. The differences were, however, disappeared with rehabilitation diet, except body weights which were not fully recovered until the end of experiment. The weights of liver, gastrocnemius muscle, kidney, small intestine, spleen, and lung were the highest in the casein group, and the lowest in gluten group at the end of restriction period. The recovery with rehabilitation diet were differ in different organs. The muscle and liver seemed to be the most affected organs by dietary protein quality and energy restriction. There were no differences in brain weights among the experimental groups during the restriction period, however, after 2 weeks of rehabilitation, rats in AC group showed lower brain weight compared to AC and AS groups, and the brain weight of PC group was lower than AC group. Protein contents in liver and muscle showed the same trends as their weights. Gluten-fed rats showed low serum protein concentration, but recovered fully with rehabilitation diet for 3 days.

액상 대두유 섭취가 영아의 적혈구 지방산 조성과 혈청 지질농도에 미치는 영향 (Effects of Soy-Based Infant Formula on the Fatty Acid Composition of Erythrocytes and Plasma Lipid Levels in Full-Term Infants)

  • In Korea, the concerns about the consumption of soy-based infant formulas have received considerable public attention. In has been known that the n-3/n-6 ratio of soy has a beneficial effect on the health. This study was conducted to investigate whether the soy-based infant formula has an effect on the composition of fatty acids in erythrocytes with regard to the plasma lipid levels in full-term infants. The infants who were delivered at Kyung Hee Medical Center were recruited. Experimental groups were the breast milk group(n=17), soy-based infant formula group(n=12) and casein-based infant formula group(n=19). Anthrophometic and dietary assessments were performed and the composition of RBC fatty acids and blood lipid levels were measured. The body weight, length, chest circumference and head circumference of the subjects were normal growth range showing no significant difference among the experimental groups. The ratio of n-3/n-6 ratio in breast milk, soy-based infant formula and casein-based infant formula were 0.11, 0.14 and 0.13, respectively. N-3/n-6 ratio of the subjects erythrocyte fatty acids for each groups were 0.18, 0.21 and 0.17, respectively. Most of the fatty acids of erythrocyte were not significantly different among the three experimental groups. The blood lipid levels of each experimental group were normal range. The total lipid level of casein-based formula group was higher than that of other experimental groups and LDL-cholesterol level of soy-based formula group was the lowest among the three groups.

나이가 다른 단계에서 식이단백질 수준이 흰쥐의 Ca 대사에 미치는 영향 (The Effects of Age and Dietary Protein Level on Ca Metabolism in Rats)

  • To study the effects of the age and the dietary protein content on Ca metabolism male rats of 1 month 6 month 12 month of age were fed experimental diets containing 5%, 15% or 50% casein for 4 weeks. Food and ca intake were higher in old rats and in high protein groups. The weight ash and Ca contents of femur and tibia were higher in old rats. The higher dietary protein level resulted in higher skeletal weigh ash and Ca contents. But high protein diet(50% casein) lead to reduced bone mineral density(ash/dry bone weight) and Ca density(Ca/dry bone weight) in 1 month old rats. Low protein diet(5% casein) on the other hand reduced the bone growth even though the bone density was higher in this group. The ill effect of low protein diet was not evident in 12 month old rats. Glomerular filteration rate(GFR) and urinary Ca excretionincreased with age and with dietary protein level especially in 12 month old rats. Serum immunoreactive parathyroid hormone(iPTH) level tended to be higher in aged rats but was not affected by dietary protein level except 1 month old rats where 50% protein group showed significantly higher value. This study showed that the dietary protein level seemed to have different effect on Ca metabo-lism in rats of different age., The low bone density in the high protein group of growing rats may be due to the higher iPTH level and increased urinary Ca. The dietary protein level however had no effects on the bone composition in aged rats even though the higher urinary Ca excretion. In conclusion this study suggests that high protein intake from young may lead to less peak bone mass and to increase the bone loss in later years, which would increase the risk for osteporosis.

불포화 고지방식을 섭취한 흰쥐의 체내지질대사에 대한 대두 단백질과 칼슘의 섭취 효과 (Effects of Soy Protein and Calcium on Lipid Metabolism in Rats Fed Unsaturated Fat Diet)

  • This study was performed to observe how soy protein and calcium in rats fed unsaturated fat-enriched diet affect the total lipid and cholesterol contents of blood and tissues male Sprague-Dawley rats weighing approximately 220g were fed four purified diets which contained 18% (w/w) corn oil, 1% (w/w) cholesterol, two sources of protein : casein or isolated soy protein (ISP) and two levels of dietary calcium : 0.1% or 1.0%, first for four weeks (Expt. 1) and second for eight weeks (Expt. 2). The contents of total lipid, cholesterol and triglyceride in blood, liver, heart and feces were determined. After four weeks feeding, the serum lipid and cholesterol concentration was not significantly different among the groups. After eight weeks feeding, these concentrations were significantly lower in ISP-High calcium group. The serum lipid concentration was influenced by dietary protein sources and calcium levels. These results indicated that the hypolipidemic effects of soy protein and high calcium intake were partly due to decrease in lipid absorption and these effects were not detected at 4 weeks feeding but 8 weeks feeding.

콩 단백 성분 및 연령이 암컷 흰쥐의 혈장 지질 농도와 인지질 지방산 패턴에 미치는 영향 (Effects of Soy Protein Concentrate and Age on Plasma Lipids and Phospholipid Fatty Acid Patterns in Female Rats)

  • 혈중 지질농도는 식이단백질 종류보다는 연령의 영향을 많이 받아, young군보다 old군의 총 콜레스테롤, TG, HDL-콜레스테롤, LDL + VLDL-콜레스테롤 및 AI 모두 높았다. 한편 콩 단백질군에서 HDL-콜레스테롤 수준이 증가하고, LDL+VLDL-콜레스테롤와 AI는 감소하여, 혈관질환에 대한 콩 단백질의 유익한 효과를 확인할 수 있었다. 식이 단백질에 의해 유의한 영향을 받은 혈장 인지질의 지방산조성중 22:0, 18:1$\omega$9, ∑MUFA 조성은 카제인군에서 콩 단백질군보다 높았으며, 반대로 ∑SFA 조성은 카제인군에서 낮았다. 연령에 의해 혈장 인지질 지방산 조성에 있어 유의한 차이를 보인 지방산 중 22:0, 18:l$\omega$9, 22:1, 18:3$\omega$3 및 22:4$\omega$6는 young군에서 높은 반면에, 22:6$\omega$3, ∑$\omega$3, 18:2$\omega$6, 20:4$\omega$6 ∑$\omega$6 및 ∑PUFA의 조성비율은 old군에서 높게 나타났다. 이와 같이 식이 단백질의 종류보다 연령의 영향력이 더 많이 나타난 것은 혈중 콜레스테롤 수준이 식이 단백질보다는 연령의 영향을 더 크게 받았기 때문으로 생각된다. 혈장 인지질 지방산의 대사지표 중 $\Delta$7-desaturation index(16:0⇒16:1$\omega$7)와 $\Delta$9-desaturation index(18:0⇒18:1$\omega$9)가 식이 단백질의 영향을 받아 카제인군에 비해 콩 단백질군에서 유의하게 낮았다. $\Delta$7-및 $\Delta$9-desaturation index만이 식이 단백질의 영향을 받은 것은 식이 단백질보다 desaturase활성에 더 큰 영향력을 미치는 콜레스테롤이 식이에 첨가되었기 때문이라 생각된다. $\Delta$-4 desaturation index (22:4$\omega$6⇒22:5$\omega$6)는 young군에 비해 old군에서 높았으며, elongation index(20:4$\omega$6⇒22:4$\omega$6)는 old군에서 낮았다. 대부분의 elongation과 desaturation 단계는 연령에 따라 유의한 차이를 보이지 못하였으나, $\omega$3계 지방산의 전체적 elongation-desaturation 단계를 나타내는 products-fatty acid($\omega$3)/a-LNA($\omega$3) 비율이 old군에서 young군보다 유의하게 높아, 연령에 따른 PUFA 대사의 변화가 관찰되었다. 이 상에서와 같이, 콩 단백질의 섭취로 흰쥐 혈장 인지질의 ∑MUFA조성은 낮고 ∑SFA 조성은 높아 다른 지질 강화성 분의 섭취와 유사한 결과를 나타내었다. 혈중 콜레스테롤 수준이 높은 old군에서 체내 막조직의 유동성을 유지하기 위해 PUFA의 합성 특히 $\omega$3계 지방산의 elongation-desaturation이 증가하여 총 PUFA 조성이 young군보다 높았다. 연구결과로 이소플라본을 포함한 콩 단백질의 혈중 지질저하 기전에 대해 체계적인 설명은 할 수 없지만, 콩 단백질이 혈장 지방산의 조성을 변화시킴으로써 간접적으로 심혈관계 질환에 대해 유익한 효과를 미침을 알 수 있었다. 한편, 이러한 혈관의 건강과 관련된 콩의 유익한 효과는 이소플라본과 같은 콩의 특정 성분의 단독효과라기보다는 콩 단백질과의 복합효과에 의한 것이라는 주장은 매우 설득력 있다고 생각된다.

대두 펩타이드의 표면소수도가 흰쥐의 혈청 콜레스테롤 농도 및 분변 스테로이드의 배설량에 미치는 영향 (Effect of Surface Hydrophobicity of Soybean Peptides on the Concentration of Serum Cholesterol and Fecal Steroid Excretion in Rats)

  • 대두 펩타이드의 표면소수도가 혈청 콜레스테롤의 농도에 미치는 영향을 조사하기 위하여, 대두 단백질(ISP), 카세인(CNP), 이들 단백질을 펩신으로 가수분해하여 pH에 따른 펩타이드 침전 획분들(SHT, SH8, SH6, SH4, CHT, CH6, CH5, CH4)을 흰쥐에 섭취시키고 혈청 콜레스테롤 농도 및 분변 스테로이드이 배설량을 측정하였다. 그리고 각 펩타이드의 표면소수도를 ANS 형광법 및 SDS 결합법으로 측정하여, 이들의 상관관계를 분석한 결과, 펩타이드의 ANS 표면소수도가 높아질수록 분변으로 배설된 스테로이드량은 증가하였으며(r=0.81), 혈청 콜레스테롤 농도는 낮아졌다.(r=-0.868). 그러나 SDS 표면소수도는 그들과 상관관계가 없었다. 또한 대두 단백질은 가수분해에 의하여 ANS 표면소수도가 증가하였다. 이상의 결과는 흰쥐의 담즙염이, 소화중 생성된 높은 표면소수도의 펩타이드와 결합하여 체외로 배출되므로서 대두단백질의 섭취에 의한 혈청 콜레스테롤 농도가 낮아짐을 시사하였다.

대두 펩타이드의 아미노산 조성 및 평균소수도가 흰쥐의 혈청 콜레스레롤 농도에 미치는 영향 (Effects of Amino Acid Composition and Average Hydrophobicity of Soybean Peptides on the Concentration of Serum Cholesterol in Rats)

  • 대두 펩타이드의 아미노산 조성 및 평균소수도가 혈청 콜레스테롤의 농도에 미치는 영향을 조사하기 위하여, 대두 단백질(ISP), 카세인(CNP), 이들 단백질을 펩신으로 가수분해하여 pH에 다른 펩타이드 침전 획분들(SHT, SH8, SH6, SH4, CHT) 및 각 단백질과 같은 조성의 아미노산 혼합물(SAA, CAA)을 흰쥐에 섭취시키고 혈청콜레스테롤 농도를 측정하였다. 각 펩타이드의 아미노산 조성을 분석하고, 이로부터 구한 Lys/Arg 비율 및 소수성 아미노산의 몰분율은 혈청 콜레스테롤의 농도와 상관관계가 없었으며, Tanford, Manavalan 및 Meirovitch법으로 구한 평균소수도와도 상관관계가 없었다. 그러나, Krigbaum법으로 구한 평균소수도와는 상관계수가 -0.736으로 음의 상관관계이어서, 식이 펩타이드의 krigbaum 평균소수도가 높을 수록 래트의 혈청 콜레스테롤 농도가 낮아졌다.
