Atopic dermatitis (AD) has become a serious epidemic in Korean children. We aimed to investigate the association between vitamin C, E and other nutrients, and serum total IgE/specific IgE levels in children with AD. A total of 119 children (0-24 mo) diagnosed with AD were recruited for this cross-sectional study from a medical center in Seoul. A 24 h recall was used to assess dietary intakes. Serum total and six food-allergen specific IgE levels were measured by CAP-FEIA. Serum vitamin E was also measured but only in 25 out of the total 119 participants. Multiple linear regression analysis was performed to estimate the coefficients between serum IgE levels and dietary intake as well as serum vitamin E. Serum vitamin E levels showed a significantly inverse association with serum total IgE and all specific IgE levels (P < 0.05). Fat intake was inversely related with specific-IgEs for egg whites, milk, buck wheat, soy, and peanuts (P < 0.05). Positive associations were found between carbohydrate (CHO) intake and total IgE and specific IgEs to egg whites, milk, soy, and peanuts (P < 0.05). Vitamin C, E and n-3/n-6 fatty acids were not related with serum total IgE and specific IgE levels except for the association between buck wheat and vitamin E. In addition, there were no significant differences between males and females in dietary intake and serum IgE levels by student's t-test. Although dietary vitamin E showed no association with serum IgE levels, serum vitamin E drew a significant inverse relationship with serum IgE levels. The evidence seems to suggest that vitamin E may possibly lower total and specific-IgEs in children with AD, and that it is important to maintain a relatively high serum vitamin E level in children with AD.
Many previous studies have proved that human allergic disease resulted from the formation of antibodies belonging to a unique immunoglobulin isotype termed immunoglobulin E (IgE). Most of IgE-producing plasma cells are found in the lymphoid tissue associated with the gastrointestinal and respiratory tracts. IgE may be found free in the mucosal secretions of these tissues, bound to local mast cells, or distributed by the systemic circulation to mast cells and basophils throughout the body. Total serum IgE concentrations tend to be higher in allergic adults and children compared with non-allergic individuals, but the value of total serum IgE as a screening test for allergic disease is limited. Total serum IgE levels are related to the probability of an individual having detectable allergen-specific IgE. Allergen-specific IgE concentrations vary with a person's age, the degree and duration of the recent allergen or cross-reactive allergen exposure. The value of quantitative assays for allergen-specific IgE has been suggested in recent studies. Serum IgE increases in many non-allergic diseases, including infectious and parasitic diseases. The IgE changes appear to be specific to the infectious agents, whereas non-specific in other diseases. The increased serum IgE in some of these conditions probably results from alterations in immune function. This review summarizes the clinical significance of total and allergen-specific IgE examinations in allergic diseases.
Atopic dermatitis is one of the most common skin disease in children and a characteristic chronically recurrent form of dermatitis with a hereditary tendency, affecting infants and may extend to the childhood or th the adult age. Environmental factors, stress, and foodstuffs such as milk, egg may cause atopic dermatitis. Nc/Nga mice were used as an animal model for human atopic dermatitis. The divided by 4group such as normal group, BMAC group, FK506 group, MR group for this study raised in conventional conditions. To investigate effect of Chamomile German on atopic dermatitis in NC/Nga mice, the serum IgE and IgG1 level were measured while the severity degree of the skin lesion was examined by the naked eyes of two volunteers who were unaware of the treatment status. The results were the followings. 1. The score on the severity degree of skin dermatitis in FK 506 and MR group was lower than that in control group. 2. The serum IgE level in control group was higher 25% than that in normal group. 3. The serum level of IgE in FK506 and MR group compares to control group was decreased. 4. The serum IgG1 level was decreased more than 3.5 times in FK506 compared to control group while MR group had significantly less the serum IgG1 than control group. From the above results, treatment of Chamomile German oil had the effect on atopic dermatitis in NC/Nga mice. If scientific researches on aroma oil are performed in various way, aroma oil will be used to cure skin dermatitis as a alternative therapy in the future.
Background: Although a skin test is the primary option for detecting allergen-specific IgE in clinics, the serum IgE immunoassay is also important because it allows for the diagnosis of allergy without any accompanying adverse effect on the patient. However, the low detection limit of IgE levels by immunoassay may restrict the use of the method in some occasions, and improving its sensitivity would thus have a significant implication in allergy-immunology clinics. Methods: In this study, we attempted to detect specific serum IgE by using immuno-polymerase chain reaction (IPCR) which combines the antigen-antibody specificity of enzyme-linked immunosorbent assays (ELISAs) with the amplification power of PCR. Results: Our results demonstrated that Blo t5-specific serum IgE can be detected by IPCR with a 100-fold higher sensitivity than ELISA, and cross-reactivity of serum IgE to other mite allergens is able to be analyzed by using only $0.3{\mu}l$ of serum sample. Use of real-time IPCR seemed to permit more convenient determination of specific serum IgE as well. Conclusion: We believe that IPCR can serve as a valuable tool in determining specific serum IgE, especially when the amount of serum sample is limited.
To assess a polential relationship of Korean ginseng to allergic reactions, the effect of Korean ginseng on the total IgE in serum as an allergologic parameter was investigated in humans. Serum total IgE levels were measured in 8 subjects who have been taking ginseng for more than 5 years, 4 subjects of weak constitution, 10 newly hired workers in the ginseng processing industry and 7 normal subjects unexposed to Korean ginseng as control group. Blood samples were taken before and after the exposure to Korean ginseng. Total IgE levels after the exposure to Korean ginseng were not significantly different from those before the exposure within each group. And also, the IgE levels of control subjects were not significantly different compared with those of other groups. These results suggest that the exposure to Korean ginseng dose not affect significantly the IgE immune response of the subjects.
Paragonimus westermani is a common fluke in Korea. The present study aimed to determine serum total IgE and specific IgG levels in experimental paragonimiasis of rats. Each Wistar rat was inoculated orally with 20~25 metacercariae of P. westermani from Cambaroides similis. Before and after infection(1,2,3,4,6,8 weeks) of P. westermani, the blood was collected from the retro- orbital venous plexus of rats and kept serum at $-70^{\circ}C$. Serum total IgE and specific IgG levels were determined by the capture and conventional enzyme-linked immunosorbent assay, respectively. The results were as follows; 1. Serum IgE values were increased to 0. 18${\pm}$0.042 at 2 weeks, $0.28{\pm}0.151$ at 4 weeks and 0.43${\pm}0.055$ at 8 weeks after infection. The absorbances of non-infected rats ranged $0.07{\pm}0.021~0.12{\pm}0.025$. 2. Specific IgG values were slightly increased at 3 weeks ($0.20{\pm}$0.032) and gradually increased up to 8 weeks($0.31{\pm}0.067$) after infection. The absorbances of non-infected rats ranged $0.11{\pm}0.035~0.18{\pm}0.019$. The present results suggested that p. westermani could elevate serum IgE and specific IgG antibodies in Wistar rats which were not a good definitive host.

Recently, it was reported that Pfaffia paniculata, known as Brazil ginseng, caused occupational asthma. To assess a potential relationship between Korean ginseng and allergic reactions, total IgE in serum, allergologic parameter, was investigated. Serum total IgE were measured in 8 takers of ginseng for more than 5 yr, 4 subjects of weak constitution, 10 workers participated in the washing of fresh ginseng in factory and 7 control subjects, Blood samples were taken before and after the scheduled exposure to Korean ginseng, sera were kept frozen until assay. Total IgE levels after the scheduled exposure to Korean ginseng were not significantly different in each group and total IgE levels were not significantly different in the control subjects compared with those of other groups at the start of experiment and at the end of experiment. These results suggest that the exposure to Korean ginseng dose not affect significantly the IgE immune response in volunteers. No changes of levels of IgE on the exposure of Korean ginseng may represent additional evidence of the lack of antigenic potential of Korean ginseng.
Poncirus trifoliata (L.) Raf (Rutaceae) fruits (PTFE) has been used for the treatment of allergic disease. IgE is normally one of the least abundant immunoglobulin (Ig) isotypes in the serum of both humans and several species of experimental animals: however a number of different stimuli can result in profound increases in IgE levels relative to other isotypes. In rodents, infection with many parasitic helminths can cause approximately 100-fold elevation in IgE within 2 wks. Immunization of mice with small amounts of protein antigens on alum also results in 10-fold to fold increase in total serum IgE, much of it specific for the immunizing antigen. In this experiment, I investigated the effect of an aqueous extract of Poncirus trifoliata (L.) Raf (Rutaceae) fruits (PTFE) on a in vivo and in vitro IgE production. PTFE dose-dependently inhibited the serum levels of IgE induced by antigens. The regulation of IgE synthesis is influenced by T cells and T cell derived factors. IL -4, a T cell-derived cytokine, has been shown to stimulate murine IgE synthesis both in vitro and in vivo. Current evidence suggests that IL-4 induces IgE synthesis in the mouse by stimulating H chain isotype switch. Lipopolysaccharide (LPS) plus IL-4 cause about l00-fold increase in IgE secretion by murine B cells. The effects of PTFE on the IL-4-dependent IgE response by mouse whole spleen cells were studied. Whole spleen cells were cultured for 7 days in the presence of LPS plus IL-4 and PTFE and the supernatants were assayed for IgE. IL-4 dependent IgE production of LPS-stimulated whole spleen cells was inhibited by PTFE. Moreover, in the present study using U266Bl human IgE-bearing B cells, I found that PTFE inhibited the production of IgE activated by LPS plus IL-4. These results indicate that PTFE have antiallergic activity by inhibition of IgE production from B cells.

Objectives : This study was aimed to evaluate the correlation between changes in disease severity and serum total IgE or eosinophil count in individual patients of atopic dermatitis depending on the change of severity. Methods : We performed a retrospective chart review of 8 cases of atopic dermatitis who underwent blood tests more than five times. Disease severity was assessed by objective SCORing of Atopic Dermatitis index (OSI). Blood tests include serum total IgE, eosinophil counts, aspartate aminotransferase (AST), alanine aminotransferase (ALT), blood urea nitrogen (BUN) and serum creatinine (SCr). In each subject, on the day that the blood test was performed, OSI was also assesed by a same physician. Correlation between OSI and total IgE or eosinophil count was analysed by nonparametric correlation analysis. Results : There was a statistically significant positive correlation between OSI and total IgE in three cases and a positive correlation between OSI and eosinophil count in two cases. In other three cases, there were no significant correlation. There were no cases that OSI correlated with total IgE and eosinophil count at the same time. In all cases, AST, ALT, BUN and SCr were stable. Conclusions : We suggest that total IgE and eosinophil count may reflect the severity of atopic dermatitis differently in individual patients. We also suggest that total IgE and eosinophil counts may be used as biomarkers to predict the severity of atopic dermatitis in some patients depending on the underlying pathology.
The change in mean absorbance values for IgA/IgE in rats and mice infected with Echinostoma hortense metacercariae was studied from the 2nd week to the 8th week after infection. Serum and intestinal luminal content (ILC) levels of IGA/IGE were measured by enzyme-linked immunosorbent assay(ELISA). The mean absorbance values obtained from IgA in the rats' ILC increased from the 2nd week to the 8th week after infection. The peak value (0.47$\pm$0.01) appeared in the 8th week. The mean absorbance values of IgE in the rats' ILC didn't increase significantly (p>0.05). The worm recovery rate decreased at a slower, pace after, infection. The duration in which the peak value of IgA in rats' ILC appeared was similar to that in which the worm recovery rate declined significantly. Serum levels of IgA/IgE in mice increased gradually from the 2nd week after infection. The peak value (0.45$\pm$0.01) of IgA appeared in the 8th week, and that (0.23$\pm$0.02) of IgE appeared in the 7th week after infection. The ILC level of IgA in mice continued to increase after infection, and reached its peak in the 8th week. The change in IgA/lgE in the serum and IgA in the ILC of mice was inversely related to worm recovery rate. As a result of this experiment, it is supposed that IgA/IgE may play an important role in the expulsion of Echinostoma hortense.
