  • Pregnant women in South Korea are a highly risk group fur iron deficiency anemia. Previous studies indicated that the 24-hour recall method was insensitive in distinguishing iron deficiency anemic women from normal women. This method is also impractical to when used at community health centers where no public health dietitians are employed. The objective of this study was to develop a convenient tool to evaluate the usual iron (Fe) intake of pregnant women. The study participants were 115 pregnant women (age 23 to 37 years) at gestational stage of 13 to 24 weeks. Anemic subjects were classified on the basis of their serum ferritin < 12.0 ${\mu}$g/L and hemoglobin < 12.0 g/dL levels. Food frequency questionnaires with 46, 29, and 15 commonly consumed food items were used to measure the usual intake of iron of the subjects. Hemoglobin and serum ferritin were measured from fasting blood samples. Nutrients intake was assessed on three consecutive days using the 24-hour recall method and the food record method. The iron index score calculated using the food frequency method showed a significantly positive correlation with iron intake for the three days dietary intake. The iron index showed a significantly difference (p < 0.05) between the normal and anemic groups. However, there was no significant difference in the iron intake between the anemic and the normal women as measured by the 24-hour recall and food record method. Our study indicated that the 29-food items questionnaire could be used as a screening tool to identify poor dietary intake of iron. (Korean J Community Nutrition 8(2) : 160170, 2003)

Effect of methyl donors supplementation on performance, immune responses and anti-oxidant variables in broiler chicken fed diet without supplemental methionine

  • Objective: Methionine (Met) is involved in methyl group transfer besides protein synthesis. As the availability is limited and cost is high for synthetic Met, reductions in its inclusion in broiler diet may be possible by supplementing the low Met diets with methyl donors (MD) like betaine (Bet), folic acid (FA), vitamin B12 (B12), and biotin (Bio). An experiment was conducted to study the effects of supplementing the MD on performance (average daily gain [ADG], daily feed intake, feed efficiency [FE]), anti-oxidant variables, immune responses and serum protein concentration in broilers fed sub-optimal concentrations of dietary Met. Methods: Maize-soybean meal diet was used as control (CD). Different MD like Bet (0.2%), B12 (0.1 mg), FA (4 mg), or Bio (1.5 mg/kg) were supplemented to basal diet (BD) having no supplemental Met. The BD without MD was kept for comparison. Each diet was fed ad libitum to 10 replicates of 25 chicks in each from 1 to 42 d of age. Results: At the end of experiment, the ADG in MD group was higher than BD and lower than CD. The FE improved with FA or Bet compared to the BD. Breast meat weight was higher in Bet compared to the BD, while it was intermediate between BD and CD in other groups. The lipid peroxidation reduced with Bio, B12, or Bet, while the glutathione peroxidase activity improved with Bio or B12 compared to the BD. Lymphocyte proliferation improved with Bet compared to the BD. The serum protein concentrations increased with FA, Bio, or Bet compared to those fed BD. Conclusion: It can be concluded that the ADG can be improved partially with supplementation of MD while the FE improved with FA or Bet. Some MD also reduced the stress indices and improved immune responses compared to the BD fed broilers.

Influence of a Single Dose of Fe Dextran Administration with Organic Trace Mineral Supplementation on the Performance of Piglets

  • This study was designed to evaluate the influence of a single or double dose of Fe dextran with organic trace mineral supplementation on the performance of piglets from dams fed diets with either inorganic (ITM) or organic trace minerals (OTM). It also determined the effect of the source of the trace minerals on the reproductive performance of sows. The trace mineral premixes were prepared using metal proteinates and the corresponding inorganic salts for the OTM and the ITM, respectively. Each mineral premix provided 100 ppm Fe/175 ppm Fe, 35 ppm Cu/170 ppm Cu, 90 ppm Zn/120 ppm Zn, and 40 ppm Mn/35 ppm Mn when added at 0.20% in sows /weaned pigs' diets, respectively. The first dose of Fe dextran was administered to piglets at 3 d and the second dose at 10 d after birth. One dose of Fe dextran supplied 100 mg of Fe. A total of 16 gestating sows (Landrace${\times}$Yorkshire) in parities 2 to 4 were randomly allocated to four treatments: 1) diet with ITM/one dose of Fe dextran to piglets, 2) diet with ITM/two doses of Fe dextran to piglets, 3) diet with OTM/one dose of Fe dextran to piglets, and 4) diet with OTM/two doses of Fe dextran to piglets. The total born alive, weaned, body weight at birth and at weaning were not affected by the sow's dietary treatment. Although organic trace mineral supplementation tended to increase the milk Fe content (p<0.10) at 7 d postpartum, piglets in all treatments performed equally from birth to weaning. The double doses of Fe dextran neither improved the average daily gain (ADG) nor influenced the survival of piglets from birth to weaning (21 d). Results suggest that a single dose of Fe dextran given to suckling pigs is adequate to sustain their needs for growth throughout the lactation period (21 d). Furthermore, there was a 21% improvement in both the ADG and the average daily feed intake (ADFI) (p<0.05) in weaned pigs fed diets with OTM. Cu and Fe in the liver (p<0.01), and Zn in both the bone (p<0.01) and the serum (p<0.01) were higher in piglets fed OTM than in those fed ITM. It would be concluded that single dose of Fe dextran administration with organic trace mineral supplementation show similar growth performance compared to 2 dose Fe dextran administration with inorganic mineral supplementation in young pigs.

Interaction between Iron and Vitamin A in Broilers

  • A $3{\times}3$ (Fe$\times$VA) experiment with repeats was designed to study the interaction between iron and vitamin A in broilers. 504 broilers were divided into 9 groups (50% males, 50% femals), each group with 4 repeats. Iron supplemental amount was 0, 30 and 60 mg/kg; Vitamin A supplemental amount was 750, 1,500 and 2,700 IU/kg. Iron concentration in liver, serum, tibia and duodenum and vitamin A concentration in liver and serum were measured, and erythrocyte count was also observed. Results showed with the increase of dietary supplemental iron levels, vitamin A concentration in liver significantly decreased lineally (p<0.05) (0.861, 0.671, 0.639 mg/100 g at the end of 4th week; 0.900, 0.765, 0.739 mg/100 g at the end of 7th week), and vitamin A concentration in serum significantly increased lineally (p<0.05) (82.725, 97.842, $109.475{\mu}g/100mL$ at the end of 4th week; 62.288, 91.900, $95.117{\mu}g/100mL$ at the end of 7th week), meaning iron could promote the mobilization of vitamin A from liver to serum. With the increase of dietary supplemental vitamin A levels, liver iron concentration decreased and serum iron concentration increased, vitamin A could promote the mobilization of iron from liver to blood. Iron concentration in Duodenum and tibia and erythrocyte count increased significantly with higher dietary vitamin A supplementation (p<0.01), vitamin A could promote iron absorption, iron mobilization from liver to target tissues and erythropoiesis. Effects of the interaction between iron and vitamin A on vitamin A concentrations in liver and serum, iron concentration in tibia and erythrocyte count were significant (p<0.05).

흡연자와 비흡연자의 영양 섭취상태와 혈중 무기질 함량 비교 (Comparison of Nutrient Intakes and Serum Mineral Levels between Smokers and Non-Smokers)

  • This study was conducted to investigate the effects of smoking on nutrient intake and blood mineral status. The subjects were composed of two groups.55 smokers and 52 non-smokers. A 24-hour recall method was used along with questionnaires and serum mineral levels were analyzed by ICP spectrometer. The average ages of the smokers and non-smokers were 55.5 and 59.3 years old, respectively. The height, obesity degree, BMI, and WHR of the smokers were significantly higher than those of the non-smokers (p < 0.05, p < 0.05, p < 0.05, p < 0.01) Approximately 45.5% of the smokers smoked 16-20 cigarettes per day. The average age that the smokers started smoking was 22.0 years old and their smoking history was 33.5 years. About 74.5% of the smokers drank alcoholic beverages, while 44.2% of the non-smokers did. The smokers tended to eat less meals and breakfast meal, but drink coffee more often compared to the non-smokers. The mean daily energy intake and CPF energy intake ratio were 1231.8 ㎉ and 69.8 : 14.8 : 14.7 in the smokers and 1210.2 ㎉ and 72.1 : 14.7 12.7 in the non-smokers, respectively. The results show that the smokers tended to consume more energy, lipid, and cholesterol compared to the non-smokers. The results also show that in both groups, nutrient intake was lower than the RDA. The two groups were not significantly different in terms of the intake frequency of green-yellow vegetables and fresh fruits. There were no significant differences in serum levels of Ca, P, Mg, Cu, Fe, Mn, and Zn. However, serum Se level of the smokers was significantly higher than that of the non-smokers. In conclusion, the subjects of this study showed a serious imbalance in the nutrient intake, and the smokers showed a more undesirable dietary intake in the light of their high intake of energy, lipid, cholesterol, alcoholic beverages, and coffee. The serum Se level of the smokers was higher than that of the non-smokers, showing that Se is involved in smoking, Therefore, it could be suggested that more systematic research be conducted with respect to Se and smoking and that increased nutrition education and guidelines for smokers are required.

Effects of Liquid Culture of Agaricus blazei Murill on Protein, Mineral Levels and Enzyme Activities in Rats

  • This study was performed to investigate the effects of liquid culture of Agaricus blazei Murill on the weight gains, food efficiency ratios, serum protein and mineral levels, and serum enzyme activities in growing male rats. Sprague-Dawley rats (7 weeks old) were given four different types of diets for a succeeding period of 6 weeks, respectively: a normal diet group, a control diet group (normal diet + 15% lard + 0.5% cholesterol), a 30% or 40% A. blazei diet groups (control diet + 30% or 40% A. blazei in water) according to the levels of A. blazei supplementation. The body weight gains, food efficiency ratios, and the liver of the rats fed control diet, 30% or 40% A. blazei diets were significantly increased compared to rats fed the normal diet, but those of rats fed the 30% and 40% A. blazei diets were similar to those of rats fed the control diet. The concentrations of total protein, albumin, glucose, and hematocrit value in serum of rats fed the control diet, 30% or 40% A. blazei diets were similar to those of rats fed the normal diet. The concentrations of urea and creatinine in serum of rats fed the 30% or 40% A. blazei diets were similar to those of rats fed the control diet. but the urea of rats fed the 30% and 40% A. blazei diets were significantly decreased compared to rats fed the normal diet. The concentrations of Ca, P, Fe and Mg, and $Ca^{++}$, $K^+$, $Na^+$ and $Cl^-$ of rats fed the control diet, 30% or 40% A. blazei diets were similar to those of rats fed the normal diet. There were no differences in the activities of aspartate aminotransferase, alanine aminotransferase, $\gamma$-glutamyltranspeptidase and alkaline phosphatase in the serum among the experimental groups. In conclusion, the rats fed the A. blazei maintained normal protein and mineral levels, and enzyme activities of serum. But the A. blazei feeding could not decrease the body and liver weights in the rats fed high cholesterol diets.

  • The purpose of this study was to investigate the effect of nutritional status on the cell-mediated and humoral immunity in female college students. The nutritional status of twenty subjects was determined by six-days food records, anthropometric measurements, and biochemical assessments of serum nutrients. Cell-mediated and humoral immunity of the subjects was analyzed by in vivo and in vitro assessments. The results were summerized as follows : First, The average daily energy intake was 1437Kcal(CHO : PRO : FAT = 61:13:26), which corresponds to 71.9% of RDA. 폐경 전·후 여성들의 체위, 영양소 섭취 상태 및 혈중 지질과의 관련성에 관한 연구 (Correlation of Anthropometric Data, Nutrient Intakes and Serum Lipids in Premenopausal and Postmenopausal Women)

  • This study was conducted to investigate the anthropometric data, nutrient intakes and serum profiles in premenopausal and postmenopausal women living in Gyeonggi-do Province, Republic of Korea. The subjects were 49 premenopausal women and 63 postmenopausal women who are not taking any hormone or cardiovascular drugs. Anthropometric measurements were taken by a trained practitioner and the data for dietary intakes were obtained by a 24-hour recall method. Serum samples were collected and analyzed for the total cholesterol, triglyceride (TG) and lipoprotein fractions. The mean age of the premenopausal women was $45.17{\pm}3.28$ years and that of the postmenopausal women was 2$62.5{\pm}4.14$ years. The height and weight were $157.86{\pm}$4.35 cm, $58.75{\pm}6.01$ kg in premenopausal women and $156.42{\pm}3.62$ cm, $57.63{\pm}5.38$ kg in postmenopausal women, respectively. WHR (waist hip ratio) in postmenopausal women was significantly higher than that of premenopausal women (p<0.05). There were no differences between the pre-and postmenopausal women in the intakes of energy, protein, fat, Ca, Fe, vitamin A, vitamin $B_1$, vitamin $B_2$, niacin, vitamin C and vitamin E. However carbohydrate and Na intakes in postmenopausal women were significantly higher than those of premenopausal women. In postmenopausal women, Ca intake was below and Na intake was extremely high considering KDRIs (Dietary Reference Intakes for Koreans). Serum triglyceride in postmenopausal women was positively correlated with age, BMI (body mass index) and WHR. Serum total cholesterol and triglyceride in postmenopausal women showed significantly negative correlations with fiber intake. These results suggest that it is necessary to help postmenopausal women maintain a healthy body weight. Postmenopausal women needs to increase Ca (calcium) intakes and diet quality by decreasing the intakes of Na (sodium). In addition, an adequate intakes of fiber is recommended for postmenopausal women to prevent cardiovascular disease.

House dust mite-specific immunoglobulin E and longitudinal exhaled nitric oxide measurements in children with atopic asthma

  • Purpose: House dust mite (HDM) has been suggested to be the most important aeroallergen responsible for atopic asthma in Korea. We aimed to investigate that specific IgE antibodies to HDM and other common indoor aeroallergens contribute differently to total serum IgE and show different relationships with longitudinal fractional exhaled nitric oxide (FeNO) measurements in Korean atopic asthmatic patients. Methods: A total of 193 children aged 8 to 16 years with intermittent or mild persistent atopic asthma were recruited. Sera were assayed for total IgE and specific IgE antibodies to HDM and other common indoor allergens. FeNO was serially measured 10 times or more over 2 years when subjects were not receiving controller medications. Results: In 152 children who completed the study, IgE antibodies to specific HDM were more prevalent than those to other common indoor aeroallergens. In addition, IgE antibody titers to HDM were the strongest contributor to total IgE increases. Furthermore, only HDM-specific IgE antibody titer significantly correlated with maximum FeNO (r=0.21, P=0.029) and the rate of FeNO higher than 21 parts per billion (ppb) (r=0.30, P=0.002). Eight patients (5%) were found to have maximum FeNO of 21 ppb or less, suggesting the presence of a low FeNO phenotype among atopic asthmatic patients. Conclusion: The quantity of HDM-specific IgE antibody provides a possible explanation for increases of total IgE and significantly correlates with the amount and frequency of FeNO increases in Korean atopic asthmatic patients.

양전자 방출핵종 $^{68}$Ga을 이용한 NOTA와 DOTA의 표지 및 시험관내 특성 연구 (Radiolabeling of NOTA and DOTA with Positron Emitting $^{68}$Ga and Investigation of In Vitro Properties)

  • 목적: $^{68}$Ge/$^{68}$Ga-제너레이터에서 생산되는 PET용 방사성동위원소인 $^{68}$Ga을 NOTA와 DOTA에 표지하는 조건을 확립하고 이의 안정성 및 단백질 결합 특성을 연구하였고, 여러 가지의 금속이온 공존시 표지효율에 미치는 영향도 관찰하였다. 대상 및 방법: 여러 가지 농도의 NOTA 3HCl과 DOTA 4HCl에 $^{68}$Ge/$^{68}$Ga-제너레이터에서 0.1 M HCl로 용출한 $^{68}$GaCl$_3$ 6.66$\sim$272.8 MBq 1.0 mL와 합치고 초산나트륨 또는 탄산나트륨 완충액을 사용하여 다양한 pH 조건에서 반응하였다. 다양한 금속이온(CuCl$_2$, FeCl$_2$, InCl$_3$, FeCl$_3$), GaCl$_3$, MgCl$_2$, CaCl$_2$)과 0.373 mM NOTA를 $^{68}$Ga(6.77$\sim$8.58 MBq)으로 표지할 때 표지효율을 관찰하였다. $^{68}$Ga의 표지효율은 ITLC-SG고정상으로 하고 아세톤과 생리식염수를 이동상으로 하여 측정하였다. 최적의 pH 조건에서 $^{68}$Ga-NOTA와 $^{68}$Ga-DOTA를 표지한 후 4 시간 동안 안정성을 확인하고, 사람 혈청에서의 단백질 결합능을 평가하였으며, 지용성 정도를 측정하기 위하여 octhanol distribulion 실험을 실시하여 log P값을 구하였다. 결과: $^{68}$Ga-NOTA와 $^{68}$Ga-DOTA의 치적 표지 pH 조건은 각각 pH 6.5와 3.5였고, NOTA는 실온에서 표지가 잘 되었으나 DOTA는 가열이 필요하였다. MgCl$_2$와 CaCl$_2$의 존재는 $^{68}$Ga-NOTA의 표지 효율에 영향을 미치지 않았으나 CuCl$_2$, FeCl$_2$, InCl$_3$, FeCl$_3$, GaCl$_3$이 존재할 경우에는 표지효율이 감소하였다. $^{68}$Ga-NOTA와 $^{68}$Ga-DOTA는 실온에 그대로 두거나 사람혈청과 37$^{\circ}C$에 두었을 때 4 시간 이상 안정하였고, 사람 혈청 단백질 결합능은 2.04$\sim$3.32%로 낮았으며, log P 값은 -3.07로 수용성을 보였다. 결론: $^{68}$Ga의 표지에는 NOTA가 DOTA에 비하여 이상적인 양기능성 킬레이트제로 쓰일 수 있음을 알았다. 또한 $^{68}$Ga-NOTA는 금속이온 존재시 표지효율이 떨어질 수 있지만 안정하고 낮은 단백질 결합을 보였다.
