Establishment of a statistically reliable sampling method and size for serological surveillance of classical swine fever (CSF) in Korea (우리나라 돼지콜레라 항체 수준 측정을 위한 표본감사의 통계학적 기준 설정)

  • To establish a statistically reliable sampling strategy for serological surveillance of classical swinefever (CSF) in Korea, antibody test data from CSF surveillance conducted during year 2005 were analyzed.The most appropriate sampling method was determined to be stratified multi-stage random sampling strategy,in which the primary sampling unit is a pig farm and the secondary are the pigs by the strata of breedersand finishers in the selected farm. The optimum sample size was 5 to 19 including 1 to 2 breeders accordingto the number of pigs in the farm. The optimum sampling strategy demonstrated in this study was veryFindings of our study provide practical guidelines for surveillance of herd immunity level to CSF in Korea.

  • We investigated the serological prevalence of avian metapneumovirus (AMPV), avian reovirus (ARV), Mycoplasma gallisepticum (MG) and Mycoplasma synoviae (MS) in 760 broiler breeder (38 flocks), in the Jeonbuk province in 2013. This study was conducted to evaluate the immune and infection status of the broiler breeder flocks against AMPV, ARV, MG, MS by an enzyme-linked immunosorbent assay (ELISA). Serological test for AMPV were positive 37 (97.3%) flocks and 712 (93.6%) broiler breeder and geometric mean antibody titers were $16,350{\pm}10,195$, ARV were high positive rate 100% (38/38) flocks and 97.8% (743/760). The seropositive flocks against MG were 71.1% (27/38) and the geometric mean antibody titers were $2,474{\pm}2,045$, whereas the rates of positive flocks against MS were 50.0% (19/38) and the geometric mean antibody titers were $1,469{\pm}1,230$.

  • In a kenel of 62 dogs, 33 were diagnosed as infected dogs Brucella canis by serological test and blood cultures in a large kennel in Chonnam area. Twenty eight of 33 dogs were treated with combined antibiotics therapy consisting of tetracycline and dihydrostregtomycin. After the first treatment, all dogs became abacteremic and serologic titers declined. Abortion due to B canis infection could be prevented by antibiotic therapy during pregnancy. However, sequential antibiotics therapy for 4 weeks did not eradicate B canis from affected bitch. According to results of serological and blood culture tests, effect of antibiotics treatment respectively revealed that 17 of the originally infected dogs were cure with second therapy schedule at 2 months and 27 of that with 6 months after second therapy.

  • Nonalcoholic steatohepatitis(NASH) may progress to advanced liver disease. The diagnosis is made on liver biopsy when investigating a patient with raised transaminases and an otherwise negative biochemical and serological work-up. The subject was a obese male patient who had unexplained raised GOT, GPT. He had no alcoholic consumption and drug ingested. On serological examination, HBsAg and Anti-HCV test are negative. The subject was diagnosed as NASH, and was treated with oriental treatment for obesity. After 2months treatment the raised GOT, GPT decreased to normal range.

  • In the indeterminate chronic period of Chagas disease (ChD) the treatment has not been conclusive, because the serological negativization requires many years. This study aims to evaluate the efficacy of nifurtimox (NF) in the treatment of chronic ChD in prolonged follow-up by serological techniques of indirect immunofluorescence assay (IFA) and enzyme-linked immunosorbent assay (ELISA) IgG comparing 2 groups of patients, treated and non treated. Mann-Whitney test was performed for ELISA and IFA, with significant difference between the groups (P<0.05). IgG levels were lower in individuals treated compared with untreated patients, indicating chemotherapeutic efficacy in prolonged follow-up.

  • As animal welfare issue becomes important, the European Union bans conventional cages for laying hens from 2012. So the alternative housing systems like floor pens, aviaries or free range systems have been suggested. From 2011 to 2014, we monitored 20 welfare-oriented laying hen farms in South Korea to figure out serological status of major viral diseases. During this period, total 3,219 blood samples were collected from the randomly selected chickens to test and evaluate the hemagglutination inhibition titers for low pathogenic avian influenza, Newcastle disease and egg drop syndrome '76. A total of 2,926 blood samples were tested through enzyme linked immunosorbent assay (ELISA) to assess the serological status of infectious bronchitis (IB). The distribution of ELISA titers for IB was various from almost 0 to 20,000 through the all weeks of age. Also, the antibody coefficient of variation for most of the diseases in this study was higher than those of typical cage layers. As this study was the first surveillance for major avian viral diseases of the animal welfare-oriented farms in South Korea, the results obtained from this study will help to determine what information and resources are needed to maintain better biosecurity and to improve the health and welfare of laying hen flocks.

  • Throughout the studies the following experimental results were obtained and so are summarized here. 1) What caused tussah silkworms terrible disease broken ous all over the Korea in 1965, was examined to be “Inclusion body of virus” through microscope. 2) The examined inclusion bodies are easily stained as purple by seller's staining. 3) The proper speed of centrifugation for the purification of inclusion bodies is 1,000 r. p. m for 10 minutes. 4) It is possible, cleanly resulted, to attempt the “Rapid Agglatination Test & Complement Fixation test” with autigen of inclusion bodies. 5) The Anti-polyhedra rabbit serum from antigen of the dilution of 2${\times}$10$\^$6//l$m\ell$ polyhedra made the Ropid Agglutination test possible even with dilution of 1/8 infected pupa blood(antigen).

  • To investigate infection rates of Anaplasma spp in Red Blood Cell(RBC) of 209 goats and to detect the Anaplasma antibody in sera of 242 goats by Complement Fixation test (CF test), the present study was practiced. The obtained results were as follows : 1. Infection rates of Anaplasma spp in RBC of blood smears, stained by the Giemsa method, were seasonally 15.3% in spring, 20.0% in summer, 39.3% In fall, 16.9% in winter, respectively. 2. The positive rates of the Anaplasma antibody in sera of goats tested by CF test were seasonally 80.2% In spring, 79.0% in summer, 85.7% in fall, 62.0% In winter, respectively.

  • Twenty Four cases of calculus, which were collected feom the patients, were tested by a serological agglutinin absorption test and the following results were obtained. 1) In all observed xases, blood group specific substances were detected, and this results were agreed with blood group of observed patients. 2) Because the α titer of O-serum is somewhat higher than B, we have come to a conclusion that O type and B type results must be reexamined with much more weighted sample than other type cases.
