두종의 Bacillus thuringiensis 내독소단백질 유전자의 융합에 의한 발현 (Expression of Fusion Products of Insecticidal Crystal Protein Genes from Two Different Bacillus thuringiensis Strains)

  • Bacillus thruingiensis subsp. kurstaki HD-1의 내독소단백질 유전자인 CryIA(a)의 N-말단 부분과 B.tk HD-73의 내독소단백질 유전자인 CryIA(c)의 C-말단 부분의 융합에 따른 독성발현 여부에 관하여 조사하였다. 융합생산물인 pSK3, pSK4 및 pSK5의 플라스미드는 각각 4.5kb, 4.8kb 그리고 5.5kb로 구성되어 있다. 형질전환체의 내독소단백질에 대한 Western blotting은 SK4 및 SK5가 77-kDa 그리고 105-kDa에서 B.t k HD-1 항체에 대하여 반응을 나타내었다. 독성검정의 공시충인 배추좀나방 및 담배나방을 사용한 결과 pSK5를 포함하는 형질전환체만이 각각 96% 및 97%의 높은 치사율을 나타냈다.

Effects of silkworm and its by-products on muscle mass and exercise performance in ICR mice.

  • In this study, we investigated the effect of silkworm, and its by-products on exercise endurance, muscle mass, and fatigue recovery using ICR mice model. Powders of silkworm, pupae, dongchunghacho, and silk powder were suspended in water and feed to mice for 2 weeks. The forced swimming time was increased by 4.3, 4.1 and 2.8 seconds after silkworm, pupae, and dongchughacho administration compared to DW fed group. Increases in exercise ability were achieved by difference mechanism according to feeding materials. Pupae increased muscle mass by 129% compared with the control group which may effect on elongation of swimming time. Dongchunghacho improved the plasma concentrations of fatigue markers such as creatinine, blood urea nitrogen (BUN), and D-lactate. Silkworm administration showed dual effect, the muscle mass induction (114% vs. control) and anti-fatigue (plasma creatinine, BUN, and D-lactate were 63, 75, and 78% vs. con) effect which contributed most elongated swimming time. In conclusion, silkworm and its by-products including pupae and dongchunghacho with the predominant protein and bioactive components improved muscle mass and showed anti-fatigue effect which could promote exercise performance ability.

서양종 꿀벌(Apis mellifera L.) 수벌번데기 동결건조 분말의 영양학적 성분 및 항산화 효과 (Analysis of Nutritional Compounds and Antioxidant Effect of Freeze-Dried powder of the Honey Bee (Apis mellifera L.) Drone (Pupal stage))

  • 본 연구에서는 양봉산물로서의 가치 평가와 제품다양화를 위한 기초자료 제공을 위해 수벌번데기(16 ~ 20일령)의 영양성분분석 및 이를 이용한 추출물을 제조하고 생리활성 효과를 검정하였다. 수벌번데기 동결건조 분말의 일반성분 분석 결과 수분 1.69 ± 0.07%, 조단백 48.52 ± 0.20%, 조지방 23.41 ± 0.14%, 조회분 4.05 ± 0.02% 였고, vitamin C 14.92 ± 0.52 mg/100 g, vitamin E 6.06 ± 0.11 mg α-TE/100 g 이었으며, 무기성분은 K과 P의 함량(mg/100g)이 각각 1,349.13 ± 34.57 및 1,323.55 ± 43.85로 가장 높게 함유되어 있고, Ca과 Fe의 함량은 각각 55.43 ± 1.51 및 5.49 ± 0.19가 함유된 것으로 분석되었다. 수벌번데기 동결건조분말 중에 함유된 지방산은 포화지방산(g/100g total fatty acids)이 약 59.62, 불포화지방산이 약 40.38을 차지하였고, 고급지방산인 palmitic acid (C16:0)가 35.49 ± 0.08, 그리고 oleic acid (C18:1, n-9)가 35.91 ± 0.22로 다량 함유되어 있었다. 구성아미노산(g/100 g)은 총 38.99 ± 2.63이 함유된 것으로 분석되었고, 유리아미노산(mg/100 g)은 총 5,129.04 였으며, 이중 proline이 1,257.68, glutamic acid가 759.12로 가장 높았다. 수벌번데기 추출물의 DPPH radical-scavenging 활성(㎍/ml)은 증류수추출 시 0.8, 50% EtOH 추출 시 3.2, 70% EtOH 추출 시 6.4, 그리고 EtOH 추출 시 32 농도에서 약 90% 이상이었다. 이와 같은 결과는 항산화능에 기여하는 주요 화합물이 극성계 화합물임으로 추정되었으며, 저분자단백질 또는 유리형 아미노산일 가능성이 높은 것으로 추측되었다. 이상의 결과와 같이 수벌번데기는 우수한 식품원료로의 사용과 새로운 기능성 소재로의 활용을 기대할 수 있을 것으로 사료된다.

Bioconversion and growth performance of Hermetia illucens in single fruit by-products

  • As agricultural production increases due to population growth, agricultural by-products that are generated at all production stages increase commensurately. Black soldier fly (BSF) (Hermetia illucens) treatment has potential as an environmentally friendly process to combat the environmental pollution caused by agricultural by-products. This study examined the utilization of BSF larvae in the decomposition of the by-products of apples and mandarins, fruits commonly produced in South Korea. The BSF test larvae were fed apple pomace or mandarin waste, and the control larvae were fed calf feed. Larval weight and size were measured at 4-day intervals until larvae reached the pre-pupal stage. Larval development time, survival rate, and BSF fecundity rate were calculated for all three substrates. Waste reduction and bioconversion ratios were also calculated. The developmental time of larvae fed with apple pomace and mandarin waste was greater than that of the control larvae. The average weight of larvae fed with the fruit by-products was less than that of the control. There was no significant difference in the survival rate of BSF larvae or the fecundity rate of BSF between the substrates used in this study. BSF larvae decomposed 48.0% and 61.5% of apple pomace and mandarin waste, respectively. The bioconversion efficiency rates of BSF larvae fed with apple pomace and mandarin waste were 9.1% and 12.1%, respectively. These results indicated that decomposition of single fruit by-products by BSF larvae is an environmentally friendly and effective bioconversion process.

Analyzing the Nutritional Value of Protaetia brevitarsis Larvae Feeding on Coffee and Oyster Mushroom Cultivation By-products

  • This study aimed to validate the nutritional superiority and safety of fermented coffee byproducts (CB) and fermented oyster mushroom cultivation by-products (OMCB) as alternative food sources for Protaetia brevitarsis larvae. Thus, we conducted a comparative analysis of the nutrient composition of the food sources and developmental characteristics of the larvae. P. brevitarsis larvae have traditionally utilized various by-products as food sources and are able to efficiently utilize these. The analysis of the nutrient composition of the food sources indicated that, in comparison to control group fermented oak sawdust (FOS), the content of crude protein was 2.2 folds higher in larvae fed with OMCB and 3.2 folds higher in larvae fed with fermented 50% CB + 50% OMCB (CB + OMCB). Moreover, the cellulose content, utilized as a nutritional source for the larvae, was 1.3 folds higher in the CB + OMCB group than in the control group. Significantly, the weight of larvae fed with OMCB and CB + OMCB increased 1.7-4.2 folds compared to those fed with FOS. Additionally, the survival rate of larvae before the formation of pupal cells was over 90% in all groups. Therefore, it was concluded that CB and OMCB contain various nutrients without harmful composition and have a larval growth-promoting effect. Consequently, they are considered appropriate dietary materials for P. brevitarsis larvae. This study enhances our understanding of by-product usages by P. brevitarsis larvae and confirms their potential as sustainable food resources.

  • Biologically active porcine Interleukin-2(poIL-2) was produced from in vitro and in viva baculovirus expression system, namely the Autographa californica nuclear polyhedrosis virus (ACNPV)-cell culture system and the Hybrid nucler polthedrosis virus (HyNPV)-sillkworm larva system. The concentration of the recombinant poIL-2(rpoIL-2) in the larvae hemolymph was 1 to 3 mg/mL, which was about 7 to 20 times those of the cell culture systems. The level of this expression efficiency is equal to that with transgenic livestock, secretion products in milk.

  • The presence of considerable amount of enzymes of TCA cycle isocitrate dehydrogenase (ICDH-NADP+, EC1.1.1.42), $\alpha$-ketogluterate dehydrogenase ($\alpha$-KGD, EC1.2.4.2) and malate dehydrogenase (MDH, EC1.1.1.37) in fresh control and in vitro starved adult Isoparorchis hypselobagri establish the functional TCA cycle in this fluke. The major metabolic end products are pyruvate, lactate, oxaloacetate and malate. The ratio of oxaloacetate/malate assess that oxaloacetate is reduced to malate and in this fluke the reverse TCA cycle is active. The pyruvate/lactate ratio shows pyruvate is reduced to lactate and the fluke is homolactate farmenters.

Strategies of Caffeine Loading into Silk Fibroin Film for Weight Loss Patch

  • Caffeine is a thermogenic agent that can be used in weight loss products. In order to achieve a sustained release of caffeine, silk fibroin (SF) film was uses as carrier. It has been shown that the loading method of caffeine into SF film affected the uniform distribution of caffeine in the SF film. When caffeine was added directly into SF solution, gelation has been occurred immediately and prevented the uniform distribution of caffeine. On the other hand, caffeine was dissolved in methanol in order to load the caffeine in SF film and crystallize the SF film at the same time. However, due to the fast evaporation of methanol, caffeine was recrystallized on the surface of SF film rather than penetrating into the film. Finally, caffeine was loaded into pre-crystallized SF film and uniform distribution of caffeine could be achieved. There was an initial burst of caffeine during the first 15 min, but after that a sustained release was achieved.

Taxonomic Characteristics of Six Species of Entomopathogenic Fungi Isolated from the Silkworm, Bombyx mori

  • Six entomopathogenic fungus isolates, Beauveria bassiana J57A, Nomuraea rileyi J125A, Paecilomyces farinosus J3A, Paecilomyces fumosoroseus J50A, Metarhizium anisopliae J88, Aspergilius sp. J64A, causing muscardine disease and aspergillosis in the silkworm, Bombyx mori were investigated for their cultural and morphological characteristics (on PDA culture media within 14 days at $24^{\circ}C$). The results showed that they differ each other from the features of cultural characteristics (colony elevation, colony color, colony growth rate) or morphological characteristics (conidiogenous cell structure, phialides, conidia size and shape). Among cultural characteristics, colony color is the easiest recognizable character between isolates. The morphological characteristics of each fungal isolate correspond to the descriptions of current system of classification.

Ultrastructural studies on the neurosecretory cells in the pars intercerebralis of larvae of tasar silkworm, Antheraea mylitta (D) eco-race Bhandara (Lepidoptera: Saturniidae)

  • The cerebral neurosecretory cells (NSC) constitute four paired groups, medial (MNC), lateral (LNC-I, LNC-II) and posterior (PNC) in the brain of larvae of tasar silkworm Antheraea mylitta (D) Eco-race Bhandara. The MNC is the largest groups of peptidergic neurosecretory cells and are located in the pars intercerebralis region. The transmission electron microscopic (TEM), ultrastructure of the NSC confirmed the presence of mitochondria, endoplasmic reticulum, Golgi bodies, lysosomes and neurosecretory granules. The median neurosecretory cells shows secretory activity and release of secretory products, the neurosecretory granules (NSG) in the axons of NSC as well as the blood sinus.
