Protective Activity of Seolitae Chungkukjang Added with Green Tea against Cellular Oxidative Stress induced by AAPH

  • The protective activity of seolitae chungkukjang added with green tea against oxidative stress was investigated under the cellular systems using LLC-$PK_1$ cells. The treatment of 2,2'-azobis(2-aminopropane) dihydrochloride (AAPH) showed increase in lipid peroxidation, and decrease in endogenous antioxidant enzymes activity and cell viability. However, the methanol extract of seolitae chungkukjang inhibited lipid peroxidation by 58.3%, and increased cell viability up to more than 60%. In addition, it enhanced superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activities. Seolitae chungkukjang improved oxidative stress-induced cellular injury through the radical scavenging activities. In particular, the addition of green tea in seolitae chungkukjang showed stronger effect against oxidative stress induced by AAPH. The more addition of green tea resulted in the greater antioxidative effect through elevation in activities of SOD and GSH-Px, and inhibition of lipid peroxidation, eventually leading to increase in cell viability. Theses results suggested that seolitae chungkukjang added with green tea have protective effects from cellular oxidative damage and could be considered as an application for the development of chungkukjang with functionality.

녹차 첨가 서리태 청국장의 항산화 증진 효과 (Increased antioxidative effects of Seolitae Chungkukjang added with green tea powders)

  • To increase antioxidative activity and minimize off-flavor of Chungkukjang, Seoliate as main ingredient and green tea as minor ingredient were selected. The protective efffect of green tea for Chungkukjang was evaluated under in vitro. Seolitae Chungkukjang added with green tea (SCG) was prepared on the basis of the protective effect of green tea from oxidative stress. Green tea showed the strong protective effect against nitric oxide (NO) and superoxide anion ($O_2{^-}$) radicals. The SCG perparation method was as follows. Seolitae was cleaned and washed, soaked in 1.5-fold water ($15^{\circ}C$) of Seolitae weight for 48 hr. Seolitae was steamed about 40 min by autoclave and then cooled about $40^{\circ}C$. Bacillus subtilis was inoculated and 5.0-fold green tea of weight was added, and then it was fermented for 60 hr at $40^{\circ}C$. According to the preparation method, Seolitae Chungkukjang (SC) and Seolitae Chungkukjang added with green tea 5.0% (SCG5.0) were prepared. Among the Chungkukjang groups SCG5.0 exerted more effective scavenging activity of NO than SC. In addition, SC added with green tea 5.0% showed high $O_2{^-}$ scavenging effect. These results suggest the antioxidative potential against free radical-induced oxidative damage of SCG.

Established marginal seeding period for green cotyledon black soybean (Seoritae) cultivation in Chungbuk province of Korea

  • Seoritae with green cotyledons refers to Korean native black beans harvested when the frost falls because the maturity is late. It is one of the beans preferred by consumers because of its softness and high sugar content. Because of late maturity, if the agricultural work is delayed by weather or agricultural schedule in green cotyledon black soybean (Seoritae), it affects seriously harvest and yield of soybeans. The aim of this study was to investigate the marginal seeding period on June 30, July 10 and July 20 in Cheongju and Jecheon area in Korea to produce stable soybean yield. The yields of green cotyledon black soybean as seeding date in Cheongju area are as follows: Seolitae (Yeoncheon), Seoltae (Goesan) and Seoritae (Gogseong), which increased by 23%, 56%, 23% and 40%, respectively, compare to July 10th to June 30th. As soybean sowing is delayed, the quality of soybean seeds has decreased due to the increase of immature seeds and fungal damaged seeds. The contents of anthocyanin in Cheongju area a functional substance of soybeans, was high on July 20 for Heukcheong, on June 30 for Seolitae (Yeoncheon), on July 10 for Seoritae (Goesan). The yields of Heukcheong and Seolitae (Yeoncheon) in Jecheon area were increased by 5% and 17%, respectively, compare to July 10th to June 30th, while Seolite (Goesan) and Seolite (Gogseong) were high in yields on June 30th. Similarly in Cheongju area, as the sowing period is delayed, the number of immature and mold damaged seeds in Jecheon area increased. The contents of anthocyanin in Jecheon area was high on July 10 for Heukcheong, on June 30 for Seolitae (Yeoncheon) and Seolitae (Goesan). From the above results, Sowing marginal date of green cotyledon black soybeans (Seolitae) in Cheongju area increased 30% in sowing on July 10 and increased 2% in sowing on July 20 compared to June 30. And Sowing marginal date of green cotyledon black soybeans (Seolitae) in Jecheon area increased 2% in July 10 compared to June 30, and the yield decreased rapidly on July 20. We have identified the seeding time limit of green cotyledon black bean in Chungbuk province. It will be possible to provide a variety of crop selection after double cropping of farmers. And by knowing the yield and seed quality of soybean according to sowing date, farmers will observe appropriate sowing period of soybeans for high quality. From the viewpoint of consumers, functional substances of Seoritae will meet the desire for health.

녹차첨가 서리태청국장의 이화학적 특성 및 유리기 소거능 (Radical Scavenging Effects and Physicochemical Properties of Seolitae Chungkukjang Added with Green Tea)

  • 본 연구에서는 청국장의 기능성과 관능적 품질개선을 위한 연구의 일환으로 서리태청국장에 녹차를 0.5%, 2.0%, 5.0% 첨가(w/w)하여 청국장을 제조한 후 특성을 비교하고 항산화능을 검토하였다. 녹차 5% 첨가한 서리태청국장이 in vitro상에서 DPPH와 ${\cdot}OH$ 소거능이 우수하였고 관능검사 결과는 향, 맛, 종합적 기호도가 일반청국장과 유사하게 높게 나타났다. 이상의 결과를 종합하여 볼 때 녹차 5% 첨가 서리태청국장이 청국장의 관능적 품질이 뛰어날 뿐만 아니라 항산화활성이 가장 높은 것으로 평가되었다.

시판되는 밥밑콩류의 이소플라본 함량, 항산화활성 및 혈전용해활성 (Isoflavone Contents, Antioxidative and Fibrinolytic Activities of Some Commercial Cooking-with-Rice Soybeans)

  • 우리나라 자생콩류중 주로 밥밑콩으로 이용되는 7품종(강남콩, 거두, 서리태, 선비콩, 울타리콩, 청태, 황태)의 isoflavone 함량과 일부 항산화 및 혈전용해활성을 조사하였다. Isoflavone 함량 분석 결과, 서리태에서는 daidzein과 genistein 모두, 강남콩과 거두에서는 daidzein이 검출되지 않았다. 청태의 daidzein 및 genistein 함량은 각각 278 mg/kg과 223 mg/kg이었으며, 황태중에는 총 1552 mg/kg으로 대두와 비교할 만한 정도였다. 청태와 황태의 경우 daizein/genistein의 비율이 1.25와 1.42로서 daidzein이 많은 것으로 나타났다. 전자공여능은 울타리콩, 황태, 강남콩이 90% 이상으로 가장 높았고, 거두와 서리태 역시 황태와 강남콩과 유의적 차이를 보이지 않았으며, 선비콩과 청태의 경우 각각 57%와 50%로 유의적으로 낮았다(p<0.001). 울타리콩과 강남콩, 거두는 SOD 유사활성도 다른 종류보다 유의적으로 높았다. 전자공여능이 컸던 황태와 서리태는 SOD 유사활성이 낮았고, 전자공여능이 낮았던 선비콩은 황태나 서리태보다 유의적으로 높은 활성을 보였다. 형전용해활성은 선비콩, 황태, 강남콩 등이 비교적 컸고, 청태는 활성을 보이지 않았다. 혈전용해효소의 저해성분을 부분적으로 제거하기 위해 추출액의 pH를 6.4로 조절한 후 18시간 동안 냉장처리한 결과 대부분의 품종에서 활성이 뚜렷이 증가하였다(비활성도 값으로 비교시 거두, 서리태, 울타리콩류는 $10{\sim}15$배, 강남콩은 2배 내외, 선비콩과 황태는 $18{\sim}19%$ 증가). pH를 3.0으로 조정시 비활성도가 선비콩과 황태, 청태, 강남콩에서는 크게 증가한 반면, pH 6.4 조절 및 냉장처리시 혈전용해활성이 컸던 거두, 서리태 및 울타리콩에서는 실활되었다. 활성을 보이지 않던 청태는 $55^{\circ}C$에서 30분간 열처리하였을 때 약한 활성을 나타내었으며, $100^{\circ}C$로 처리시 실활되었다. 거두와 서리태, 선비콩, 황태, 강남콩은 $55^{\circ}C$에서 30분 및 $100^{\circ}C$에서 10분의 열처리로 활성이 점차 증가하였으나 $100^{\circ}C$에서 30분 처리시 활성을 대부분 혹은 완전 상실하였다. 울타리콩의 혈전용해활성은 $100^{\circ}C$에서 30분간 가열시에도 높은 비활성도를 유지하여 내열성이 매우 큰 것으로 나타났다.
