  • New powder compaction process, in which a Bingham semi-solid/fluid mold is utilized, is developed to fabricate micro parts. In the present process, a powder material is filled as slurry in a solid wax mold, dried and compressed. The wax is heated during compaction and becomes semi-solid state, which can acts as a pressurized medium for isostatic compaction. Since the compacted micro parts are very fragile, the mold's temperature is controlled to higher than its melting point during unloading, to avoid breakage of the compacts. To demonstrate effectiveness of this process, some micro compacts of alumina are shown as examples.

CLSI M40-A2 기준에 따른 수송배지의 흡수 및 배출, 생존 효율, 회수율 평가 (Evaluation of Absorption and Release, Survival Efficiency and Recovery Rate of Transport Medium according to the CLSI M40-A2 Standard)

  • 5종류의 수송배지를 흡수 및 배출 능력, 생존 효율, 회수율을 CLSI (Clinical and Laboratory Standards Institute) M40-A2의 Swab elution method (Quantitative) 기준으로 평가하였다. 액체배지가 반유동 배지보다 3가지 평가에서 대부분 우수한 결과값을 보여주었다. Flocked swab이 standard swab 형태보다 균의 흡수 및 배출의 능력 또한 우수하였다. 생존 효율에 대한 평가 결과는 액체배지(S4)가 가장 우수한 결과값을 보였다. 성장이 좋지 않은 S. pneumoniae는 액체배지(S4, S5)에서 생존효율과 회수율이 높았다. 균 회수율 평가 결과는 S. pyogenes는 모든 배지에서 CLSI 기준에 적합하였다. S. pneumoniae는 반유동 배지(S2, S3)에서 부적합하였고, 나머지 배지는 모두 기준에 적합하였다. H. influenzae는 반유동 배지(S1, S3)에서 부적합하였고, 반유동 배지(S2), 액체배지(S4, S5)에서 기준에 적합하였다. 호흡기 질환을 유발하는 S. pneumonia, H. influenzae의 생존 능력은 대부분의 배지에서 좋지 않았다. P. aeruginosa는 실온에서 과성장이 관찰되었다. 액체배지와 flocked swab의 조합이 3가지 평가 방법에서 가장 뛰어난 결과를 평가를 통해 확인하였다.

The Effect of Cultural Factors on Anther Culture in Spring Wheat (Triticum aestivum)

  • Effects of shaking, medium consistency and anther density on polyhaploid production in two wheat cultivars, Pavon and Chris, were studied using a modified 85D12 medium. Pavon produced more calli in shaking and more albino plants tban Chris. However, Chris produced threefold more green plants than Pavon in non-shaking treatment. More calli and green plants were derived from non-shaking treatment than those from shaking treatment. Anthers were cultured on both liquid and semi-solid 85D12 media, using two anther densities, 48 and 96 anthers per plate. Although Pavon generally produced more calli and albino plants than Chris, Chris produced more green plants than Pavon. More green plants were derived from semi-solid medium than those from liquid medium. A factor that may affect plant regeneration from anthers is the length of time on initiation medium. Most of the calli for both genotypes were transferred during the first two time periods. Fertility, as measured by seed set, was determined for all surviving regenerated plants. About 24% of Chris and Pavon anther-derived green plants in the experiment of medium consistency and anther density produced seed.

액상 및 반고체배지 발효에 의한 Bacillus thuringiensis 살충제의 제조 (Formulations of Bacillus thuringiensis Insecticides by Liquid and Semi-Solid Fermentations.)

  • Microbial insecticide formulations were prepared by liquid and semi-solid fermentations using Bacillus thuringiensis subsp. kurstaki, HL-106 (BTK-HL106), B. thuringiensis subsp. israelensis HL-63 (BTI-HL63) and B. sphaericus 1593 (BS-1593) strains. The liquid fermentation medium contained molasses 2%, dextrose 1.5%, peptone 2%, D-xylose 0.025%, CaCl$_2$ 0.1%, K$_2$HPO$_4$ 0.1%, KH$_2$PO$_4$ 0.1%, MgSO$_4$$.$7H$_2$O 0.03%, FeSO$_4$$.$7H$_2$O 0.002%, ZnSO$_4$$.$7H$_2$O 0.02%. The composition of the semi-solid fermentation medium was rice bran 45.2%, zeolite 31%, yeast powder 0.02%, corn powder 5%, dextrose 3%, lime 0.3%, NaCl 0.06%, CaCl$_2$ 0.02%, and H$_2$O 15.42%. Insecticide formulations produced in the liquid fermentation named BTK-HL106, BTI-HL63 and BS-1593 pesticides and those in the semi-solid fermentation were designated as BTK-HL106-1, BTI-HL63-1 and BS-1593-1 pesticides, respectively. The number of spore (endotoxin crystals) was 2.65${\times}$10$\^$9/ spores per $m\ell$ in the BTK-HL106 and 3.5${\times}$10$\^$10/ in the BTK-HL106-1 3.8${\times}$10$\^$9/ spores in the BTI-HL63 and 7.0${\times}$10$\^$10/ in the BTI-HL63-1, and 7.5${\times}$10$\^$9/ in the BS-1593 and 1.4${\times}$10$\^$10/ in the BS-1593-1. The spores in the BS-1593 formulation was produced two times more than the other formulations. The spores in the BTI-HL63-1 were contained twice than those in the BTK-HL106-1, and five times than those in the BS-1593-1. The results indicated that spore (endotoxin crystals) productions in the semi-solid fermentation increased about ten times than those in the liquid fermentations. $LC_{50}$s of the BTI-HL63 and BS-1593 were 4.5 $\mu\textrm{g}$, and those of the BTI-HL63-1 and BS-1593-1 were 1.5 $\mu\textrm{g}$. $LC_{50}$ of the BTK-HL106 was 1.5 mg and that of the BTK-HL106-1 was 0.9 mg. The $LC_{50}$s of the formulations in the semi-solid fermentations showed about two to three times higher than those in the liquid fermentations.

  • A relationship between stress and stain is very important to design a die to avoid defects of products during semi-solid forming process. Since the liquid will be of eutectic composition in alloys liquid segregation will result in significant or undesirable situation. The materials used in this experiment are A 357. A390, Al2024 alloys that is fabricated by the electro-magnetic stirring process from Pechiney in France. The compression test was performed by induction heating equipment and MTS. In order to prevent the liquid segregation these measured temperature would be useful to control of strain rate during compression test. The liquid segregation is controlled as change of the strain rate and solid fraction during the compression process, The characteristics of flow between solid and liquid phase considering liquid segregation is examined through the above experiments. In the case of medium and high volume fractions of solid the distribution of strain rate is calculated by using compression test data of semi-solid materials (SSM). The thixoforming experiments with the designed die are carried out successfully. The die filling patterns of SSM for variation of die temperature and pressing force have been investigated. The hardness of the thixoformed scroll products is evaluated in terms of the microstructure for each position.

알팔파(Medicago sativa L.)의 원형질체로부터 다경 유도와 식물체의 구분화 (Induction of Multi Shoots and Plant Regeneration From Protoplasts of Alfalfa(Medicago sativa L.))

  • A system was established for induction of multi-shoots and plant regeneration from mesophyll protoplasts of alfalfa, Medicago sativa L. cv. Vernal. Different hormonal effects were tested at each step of protoplast culture, i.e. cell division in modified Kao's liquid medium (K566-7). calli formation on SH semi solid medium, and multi-shoot regeneration from calli on SHa and SHb solid media. Frequency of multi-shoots and plant regeneration was affected by various combinations of phytohormones in final step. The evaluation of multi-shoots induction systems via protoplast culture was discused.

현사시나무 기내배양(器內培養) 엽육조직(葉肉組織)에서 분리(分離)된 원형질체(原形質体) 배양(培養) 및 식물체(植物体) 재분화(再分化) (Culture and Regeneration of Populus alba × glandulosa Leaf Protoplasts Isolated from in vitro Cultured Explant)

  • 현사시(Populus alba ${\times}$ glandulosa) 기내배양(器內培養)한 엽육조직(葉肉組織)에서 원형질체(原形質體)를 분리(分離), 배양(培養)하여 식물체(植物體) 재분화(再分化) 과정(過程)에 관(關)해 조사(調査) 검토(檢討)하였다. $NH_4NO_3$를 뺀 MS배지(培地)에 BAP 0.5mg/l와 2.4-D 2.0mg/l를 첨가(添加)하여 liquid plating 법(法)으로 원형질체(原形質體)를 배양(培養)하였을때 비교적(比較的) 높은 세포분열(細胞分裂)이 일어났다. colony 형성율(形成率)은 gauze를 넣은 semi-solid agar plating 법(法)에서 가장 높게 일어나서 배양(培養) 5주후(週後)에는 micro-callus가 형성(形成)되었다. gauze를 제거(除去)한 후(後)에는 원형질체(原形質體) 분리배양후(分離培養後) 8주(週)가 되었을때 mini-callus가 형성(形成)되었으며 이들 callus는 2, 4-D 0.5mg/l와 BAP 0.1mg/l를 첨가(添加)한 MS배지에서 증식되었다. shoot 형성(形成)은 zeatin 1.0mg/l를 첨가(添加)한 배지에서 이식한지 3주(週)만에 일어나기 시작했으며 6주후(週後)에 뿌리발달을 도모하고져 생장조절물질(生長調節物質)이 첨가(添加)되지 않은 1/2MS배지로 이식하였다. 이식한지 4주후(週後)에 많은 뿌리가 형성(形成)되었다. 발근(發根)이 된 개체(個體)는 온실(温室)에 이식(移植)하였는데 원형질체(原形質體)를 분리(分離)하여 완전식물체(完全植物體)로 분화(分化)시켜 온실(温室)에 이식(移植)하기 까지는 22주(週)가 걸렸다.

  • Rotor in small-medium induction motor has been usually manufactured by aluminum diecasting. In order to improve the efficiency of induction motors, newly developed Cu-Ca alloys have been investigated. The electrical conductivity in the Cu alloys containing Ca less than $1.0wt\%$ was higher than $80\%$ IACS. Cu-Ca alloy is desirable for the thixoforming process because it has wide semi-solid range over $150^{\circ}C$. In this study, Cu-rotor with thixoforming process was developed to replace the conventional aluminum diecasting rotor. Analysis was performed for the microstructure of thixoforming rotor. Effect of incomplete filling on the efficiency of induction motor was discussed.

  • 고상 발효의 기초적인 애커니즘을 규명하기 위하여 고행화된 배지속에 8앙 효모을 증식시켰마. 이때 실험 배지는 Maxon ] Johnson배지 이었으며, 고형화 물질로는 갈근에서 얻은 전분을 푸깨비로 6-12%를 첨가하였다. 전분이 6-8% 첨가 될때 보 다도 10←12% 첨가될 때 효모의 증식이 8-24시간에서 훨씬 빨랐으며, 단백잘의 분바양도 전분을 10% 첨가 하었을때 10-24시 간에서 가장 높았다.
