KINETICS OF AUTOTROPHIC DENITRIFICATION FOR THE BIOFILM FORMED ON SULFUR PARTICLES : Evaluation of Molecular Technique on Monitoring Biomass Growth

  • Characteristics of sulfur-based autotrophic denitrification in a semi-continuous type reactor and the kinetic parameters were studied. Enriched autotrophic denitrifying culture was used for the reactor operation. Biomass growth on sulfur particles and in the liquid medium was monitored using the DAPI staining method. From the result of ion concentration changes and the biomass growth, maximum specific growth rate, ${\mu}_{max}$, and the half velocity constant, $K_M$, were estimated as $0.61\;d^{-1}$ and 3.66 mg/L, respectively. Growth yield coefficient, Y values for electron acceptor and donor were found as 0.49 gVSS/g N and 0.16 gVSS/g S. The biomass showed specific denitrification rate, ranging 0.86-1.13 gN/g VSS-d. A half-order equation was found to best simulate the denitrification process in the packed bed reactor operated in the semi-continuous mode.

  • Butyl acetate is produced from acetic acid and butanol via an esterification reaction in a reactive distillation column. Entrainer can be used for efficient removal of produced water from the reaction region, leading to significant improvement of the column performance. Batch reactive distillation has clear advantages over continuous one in terms of flexibility and adaptability in a small plant. We studied batch and semi-batch reactive distillation processes through process simulation and pilot-scale experiments. We investigated process configuration and type of entrainer for improvement of the column performance and suggested a novel cyclic operation strategy using the semi-batch reactive distillation column. The cyclic strategy was shown to give relatively high production rate and stable operation.

  • The above described semi-transparent monochromators have been operational since 1997. Four units are permanently operating at the ESRF beam line ID14. Two units are in continuous operation at ID 10 and one unit is installed at the APS beamline 8-ID in USA. The water cooling of the crystals is currently being revised and above we showed that improvements most likely are possible by using micro fluidics techniques. Further tests will be performed in collaboration with the CEA-France and tested at ESRF. Parallel developments including nanofluids as coolants are under evaluation. Combination of nanofluidics and microfluidics cooling devices are under study. The authors are grateful to C. Gillot and J-A.   • This study is intended to provide navigator with specific information necessary to assist the avoidance of collision and the operation of ships to evaluate the maneuverability of dead weight tonnage 8,000 tons Oil/Chemical tanker. The actual maneuvering characteristics of ship can be adequately judged from the results of typical ship trials. Author carried out sea trials based full scale for turning test in ballast condition and full load condition, semi balanced rudder and flap rudder. The turning circle maneuvering were performed on the starboard and port sides with $35^{\circ}$ rudder angle at the normal continuous rating. The results from tests could be compared directly with the standards of maneuverability of IMO and consequently the maneuvering qualities of the ship is full satisfied with its.

반연속 흐름 2단 토양 컬럼에서의 사염화 에틸렌(PCE)의 혐기성 완전탈염소화 환원 생분해

  • Anaerobic reductive dechlorination of tetrachloroethylene(PCE) to ethylene was investigated by performing laboratory experiments using semi-continuous flow two-in-series soil columns. The columns were packed with soils obtained from TCE-contaminated site in Korea. Site ground water containing lactate(as electron donor and/or carbon source) and PCE was pumped into the soil columns. During the first operation with a period of 50 days, injected mass ratio of lactate and PCE was 620:1 and incomplete reductive dechlorination of PCE to cis-DCE was observed in the columns. However, complete dechlorination of PCE to ethylene was observed when the mass ratio increased to 5,050:1 in the second operation, suggesting that the electron donor might be limited during the first operation period. During the degradation of cis-DCE to ethylene, the concentration of hydrogen was $22{\sim}29mM$. These positive results indicate that the TCE-contaminated groundwater investigated in this study could be remediated through biological anaerobic reductive dechlorination processes.

  • This paper describes the implementation and the evaluation of the speech recognition automatic exchange system. The system provides government or public offices, companies, educational institutions that are composed of large number of members and parts with exchange service using speech recognition technology. The recognizer of the system is a Speaker-Independent, Isolated-word, Flexible-Vocabulary recognizer based on SCHMM(Semi-Continuous Hidden Markov Model). For real-time implementation, DSP TMS320C32 made in Texas Instrument Inc. is used. The system operating terminal including the diagnosis of speech recognition DSP and the alternation of speech recognition candidates makes operation easy. In this experiment, 8 speakers pronounced words of 1,300 vocabulary related to automatic exchange system over wire telephone network and the recognition system achieved 91.5% of word accuracy.

  • As a basic studies for the laboratory scale production of alfalfa inoculum, Rhizobium meliloti M14 was characterized for its carbon and nitrogen sources, and some parameters for broth cultivation in a chemostat were studied by semi-continuous operation. The result s obtained were as follows. 1. Growth rate of the strain was increased by disaccharides than by monosaccharides tested, and pentoses resulted in poor growth than hexoses. Sugar alcohols including inositol supported the best growth among sugars. 2. Mannitol in the yeast-mannitol-broth was substituted by natural carbon sources such as malt extract or molasses. 3. Ten per cent of fresh yeast water appeared to supply enough amount of growth factor s for the strain, and the effect was equivalent to 0.24 percent of the commercial yeast extract powder. 4. Batch growth of the stain in a chemostat, New Brunswick Micro Ferm 28L, reached in the early stationary growth phase of $5{\sim}7{\times}10^9cells/ml$ after 36 hours of incubation. The culture at this stage was switched to semi-continuous cultivation, and the culture broth of four-fifth of the working volume was recovered every 24 hours when the maximal count was obtained.

Anaerobic Reductive Dechlorination of Tetrachloroethylene (PCE) in Two-in-series Semi-continuous Soil Columns (반연속 흐름 2단 토양 컬럼에서의 사염화 에틸렌(PCE)의 혐기성 환원탈염소화)

Dechlorination rate of PCE to cis-DCE was $0.62{\sim}1.94\;{\mu}mol$ PCE/L pore volume/d and $2.76\;{\mu}mol$ cis-DCE/ L pore volume/d for that for cis-DCE to ethylene, resulting that net dechlorination rate in the system was 1.43 umol PCE/L pore volume/d. During the degradation of cis-DCE to ethylene, the concentration of hydrogen in column groundwater was $22{\sim}29\;mM$ and $10{\sim}64\;mM$ for the degradation of PCE to cis-DCE. These positive results indicate that the TCE-contaminated groundwater investigated in this study could be remediated through in-situ biological anaerobic reductive dechlorination processes.

Production of Sorbitol Using Dried and Immobilized Zymomonas mobilis (건조 고정화 Zymomonas mobilis에 의한 sorbitol 생산)

  • The purpose of this study is to develop a continuous process for sorbitol production using dried Zymomonas mobilis immobilized in K-carrageenan. The methods of glularaldehyde cross-linking of enzymes in CTAB (celyltrimetylammoniumbromide) treated cells immobilized in K-carrageenan showed stability for the production of sorbitol for 30 days of operation. K-carrageenan beads entrapping permeabilized cells were dried to Improve bead rigidity and storage stability. A semi-batch process with dry beads was carried out and only a small loss of enzyme activity (less than 8%) was observed during 72h. The value of Vmax for the dry K-carrageenan beads was found to be one half or that for free cells. It was shown that the productivities of the continuous process with wet K-carrageenan beads and dry beads at a dilution rate 0.1h-1 were 3.4g/L-h and 2.88h/L-h, respectively.

A Feasibility Study on Biogas Production from Anaerobic Digestion of Straw (볏짚의 혐기성소화시 소화가스 생성에 관한 연구)

  • Quantity and composition of biogas from semi-continuous anaerobic digestion of straw were obtained experimentally in laboratory scale digesters fed with 1 liter of 5% straw-water mixture and maintained at 35$^{\circ}C$. Experiments were carried out for hydraulic retention time(HRT) of 8, 10 and 15days, respectively. The amount and composition of biogas produced were measured until steady-state was achieved for each run. The amount of biogas and methane percent go through a maximum and decrease continuously towards the steady-state after three times operation of hydraulic retention time(HRT). Methane gas production rates at steady-state increase with the increasing of HRT. Biogas production of 0.45 liter/day with 25% methane, 0.42 liter/day with 33.7% methane and 0.492 liter/day with 31.7% methane were obtained for 8, 10 and 15days of HRT, respectively. The high proportion of soluble carbohydrates present in straw makes the volatile fatty acids to build up within the digester causing a drop in pH that inhibits digestion. Regular control of pH is therefore necessary by adding alkalinity. Reductions in COD increase with increase in HRT. The stratification of plant material within the digester is different from that of manure, and modifications in design and operation of digesters may be necessary if they are fed with plant matter.
