In this study, the performance of five selective media for coliform bacteria was evaluated. In total, 83 coliform isolates from ready-to-eat food and 21 reference strains were inoculated in five agar media : Chromocult coliform agar (Merck Millipore), HiCrome coliform agar (Sigma), CHROMagar ECC chromogenic media, Brilliance E. coli/coliform selective agar (OXOID), and endo agar (Merck Millipore). Coliform isolates and reference strains were inoculated on the selective media to test media sensitivity and specificity. The tested media showed the following sensitivities for the isolated strains: Chromocult coliforms agar and HiCrome coliform agar, 94%; Brilliance E. coli/coliform selective agar, 93%; CHROMagar ECC chromogenic media, 92%; and endo agar, 74%. In addition, all media showed 100% specificity, except for endo agar (71%). Moreover Chromocult coliform agar and HICrome coliform agar showed high levels recovery. Taken together, these results identified Chromocult coliform agar and HICrome coliform agar as an effective selective medium for coliforms with higher sensitivity and specificity compared to other media tested in this study.
An enterococci selective medium was developed to detect the presence of enterococci for use as a fecal contamination indicator. Among several media which have been known to detect enterococci, the following 9 different kinds of media were selected: Enterococci Confirmatory agar, Azide dextrose agar, Bromocresol-purple azide agar, Esculin bile agar, Citrate azide tween carbonate agar, KF Streptococcus agar, BROLACIN agar, Kanamycin esculin azide agar, and Membrane filter Enterococcus selective agar. Various components from the nine media were mixed to develop a more effective enterococcus selective medium. The newly developed medium named as 'Enterococcus Mixed medium' was more effective than the previous 9 media. Enterococci strains (Enterococcus avium KACC 10788, Enterococcus faecium KACC 11954, Enterococcus saccharolyticus KACC 10783, Enterococcus durans KACC 10787, Enterococcus faecalis KACC 11304, and Enterococcus hirae KACC 10779) and non-enterococci strains (Escherichia coli KACC 10005, Staphylococcus aureus subsp. aureus KACC 10768, and Bacillus subtilis KACC 10111) were used to test the new medium. As a result, the enterococci strains grew well on the Enterococcus Mixed medium whereas the non-enterococci strains did not grow well on it. Additionally, growth of enterococci with freshwater and seawater samples was observed to be good on the Enterococcus Mixed medium. The result of this study confirmed that the Enterococcus Mixed medium was effective in detecting the target enterococci.
A selective agar medium was developed and tested for the isolation of Acidovorax avenae subsp. avenae, the causal bacterial pathogen of bacterial brown stripe, from rice seeds. The new selective agar medium, designated sorbitol pyroglutamic acid agar (SPA) medium, contained 0.5 g of $K_2$HPO$_4$, 3.0 g of Na$_2$HPO$_4$, 2.0 g of D-sorbitol, 0.2 g of L-pyroglutamic acid, 10.0 $m\ell$ of tween 80, 40.0 mg of victoria blue B, 15.0 g of agar, 150.0 mg of ampicillin and 25.0 mg of vancomycin per litter. Colonies of A. avenae subsp. avenae on SPA medium were smooth, round, convex, shiny, blue and 1.5-2.0 mm in diameter 4 days after incubation at 28$^{\circ}C$. Blue colored colony having dark blue zone was typical type of A. avenae subsp. avenae colonies on the medium. Mean recovery of 8 isolates of A. avenae subsp. avenae on the selective SPA medium was 95.8% in comparison to that on KB medium. The saprophytic bacteria were reduced to 97.9% on SPA medium compared to those on KB medium. Most of other rice seedborne bacteria as well as reported pathogenic bacteria were failed to grow on SPA medium. This medium was highly selective for recovering A. avenae subsp. avenae from rice seed samples, and it could be used to enhance the recovery of this bacterium from rice seed samples, which may be contaminated with large numbers of competing microorganisms.
For the more effective isolation of soil actinomycetes, we have developed HHV (Hair hydrolysate-vitamin) agar medium, containing hair as the sole source of carbon and nitrogen. The HHV agar medium was superior to other media such as colloidal chintin agar, glycerol-arginine agar and starch-casein-nitrate agar, and HV (humic acid-vitamin) agar. The maximum effect of this medium has been shown in hair dry weight 0.4 g/l medium. Of each soil sample, the greatestest number of actinomycetes was isolated from the potato annual planted soil among the tested samp- les. The genus of actinomycetes isolated from the potato annual planted soil sample was identified such 5 group as Stretomyces, Micromonospora, Microbispora, Nocardia and Saccharopolyspora.
A semi-selective agar medium was developed for detecting and enumerating Colletotrichum scovillei in chili (Capsicum annum) plant material. Potato-dextrose-agar(PDA) was used as the basic medium. The composition of the semi-selective agar medium was established after several attempts to favor the development of C. scovillei and inhibit the growth of other fungi and bacteria. The semi-selective agar medium contained PDA amended with pyribencarb and pydiflumetofen at 40 ㎍ mL-1 each and streptomycin at 100㎍ mL-1 for preventing bacterial growth. The pH was adjusted to 4.8 with 85% lactic acid. The inhibition of the mycelial growth of C. scovillei was significantly less than that of most other fungi including Fusarium species when grown on the semi-selective medium. C. scovillei was detected from naturally infected chili plants by plating fruit and stem tissue suspensions on the semi-selective medium, which was found to be reliable and quantifiable. This was the first report of a semi-selective agar medium to detect the presence of C. scovillei in naturally infected chili tissue.
This study aimed to evaluate the appropriateness of MRS-C (0.05% L-cystein; pH 5) and BHI-CM (0.05% L-cystein, 0.5% mucin) agars for the selective isolation of bifidobacteria in fecal samples compared to blood-liver-NPNL (BL-NPNL) agar. Over 200 isolated colonies were characterized morphologically and biochemically. Genomic DNA was extracted from pure cultures of the isolated strains, followed by PCR amplification of the 16S rRNA gene. Bifidobacterium longum and B. animalis were selectively isolated from MRS-C agar and Lactobacillus acidophilus and Enterococcus avium were also isolated. B. longum, B. faecale, and B. animalis were isolated from feces on BHI-CM agar; however, different Bacteroides strains (including Bac. fragilis, Bac. kiribbi, Bac. ovatus, Bac. koreensis, and Bac. salyersiae) were also detected. BL-NPNL agar successfully isolated B. longum and Bacillus, while other Bifidobacterium and Bacteroides species could not grow owing to the presence of antibiotics in the medium. The use of antibiotics in a medium can enhance the selectivity; however, antibiotics may inhibit the growth of certain bacteria in a sample. Hence, adjusting pH or adding non-antibiotic nutrients to the medium is more advantageous, than relying on antibiotics.
The objective of this study was to develop a differential medium with improved selectivity for the isolation of Bacillus cereus. Mannitol egg yolk polymyxin medium supplemented with D-galactose allowed the differentiation of diarrheal- and emetic-type B. cereus through pH monitoring. The pH of the medium decreased significantly when incubating the emetic-type B. cereus, whereas the pH change was not significant when incubating the diarrheal-type. The addition of pH indicators, such as methyl red and phenol red, to the medium allowed visual differentiation between diarrheal- and emetic-type B. cereus. A solid agar medium was also developed by optimizing the concentrations of medium components such as monosaccharides, agar, egg yolk enrichment, pH indicators, and antibiotics. This study indicates the possibility of applying selective media for the differentiation of diarrheal- and emetic-type B. cereus.
Phage utilizing medium and BL agar supplemented with antibiotic Tc(tetracycline) were developed as selective media for the isolation and counting of bifidobacteria in dairy products. The former was based on the host specificity of phage. When bifidobacteria and Lactobacillus casei HY 2782 were mixed together in dairy product, L. casei HY 2782 was laysed by J1 phage which has host specificity to L. casei HY 2782 whereas bifidobacteria grew well on the selective medium added wIth J1 phage. The latter was found to inhibit the growth of S. salivarius subsp. thermophilus, L. acidophilus, and L. casei, but commercial bifidobacteria grew well in Tc-containing BL agar.
Selective agar media were constructed for the counting of bifidobacteria in dairy produ- cts containing bifidobacteria, lactobacilli and streptococci. This media containing antibiotics inhibited the growth of lactobacilli and streptococci at less than 10$^{5}$ cfu/ml, but had no influence on the recovery of bifidobacteria. In order to inhibit the growth of 10$^{5}$ cfu/ml of lactobacilli and streptoco- cci, the addition of 1.0~2.0 $\mu$g/ml of tetracycline in BL agar medium was needed. When 25 $\mu$g/ml of neomycin and paromomycin were mixed with 1.0 $\mu$g/ml of tetracycline in BL agar medium, it was able to inhibit the growth of 10$^{6}$ cfu/ml of lactobacilli and streptococci but had a little negative effect on the recovery of colonies of bifidobacteria. The results revealed that the BL agar medium containing 1.0 $\mu$g/ml of tetracycline was suitable to count the cell number of bifidobacteria selecti- vely in the presence of a 1 to 10$^{5}$-fold excess of L. casei, L. acidophilus and S. salivarius subsp.thermophilus.
Selective culture of actinomycetes from soil microbes and their antibiotic producing characters by agar-disk method were examined. Some of the organisms which produced antibiotics on agar disk did not produce antibiotics in liquid culture. Further examination indicated that production of antibiotic was dependent on the composition of medium. Many streptomycestes produced antibacterial substances in tryptic soy broth but others produced antifungal antibiotics in V-8 broth. Production of antibacterial substances by Streptomyces sp. was also dependent on the medium composition.
