Glucosinolates (3-Butenyl Isothiocyanate, Total Glucosinolates) Change in Seeds and Young Seedlings on Chinese Cabbages

  • The main objective of this study was to investigate the patterns of glucosinolates (3-butenyl isothiocyanate, total glucosinolates) in different parts of young seedling and seeds on Korean Chinese cabbages 55 days and Winter pride cultivars. For determination of glucosinolates, two cultivars of Chinese cabbages seeds and different parts of 1-day-old, 3-day-old, 5-day-old, and 7-day-old seedlings were used for analytical sample preparation provided with an anion exchanges column, and measured by GC and UV-Visible Spectrophotometer. 3-Butenyl ITC concentration was the highest in the cotyledon and the hypocotyl parts of 55 days cultivar seedling. In the cotyledon part of Winter pride cultivar seedling, 3-butenyl ITC amount was increase to 3-day-old seedling and then reduced. The cotyledon of 55 days cultivar contained the highest concentration of total glucosinolates while those were increased in the hypocotyl and decreased by degrees in the root. Total glucosinolates amounts were increased to 5-day-old seedling and then decreased in the cotyledon and the hypocotyl parts of seedling on Winter pride cultivar. There was no significant difference in the root part. In the seeds, both of 3-butenyl ITC and total glucosinolates, 55 days cultivar concentration was higher than Winter pride cultivar. The study has shown the variability in 3-butenyl ITC and total glucosinolates concentrations among cultivars, growth stages, and parts.

Variation of Glucosinolate Composition during Seedling and Growth Stages of Brassica rapa L. ssp. pekinensis

  • The objective of this study was to evaluate glucosinolate (GSL) profiles and variation of total and individual GSLs concentrations within seedling (0-14 days) and growth stages (0-15 weeks) of Korean Chinese cabbage (Brassica rapa L. ssp. pekinensis). Ten GSLs (progoitrin, glucoraphanin, glucoalyssin, gluconapin, glucobrassicanapin, 4-hydroxyglucobrassin, glucobrassicin, 4-methoxyglucobrassicin, neoglucobrassicin and gluconasturtiin) were identified from Korean Chinese cabbage. In general, total GSL content significantly decreased during seedling (from 92.89 to $35.26{\mu}mol{\cdot}g^{-1}$ DW) and g rowth stages ( from 74.11 to $1.97{\mu}mol{\cdot}g^{-1}$ DW). Gluconapin was the highest in seeds and in the germination period ($73.1{\mu}mol{\cdot}g^{-1}$ DW) and declined gradually from 73 to 15% during seedling stages. The level of the major aliphatic GSLs, gluconapin and progoitrin, tended to decrease sharply, whereas levels of indolic GSLs (4-methoxyglucobrassicin, glucobrassicin) and aromatic GSLs (gluconasturtiin) were found to increase generally at the beginning of growth stages.

Influence of Thickness of Styrofoam-Panel on Tobacco Seedling Growth in Newly Developed Floating System (새로 개발된 부상형 육묘에 있어서 스티로포움 판의 두께가 연초 묘 생육에 미치는 영향)

  • The inluence of styrofoam-panel thickness of floating frame on the seedling growth of flue-cured tobacco(Nicotiana tabacum L.) in the newly developed direct-seed float system was investigated . Floating frame used instead of styrofoam-tray consisted of styrofoam-panel (106×108cm) for floating, eight holes(1.Sx2S.0 cm) to uniform the water, and nutrition supply far seedling and water and nutrition absorption cloth placed on the styrofoam-panel. Each floating-frame may lay eight plastic-trays on it. Eight grades of styrofoam-panel thickness from 26 mm to 46mm and two kinds of media were used in the greenhouse system. Dry cells were found from the styrofoam-panel thickness of 43mm in carbonized chaff, compost and original soil rate of L3-3(v/v) media, and of 37mm in peat and perlite rate of 8-2(v/v) media. The thinner styrofoam-panel produced more tender and succulent seedling with the more trunk and the less root weight. When considering the appearance of dry cell, seedling growth, producing healthy tobacco seedlings, and utilization of styrofoam-panel for two kinds of medias the ideal styrofoam-panel thickness were suggested to be around 34mm for flue-cured tobacco reeling production in the newly developed direct-seeding float system. Studies on the Development of Stevia (Stevia rebaudiana Bertoni M.) as a New Sweetness (신감미(新甘味) 자원식물(資源植物) Stevia의 개발(開發)에 관(關)한 연구(硏究))

  • The consumption of sugar in our diet increased so great that much U. S. dollars are spended to import sugar source from foreign contries. To replace a part of it we adopted mother plant and seeds of the well-known Stevia (Stevia rebaudiana Bertoni M.) originated from south America, from the Agricultural Experimental Station of Hokkaido, Japan, in 1974. In order to determine the growth condition of Stevia in our climate and soil condition, the results of survey and determination were as follows: 1. Germination rate of Stevia was averaged 33.6% at 35 th day after seedling and the maximum germination period was between 5 and 20 days after seedling. 2. The seedling rate of Stevia was about 8%. 3. Establishment of roots of Stevia propagated by cutting method was above 90% but only S-1 strain showed lower rate., 75%. 4. S-2 strain showed the best growth rate that was about 160cm by plant height and it was decreased by the following order : S-4, S-3, S-1, S-6 and S-5 strains. 5. S-4 strain resulted in 45 branches that was the highest number among strains tested and S-2 strain was the next. 6. The content of sweetening compound of Stevia, stevioside, was 5.3% in S-1, S-2, S-5 and S-4 strains but S-3 and $S^*$-6 strains were relatively low content, 4.9%. From the above results we obtained Stevia could be propagated by cutting method and the growth conditions including plant height, number of branches and content of stevioside were relatively better in S-2 and S-4 strains that were differentiated by the shape of leaf.

Growth, Carbon and Nitrogen Status of Container Grown Black Pine (Pinus thunbergii) Seedlings at Various Levels of Foliar Fertilization

  • The growth, carbon and nitrogen status of container grown black pine (Pinus thunbergii) seedlings were examined at various levels of foliar fertilization (control, 0.1%, 0.2%, 0.3%). Root collar diameter, height and dry weight of black pine seedlings increased significantly with increasing levels of foliar fertilization (P<0.05). Carbon concentration in needle of black pine seedlings was significantly higher in the foliar fertilization than in the control treatments (P<0.05), while other seedling components such as stem and roots were not significantly different (P>0.05) between the foliar fertilization and the control treatments. Nitrogen concentration and content were significantly greater in the foliar fertilization than in the control treatments (P<0.05). Shoot/root ratio of black pine seedlings (needle+stem dry weight/root dry weight) was greater in the foliar fertilization (2.40-2.89) than in the control treatments (1.87). However, nitrogen use efficiency was significantly lower (P<0.05) in the foliar fertilization (28-46) than in the control (111) treatments. The results indicate that morphological characteristics and nutritional status on container grown black pine seedlings were enhanced by various levels of foliar fertilization.

  • The mobilization of food reserves and ultrastructural changes in th cotyledons of germinating soybean seeds (Glycine max L. Mer. Cultivar Amsoy) and seedlings were studied by using light and transmission electron microscopy. When germinating began, the cotyledon tissues were packed with protein an lipid bodies. Mobilization of the reserves started in epidermis and vascular bundles. After three days of seedling growth, significant reductions of protein and lipid bodies were observed; concurrently, the numbers of starch grains, glyoxysoms, and mitochondria were increased. These ultrastructural changes are discussed with reference to the metabolism of the germinating soybean seeds and seedlings.

  • Paper sludges collected from three different paper mills were physico-chemically analyzed in order to use them as raw materials for making bed soils and seedling pots. The sludge from a fine paper mill contained lots of inorganic pigment particles used for coating, as those from a newsprint mill and a tissue mill had not. It was clearly through XRD analysis confirmed that all sludges included calcium carbonate. The paper sludge from the tissue mill contained the greatest amount of particles, which would contribute to water absorption and nutrient storage. The sludge from the fine paper mill had the highest density due to many inorganic elements. While the ash content and the total nitrogen content were the highest in the sludge from the fine paper mill, the C/N ratio was the lowest in the fine paper mill sludge. All sludges seemed to have insufficient contents of potassium. The sludges from the newsprint mill and the tissue mill showed more silicon contents than that from the fine paper mill. It was concluded that the sludge from the fine paper mill would be able to be the most efficient raw materials for making bed soils and seedling pots and the other two sludges would be more efficient for intensive culture for crops such as rice and grain with additional supplement of nitrogen and other nutrients.

The Effects of Calcium Nutrition on the Activities of Lactate Dehydrogenase, Alcohol Dehydrogenase and Other Enzymes in Melon (Cucumis melo L.) Seedlings Subjected to Flooding

  • With transient flooding followed by poor or slow drainage plant roots may become reduction conditions because the root zone was fully filled with water. This study was examined the effects of calcium treatment in the early growth stage on biochemical changes in leaves and roots of melon (Cucumis melo L.) seedlings kept under flooding condition for 72 h. The activities of lactate dehydrogenase more gradually enhanced in the roots than those of leaves of melon seedlings treated with calcium. The activities of alcohol dehydrogenase associated with alcohol fermentation under low oxygen conditions continuously increased in the leaves and roots of seedlings untreated with calcium under flooding at least 72 h but those was constant within at least 12 h in treated with calcium. These results showed that calcium supplying in the early growth stage mitigated alcohol fermentation of melon seedlings kept under flooding condition for 72 h. Activities of nitrate reductase and acid phosphatase in the leaves and roots of seedlings in treated with calcium somewhat higher than those of non-treated with calcium. The activities of sucrose phosphate synthase and fructose-1,6-bisphosphatase of leaves of seedlings in treated with calcium more higher than those of non-treated with calcium. These results indicated that calcium nutrition mitigate the reduction of activities of some enzymes of melon seedling kept under flooding condition for 72 h.

Effects of Artificially Acidified Soils on the Growth and Nutrient Status of Pinus densiflora and Quercus acutissima Seedlings (토양산성화가 소나무, 상수리나무 묘목의 생장 및 영양상태에 미치는 영향)

  • The effects of soil acidification on the seedling growth and nutrition of Pinus densiflora and Quercus acutissima were investigated. The relationship between the seedling growth and molar (Ca+Mg+K)/Al ratio of in soil solution was examined. The results suggested that growth inhibition of seedling Pinus densiflora and Quercus acutissima was due to the low pH of soil solution, which was followed by leach of Al into soil solution, and decrease of essential elements, such as Ca in aerial pant of the seeding caused by the increase of Al concentration in subterranean pant of the seedlings. The level of growth inhibition was determined not only by Al concentration, but also by the balance of inorganic elements, Al, Ca, Mg and K. The growths of two species in total dry weight were clearly inhibited when molar (Ca+Mg+K)/Al ratio of the soil was lower than 6.0. The growth in dry weight, in the condition of the molar ratio was 0.8, was decreased 60% or 50% for the seedling of Pinus densiflora or Quercus acutissima respectively. It was concluded that the molar (Ca+Mg+K)/ Al ratio could be an important index for evaluation of the effects of soil acidification, due to acid deposition such as acid rain, on growth of trees and nutrition. And it might be a more useful indicator for evaluation of critical load of acid deposition on forest ecosystems.

The Energy Expenditure of Female Farmers in Kyungki Area (경기일부지역 농촌주부의 농작업에너지 대사)

  • This study was carried out on 10 persons of female farmers to determine the energy expenditure of agricultural work. Their mean age was 44 year-old, mean daily urinary nitrogen excretion was 11.3g, and creatinine excretion was 0.183g. The energy metabolism was determined by indirect calorimetry using Douglas bag and Scholander's gas analyzer. The measured basal metabolism was 1,273 kcal per day and rest metabolism was 1,614 kcal per day. To remove seedling from the seedplot, they spent 2.479kcal per minute, to weed 2.805kcal/min, and to reap with sickle 4.58kcal/min. Energy expenditure of the same work, only sitting on the stool, was significantly reduced. Prolonged work did not change energy expenditure per minute significantly.
