  • A total of 10,753 oligotrophic bacteria were isolated from the cultivated soils of red-pepper infected by Phytophthora blight disease in various regions of Korea (Chungju, Anmyon, Taean, Andong, Eumsung and Goesan). Seven bacteria isolates among these collected resources were selected by the first screening of in vitro antagonistic assay against major several plant pathogenic fungi including Phytophthora capsici. Finally, strain 7F was selected by pot assay for a possible biological control agent against Phytophthora blight disease of pepper seedling in the greenhouse. Strain 7F was identified as Bacillus subtilis on the basis of its 16S rDNA sequence analysis and as standardized biochemical characteristics assay kits such as API20 NE. In the experiment of P. capsici zoospore infected red-pepper on the pot test, infection rate of red-pepper with nonetreatment to Phytophthora blight disease was 87%, while the rate was only 6% in the pot treated with strain 7F. This result indicated that the Bacillus subtilis strain 7F will be useful as a potential biocontrol agent for Phytophthora blight disease of red-pepper.

  • The great economic losses caused by Cucumber mosaic virus (CMV) infection of peppers has led to the development of genetically modified (GM) CMV-resistant peppers. We developed virus-resistant pepper plants using Agrobacterium tumefaciens -mediated transformation. The expressed recombinant protein was purified using nickel-nitrilotriacetic acid resin and immunoaffinity chromatography, and purity was assessed by sodium dodecyl sulfate polyacrylamide gel electrophoresis. Immunoblot analysis revealed the purified CMV coat protein (CMV-CP) had a molecular mass of 25 kDa. After in-gel digestion and desalting, the internal peptide fragments of CMV-CP were sequenced by matrix-assisted laser desorption/ionization-time of flight. Most GM pepper and Escherichia coli BL21 internal peptides had identical peptide sequences and contained 137 of 183 whole peptides in CMV-CP. A quantitative enzyme-linked immunosorbent assay was performed to detect CMV-resistant GM peppers. We also provide basic information about the expressed protein in GM peppers for further safety assessment. The contents of soluble protein and CMV-CP were measured in GM and control peppers cultivated in three different areas of Korea. Statistical significance in terms of cultivation areas, harvest times, generations, and plant tissue origin were determined based on a P value of 0.05. The highest amount of CMV-CP was detected at the seedling stage from plant grown in each region. T3 and T5 showed significantly different levels of CMV-CP from T4 in leaves in the whorl stage. No statistical differences were observed among GM peppers at different stages of maturity in any cultivation area. The results from this study contribute to the safety evaluation of newly designed CMV-resistant GM peppers and provide a standard against which to compare other virus-resistant GM peppers.

  • Powdery mildew caused by Podosphaera fusca is one of the most important diseases in leafy lettuce (Lactuca sativa L.). Since the disease has been a threat to safe cultivation of leafy lettuce, its control methods have to develop to produce good quality of lettuce for farmer and consumer. Occurrence of lettuce powdery mildew is increasing more and more due to continuous cultivation of lettuce all through the year, non-removal of diseased plant parts of lettuce, spray of inadequate fungicides by mistaken acknowledge of lettuce powdery mildew for lettuce downy mildew, etc. The control effect of five fungicides against lettuce powdery mildew was examined in a plastic greenhouse located in Suwon. When fungicides were sprayed three times at 10 days-intervals in the early stage of occurrence of powdery mildew, the incidence of powdery mildew in the plants treated with kresoxim-methy SC, azoxystrobin SC, Ampelomyces quisqualis AQ94013 WP, Paenibacillus polymyxa AC-1 SC and Bacillus subtilis Y 1336 WP was 0.7%, 0.7%, 26.0%, 36.7% and 42.0%, respectively, whereas the incidence of non-treated control was 55.3% on eight days after final application. Phytotoxicity of five fungicides tested was not observed in lettuce seedling plants.

  • Develoment precess and characteristics of eggs of the geoduck clam, Panope japonica are reporting in this study. Eggs and sperm were excised from gonad, artificially fertilized in an aquarium, reared under various temperature regimes, and record and record the larval period and the time need to reach a certain larval stage from ferilization. Unfertilized eggs of P. japonica appeared to be oval with a mean diameter of $70\mu$m and they became spherical after fertilization. The eggs of P. japonica can be classified as demersal. At a constant water temperature of $ 11^{citc}C$, it took 4 hours form fertilization to become four-cell stage, two days to become trochophore larvae, three days to become D-shape larvae, twenty-three days to become umbo stage, and thirty-six days to become fully grown veliger ready form settlement. A negative correlation was observed between the water temperature and the larval period of P. japonica. From fertilization to D-shape larvae, it took five days at 8$^{\circ}C$, while it was only two days to become D-shape larvae at $ 17^{citc}C$. Time required to D-shape larvae from fertilization was proportional to temperature, and the relationships were expressed as follows : To 8-cell stage, 1/t=0.0209 w-0.1167 (r=0.9967) To blastula stage, 1/t=0, 0055 w-0.0192 (r=0.9825) To trochophore stage, 1/t=0.0034 w-0.0155 (r=0.9907) To D-shape larvae stage, 1/t=0.0014 w-0.0023 (r=0.9843) (t, time in hours ; w, water temperature) Bioligical minimum temperature for egg development was calculated as 3.82$^{\circ}C$ in average.

  • This study was launched to verify the effective composition of plant species and its management program most suited for the dredged soiled area near Nakdong River Site 14. The improvement methods of planting base and the composition of plants such as silver grass, reed, and some colonies in aesthetic effect were studied. To search the management methods to decrease the confining pressure risen from the burried seeds which would consequently harm the previously seeded plants, experimental construction process was measured on the site. The purpose of this experiment was to figure out which part of the improvement on the plant base has the most significant effect for the revegetation of infertile, dredged soil, to verify the easily seeded, developing plants among seeded plants, and finally, to find the restoration model using plants near the dredged soil around riverside. 8 seeded plants and 23 invaded species were appeared which among the emerged plants, development of Aster yomena MAKINO, Lotus corniculatus var. japonica Regel, Trifolium repens L, and Dianthus longicalyx Miq were proved to be brought up well. Difference risen from the seed composition were not noticeable until 150day since the germination was proceeded mainly by Aster yomena MAKINO. The experimental plot with dredging sand+organic fertilizer method of construction and dredging sand+soil conditioner method showed most development while the effect of the plot with only the soil base of dredging sand stayed low. Another important method for the management of infertile, dredged soil base would be the removal of disturbing species which the experiment showed the tied relationship between the removal of disturbing species and development of seeding plants. Although this study was carried out focused on the Nakdong River Project, the study suggests the general management program that the removal of disturbing species such as Humulus japonicus Sieboid & Zucc. and Pueraria lobata (Willd.) Ohwi in times around rainy season(60days after seedling) would be effective for the easy growth of revegetation plants.

  • Root-knot disease caused by Meloidogyne incognita is major soil pathogen and cause severe economic damages to vegetable crops. In this study, we selected rhizobacteria for biocontrol of the root-knot nematode, M. incognita, and identified; performed bioassay of the bacterial extracts in cucumber seedlings. The crude extracts of strains CT16 and JC05 out of 180 strains inhibited egg hatching and increased juvenile mortality in vitro assay; based on 16S rRNA sequences analysis, the two strains were identified as Bacillus sp. CT16, and Neobacillus sp. JC05. After extracting the bacterial supernatants by using various organic solvents, n-butanol and n-hexane extracts of strain CT16 and n-butanol extract of strain JC05 showed inhibitory activity of egg hatching depending on concentrations. Subsequently, n-butanol extracts of two strains significantly suppressed formation of egg masses in cucumber seedling. Therefore, these results indicated that strains CT16 and JC05 could be used as potential biocontrol agents against M. incognita.

  • Callus induction and somatic embryogenesis are fundamental to cotton tissue culture biotechnology. An efficient protocol for callus induction, somatic embryogenesis and their maturation have been developed to regenerate plantlets from cotton (Gossypium hirsutum L.) variety coker 312. Embryogenic callus was initiated from hypo-cotyl region that was used as an explant at seedling stage when it was about 7-8 days old. Callus induction was achieved through culturing hypocotyls (5-7mm) on $MS_{1a} medium supplemented with 2,4-D (0.1 mg/L) and KT (0.5 mg/L) for six weeks. A friable, colorless, bulky and well proliferating callus becomes greenish with the addition of NAA (2.0 mg/L), ZT (0.1 mg/L) and removal of 2,4-D (M $S_{1b}$) cultured for two weeks then again transferred to $MS_{1a}. 2,4-dichlorophenoxyacetic acid (2,4-D) promoted the proliferation of embryogenic callus, but had a negative effect on the differentiation and germination of somatic embryos. ZT (0.1mg/L) and activated charcoal (2g/L), both hormones play an important role in differentiation and germination of somatic embryos in hypocotyls derived embryogenic callus but in case of cotton, such a capability have been observed on MS medium with 1.92 g/L $KNO_3$, but it is considered to attain somewhat more improvement. High embryogenesis frequency was achieved through nutrient deficient stress treatment. The frequency of globular embryogenesis (two-three folds) was achieved when well proliferating callus was (from $MS_{1a}$ media) cultured on MS (1/5 strength) medium for four weeks. Here the development of anthocyanins is the best indicator for somatic embryogenesis. However, when embryoid callus was cultured on MS (full strength) medium, the globular embryos were developed into normal plantlets immediately. In this procedure 27.49% cotyledenary embryos were developed. Of that 70% cotyledenary embryos were developed not only into normal plantlets but rooted simultaneously, when cultured on MS (with 0.05 mgg/L giberrelic acid) medium. So complete plants could be regenerated through somatic embryogenesis from hypocotyl explants within 6 months.s.

  • Dandelion has been widely utilized for medicinal and edible purposes. This research was conducted to evaluate the effect of supplemental LED (light-emitting diode) light on germination, growth characteristics and anthocyanin content of dandelion (Taraxacum officinale) seedling using LED blue (460 nm), red (660 nm, R), blue + red (B:R=6:4) and fluorescent lamp light treatment. By LED illumination to T. officinale seed germination speed was delayed, and germination rate was the highest in the fluorescent light. The growth characteristics (plant height, number of leaves, root length and fresh weight) were greatly influenced by supplemental LED light compared with control treatment, and the growth promotion was the most effective in the red LED illumination. After 60 days of red and mixed LED light treatments, anthocyanin content of dandelion plants was significantly changed. The anthocyanin content was increased by 12~19 mg/100 g under the red LED and the mixed light conditions compared with the control and the blue LED. Results indicate that illumination with red and mix LEDs, compared with other light treatments, is beneficial for promotion of growth and anthocyanin content in dandelion.

  • Lisianthus (Eustoma grandiflorum) is relatively sensitive to temperature and light conditions. For year round production of good quality potted plants and energy saving, it is necessary to understand the growth and flowering response to the combined conditions of these environmental factors. This study was conducted to examine the growth and flowering responses to temperature, photoperiod, and light intensity during the post-seedling stage. 'El Paso Deep Blue' lisianthus plants with four true leaf pairs were grown in growth chambers maintained at average daily temperatures (ADT) of 14, 20, and $26^{\circ}C$ and provided with three photosynthetic photon fluxes [PPF; 100, 200, and $400{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$] for 8 (08:00-16:00) and 16 hours (08:00-24:00) by fluorescent and incandescent lamps, resulting in four daily light integrals (DLI): about 3, 6 (two photoperiods), 12 (two photoperiods), and $24mol{\cdot}m^{-2}{\cdot}d^{-1}$. After treatment for three weeks followed by growth for one week in a greenhouse of $20{\pm}3^{\circ}C$, growth and development were measured. Higher temperature, higher PPF, and longer photoperiod promoted plant growth and flowering; however the impacts of PPF and photoperiod were smaller than those of temperature. As ADT and DLI increased, the number of leaves, number of flowers, lateral shoot length, and shoot dry weight increased. An increase of about $1mol{\cdot}m^{-2}{\cdot}d^{-1}$ DLI could constitute an increase of 0.40 to $0.76^{\circ}C$ ADT depending on these crop characteristics when ADT and DLI are above $20^{\circ}C$ and $12mol{\cdot}m^{-2}{\cdot}d^{-1}$, respectively. Therefore, growers can select a regimen of heating or supplemental lighting without delaying harvesting time or decreasing crop quality.

  • As for chemical characteristics of cultivated media waste, the total content of nitrogen, phosphoric acid, and potassium was 6.93%; organic matter content was 67.78%; pH and EC were 7.20 and 1.46 ds/m, respectively. Organic acids identified during stabilization included citric acid, oxalic acid, fumaric acid, and acetic acid, which was highest. During the stabilization of cultivated media waste, temperature decreased and pH increased on the 15th day, and the germination index was 88 after the period, indicating that at least 15 days of stabilization is needed when cultivated media waste is used for growth material of crop. As for germination characteristics, when the mixture rate of cultivated media waste was 20% and under, germination of cucumber and oriental melon was favorable without difference compared to the control plots. For lettuce and Chinese cabbage, no statistically significant difference was found when the rate was 10% and under compared to the control plots. As for seedling quality after 20 days of sowing, cucumber, oriental melon, and Chinese cabbage showed favorable growth only when the mixture rate of cultivated media waste was 10% and under; lettuce showed lower growth characteristics regardless of mixture rate when compared to the control plots. Cultivated media waste may be significantly useful for cucumber and oriental melon, but not for Chinese cabbage and lettuce.
