Studies on Seed Germination of Miscanthus sinensis Native to Jeju Island (제주도 자생 참억새의 종자발아에 관한 연구)

  • The study was conducted to find out the harvesting time, storage duration, cold treatment and sterilization on seed germination of Miscanthus sinensis. The optimum harvesting time of the Miscanthus seed in native to Jeju island was December. And the seed germination passible 3 years after harvesting. When the seeds had cold treatment, seed germination nate was 74% rather than 63% of non-treatment. For seed sterilization treatment, soaking in solution of Benomyl 7% during from 3 hours to 12 hours germinated 9% over. But the non-treatment on 24 hours treatment reduced the seed germination of Miscanthus sinensis.

Enhancement of seed germination and microbial disinfection on ginseng by cold plasma treatment

  • Background: This study aimed to investigate the effect of cold plasma treatment on the improvement of seed germination and surface sterilization of ginseng seeds. Methods: Dehisced ginseng (Panax ginseng) seeds were exposed to dielectric barrier discharge (DBD) plasma operated in argon (Ar) or an argon/oxygen mixture (Ar/O2), and the resulting germination and surface sterilization were compared with those of an untreated control group. Bacterial and fungal detection assays were performed for plasma-treated ginseng seeds after serial dilution of surface-washed suspensions. The microbial colonies (fungi and bacteria) were classified according to their phenotypical morphologies and identified by molecular analysis. Furthermore, the effect of cold plasma treatment on the in vitro antifungal activity and suppression of Cylindrocarpon destructans in 4-year-old ginseng root discs was investigated. Results: Seeds treated with plasma in Ar or Ar/O2 exhibited a higher germination rate (%) compared with the untreated controls. Furthermore, the plasma treatment exhibited bactericidal and fungicidal effects on the seed surface, and the latter effect was stronger than the former. In addition, plasma treatment exhibited in vitro antifungal activity against C. destructans and reduced the disease severity (%) of root rot in 4-year-old ginseng root discs. The results demonstrate the stimulatory effect of plasma treatment on seed germination, surface sterilization, and root rot disease suppression in ginseng. Conclusion: The results of this study indicate that the cold plasma treatment can suppress the microbial community on the seed surface root rot in ginseng.

Status of Plasma Technology Applied to Agriculture and Foods (플라즈마 기술의 농식품 분야 활용)

  • Recently, plasma technology has been increasingly applied to agriculture and foods. Owing to the three important plasma characteristics which are activation, sterilization, and catalysis, the plasma technology can be properly applied to agriculture, in the other word, to the whole cycle of agriculture from farm to table: seed germination, plant growth, harvest and storage, washing, packaging, transport, in store, household, cooking, garbage, etc. Some representative case studies for plasma activation, sterilization, and catalysis show well that plasma technology can be successfully applied to the whole cycle of agriculture.

Survey of Diease and Weed Control in Organic and Free-pesticide Cultivation of Chunnam Area 'Ssam' Vegegable (전남지역 쌈채류 무농약.유기재배농가의 잡초, 병해충관리 실태분석)

  • For developing standard method for diease, pest and weed control in environmental friendly 'Ssam' vegetable cultivation, this study was carried out to investigating agriculture material use in organic agriculture and no pesticide cultivation for lettuce, kale, leafy perilla and korean cabbage. The 28.6% of investigated farmer carried out seed sterilization by seed selection with salt solution and soaking in chitosan that not validated. For raising seedling periods, the 55.6% of farmer did not use environmental-friendly agriculture material for, diease control and the 50% of farmer used one time for. pest control. Therefore, the control of disease and pest could be achieved with one or two times use of environmental-friendly agriculture material. Seed sterilization was carried out by soil solar sterilization, one time per year in 71.4% of farmer. Weed was controled by black PE film for weed germination of furrow in many farmer, by man-power weeding for weed of ridge in 85% of farmer and by machine weeding and mulching in some farmer. During cultivation period, the major pest were Aphis gossypii in lettuce, Plutella xylostella in kale, Plutella xylostella and Phyllotreta striolata (Fabricius) in korean cabbage and Pyrausta panopealis (Walke) in feat perilla. The many farmers used environ-mental-friendly agriculture material for control of pest over 10 times for spring season, and more used sold materials in market than home-made materials. In result, it needs to develop standardized method and validate cultivation methods for control of disease and pest, and seed sterilization treatment environmental-friendly 'Ssam' vegetable.

Effects of High Voltage pulse on Seed Germination and Plant Growth

  • It is generally known that electron beam has sterilization effects and can activate plant germination and growth. Compared to electron beam, electrical pulse has not been frequently studied with respect to the biological application. In this study, we have analyzed the effects of high voltage pulse on seed germination and growth using various plant species. We have used the high voltage generator for examining seed's responses to the high voltage pulse. The operating voltage and currents of the generator are about 300 kV and 30 kA, respectively. Pulse width is 60 ns. High voltage pulse has slightly activated germination and growth of radish during early stage. Various levels of germination and growth are observed in different plant species after treated with high voltage pulse.

Studies on the Germination of Korean Ginseng (Panax ginseng C.A.Meyer) Seed II. Influences of Temperature and Seed Treatment on Embryo Growth and Germination (인삼 종자의 발아에 관한 연구 II. 온도 및 종자처리가 배생장 및 발아에 미치는 영향)

  • Freshly harvested and depulped Korean ginseng seeds were subjected to the seed treatment of removing endocarp plus surface sterilization with sodium hypocloride, surface sterilization only, and nontreated control. These seeds were stratified at temperatures of 5$^{\circ}$, 10$^{\circ}$, 15$^{\circ}$ and 20$^{\circ}C$ for 20, 40, 60, 80 and 100 days. Embryo growth of the ginseng seeds of which endocarp was removed was most rapid in each stratification temperature and that of sterilized seeds was slower than unsterilized seeds after 80 days stratification at 15$^{\circ}$ and 20$^{\circ}C$. About 15$^{\circ}C$ was an optimal stratification temperature for embry growth in ginseng seeds. Chilling treatment at 5$^{\circ}C$ for 100 days was needed for better germination of dehisced ginseng seeds. An optimal germination temperature for the ginseng seed following chilling treatment was about 15$^{\circ}C$.

  • The effect of calcium cyanamide (China-made) soil fumigation on the growth of the ginseng pathogen Rhizoctonia, Pythium), soil microbes, and seed germination of lettuce and radish was investigated. At twice the recommended level (2S0-ppm $CaCN_2$), the growth of Rhizoctonia and Pythium, and the seed germination, were not inhibited. Rather, the effective level was 10,000 ppm. The powder form was more effective than the granular form in inhibiting pathogen growth and seed germination. The lettuce seed was also more sensitive than the radish seed. Calcium cyanamide appearedto decrease the fungi population and to increase Actinomycetes in the soil.

Comparison in Mycelial Growth and Fruit Body Development According to Sterilization Condition of Substrate in Pot Cultivation of Pleurotus ostreatus (느타리버섯 봉지재배시 배지 살균 조건에 따른 균사생장 및 자실체 생육 비교)

  • The suitable sterilization conditions in pot cultivation of varieties of Chunchunutari-2-ho and Suhannutari-1-ho Pleurotus ostreatus were investigated. The substrate formulations for the cultivation consisted of 40% poplar sawdust, 40% waste cotton and 20% beet pulp (PWB), and 40% poplar sawdust, 40% cotton seed hull and 20% beet pulp (PCB). The increase in the sterilization temperature caused decrease in the pH of the substrate. The moisture content of PWB substrate increased by 1.8%, but that of PCB substrate decreased by 4.1 % after low temperature sterilization ($12hr\;at\;60^{\circ}C$). However, a little change in the sterilization at higher temperature ($6hr\;at\;100^{\circ}C\;and\;90min\;at\;121^{\circ}C$) did not alter the moisture content of both substrate appreciably. The duration of mycelial growth was longest and its density was lowest in low temperature sterilization ($12hr\;at\;60^{\circ}C$). The high pressure sterilization ($90min\;at\;121^{\circ}C$) of PCB substrate produced high yield (236.6 g/pot) and the highest biological efficiency (96.6%) for Chunchunutari-2-ho. On the contrary the high temperatures sterilization ($6hr\;at\;100^{\circ}C$) of PCB substrate resulted in the yield of 259.1 g/pot with the biological efficiency 94.9% for Suhannutari-2-ho.

Optimization of in vitro seed germination of Taraxacum platycarpum

  • Dandelion (Taraxacum platycarpum) has been widely utilized for medicinal purposes. However, the dandelion seeds are relatively difficult to germinate under cultivation conditions, which hampers seedling propagation of dandelion plants and reduces the opportunity of usage of such a useful medicinal plant. Thus, in this study, in vitro conditions for the dandelion seed germination were optimized to enhance the germination rate. In seed washing steps, the sequential treatments with 20% of ethanol, 20% of NaOCl, and distilled water avoided microbial contamination with the highest in vitro germination rate (67.5%) from seeds sown in germination media. The media supplemented with 1.4 g/L of MS salts and 1% of sucrose significantly enhanced the germination rate compared to the media with 4.4 g/L of MS and 3% of sucrose. Sowing the seeds vertically in the optimized media supplement conditions, 1.4 g/L of MS salts and 1% of sucrose, gave the maximum in vitro germination rate (61%), which was almost three times higher than sowing seeds on a soil pot (23%). Our results indicate that the seed washing and sowing methods including germination medium supplements can be optimized to enhance in vitro seed germination of dandelion.

Screening of Seed Treatment Agents against Leaf Blight and Black Root Rot for Carrot Organic Cultivation (당근 유기재배를 위한 검은잎마름병과 검은무늬병 종자소독제 선발)

  • This study was conducted to control the seed borne pathogens (Alternaria spp.) of carrot and to examine the seed germination rate by using 13 environment friendly agricultural materials instead of conventional chemicals for organic cultivation. The growth inhibiting effects on pathogens showed the different responses according to each agricultural material and effective 7 materials against seed borne pathogens were selected. Among 7 materials, the carrot seeds sterilized with plant extracts, Tanger Stop and Land Saver were not germinated at all. The germination rate of seeds sterilized with other materials showed the similar levels with reference chemical (Benlate-T) and non-treated seeds. Infection rates of seeds sterilized with seaweed extract, Bellopper for controlling A. radicina and plant extract, Ssial-100 for A. dauci were similar or low infection rate compared with reference chemical. The germination rate and root length of seeds sterilized with Bellopper and Ssial-100 was similar or superior to those of chemical sterilized seeds. The optimal condition seemed to be a little bit different depending on the concentration of materials. As a result, the sterilization of carrot seeds by using the environment friendly materials could be effectively utilized as a technology to inhibit the infection of seed-borne pathogens.
