Efficacy of Fluazinam and Iprodione+Propineb in the Suppression of Diaporthe phaseolorum, Colletotrichum truncatum and Cercospora kikuchii, the Causal Agents of Seed Decay in Soybean

  • Seed decay of soybean caused by Diaporthe phaseolorum, Colletotrichum truncatum and Cercospora kikuchii is a serious disease when soybean is harvested under warm and wet weather conditions. Benomyl has been used for controlling the disease, however, benomyl application may be limited due to common occurrence of resistance. The efficacy of 21 fungicides against the pathogens was evaluated in vitro. Among the fungicides tested, benomyl, carbendazim, fluazinam, iprodione+propineb, thiophanate-methyl, and triflumizole were found effective and were evaluated for their ability to control the seed pathogens. Fluazinam completely inhibited mycelial growth at a concentration of 100 $\mu\textrm{g}$/$\textrm{m}{\ell}$ for D. phaseolorum; and at a concentration of 500 $\mu\textrm{g}$/$\textrm{m}{\ell}$ for C. truncatum and C. kikuchii. $EC_90$ values of fluazinam were similar to that of benomyl. Because fluazinam, iprodione+propineb, and triflumizole were found effective against the seed pathogens, these were subjected for field-testing. Suppression of pod and seed infection by fluazinam and iprodione+propineb was as high as that of benomyl without any reduction in agronomic characters of soybean. This study shows that fluazinam and iprodione+propineb may be used in combination with benomyl to control seed pathogens, manage resistance, and ensure production of high quality soybean seeds.

넙치의 바이러스성(性) 질병(疾炳) (Viral diseases of Japanese flounder(Paralichthys olivaceus) in Japan)

  • With the rapid progress in seed production techniques, aquaculture production of economically important species of marine fish has been accelerated in Japan. Howecer, mass mortalities due to viral infections as well as other microbial infections have often occurred during the seed production and grow-out stages. Among these diseases, four viral diseases have been known in cultured Japanese flounder (Paralichthys olivaceus) since around 1980. In this paper, viral diseases of cultured flounder in Japan are briefly reviewed, with special attention to two viral diseases. viral epidermal hyperplasia and rhabdovirus infection which are relatively important because of their frequent occurrence. Viral epidermal hyperplasia is characterized by fin opacity and associated with high mortality in larval flounder Electron microscopy of affected epidermal cells and transmission experiments with tissue filtrates demonstrated that the disease was caused by a herpesvirus but the agent has not been isolated in fish cell lines. On the other hand, rhabdovires infection occurrs in juvenile and production size fish with hemorrhage in the skeltal muscle and fins, congestion of the gonads, and ascites. A rhabdovirys was isolated in RTG-2 cells from the diseased flounder as a causative agent, which was designated hirame rhabdovirus (HRV) or Rahbdovirus olivaceus. HRV is serologically distinguishable from other known fish rhabdoviruses. Intensive researches on these viral diseases started in 1980th. but properties of the causative agents and infection mechanisms have not been fully investigated. This results in difficulty in controlling these diseases.

참깨 종자전염진균(種子傳染眞菌)의 검정(檢定) 및 Corynespora cassicola의 병원성(病原性) (Detecting Seed-borne Fungi of Sesame and Virulance of Corynespora cassicola)

  • 충남농촌진흥원으로부터 분양받은 참깨종자의 종자전염진균을 표준 Blotter 법으로 조사하였던 바 12종의 균류가 검출되었다. 그 중 Alternaria sesamicola, A. tenuis, Corynespora cassicola가 많은 비율로 검출되었다. Chlorine으로 종자처리를 하였을 경우, C. cassicola를 제외한 모든 균이 거의 완전히 제거되었다. 참깨 종자내의 감염부위는 C. cassicola가 가장 깊숙이 배에까지 감염되어 있었다. C. cassicola의 병원성을 조사하였던 결과 참깨의 잎과 줄기에 심한 반점을 형성하고 병반이 급격히 확대되며 말라죽었다.

수집재래종대두의 자주빛무늬병(Cercospora kikuchii)에 대한 저항성검정 (Evaluation of Native Soybean Collection for Resistance to Purple Blotch)

  • 대두 자주빛무늬병에 대한 저항성인자원 선발을 위하여 한국에너지연구소, 방사선농학연색실이서 보존하고 있는 수집재내종을 자연감염율과 인공접종법에 의한 저항성검정을 실시하였다. 공시한 수집재래종 467 계통중 $28.9\%$$0.1\%$이하의 감염율을, 약 $13.4\% $$2\%$ 이상의 감염율을 보였으며 이 자연감염율은 결협초기의 기상조건과 밀접한 상관을 보였다. 종자접종에 의한 자반의 형성은 자연감염율과 정의 상관을 보여 일차적인 저항성 검정방법으로서 효과적이었으며 만숙대두계통은 병해도피에 의한 저항성을 보였다. 그리고 갈색무늬병균의 병원성과 관계되는 종자의 자반정도는 배양기의 착색정도와 높은 상관을 보였다.

Timing of Fusarium Head Blight Infection in Rice by Heading Stage

  • Fusarium graminearum causes the devastating plant disease Fusarium head blight and produces mycotoxins on small cultivated grains. To investigate the timeframe of F. graminearum infection during rice cultivation, a spore suspension of F. graminearum was applied to the rice cultivars Dongjin 1 and Nampyeongbyeo before and after the heading stage. The disease incidence rate was the highest (50%) directly after heading, when the greatest number of flowers were present, while only 10% of the rice infected 30 days after heading showed symptoms. To understand the mechanism of infection, an F. graminearum strain expressing green fluorescent protein (GFP) was inoculated, and the resulting infections were visually examined. Spores were found in all areas between the glume and inner seed, with the largest amount of GFP detected in the aleurone layer. When the inner part of the rice seed was infected, the pathogen was mainly observed in the embryo. These results suggest that F. graminearum migrates from the anthers to the ovaries and into the seeds during the flowering stage of rice. This study will contribute to uncovering the infection process of this pathogen in rice.

고추 탄저병균(炭疽病菌)의 종자전염(種子傳染)에 관(關)한 연구(硏究) (Studies on the Seed Transmission of Colletotrichum spp. in Red Pepper (Capsicum annuum))

  • 고추 종자에서 Colletotrichum acutatum, C. coccodes, C.dematium 및 C. gloeosporioides 등 4종의 탄저병균(炭疽病菌)이 검출(檢出)되었다. 그중 C.dematium과 C.gloeosporioides가 높은 비율(比率)로 검출(檢出)되었으며, 감염율(感染率)이 높은 종자시료(種子試料)는 각각(各各) 최고 84% 및 28%의 감염율(感染率)을 나타내었다. 반면에 C.acutatum과 C.coccodes는 각각 1개의 종자 시료(試料)에서 1% 및 2%의 낮은 비율(比率)로 검출(檢出)되었다. 표준습지법(標準濕紙法)이 냉동습지법(冷凍濕紙法)이나 한천배지배양법(培地培養法)에 비하여 종자상의 탄저병균(炭疽病菌) 검출(檢出)에 효과적(效果的)이었다. C.dematium과 C. gloeosporioides의 종자내 감염부위(感染部位)를 조사(調査)하였던 바, 종피(種皮)뿐 아니라 종자내 깊숙이 배유(胚乳)와 자엽(子葉)에 까지 감염(感染)되어 있음이 확인되었으나 종피(種皮) 감염율(感染率)이 높았다. C.dematium과 C.gloeosporioides 감염종자(感染種子)를 한천배지와 살균토양에 옮겨 발아율과 유묘생육상황(幼苗生育狀況)을 조사한 결과, 감염종자(感染種子)는 종자부패(種子腐敗), 모잘록, 유묘고사(幼苗枯死) 및 자엽(子葉)과 하배축(下胚軸)의 변색(變色)을 초래하였다. 접종실험(接種實驗) 결과 C.acutatum은 고추의 적과(赤果)에 병원성(病原性)이 있었고 C.coccodes는 고추 적과(赤果)에 강한 병원성(病原性)이, 고추 잎에 약한 병원성(病原性)이 있었다. C.dematium은 고추 잎과 청과(靑果)에 강한 병원성(病原性)이 있었고, C.gloeosporioides 는 고추 잎과 청과(靑果) 및 적과(赤果)에 병원성이 있었다. 이들 4종의 종자전염성(種子傳染性) 탄저병균(炭疽病菌)의 고추 이외의 다른 작물(作物)에 대한 병원성도 조사하였다.

Distribution of Seed-borne fungi on Rice Seeds Affecting Rice Grain Qualify

  • Seed-borne fungal pathogens on rice seeds were investigated in order to evaluate their effect on rice grain quality. Rice seeds of two cultivars, Ilmibyeo and Daesanbyeo were collected from 27 areas of Korea and the fungal parasites on seeds were isolated by using a blotter method. Isolation frequency and number of species were varied from collection areas of seed samples. A total 13 species of fungi were identified from the seeds. Among them, Altemana alternata was the most frequent over the country. Bipoiaris oryzae most commonly from Gyeongbuk and Jeonbuk, Alternaria padwickii from Jeonbuk, and Nigrospora oryzae from Choongnam. However Bipolaris. oryzae, Alternaria padwickii, and Nigrospora oryzae were the most frequently isolated fungi from Gyeongbuk and Jeonbuk, and Chungnam, respectively. B. oryzae, A. alternata, A. padwickii, and N. oryzae were dominants on Ilmibyeo showing 10.3%, 10.2%, 5.2%, and 5.2% infection rate, respectively. While, N. oryzae, A alternata, and Cladosporium sp. were most frequently isolated fungi from Deasanbyeo revealing 15.1%, 9.6%, and 7.5% infection rate, respectively. These fungi inhabiting on hulls or endosperms of rice seed might be considered as potential factors decreasing rice grain quality. Further investigation of the fungi on grain rice quality are undergoing.

맥류(麥類)에 발생(發生)하는 Ustilago nuda 및 Pyrenophora graminea에 대한 종자(種子) 소독(消毒)에 관한 연구(硏究) (Control of Seed-borne Infection of Ustilago nuda and Pyrenophora graminea on Barley)

  • 맥류(麥類)의 겉깜부기병(라흑종병(裸黑種病))과 줄무늬병(반엽병(班葉病))에 대한 종자(種子) 소독(消毒) 효과(效果)를 보기 위하여 자연이병(自然罹病)된 종자(種子)에 Baytan외(外) 13종(種)의 엽제(葉劑)를 처리(處理)한 다음 포장(圃場)에 파종(播種)하여 발병상황(發病狀況)을 조사(調査)하였다. Sisthane 및 Benlate T는 겉깜부기병과 줄무늬병에 대하여 Vitathiram과 같은 방제효과(防除效果)를 나타냈으며, Baytan U는 겉깜부기병에는 효과적(效果的)이었으나 줄무늬병에는 효과(效果)가 떨어졌다. P242는 줄무늬병에는 좋은 약효(藥效)를 나타냈으나 겉깜부기병의 발생(發生)을 막는데는 효과(效果)가 적었으며, Busan 30은 겉깜부기병과 줄무늬병에 대하여 Vitathiram에 비해서 효과(效果)가 불안정(不安定)하였다. 홍시약제(洪試藥劑)로 처리한 보리 쌀보리 및 밀을 실내(室內)서에 발아상(發芽箱)에 키웠을 때 Sisthane과 Baytan U의 고농도처리구(高濃度處理區)에서는 뿌리뒤틀림 현상으로 생육(生育)의 저해(沮害)가 나타나지 않았다.

벼 갈색잎마름병균(Gerlachia oryzae)의 종자감염과 종자감염이 벼종자 및 유묘에 미치는 피해 (Seed Infection and Damage to Rice Seeds and Seedlings by Seed-Borne Gerlachia oryzae)

  • 1. 영남작물시험장, 호남작물시험장 및 충남농촌진흥원에서 분양받은 18개의 벼종자시료와 충남지역 일반농가에서 수확한 8개의 벼종자시료 등 모두 26개의 종자시료를 공시하여 표준습지법으로 조사하였던 바 21종의 균류가 검출되었으며, 벼 갈샐잎마름병균(Gerlachia oryzae)은 22개의 벼 종자시료에서 $1.0\~45.0\%$의 범위로 검출되었다. 2. 종자전염성 G. oryzae의 효과적인 검출방법을 찾기 위하여 표준습지법, 냉동습지법, 물한천법 등으로 비교 조사한 결과, 냉동습지법에서 검출율이 가장 높았다. 3. 벼종자의 각 부위를 무균적으로 분리하여 부위별로 G. oryzae의 감염여부를 표준습지법으로 조사한 결과, 벼 종자의 껍질에서 가장 많이 검출되었으며, 심하게 이병된 종자는 배유 및 종피 뿐 만 아니라 배에 까지 감염되어 있었다. 4. G. oryzae에 감염된 벼 종자를 파종하여 심하게 이병된 종자는 종자부패 및 묘입고를 일으켰으며, 약하게 이병된 종자는 초엽, 1엽, 2엽에 갈색의 병징감염을 초래하였다.

Incidence and variability of Hosta virus X and seed-transmission in Hosta plants

  • This study was performed to investigate the incidence of Hosta uirus X (HVX), a Potexvirus, from cultivated hosta ornamental plants in Korea and to ascertain seed transmission of the virus from infected parent plant to progeny ones for breeding program of hosta plants. Infection rate of HVX in cultivated hostas was 25.6 % (11 out of 43 collected samples contained HVX) based on Western blot and RT-PCR detection methods. Most of HVX-infected hostas showed visible systemic leaf symptoms (mosaic, mottle, curling, stunting or combinations). Variability of HVX was confirmed by sequences of coat protein gene of individual isolates from different hostas. HVX was seed-transmitted on Hosta 'Blue Cadet'. The virus was detected from seeds, and sprouts and seedlings from the virus-contaminated seed sources. Over 7.5 % of seeds were HVX-contaminated surveyed in this study, Our data suggest that HVX can be transmitted by seed source, and indexing of the virus should be done for breeding program of Hosta.
