The plasma membrane-localized BRASSINOSTEROID-INSENSITIVE1 (BRI1) and BRI1-ASSOCIATED KINASE1 (BAK1) are a well-known receptor pair involved in brassinosteroids (BR) signaling in Arabidposis. The formation of a receptor complex in response to BRs and the subsequent activation of cytoplasmic domain kinase activity share mechanistic characteristics with animal receptor kinases. Here, we demonstrate that BRI1 and BAK1 are BR-dependently phosphorylated, and that phosphorylated forms of the two proteins persist for different lengths of time. Mutations of either protein abolished phosphorylation of the counterpart protein, implying transphosphorylation of the receptor kinases. To investigate the specific amino acids critical for formation of the receptor complex and activation of BAK1 kinase activity, we expressed several versions of BAK1 in yeast and plants. L32E and L46E substitutions resulted in a loss of binding of BAK1 to BRI1, and threonine T455 was essential for the kinase activity of BAK1 in yeast. Transgenic bri1 mutant plants overexpressing BAK1(L46E) displayed reduced apical dominance and seed development. In addition, transgenic wild type plants overexpressing BAK1(T455A) lost the phosphorylation activity normally exhibited in response to BL, leading to semi-dwarfism. These results suggest that BAK1 is a critical component regulating the duration of BR efficacy, even though it cannot directly bind BRs in plants.
The objective of this study was to rapidly evaluate fatty acids in a collection of millet (Panicum miliaceum subsp. miliaceum) of different origins so that this information could be disseminated to breeders to advance germplasm use and breeding. To develop the calibration equations for rapid and nondestructive evaluation of fatty acid content, near-infrared reflectance spectroscopy (NIRs) spectra (1104-2494 nm) of samples ground into flour ($n$=100) were obtained using a dispersive spectrometer. A modified partial least-squares model was developed to predict each component. For foxtail millet germplasm, our models returned coefficients of determination ($R^2$) of 0.89, 0.89, 0.89, and 0.92 for palmitic acid, oleic acid, linoleic acid, and total fatty acids, respectively. The prediction of the external validation set (n=10) showed significant correlation between references values and NIRs values ($r^2$=0.64, 0.90, 0.79, and 0.89 for palmitic acid, oleic acid, linoleic acid, and total fatty acids, respectively). Standard deviation/standard errors of cross-validation (SD/SECV) values were close to 3 (2.62, 2.40, 1.85, and 2.23 for palmitic acid, oleic acid, linoleic acid, and total fatty acids, respectively). These results indicate that these NIRs equations are functional for the mass screening and rapid quantification of the oleic and total fatty acids characterizing millet germplasm. Among the samples, IT153514 showed an especially high content of fatty acids ($48.14mg\;g^{-1}$), whereas IT123909 had a very low content ($34.44mg\;g^{-1}$).

AT-hook proteins of plant have shown to be involved in growth and development through the modification of chromatin architecture to co-regulate transcription of genes. Recently, many genes encoding AT-hook protein have been identified and their involvement in senescence delay is investigated. In this study, soybean transgenic plants overexpressing chromatin architecture-controlling ATPG7 gene was produced by Agrobacterium-mediated transformation and investigated for the positive effect on the important agronomic traits mainly focusing on yield-related components. A total of 27 transgenic soybean plants were produced from about 400 explants. $T_1$ seeds were harvested from all transgenic plants. In the analysis of genomic DNAs from soybean transformants, ATPG7 and Bar fragments were amplified as expected, 975 bp and 408 bp in size, respectively. And also exact gene expression was confirmed by reverse transcriptase-PCR (RT-PCR) from transgenic line #6, #7 and #8. In a field evaluation of yield components of ATPG7 transgenic plants ($T_3$), higher plant height, more of pod number and greater average total seed weight were observed with statistical significance. The results of this study indicate that the introduction of ATPG7 gene in soybean may have the positive effect on yield components.

These results indicate that these NIRs equations are functional for the mass screening and rapid quantification of the oleic, linolenic, and total fatty acids characterizing foxtail millet germplasm. Among the samples, IT153491 showed an especially high content of fatty acids ($84.06mg\;g^{-1}$), whereas IT188096 had a very low content ($29.92mg\;g^{-1}$).
Plant height is an important component of plant architecture and significantly affects crop breeding practices and yield. We studied DNA variations derived from F5 recombinant inbred lines (RILs) with 96.8% homozygous genotypes. Here, we report DNA variations between the normal and dwarf members of four lines harvested from a single seed parent in an F6 RIL population derived from a cross between Glycine max var. Peking and Glycine soja IT182936. Whole genome sequencing was carried out, and the DNA variations in the whole genome were compared between the normal and dwarf samples. We found a large number of DNA variations in both the dwarf and semi-dwarf lines, with one single nucleotide polymorphism (SNP) per at least 3.68 kb in the dwarf lines and 1 SNP per 11.13 kb of the whole genome. This value is 2.18 times higher than the expected DNA variation in the F6 population. A total of 186 SNPs and 241 SNPs were discovered in the coding regions of the dwarf lines 1282 and 1303, respectively, and we discovered 33 homogeneous nonsynonymous SNPs that occurred at the same loci in each set of dwarf and normal soybean. Of them, five SNPs were in the same positions between lines 1282 and 1303. Our results provide important information for improving our understanding of the genetics of soybean plant height and crop breeding. These polymorphisms could be useful genetic resources for plant breeders, geneticists, and biologists for future molecular biology and breeding projects.
Adzuki beans have gained popularity in recent years due to their health benefits. Breeding of Adzuki beans is less favorable than with other legumes due to low genetic diversity. This study aimed to evaluate the genetic diversity of 252 adzuki bean germplasms from China, Japan, and Korea using 18 agro-morphological parameters and comparing their performance to three prominent Korean cultivars: Geomguseul, Arari, and Chungjupat. Leaf shape, pod color, and seed coat color were among the qualitative traits that showed wide variations. The quantitative variables also showed wide variations among adzuki bean germplasms. Although there was no significant difference (p < 0.05), the average rate of germination declined in the order of Korean (91.44%) > Chinese (91.31%) > Japanese (87.47%) adzuki beans. Chinese adzuki beans needed fewer days to flower (DF, 58.22 days) and days to mature (DM, 107.13 days), which varied significantly compared to the Korean and Japanese adzuki beans (p < 0.05). The average number of pods per plant (PPP) and one-hundred seeds weight (HSW) were higher in Japanese adzuki beans compared to the Korean and Chinese adzuki beans although the variation of each was not significant. Almost 29.76% of the accessions had early-blooming flowers, 3.97% were premature, 21.43% produced more PPP, and 3.97% yielded more SPP compared to control cultivars. Results of hierarchical cluster and principal component analyses revealed three clusters with significant variation in all quantitative variables except for RG (p < 0.05). The key factors in multivariate analyses were DF, DM, and HSW. Our study investigated the genetic diversity of adzuki bean accessions and identified ten early maturing and ten high PPP-yielding accessions. Our findings would help farmers and breeders to select the top-performing accessions that can provide them with various options.
Traditional germplasms are unsuitable for mechanized production, limiting adzuki bean production. The creation of cultivars that can be harvested by mechanized means is an urgent task for breeders. The bottom pod height (BPH), lodging resistance, and synchronous maturing of adzuki beans are critical factors for the reduction of losses due to mechanized harvesting. In this study, 14 traits of 806 adzuki bean accessions were analyzed. All growth stages and the yield, lodging score, and synchronous maturing correlated negatively with the BPH. These negative correlations reflect the increased difficulty of breeding to simultaneously satisfy the needs for no lodging, high synchronous maturing rates, BPHs > 10 cm, and high yield. We screened three germplasms with no lodging, high synchronous maturing rates, and BPHs > 10 cm that were used as mechanization-adapted breeding material for crossing with high-yield cultivars. Agronomic trait diversity in adzuki beans was also examined in this study. Principal component and cluster analyses were conducted for 806 germplasms resulting in three clusters with the yield and three growth stage traits serving as the main discriminating factors. Cluster 1 included high-yield germplasms with the number of pods per plant and the number of seeds per pod being the major discriminant factors. Cluster 2 included germplasms with long growth periods and large 100-seed weights while cluster 3 contained germplasms with high BPHs. In general, the characteristics that make mechanical harvesting feasible and those assessed in this study could be utilized to choose and enhance adzuki beans production.

This study selected 9 species of domestic native plants (Baechohyang, Red Pine, Turmeric, Ginger, San parsley seed, Bulsugam, Hwangchil, Tangja, Valerian) that have been proven effective for depression, and based on the analysis results of each fragrance component, 9 species were selected. Antioxidant (DPPH, ABTS), cytotoxicity (MTS), and anti-inflammatory (Nitric oxide) experiments were performed by combining essential oils. As a result of the analysis of fragrance components, DL-Limonene (38.44%), g-Terpinene (8.9%), Estragole (5.18%), and a-Pinene (1.73%), which were previously studied in combination oil, were identified. DPPH radical scavenging ability, which is an antioxidant activity, showed 75.4%, ABTS radical scavenging ability was 74.04%, and NO production inhibition was 33.14% at a concentration of 5uL/ml with no cytotoxicity confirmed. Through this, the effect of domestic blending essential oils on the improvement or prevention of depression is verified, and scientific efficacy and ingredient studies are conducted in a mutually cooperative manner to seek solutions to depression and provide basic data to confirm whether or not depressive symptoms are improved. want to do.
Wolfiporia cocos is a wood-decay brown rot fungus belonging to the family Polyporaceae. While the fungus grows, the sclerotium body of the strain, dubbed Bokryeong in Korean, is formed around the roots of conifer trees. The dried sclerotium has been widely used as a key component of many medicinal recipes in East Asia. Wolfiporia cocos strain KMCC03342 is the reference strain registered and maintained by the Korea Seed and Variety Service for commercial uses. Here, we present the first draft genome sequence of W. cocos KMCC03342 using a hybrid assembly technique combining both short- and long-read sequences. The genome has a total length of 55.5 Mb comprised of 343 contigs with N50 of 332 kb and 95.8% BUSCO completeness. The GC ratio was 52.2%. We predicted 14,296 protein-coding gene models based on ab initio gene prediction and evidence-based annotation procedure using RNAseq data. The annotated genome was predicted to have 19 terpene biosynthesis gene clusters, which was the same number as the previously sequenced W. cocos strain MD-104 genome but higher than Chinese W. cocos strains. The genome sequence and the predicted gene clusters allow us to study biosynthetic pathways for the active ingredients of W. cocos.

Tuber yield and content of general component and diosgenin which is a main bioactive property were investigated in order to determine the growth characteristics of round typed yam(Dungeun-ma, Dioscorea opposita L.) and the potential of artificial culture at Suwon, Korea. Sprouting rates of segmented small tubers such as 3, 5, 7, 9, and 12g were respectively 60.3, 80.5, 85.3, 98.6 and 99.3% when they were investigated 30 days after planting. Segmented small tubers of 3, 5, 7, 9, and 12g enlarged respectively to 66, 87, 98, 120, and 140g in the green-house cultivation. Segmented small tubers of 3, 7, 12, 18, and 23g enlarged respectively to 35.2, 124.7, 142.7, 174.8, and 200,7g in the open-air field cultivation.
