Stimulation of Actinorhodin Production by Streptomyces lividans with Chromosomally-Integrated Antibiotic Regulatory Gene, afsR2

  • Streptomyces lividans is one of the most commonly-used streptomyetes strain as a molecular cloning and expression host. Unlike its close relative S. coelicolor, however, S. lividans rarely produces secondary metabolite such as actinorhodin in a typical glucose-containing culture condition due to insufficient expression of some antibiotic regulatory genes including afsR2. Although multiple copies of afsR2 or a glycerol-specific culture condition stimulated actinorhodin production in S. lividans, both failed to stimulate actinorhodin production in S. lividans cultured in a typical glucose-containing medium. To generate a culture-condition-independent actinorhodin-overproducing S. lividans strain the afsR2 gene was integrated into the S. lividans TK21 chromosome via homologous recombination, followed by the genetic confirmation. This S. lividans strain produced a significant amount of actinorhodin in both glucose-containing liquid and plate cultures, with higher actinorhodin productivity compared to the S. lividans containing multiple copies of afsR2. These results suggest that a chromosomal integration of a single copy of an antibiotic regulatory gene is a promising method for the development of a stable antibiotic-overproducing streptomycetes strain.

A Dispatching Method for Automated Guided Vehicles to Minimize Delays of Containership Operations

  • There is a worldwide trend to automate the handling operations in port container terminals in an effort to improve productivity and reduce labor cost. This study iscusses how to apply an AGV(automated guided vehicle) system to the handling of containers in the yard of a port container ter-minal. The main issue of this paper is how to assign tasks of container delivery to AGVs during ship operations in an automated port container terminal. A dual-cycle operation is assumed in which the loading and the discharging operation can be performed alternately. Mixed integer linear program-ming formulations are suggested for the dispatching problem. The completion time of all the dis-charging and loading operations by a quayside crane is minimized, and the minimization of the total travel time of AGVs is also considered as a secondary objective. A heuristic method using useful properties of the dispatching problem is suggested to reduce the computational time. The perfor-mance of the heuristic algorithm is evaluated in light of solution quality and computation time.

  • A remarkable improvement of the productivity in barium titanate by slurry spray pyrolysis process was realized by supplying solid source slurry into the rector. The produced barium titanate powders showed uniform powder properties, and reproducibility with higher tetragonality in the range of 80$\sim$200 nm, case by case. The secondary calcination experiments of the as-prepared powders by spray pyrolysis revealed that the powders as-prepared over 700$^{\circ}C$ showed perfectly different behavior with the lower temperature's ones and the solid state reaction’s case. The result was discussed in terms of the reaction mechanism based on the activation energy analysis.

  • Insert-injection molding can inject two different materials or two colors in the same mold and process. If this injection process use, product has ability because the base part maintain strength and specified part can inject soft-material. It makes the cost down by single operation automatically for saving wages. In this paper, we designed double-injection mold for automobile remote control to inject secondary using this part as insert after inject external appearance of product. CAE analysis was progressed gate location and runner size as variable and analysis result is reflected in mold design process. As a result, it could solved badness that is generated at the conventional mold. Additionally, cost is downed by reducing loss of runner as well as could omit painting process because surface of finished product is improved through new mold.

Nutrient removal from secondary effluent using filamentous algae in raceway ponds

  • In this study, we investigated the cultivation possibility using Hydrodictyon reticulatum in a continuous raceway pond as a tertiary sewage treatment plant. The cultivation possibility was evaluated by varying the light quantity, wavelength, and hydraulic retention time (HRT). Experimental results showed that the growth rates of algae and the removal efficiencies of nutrients increased as the light quantity increased, and the maximum photosynthetic rate was maintained at $100{\mu}mol/m^2{\cdot}s$ or higher. When wavelength was varied, nutrient removal efficiency and growth rate increased in the following order: green light, red light, white light, and blue light. The nutrient removal efficiencies and algae productivity in HRT 4 d were better than in HRT 8 d. We conclude that if Hydrodictyon reticulatum is cultivated in a raceway pond and used as a tertiary treatment facility in a sewage treatment plant, nutrients can be effectively removed, and production costs can be reduced.

  • A ceramic vacuum chamber feedthrough ceramic insulator is made of Al2O3; the manufacturing process involves filling alumina powder into a urethane mold and pressing it with a rubber press to produce a molded body. Thereafter, manufacturing is completed through primary shape processing, sintering, and secondary shape processing in the green body, which is a pressurized molding body, This work is intended to prevent defects in the first shape processing by improving the ceramic insulator in the green body, and to improve the productivity of the ceramic insulator by determining the optimal processing conditions.

  • Cyclosporin, an immunosuppressant, is a representative group of biologically active secondary metabolites produced by the fungus Tolypocladium inflatum. The amount and ratio of cyclosporin derivatives in the culture broth are an important factors for the production of cyclosporin A and the purification in the industrial process. Therefore, we studied the effect of amino acids and complex organic nitrogen sources using Tolypocladium inflatum mutants on the productivity of cyclosporin A and the ratio of cyclosporin derivatives. Overproducing mutant YHC-004 having seven times higher productivity than mother strain's could be obtained through the artificial mutation by UV irradiation. The concentration and kind of organic nitrogens and amino acids shows the profound effect on the productivity of cyclosporin A and ratio of cyclosporin derivatives. As a result, it was possible to raise the productivity and the ratio of cyclosporin A up to 3,430 mg/L and 93% respectively, but on the other hand the other cyclosporin derivatives decreased less than 2% in the culture broth.

Enhanced Production of Shikonin by Using Polyurethane-entrapped Lithospermum erythrorhizon Cells (Polyurethane Foam 에 포괄시킨 Lithospermum erythrorhizon 세포에 의한 Shikonin 생산)

  • Production of shikonin derivatives by Lithospermum erythrorhizon cells by using polyurethane foam was invesliigated. Shikonin derivatives were effectively adsorbed mostly by phase distribution to polyurethane matrices and their production increased significantly compared to the suspension culture. The enhanced production of shikonin was probably due to more facilitated cell to cell con-tact and lowered intracellular shikonin concentration, both of which are known to be favorable for plant secondary metabolite production. In order to improve the process productivity, tell culture was conducted under various culture conditions: Of them, Schenk and Hildebrandt medium containing indole-3-acetic acid (1.75mg/ι) and kinetin (0.1mg/ι) was considered most appropriate for shikonin production. Production of shikonin increased about 4.5 times in the Schenk and Hildebrandt medium containing indole-3-acetic acid (1.15mg/ι) and kinetin (0.1mg/ι) when compared to the same medium containing p-chlorophenoxyacetic acid (2.0mg/ι) and kinetin (0.1mg/ι). When poly-urethane was used as the support material, a single-stage system was more preferred to the conventional two-stage culture system in terms of shikonin productivity.

Computerization of Nurse Staffing and Scheduling according to Patient Classification (환자분류에 의한 간호인력 산정 및 배치과정 전산화)

  • Even though Korean medical law stipulates that number of patients attended by a nurse is 2.5 for hospitalization and 30 for ambulatory care, the number of patients cared by a nurse per day is much greater than the standard prescribed by the medical law. Current nursing productivity of nurses is not desirable unless the quality of care considered. Moreover. nursing manpower staffing based on neither current nurses' productivity nor standard of medical law cannot respond properly to dynamic situation of the medical services. As for the nurse scheduling, the critical problem of it in the hospital is determining the day-to-day shift assignments for each nurse for the specified period in a way that satisfies the given requirements of the hospital. Nurse scheduling, however, involves many factors and requirements, manual scheduling requires much time and effort to produce an adequate schedule. Under these backgrounds, the necessity of more efficient management of nursing manpower occupying 1/3 of total hospital workers has been recognized by many nursing administrators. This study was performed to develop a system computerizing nurse staffing and scheduling based on the patient classification. As a preliminary step for the system development, nursing workload in a secondary hospital was measured from Sep. to Oct. 1994. On the grounds of this result, computerization of nurse staffing and scheduling was proceeded with three options. First one is based on the current medical law. Second one is based on the assigned number of nursing staff. And the last is based on the request by patient classification. Computer languages used in this study were MS Visual Basic 3.0 for the staffing and Access 2.0 for the scheduling, respectively. Prospective users may operate this system easily because icons and mouse are used for easier graphic user interface and reducing the need for typing efforts. This system can help nurse administrators manage nursing manpower efficiently and nurses develop quick and easy schedule generation and allow more time for the patient care.

A Bioactive Fraction from Streptomyces sp. Enhances Maize Tolerance against Drought Stress

  • Drought stress is threatening the growth and productivity of many economical crops. Therefore, it is necessary to establish innovative and efficient approaches for improving crop growth and productivity. Here we investigated the potentials of the cell-free extract of Actinobacteria (Ac) isolated from a semi-arid habitat (Al-Jouf region, Saudi Arabia) to recover the reduction in maize growth and improve the physiological stress tolerance induced by drought. Three Ac isolates were screened for production of secondary metabolites, antioxidant and antimicrobial activities. The isolate Ac3 revealed the highest levels of flavonoids, antioxidant and antimicrobial activities in addition to having abilities to produce siderophores and phytohormones. Based on seed germination experiment, the selected bioactive fraction of Ac3 cell-free extract (F2.7, containing mainly isoquercetin), increased the growth and photosynthesis rate under drought stress. Moreover, F2.7 application significantly alleviated drought stress-induced increases in H2O2, lipid peroxidation (MDA) and protein oxidation (protein carbonyls). It also increased total antioxidant power and molecular antioxidant levels (total ascorbate, glutathione and tocopherols). F2.7 improved the primary metabolism of stressed maize plants; for example, it increased in several individuals of soluble carbohydrates, organic acids, amino acids, and fatty acids. Interestingly, to reduce stress impact, F2.7 accumulated some compatible solutes including total soluble sugars, sucrose and proline. Hence, this comprehensive assessment recommends the potentials of actinobacterial cell-free extract as an alternative ecofriendly approach to improve crop growth and quality under water deficit conditions.
