Molecular cloning and sequence Analysis of the Gene for SecY from Streptomyces coelicolor (Muller) (Streptomyces coelicolor에서 secY 유전자의 클로닝과 염기서열 결정)

  • SecY is a central component of the protein export machinery that mediate the translocation of secretory proteins across the plasma membrane of Escherichia coli. In order to study the mechanism of protein secretion in Streptomyces, we have done cloning and sequencing of the Streptomyces coelicolor secY gene by using polymerase chain reaction method. The nucleotide sequence of the gene for SecY from S. coelicolor showed over 58% identity to that of M. luteus. The deduced amino acid sequences were highly homologous to those of other known SecY polypeptides, all having the potential to form 10 transmembrane segments, and especially second, fifth, and tenth segments were particularly conserved, sharing greater than 75% identity with W. lute s SecY. We propose that the conserved membrane-spanning segments actively participate in protein export. In B. subtilis and E. coli, the secY gene is a part of the spc operon, is preceded by the gene coding for ribosomal protein L15, and is likety coupled transcriptionally and translationally to the upstream L15 gene. In the other hand, secY gene of S. coelicolor and M. luteus have its own promoter region, are coupled translationally with adk gene and pr sented in adk operon.

Streptomyces lividans에서 secE 유전자의 클로닝과 염기서열 결정

  • The secE gene of Streptomyces lividans TK24 was cloned by the polymerase chain reaction method with synthetic oligonucleo- tide primers designed on the basis of the nucleotide sequences of Streptomyces coelicolor secE-nusG-rplK operon. The deduced amino acid sequences of the SecE were highly homologous to those of other known SecE protein, that is 36.8%, 30.4%, 80.0%, and 80.9%, similarity to E. coli, Bacillus subtilis, Streptomyces griseus, Streptomyces virginiae SecE, respectively and exactly same with Streptomyces coelicolor SecE. It means that in spite of evolutionary differences, the genes for protein translocation machinery are highly conserved in eubacteria. The gene organization of secE-nusG-rplK is also similar to that of E. coli, B. subtilis, and streptomycetes.

  • Thickness of intermetallic compounds and consumption rates of under bump metallurgies (UBMs) were investigated in wafer-level solder bumping with variations of UBM materials and reflow times. In the case of Cu UBM, $0.6\;{\mu}m-thick$ intermetallic compound layer was formed before reflow of Sn solder, and the average thickness of the intermetallic compound layer increased to $4\;{\mu}m$ by reflowing at $250^{\circ}C$ for 450 sec. On the contrary, the intermetallic layer had a thickness of $0.2\;{\mu}m$ on Ni UBM before reflow and it grew to $1.7\;{\mu}m$ thickness with reflowing for 450 sec. While the consumption rates of Cu UBM were 100nm/sec fur 15-sec reflow and 4.50-sec for 450-sec reflow, those of Ni UBM decreased to 28.7 nm/sec for 15-sec reflow and 1.82 nm/sec for 450-sec reflow.

Energy Absorption Characteristics for Spot Welded Hat-shaped Section Members at Various Velocities (속도변화에 따른 점용접된 모자형단면부재의 에너지흡수 특성)

  • Front-end side members of vehicles are structures with the greatest energy absorbing capability in a front-end collision of vehicles. This paper was undertaken to analyze the energy absorption characteristics of spot welded hat and double hat-shaped section members under the axial collapse. The experiments were performed with respect to the various collapse velocities. It was expected that para-closed sections would show collapse characteristics which be quite different from those of perfectly closed sections. The collapse velocities were selected as follows: the velocities in the hat-shaped section members were 0.00017m/sec, 0.017m/sec, 4.7m/sec, 6.5m/sec, 6.8m/sec, 7.2m/sec, and 7.3m/sec those in the double hat-shaped section members were 0.00017m/sec, 0.017m/sec, 6.5m/sec, 6.8m/sec, 7.2m/sec 7.3m/sec, and 7.9m/sec. In the program system presented in this study, an explicit finite element code, LS-DYNA3D, is adopted for simulating complicated collapse behavior of the hat and double hat-shaped section members under the same condition of the collapse test. The validity of simulation was confirmed by the comparison between the simulation result and the collapse experiment.

A Study on Effects of the Transient Compression by Tightly Tied Necktie on Blood Flow in the Internal Jugular Veins Using 2D-PC MRA (2D-PC MRA를 이용한 넥타이의 순간적인 압박(壓迫)이 내경정맥의 혈류에 미치는 영향에 대한 연구(硏究))

  • The 25 healthy male volunteers aged from 20 to 50years old have been employed in this study. 2D-PC MRA was performed to measure the velocity of the blood flow in the internal carotid artery and internal jugular veins using 3.0T MRI Whole body (signa VH/i GE). ECTRICKS-CEMRA was performed to evaluate the pattern of blood circulation from internal carotid artery to internal jugular vein. Using 2D-PC MRA, the cross-section of the 4th and 5th cervical discs was scanned with 24cm FOV. Then the speed of blood flow was measured for internal carotid artery and internal jugular vein when the subject wears a necktie tightly and no tie. The average of maximum velocity of internal carotid arteries without a necktie was 72.13cm/sec in the right side and 74.96cm/sec in the left side(average 73.54cm/sec in both sides) while the average of maximum velocity of internal jugular veins without a necktie was -34.45cm/sec in the right side and -24.99cm/sec in the left side (-29.72cm/sec in both sides). However, when wearing a necktie tightly, the average of maximum velocity of internal carotid arteries was 61.35cm/sec in the right side and 65.19cm/sec in the left side(average 63.27cm/sec in both sides) while the average of maximum velocity of internal jugular veins was -22.14cm/sec in the right side and -17.93cm/sec in the left side(-20.03cm/sec in both sides). With the necktie tightly knotted, the average blood flow speed of both internal carotid arteries slightly decreased to 86% (63.27/73.54cm/sec) compared to no tie case in which both internal jugularveins significantly went down to 67% (-20.03/-29.72 cm/sec). Thus it is suggested that wearing a necktie affects the circulation of internal jugular veins(33% decrease in blood flow speed) more significantly than that of internal carotid artery(14% decrease in blood flow speed). Without a necktie, ECTRICKS-CEMRA showed natural blood circulation patterns of internal carotid arteries and internal jugular veins without any disturbances or compressions. However, when wearing a necktie tightly, ECTRICKS-CEMRA showed severe compression onto both internal jugular veins in all 25 volunteers. In conclusion, the result of the study showed that the tightly worn necktie instantly presses more internal jugular veins than internal carotid arteries, thereby significantly reducing the blood flow speed and leading to the temporary occlusion. Thus, the defecation or washing the face under the tightly tied necktie situations can cause the unexpected and temporary compression or occlusion of the internal jugular veins, subsequently leading to the occurrences of the stroke due to the secondary intracranial venous hypertension.

The Effects of Isokinetic Exercise Program on the Muscle Strength and Blood Constituent by the Relatively Angular Velocities

  • The purpose of this study was to investigate the effects of angular velocity on muscle strength and blood constituent. Subjects was classified into two groups, which were $60^{\circ}/sec$ angular velocity group (n=8), and $240^{\circ}/sec$ angular velocity group (n=8). Each group was applied to perform the isokinetic exercises on extensor muscle group for each 7 times in 10 set (3 days per a week for 3 weeks). Muscle strength was measured using peak torque of quadriceps femoris and blood constituent was measured using RBC, WBC and Hb. The peak torque was more significantly increased after 3 weeks application of $60^{\circ}/sec$ angular velocity group than $240^{\circ}/sec$ angular velocity group. The RBC and Hb were more significantly increased after 3 weeks application of $60^{\circ}/sec$ angular velocity group than $240^{\circ}/sec$ angular velocity group. But WBC was more significantly increased after 3 weeks application of $240^{\circ}/sec$ angular velocity group than $60^{\circ}/sec$ angular velocity group. These results indicate that $60^{\circ}/sec$ angular velocity isokinetic exercise application were effective treatment strategy on increase of muscle strength. But $240^{\circ}/sec$ angular velocity isokinetic exercise application were effective treatment strategy on immune protect system.

An Analysis of the down stream impact of dam Characteristics of Dam Using Indicators of Hydrologic Alterations (수문변화지표법을 이용한 댐 특성요소별 하류하천에 미치는 영향분석)

  • 본 연구에서는 수문변화 지표법을 적용하여 광동댐 등 12개 용수 전용댐의 연 최소 최대유량 크기와 지속기간, 고 저맥파 빈도와 주기, 수문곡선 변화 비율과 빈도, 유량변화정도 등을 분석하여 댐이 하류하천에 미치는 영향을 수치로 제시 하였다. 광동댐 등 12개 댐의 월 유량변화를 산정한 결과, 갈수기에 해당하는 10월부터 다음해 6월까지 8개월간의 댐건설전 월평균 방류량은 $0.64\;m^3/sec{\sim}3.05\;m^3/sec$이었으나, 댐건설후 월평균 방류량은 $0.33\;m^3/sec{\sim}2.99\;m^3/sec$$-69.7%{\sim}167.8%$의 분포를 나타내고 있으며, 12월과 1월을 제외하면 댐건설 이후 오히려 $6.8%{\sim}69.7%$가 감소하였다. 우기철인 7월부터 9월까지 3개월간은 댐건설전 월평균 방류량이 $6.82\;m^3/sec{\sim}7.67\;m^3/sec$이었으나, 댐건설후 월평균 방류량은 $4.12\;m^3/sec{\sim}4.35\;m^3/sec$$51.0%{\sim}55.1%$가 감소하여 저수지 공용량을 이용한 저류효과가 다소 있었던 것으로 분석되었다. 연 최소 최대유량 크기와 지속기간 분석결과 1일, 3일, 7일, 30일, 90일 최소유량은 광동댐 등 10개 댐이 댐건설 전후 모두 $0.0\;m^3/sec$로 증감이 없었으며, 달방댐의 경우 30일 최소유량이 $0.026\;m^3/sec$에서 $0.000\;m^3/sec$로 99.11%가 감소하였다. 영천댐의 경우도 30일 최소유량이 $0.101\;m^3/sec$에서 $0.067\;m^3/sec$로 33.73%가 감소 하였으며, 90일 최소유량의 경우 달방댐은 $0.067\;m^3/sec$에서 $0.031\;m^3/sec$로 53.48%가 영천댐은 $0.219\;m^3/sec$에서 $0.150\;m^3/sec$로 31.29%가 감소하였다. 12개 용수 전용댐중 유지용수공급이 계획되어 있는 영천댐과 달방댐을 제외하고는 댐하류 방류량이 없이 건천화 되었다는 사실을 알 수 있다. 고 저맥파의 빈도와 주기 분석결과 댐건설 이후 수문곡선이 25% 이하로 떨어지는 저맥파는 $72.99%{\sim}98.52%$가 감소하였으며, 저맥파의 기간은 $-80.0%{\sim}4,423.3%$까지 분포하여 상당히 증가하였다. 수문곡선이 75%를 초과하는 고맥파의 발생 횟수는 댐건설 이후 $34.28%{\sim}91.67%$가 감소하였으며, 고맥파의 기간은 $-64.31%{\sim}135.62%$로 증가 또는 감소하였다.

Resistance Curves of Concrete CLWL-DCB Specimens (콘크리트 CLWL-DCB 시험편의 저항곡선)

  • The resistance curves (R-curves) for 381 m crack extension of CLWL-DCB specimens had been determined. The average velocities of the crack extension measured with strain gages were 0.70 and 55 ㎜/sec. The measured rotation angle of the notch faces showed the existence of the singularity at least before 171 and 93 mm crack extensions for the 0.70 and 55 ㎜/sec crack velocities, respectively. The maximum slopes of the R-curves occurred between 25 and 89 ㎜ crack extensions for 0.70 ㎜/sec crack velocity and between 51 and 127 ㎜ crack extensions for 55 ㎜/sec crack velocity During the maximum slopes of the R-curves, the micro-crack localization can be expected, and faster crack velocity may form longer micro-cracking and micro-crack localizing zones. The fracture resistance of 0.70 ㎜/sec crack velocity reached a roughly constant maximum value of 143 N/m at 152 ㎜ crack extension, while that of 55 ㎜/sec crack velocity increased continuously to 245 N/m at 254 ㎜ crack extension and then decreased to the value of 0.70 ㎜/sec crack velocity. The R-curve of 55 ㎜/sec crack velocity was similar to that of the small size three-point bend test, and it showed that small size specimen or fast crack velocity could cause more brittle behavior.

Estimation of Boundary Shear Velocities from Tidal Current in the Gyeonggi Bay, Korea (한국 경기만에서 조류자료에 의한 경계면 전단속도 산출)

  • From tidal current measurements on a tidal sand ridge in the Gyeonggi Bay from August 24 to September 29, 1987, tidal current velocities at 1.0 m above bottom (U/SUB 100/) and boundary shear velocities (U/SUB */) are calculated. The mean speeds of tidal current for flood and ebb over the entire period are 56.3 cm/sec and 63.7 cm/sec in mid-depth (9.0 m above bottom), and 43.9 cm/sec and 43.8 cm/sec in near-bottom (1.5 m above bottom). The exponent(P) in "power law", which is generally used for extrapolation from the mid-depth current velocity to that at the top of nationally logarithmic layer, is estimated to be 0.15 in the study area. Using logarithmic velocity profile assumption, mean values of U/SUB 100/ and U/SUB */ are calculated to be 41.4 cm/sec and 2.39 cm/sec, respectively. The mean value of U/SUB */ (2.39 cm/sec) is much higher than the critical shear velicity (U/SUB *c/) of 1.40 cm/sec reported by Choi (1990). and thus, it can be suggested that the most of sands on the tidal sand ridge in the study area are easily eroded and transported for the greater part of tidal period.

Luminescent Patterns of the Firefly Hotaria papariensis from Korea (한국산 파파리반딧불이의 발광양상)

  • Luminescent patterns of the firefly Hotaria papariensis were classified into routine flashing and courtship flashing, and they were analyzed into duration and period. Flash period of male was changed from 1.26 sec (routine flashing) to 1.12 sec (courtship flashing) and the female's was changed from 2.99 sec to 1.06 sec in this species. Duration time of male increased approximately 1.4-fold from 0.12 sec (routine flashing) to 0.17 sec (courtship flashing) and period of male decreased 0.88-fold from 1.26 sec (routine flashing) to 1.12 sec (courtship flashing). Duration of female increased about 1.5-fold from 0.15. sec (routine flashing) to 0.19 sec (courtship flashing) and period of female decreased almost 0.35 from 2.99 sec (routine flashing) to 1.06 sec (courtship flashing). Hertz frequencies of them were 0.8 (routine flashing of male), 0.9 (courtship flashing of male), 0.3 (routine flashing of female) and 0.9 (courtship flashing of female), respectively. The spectral emission of H. papariensis was detected at 400 nm to 700 nm, and the highest peak wave length of this species was approximately 600 nm, but some 500 to 600 nm wave lengths were present.
