Analysis of oligosaccharides from Panax ginseng by using solid-phase permethylation method combined with ultra-high-performance liquid chromatography-Q-Orbitrap/mass spectrometry

  • Background: The reports about valuable oligosaccharides in ginseng are quite limited. There is an urgent need to develop a practical procedure to detect and analyze ginseng oligosaccharides. Methods: The oligosaccharide extracts from ginseng were permethylated by solid-phase methylation method and then were analyzed by ultra-high-performance liquid chromatography-Q-Orbitrap/MS. The sequence, linkage, and configuration information of oligosaccharides were determined by using accurate m/z value and tandem mass information. Several standard references were used to further confirm the identification. The oligosaccharide composition in white ginseng and red ginseng was compared using a multivariate statistical analysis method. Results: The nonreducing oligosaccharide erlose among 12 oligosaccharides identified was reported for the first time in ginseng. In the comparison of the oligosaccharide extracts from white ginseng and red ginseng, a clear separation was observed in the partial least squares-discriminate analysis score plot, indicating the sugar differences in these two kinds of ginseng samples. The glycans with variable importance in the projection value large than 1.0 were considered to contribute most to the classification. The contents of oligosaccharides in red ginseng were lower than those in white ginseng, and the contents of maltose, maltotriose, maltotetraose, maltopentaose, maltohexaose, maltoheptaose, maltooctaose, maltononaose, sucrose, and erlose decreased significantly (p < 0.05) in red ginseng. Conclusion: A solid-phase methylation method combined with liquid chromatography-tandem mass spectrometry was successfully applied to analyze the oligosaccharides in ginseng extracts, which provides the possibility for holistic evaluation of ginseng oligosaccharides. The comparison of oligosaccharide composition of white ginseng and red ginseng could help understand the differences in pharmacological activities between these two kinds of ginseng samples from the perspective of glycans.

  • In this paper, signed-rank based nonparametric detectors are used for direct sequence code division multiple access pseudo-noise code acquisition systems in frequency-selective Rician fading channels. We first derive the locally optimum rank detector, and then propose the locally suboptimum rank (LSR) and k-th order modified signed-rank (MSRk) detectors using approximate score functions. We compare the serial and hybrid parallel double-dwell schemes using the LSR and MSRk detectors with those using the conventional squared-sum (SS) using the cell averaging constant false alarm rate processor and modified sign detectors. From the simulation results, it is shown that the LSR and MSRk detectors perform better than the SS detector using the cell averaging constant false alarm rate processor.

Performance Improvement in Speech Recognition by Weighting HMM Likelihood (은닉 마코프 모델 확률 보정을 이용한 음성 인식 성능 향상)

  • In this paper, assuming that the score of speech utterance is the product of HMM log likelihood and HMM weight, we propose a new method that HMM weights are adapted iteratively like the general MCE training. The proposed method adjusts HMM weights for better performance using delta coefficient defined in terms of misclassification measure. Therefore, the parameter estimation and the Viterbi algorithms of conventional 1:.um can be easily applied to the proposed model by constraining the sum of HMM weights to the number of HMMs in an HMM set. Comparing with the general segmental MCE training approach, computing time decreases by reducing the number of parameters to estimate and avoiding gradient calculation through the optimal state sequence. To evaluate the performance of HMM-based speech recognizer by weighting HMM likelihood, we perform Korean isolated digit recognition experiments. The experimental results show better performance than the MCE algorithm with state weighting.

A Study on Parenting Stress of Disabled Children's Fathers in IT era (IT 시대 장애아동 아버지의 양육 스트레스에 관한 연구)

  • The purpose of this study is to investigate the parental distress of disabled children's fathers. The survey was conducted targeting 50 fathers with disabled children who are under 13 years old and undergoing rehabilitation treatment in G city And 38 questionnaires excluding data with inadequate answer were adopted. The questionnaire was composed of general peculiarities of disabled children and their fathers, and K-PSI-SF of parents. The results are as followings. First of all, among the parenting stress of disabled children's fathers, parental distress got the highest score and difficult child and dysfunctional interaction are next in sequence in the sub items of K-PSI-SF. Secondly, fathers' stress upon the children's general characteristics showed the significant difference depending on diagnostic categories and paralysis area. Thirdly, fathers' stress upon fathers' general characteristics showed the significant difference depending on their education level and age.

  • In this paper we present a clustering procedure based on the use of HMM in order to get multiple statistical models which can well absorb the variants of each speaker with different ways of saying words. The HMM-clustered models obtained from the developed technique are applied to the speaker independent isolated word recognition. The HMM clustering method splits off all observation sequences with poor likelihood scores which fall below threshold from the training set and create a new model out of the observation sequences in the new cluster. Clustering is iterated by classifying each observation sequence as belonging to the cluster whose model has the maximum likelihood score. If any clutter has changed from the previous iteration the model in that cluster is reestimated by using the Baum-Welch reestimation procedure. Therefore, this method is more efficient than the conventional template-based clustering technique due to the integration capability of the clustering procedure and the parameter estimation. Experimental data show that the HMM-based clustering procedure leads to $1.43\%$ performance improvements over the conventional template-based clustering method and $2.08\%$ improvements over the single HMM method for the case of recognition of the isolated korean digits.

Novel SLC5A2 Mutations and Genetic Characterization in Korean Patients with Familial Renal Glucosuria

  • Purpose: Familial renal glucosuria (FRG, OMIM #233100) is a rare but relatively benign genetic condition characterized by persistent isolated glucosuria with a normal blood glucose level. We report three additional SLC5A2 mutations and examine their phenotypic and genetic characteristics in a Korean FRG cohort. We also reviewed the literature and summarized the genotypes of all Korean patients with FRG. Methods: A genetic analysis was conducted by directly sequencing all 14 exons of the SLC5A2 gene and their flanking regions in six unrelated Korean children with FRG and their family members. Novel non-synonymous single-nucleotide polymorphisms were identified and compared with known mutations that are repeatedly detected in the Korean population. Results: We found two novel mutations [c.274G>A (G92S) and c.1168C>T (L390F)] and one known [c.1382G>A (S461N)] mutation in each family and one recurrent mutation [c.1346G>A (G449D) (rs768392222)] in two pedigrees. The recurrent G449D was predicted to be "possibly damaging," with a score of 0.883 in Polyphen-2, while G92S, L390F, and S461N were predicted to be "probably damaging," with scores of 1.000, 0.999, and 0.996, respectively. Conclusions: Two novel, one previously reported, and one recurrent mutation were identified in six Korean FRG pedigrees as causative mutations of renal glucosuria. Sequence variations in the SLC5A2 gene were frequently detected in children with persistent isolated glucosuria. A long-term follow-up of this FRG cohort is needed to understand how these specific SGLT2 mutations impair kidney function and energy homeostasis.

  • Purpose: The purpose of this study was to analysis of effectiveness between cognitive function assessment scores and gaming cognitive rehabilitation system in children with intellectual impairment. Methods: Five children (male=5, $age=10.00{\pm}0.80$) with intellectual impairment participated in this study and were randomly assigned to the experiment that played (received) gaming cognitive rehabilitation system (Neuroworld). The children were applied 2 times a week for 30 minutes during 3 months. The children were assessed K-WSIC-VI (Korean-Wechsler intelligence scale for children-fourth edition) and recorded that gained score in gaming cognitive rehabilitation system before and after intervention. K-WSIC-VI contained five primary index scores: verbal comprehension index, visual spatial index, fluid reasoning index, working memory index, and processing speed index. Gaming cognitive rehabilitation system scoring was composed visual recall, target recall, sequence recall, selective attention, continuous attention, and exploration. Results: In the intelligence quotient (IQ) of K-WSIC-VI, there were significant increased in all children. The visual recall item was highest effective in all children. However, sequential recall showed the lowest improvement in all children. The performance speed of selective attention item was decreased, this means that children's skills have improved. Also, their ability to explore has improved significantly. Conclusion: In conclusion, gaming cognitive rehabilitation system was significant effectiveness in cognitive function in some categories for children with intellectual impairment. However, the visual recall and performance speed don't represent of all cognitive function. Therefore, further studies will need to verify by applying more subject and longer duration.

  • 1. Objective : This study aims to report symptomatic improvement of a Soyangin patient reporting general weakness, dizziness, gait disturbance, cold sensitivity, nocturnal sweating, and anorexia after treatment with Hyeongbangjihwang-tang. 2. Method : The patient's subjective and objective symptoms were observed daily, and the VAS scores for the main symptoms were recorded daily throughout the hospitalization period. 3. Result : The general condition of the patient improved by the end of the hospitalization period, where the VAS score dropped from VAS 10 to VAS 5 and the symptoms of dizziness, general weakness, cold sensitivity, and nocturnal sweating disappeared by day 4 of admission without recurrence. 4. Conclusion : Although the patient did not report diarrhea, one of the main Mangeum symptoms, the general symptoms improved with Hyeongbangjihwang-tang medication. This indicates that Hyeongbangjihwang-tang can be used to treat chief and subsidiary symptoms related to Mangeum symptomatic pattern when applied to the appropriate base and manifesting symptomology.

  • This paper proposes a system which extracts necessary information from call-for-paper (CFP) documents using a hidden Markov model (HMM). Even though a CFP does not follow a strict form, there is, in general, a relatively-fixed sequence of information within most CFPs. Therefore, a hiden Markov model is adopted to analyze CFPs which has an advantage of processing consecutive data. However, when CFPs are intuitively modeled with a hidden Markov model, a problem arises that the boundaries of the information are not recognized accurately. In order to solve this problem, this paper proposes a two-phrase hidden Markov model. In the first step, the P-HMM (Phrase hidden Markov model) which models a document with phrases recognizes CFP documents locally. Then, the D-HMM (Document hidden Markov model) grasps the overall structure and information flow of the document. The experiments over 400 CFP documents grathered on Web result in 0.49 of F-score. This performance implies 0.15 of F-measure improvement over the HMM which is intuitively modeled.

  • It is possible to indicate the complex design and execution of object-oriented program with dynamic UML diagram. This paper shows the way how to make several test cases from sequence, state, and activity diagram among dynamic UML diagram. Three dynamic UML diagrams about withdrawal work of ATM simulation program are drawn. Then different test cases are created from these diagrams using previously described ways. To evaluate effectiveness of test cases, mutation testing is executed. Mutants are made from MuClipse plug-in tool based on Eclipse which supports many traditional and class mutation operators. Finally we've got the result of mutation testing and compare effectiveness of test cases, etc. Through this document, we've known some hints that how to choose the way of making test cases.
