Quantum Dot LED를 이용한 상추 주요 병원성 곰팡이 및 세균의 생장억제효과 기내실험 (In Vitro Quantum Dot LED to Inhibit the Growth of Major Pathogenic Fungi and Bacteria in Lettuce)

  • Quantum Dot LED (QD LED) 조명은 소형의 크기, 좁은 대역파장, 긴 수명, 전자 시스템을 통한 제어가 용이하여 현재 시설재배에 이용되는 형광등, 할로겐램프, HID, HSP 램프의 단점을 보완할 수 있는 작물생육에 이상적인 광원으로서 잠재력을 가지고 있다. QD LED 조명을 이용하여 식물 병원성 미생물의 방제가 가능하다면 작물재배에 사용되는 인력 및 비용을 절감하고 화학적 방제제를 사용하지 않은 안전성 높은 생산물을 얻을 수 있다는 장점이 있다. 본 연구에서는 식물공장 및 온실에서 많이 재배되고 있는 상추에 큰 피해를 입히는 주요 식물 병원성 곰팡이에 대한 QD LED 조명의 영향과 생장억제효과를 확인하기 위해 시행하였다. 상추에 주로 발생하여 작물에 피해를 입히는 Rhizoctonia solani, Phytophthora drechsleri, Sclerotinia sclerotiorum, Sclerotinia minor, Botrytis cinerea, Fusarium oxysporum, Pectobacterium carotovorum, Xanthomonas campestris균을대상으로 QD LED 조명에 의한 균사생장억제 효과를 조사하였으며 처리한 6종류의 조명 중 BLUE (450 nm) 조명은 Sclerotinia sclerotiorum는 50 cm 거리에서 16.7%의 억제율을 보였으며 30 cm 거리에서 94.1%의 균사생장억제율을 보였다. S. minor는 50 cm 거리에서 80.4%, B. cinerea는 50 cm 거리에서 36.3%의 균사생장이 억제되었으며 30 cm 거리에서 S. minor와 B. cinerea는 100%의 균사생장억제율을 보였다. 15 cm 거리에서는 3종의 병원균 모두 100%의 억제율을 보였다. QD RED (M1), QD RED (M2)조명은 30 cm와 15 cm 거리에서 Sclerotinia minor와 Botrytis cinerea의 균사생장을 100% 억제했으며 Sclerotinia sclerotiorum의 경우 30 cm 거리에서 QD RED (M1)과 QD RED (M2)조명에 대해 각각 75.2%, 100%의 억제율을 보였으나 15 cm 거리에서는 각각 5.8%, 36.3%의 억제율을 보였다. 상추에 병원균을 접종하여 LED 광원 하에 생장을 확인한 결과 QD RED (M2)광원에서 S. sclerotiorum의 감염을 59.9% 억제하였고 S. minor는 BLUE (450 nm), QD RED (M1), QD RED (M2) 광원에서 59.9%의 억제율을 보였다. B. cinerea의 경우 BLUE (450 nm) 광원에서 84%의 높은 억제율을 보였다.

  • Sclerotinia sclerotiorum(Lib.) de Bary의 상치, 오이 및 유채의 3균주에 대하여 PDA를 기본부지로하여 광선이 이들의 균핵형성에 미치는 영향을 연구하였다. 광원으로서는 주광색 형광등을 사용하였다. 그 결과를 요약하면 다음과 같다. 1) 계속광처리에서 광도 480Lux까지는 광도가 증가함에 따라 증가했지만 성숙균핵의 건물중은 이와는 반대로 감소하였다. 그리고 800 Lux 처리에서는 균핵시원체의 유기가 크게 억제되었고 성숙균핵은 거의 형성되지 않았다. 2) 5000Lux의 고광도라 하더라도 48시의 단시간처리는 균핵시원체수 및 성숙균핵수를 증가시켰다. 그러나 성숙균핵의 건물중은 160 Lux에서만 다소 증가하는 경향이 있었다. 3) 광 shock 즉 1분간치의 계속광 48시간까지의 160 Lux, 500Lux 처리는 모두 균핵형성수와 건물중을 증가시켰는데 균핵형성수와 건물중을 증가시켰는데 균핵형성수는 500Lux에서 균핵건물중은 160Lux에서 더 높았다.

Biocontrol of Potato White Mold Using Coniothyrium minitans and Resistance of Potato Cultivars to Sclerotinia sclerotiorum

  • This study was conducted in Bahar and Lalehjin, Hamadan, Iran to assess the biocontrol efficacy of Coniothyrium minitans Campbell against potato white mold caused by Sclerotinia sclerotiorum (Lib.) de Bary under field and greenhouse conditions. In addition, the resistance of common potato cultivars against S. sclerotiorum was determined in a greenhouse experiment. After straw inoculation of six potato cultivars (Pashandi, Istambouli, Agria, Marfauna, Alpha and Spartaan) with S. sclerotiorum, the least disease severity was observed in Spartaan and Marfauna. Agria showed the most susceptibility to S. sclerotiorum. Compared with the healthy control, different concentrations of C. minitans conidia ($10^7$, $10^8$ and $10^9$ conidia/mL) reduced disease severity under greenhouse condition, and a concentration $10^9$ was the most effective treatment. During 2008 and 2009, four field trials were conducted to evaluate the efficacy of C. minitans in different soil and aerial applications on disease incidence of potato white mold. In 2008, soil application of $Contans^{(R)}$ WG (a commercial product of C. minitans) showed the greatest biocontrol capacity whereas soil application of solid-substrate C. minitans was found inferior when compared with other treatments in both Bahar and Lalehjin field sites. In 2009, benomyl application was the most effective treatment in reducing disease incidence in both tested field sites.

Cloning and molecular characterization of a new fungal xylanase gene from Sclerotinia sclerotiorum S2

  • Sclerotinia sclerotiorum fungus has three endoxylanases induced by wheat bran. In the first part, a partial xylanase sequence gene (90 bp) was isolated by PCR corresponding to catalytic domains (${\beta}5$ and ${\beta}6$ strands of this protein). The high homology of this sequence with xylanase of Botryotinia fuckeliana has permitted in the second part to amplify the XYN1 gene. Sequence analysis of DNA and cDNA revealed an ORF of 746 bp interrupted by a 65 bp intron, thus encoding a predicted protein of 226 amino acids. The mature enzyme (20.06 kDa), is coded by 188 amino acid (pI 9.26). XYN1 belongs to G/11 glycosyl hydrolases family with a conserved catalytic domain containing $E_{86}$ and $E_{178}$ residues. Bioinformatics analysis revealed that there was no Asn-X-Ser/Thr motif required for N-linked glycosylation in the deduced sequence however, five O-glycosylation sites could intervene in the different folding of xylanses isoforms and in their secretary pathway.

Antifungal activities of β-thujaplicin originated in Chamaecyparis obtusa

  • Environment-friendly, commercially-available agricultural products were investigated for antimicrobial activity against Sclerotinia sclerotiorum, as a pathogen of sclerotium disease. Then ${\beta}$-thujaplicin from Chamaecyparis obtuse was investigated for antifungal activity against six kinds of pathogenic fungi. It showed a statistically significant (p <0.001) growth inhibition effect on Sclerotinia sclerotiorum as a pathogen of sclerotium disease, Rhizoctonia solani AG-4 as a pathogen of damping off, Phytophthora capsici as a pathogen of phytophthora blight, and Colletotrichum coccodes as a pathogen of anthracnose at a concentration of 50 ppm and on Stemphylium solani as a pathogen of spotting disease and Alternaria alternata as a pathogen of black mold at a concentration of 100 ppm. In conclusion, these results indicate that it may be possible to develop environment-friendly agricultural products using ${\beta}$-thujaplicin compounds.

Bacillus subtilis C4와 B. cereus D8에 의한 유채의 생육증대 및 무름병과 균핵병 방제효과 (Effect of Bacillus subtilis C4 and B. cereus D8 on Plant Growth of Canola and Controlling Activity Against Soft Rot and Stem Rot)

  • 선발된 PGPR 균주인 Bacillus subtilis C4와 Bacillus cereus D8 균주의 유채에 대한 생육촉진 및 무름병균인 Erwinia carotovora와 균핵병균인 Sclerotinia sclerotiorum에 대한 방제효과를 검정하기 위하여 실내검정과 온실검정을 실시하였다. 실내검정 결과, C4와 D8균주처리에 의하여 유채의 생육이 40.3%~74% 증가하였으며 무름병이 대조구에 비하여 80% 감소하였다. 실내검정에서 C4와 D8균주를 종자에 처리하였을 때 뿌리가 크게 신장되었다. 주요 식물병원균 Sclerotinia sclerotiorum, Rhizoctonia solani, Botrytis cinerea, Fusarium oxysporum, Phytophthora capsici, Colletotrichum acutatum에 대하여 항균활성시험을 수행한 결과 두 균주 중 C4균주는 모든 병원성 곰팡이에 대하여 항균활성을 나타내었다. 온실검정에서 C4와D8균주처리는 대조구에 비하여 유채의 초장, 엽폭 및 엽장을 19.5%~24.9%, 11.3%~15.3%, 14.1%~20.7% 각각 증가시켰으며 균핵병균인 Sclerotinia sclerotiorum에 대한 억제효과가 우수하였다. 이와 같은 결과를 볼 때 C4, D8 균주 처리는 유채의 생육을 촉진시키며 유채에 저항성을 유도하므로 친환경생물방제에 적용할 수 있을 것으로 생각된다.

Characterization of Sclerotinia sclerotiorum, an Emerging Fungal Pathogen Causing Blight in Hyacinth Bean (Lablab purpureus)

  • Stems and pods of hyacinth bean cultivated in a farmer's field in Gazipur District, Bangladesh, were found rotted in nearly 5% hyacinth bean plants. A fungus having fluffy mycelium and large sclerotia was isolated from affected tissues. Combined results of morphological, molecular and pathological analyses identified the fungus as Sclerotinia sclerotiorum (Lib) de Bary. Inoculating the fungus on healthy hyacinth bean plants and pods reproduced the symptoms previously observed in the field. The three isolates obtained from naturally infected plants were cross inoculated in hyacinth bean, okra and African-American marigold and they were pathogenic to these hosts. The optimum temperature and pH for its growth were $20^{\circ}C$ and pH 5.0, respectively. Sclerotial development was favored at pH 5.0. Sucrose and mannitol were the best carbon sources to support hyphal growth, while glucose was the most favourable for sclerotial development. The hyacinth bean genotypes, HB-82 (Rupban Sheem) and HB-102 were found highly resistant, while HB-94 (Ashina) was moderate resistant to the fungus. Finally, S. sclerotiorum was sensitive to Bavistin, Dithane M-45 and Rovral fungicides and Ca in the form of $CaCl_2$. This observation could possibly aid in eliminating field loss in hyacinth bean caused by an emerging pathogenic fungus S. sclerotiorum.

Chemical and Biological Controls of Balloon Flower Stem Rots Caused by Rhizoctonia solani and Sclerotinia sclerotiorum

  • Stem rots caused by Rhizoctonia solani and Sclerotinia sclerotiorum have been known as devastating diseases in balloon flower plants. Antifungal activities of four fungicides, azoxystrobin, polyoxin B, trifloxystrobin and validamycin A were evaluated in vitro, showing effective suppression with mycelial growth of the fungal isolates on PDA media. Efficacies of the four fungicides were also demonstrated in stem tissues of balloon flower plants against R. solani and S. sclerotiorum. A commercially available Bacillus subtilis strain Y1336 was tested in terms of antagonistic biological control of stem rot disease of balloon flower plants. The bacterial strain revealed its antifungal activities against R. solani and S. sclerotiorum demonstrated by dual culture tests using paper discs and two plant pathogenic fungi on PDA media, as well as by plant inoculation assay, indicating that this antagonistic bacterial strain can be incorporated into disease management program for balloon flower stem rot diseases together with the four chemical fungicides.
