Examination of Required Functions in the PBNM Scheme for Multiple Domains as Cyber Physical System that Utilizes Data Science and AI

  • In the current Internet system, there are many problems using anonymity of the network communication such as personal information leaks and crimes using the Internet system. This is why TCP/IP protocol used in Internet system does not have the user identification information on the communication data, and it is difficult to supervise the user performing the above acts immediately. As a study for solving the above problem, there is the study of Policy Based Network Management (PBNM). This is the scheme for managing a whole Local Area Network (LAN) through communication control for every user. In this PBNM, two types of schemes exist. As one scheme, we have studied theoretically about the Destination Addressing Control System (DACS) Scheme with affinity with existing internet. By applying this DACS Scheme to Internet system management, we will realize the policy-based Internet system management. In this paper, required functions in the PBNM Scheme for multiple domains as cyber physical system that utilizes data science and AI is examined.

Sequence Homologies of GTP-binding Domains of Rab and Rho between Plants and Yeast/Animals Suggest Structural and Functional Similarities

  • Small GTP-binding proteins are divided into three major group: Ras, Rho and Ypt/Rab. They have the conserved regions designed G1 to G5 that are critical in GDP/GTP exchange, GTP-induced conformational change and GTP hydrolysis. We isolated and characterized genomic DNA or cDNAfragments encoding G1 to G3 domains of small GTP-binding protein Rab and Rho from several plant species using two different PCR-based cloning strategies. Seven rab DNA fragments were isolated from 4 different plants, mung-bean, tobacco, rice and pepper using two degenerate primers corresponding to the GTP-binding domain G1 and G3 in small GTP-binding proteins. The amino acid sequences among these rab DNA fragments and other known small GTP-binding proteins shows that they belong to the Ypt/Rab family. Six rho DNA fragments were isolated from 5 different plants, mung-bean, rice, Arabidopsis, Allium and Gonyaulax using the nested PCR method that involves four degenerate primers corresponding to the GTP-binding domain G1, G3 and G4. The rho DNA fragments cloned show more than 90% homology to each other. Sequence comparison between plant and other known Rho family genes suggests that they are closely related (67 to 82% amino acid identity). Sequence analysis and southern blot analysis of rab and rho in mung-bean suggest than thses genes are encoded by multigene family in mung-bean.

Development of a Breast Feeding Adaptation Scale (BFAS) (모유수유 적응 측정 도구 개발)

  • Purpose: The aim of this study was to develop a breast feeding adaptation scale (BFAS) to evaluate adaptation to breastfeeding for breastfeeding mothers and their infants and to test the validity and reliability of the instrument. Methods: The study was conducted as follows: application of the conceptual framework, identification of the content domains, items generation, and test of validity and reliability. In order to test validity and reliability, two panels of experts reviewed items and subcategories of the preliminary questionnaire and then data were collected from 329 mothers who were up to 4 weeks postpartum and breastfeeding. Descriptive statistics, t-test, factor analysis, and Cronbach's alpha were used to analyze the data. Results: The conceptual framework was based on the Roy adaptation model. The content domains were developed via literature review, review of instruments, and data acquired from the interviews of breastfeeding mothers and nurses. A total of 69 items belonging to 8 domains were generated. A reduction to 44 preliminary items was accomplished through content validity analysis. Factor analysis extracted 8 factors with a total of 27 items on a 5-point Likert scale. Content validity, construct validity, criterion validity, and reliability of the BFAS were established. Conclusion: The newly developed BFAS is a reliable and valid instrument with which the adaptation of breastfeeding mothers and their infants to the breastfeeding behavior can be evaluated.

Development of a Measurement of Intellectual Capital for Hospital Nursing Organizations (병원 간호조직의 지적자본 측정도구 개발)

  • Purpose: This study was done to develop an instrument for measuring intellectual capital and assess its validity and reliability in identifying the components, human capital, structure capital and customer capital of intellectual capital in hospital nursing organizations. Methods: The participants were 950 regular clinical nurses who had worked for over 13 months in 7 medical hospitals including 4 national university hospitals and 3 private university hospitals. The data were collected through a questionnaire survey done from July 2 to August 25, 2009. Data from 906 nurses were used for the final analysis. Data were analyzed using descriptive statistics, Cronbach's alpha coefficients, item analysis, factor analysis (principal component analysis, Varimax rotation) with the SPSS $PC^+$ 17.0 for Windows program. Results: Developing the instrument for measuring intellectual capital in hospital nursing organizations involved a literature review, development of preliminary items, and verification of validity and reliability. The final instrument was in a self-report form on a 5-point Likert scale. There were 29 items on human capital (5 domains), 21 items on customer capital (4 domains), 26 items on structure capital (4 domains). Conclusion: The results of this study may be useful to assess the levels of intellectual capital of hospital nursing organizations.

EF-hands in CBP7 are Important in the Process of Development

  • Calcium ions play an important role in development and intracellular signaling. Dictyostelium discoideum has 14 genes encoding calcium -binding proteins (CBPs), but the function of most CBPs during development has not yet been studied. In this study, we investigated the specific functions of CBP7, one of 14 CBPs, in development using RNA interference cell lines of CBP7, cell lines overexpressing CBP7, cell lines with point mutations in the EF-hand domain, and cell lines expressing fragment proteins. was intended to reveal. CBP7 consists of 169 amino acids and contains 4EF-hand domains. The CBP7-overexpressing cells showed complete loss of developmental process. These cells remained in the single-cell growth stage under development -inducing conditions, while wild-type cells formed aggregations within 6-8h of development and eventually formed fruiting bodies. The experiments using point-mutated CBP7 protein showed that all EF-hand domains of CBP7 were important for CBP7 to function during developmental process. These results suggest that CBP7 plays an important role in developmental processes across all EF-hand domains.

Research Progress on Strategies for Improving the Enzyme Properties of Bacteriophage Endolysins

  • Bacterial resistance to commonly used antibiotics is one of the major challenges to be solved today. Bacteriophage endolysins (Lysins) have become a hot research topic as a new class of antibacterial agents. They have promising applications in bacterial infection prevention and control in multiple fields, such as livestock and poultry farming, food safety, clinical medicine and pathogen detection. However, many phage endolysins display low bactericidal activities, short half-life and narrow lytic spectrums. Therefore, some methods have been used to improve the enzyme properties (bactericidal activity, lysis spectrum, stability and targeting the substrate, etc) of bacteriophage endolysins, including deletion or addition of domains, DNA mutagenesis, chimerization of domains, fusion to the membrane-penetrating peptides, fusion with domains targeting outer membrane transport systems, encapsulation, the usage of outer membrane permeabilizers. In this review, research progress on the strategies for improving their enzyme properties are systematically presented, with a view to provide references for the development of lysins with excellent performances.

Heat Shock Protein $90{\beta}$ Inhibits Phospholipase $C{\gamma}-1$ Activity in vitro

  • Phospholipase $C-{\gamma}1\;(PLC-{\gamma}1)$ is an important signaling molecule for cell proliferation and differentiation. $PLC-{\gamma}1$ contains two pleckstrin homology (PH) domains, which are responsible for protein-protein interaction and protein-lipid interaction. $PLC-{\gamma}1$ also has two Src homology (SH)2 domains and a SH3 domain, which are responsible for protein- protein interaction. To identity proteins that specifically binds to PH domain of $PLC-{\gamma}1$, we prepared and incubated the glutathione S-transferase(GST)-fused PH domains of $PLC-{\gamma}1$ with COS7 cell lysate. We found that 90 kDa protein specifically binds to PH domain of $PLC-{\gamma}1$. By matrix-assisted laser desorption ionization time of flight-mass spectrometry, the 90 kDa protein revealed to be heat shock protein (Hsp) $90{\beta}$. Hsp $90{\beta}$ is a molecular chaperone that stabilizes and facilitates the folding of proteins that are involved in cell signaling, including receptors for steroids hormones and a variety of protein kinases. To know whether Hsp $90{\beta}$ affects on $PLC-{\gamma}1$ activity, we performed $PIP_2$ hydrolyzing activity of $PLC-{\gamma}1$ in the presence of purified Hsp $90{\beta}$ in vitro. Our results show that the Hsp $90{\beta}$ dose-dependently inhibits the enzymatic activity of $PLC-{\gamma}1$ and further suggest that Hsp $90{\beta}$ regulates cell growth and differentiation via regulation of $PLC-{\gamma}1$ activity.

Friendship Expectation Perceived by Science-Gifted and Non-Gifted Elementary Students (초등 과학영재와 일반학생이 지각하는 교우기대감)

  • This study examined the friendship expectation that science-gifted and non-gifted elementary students perceived in gifted class and regular class. In 233 science-gifted elementary students and 329 non-gifted elementary students, we measured the friendship expectation that sub-domains were intimacy, ability similarity, and morality. The results of this study were as follows: First, according to the results of comparing the friendship expectations of science-gifted and non-gifted students at the regular class, there was statistically significant intergroup difference in the sub-domains of intimacy and morality, but there was no significant difference in the sub-domain of ability similarity. Second, according to the results of comparing the friendship expectations of science-gifted at the gifted class and at the regular class, there was statistically significant difference in the sub-domain of intimacy, but there was no significant difference in the sub-domains of morality and ability similarity. Based the results, the implications to understand the friendship of the science gifted elementary students were suggested in depth.

CHD4 Conceals Aberrant CTCF-Binding Sites at TAD Interiors by Regulating Chromatin Accessibility in Mouse Embryonic Stem Cells

  • CCCTC-binding factor (CTCF) critically contributes to 3D chromatin organization by determining topologically associated domain (TAD) borders. Although CTCF primarily binds at TAD borders, there also exist putative CTCF-binding sites within TADs, which are spread throughout the genome by retrotransposition. However, the detailed mechanism responsible for masking the putative CTCF-binding sites remains largely elusive. Here, we show that the ATP-dependent chromatin remodeler, chromodomain helicase DNA-binding 4 (CHD4), regulates chromatin accessibility to conceal aberrant CTCF-binding sites embedded in H3K9me3-enriched heterochromatic B2 short interspersed nuclear elements (SINEs) in mouse embryonic stem cells (mESCs). Upon CHD4 depletion, these aberrant CTCF-binding sites become accessible and aberrant CTCF recruitment occurs within TADs, resulting in disorganization of local TADs. RNA-binding intrinsically disordered domains (IDRs) of CHD4 are required to prevent this aberrant CTCF binding, and CHD4 is critical for the repression of B2 SINE transcripts. These results collectively reveal that a CHD4-mediated mechanism ensures appropriate CTCF binding and associated TAD organization in mESCs.

Suggesting the Conceptual Framework of the Nature of Technology(NOT) and Examining the Conceptions of Experts of Science, Technology, and Engineering Fields regarding the NOT (기술의 본성(NOT) 개념 틀 제안 및 과학기술공학 관련 전문가들의 인식 분석)

  • This study aims to suggest the conceptual framework for the Nature of Technology (NOT) by examining the current researches regarding the NOT in science and technology education and to analyze the conceptions of experts in science, technology, and engineering fields based on the framework of NOT. The NOT conceptual framework developed in the study consisted of the six domains of NOT, which are 1. Technology as artifacts, 2. Technology as knowledge, 3. Technology as practice, 4. Technology as system, 5. The role of technology in society, and 6. History of technology. Also, the results of analyzing the conceptions for the 30 experts in science, technology, and engineering fields emphasize on the three domains of the 3. Technology as practice, 4. Technology as system, and 5. The role of technology in society among the 6 domains of the framework of the six domains of the NOT framework. This findings are different from the results of previous researches conducted in science and technology education for exploring the conceptions of NOT in the POV of the public and students. As such, the results show that while the public and students possess naive ideas on NOT only focusing on the technology as products and tools, experts in science, technology, and engineering fields possess the authentic views of the NOT reflecting the complex and abstract concepts of technology in terms of the perspectives of philosophy of technology.
