Effect of Brown Algae (Undaria pinnatifida)-Noodle on Oxygen Radicals and Their Scavenger Enzymes in Liver of Sprague-Dawley Rats (미역 (Undaria pinnatifida) 국수의 투유가 랫트 간장중의 활성산소 및 제거효소에 미치는 영향)

  • This study was designed to investigate the effects og $10{\%},\;20{\%}\;and\;40{\%}$-addition of functional brown algae (FBA)-noodles on oxygen radicals and their scavenger enzymes in liver of Sprague-Dawley(SD) male rats. Hydroxyl radicals$({\cdot}OH)$ formations were significantly inhibited$(20{\~}35{\%}\;and\;12{\~}20{\%})$ in liver mitochondria and microsomes of rats administered $0{\%},\;20{\%}\;and\;40{\%}$ FBA-noodles compared with that of control group. Significant differences in $H_2O_2$ formations of liver microsome in these FBA-noodles fed groups could not be obtained, but superoxide-radical $(O_2^({\cdot}-))$ formations of liver cytosol resulted in a significant decrease about $10{\%}\;in\;20{\%}\;and\;40{\%}$ FBA-noodles compared with control group. Mn-SOD activities in liver mitochondria were significanlty increased $(10{\~}15{\%})$ in the groups fed $10{\%},\;20{\%}\;and\;40{\%}$ FBA-noodles, while a group administered $40{\%}$ FBA-noodle only resulted In a significant increases $(about 12{\%})$ in Mn-SOD activity of liver microsomes compared with control group. Cu, Zn-SOD activities in liver cytosol were significantly increased $(10{\~}20{\%})\;in\;10{\%},\;20{\%}\;and\;40{\%}$ FEA-noodles compared with control group. Administration of $10{\%},\;20{\%}\;and\;40{\%}$ FBA-noodles resulted in a marked increases$(20{\~}40{\%})$ in liver cytosolic glutathione peroxidase (GSHPx) compared with control group. Significant differences in lipid peroxide (LPO) levels of mitochondria and microsomes in $10{\%}$ FBA-noodle could not be obtained, while LPO levels of $20{\%} and 40{\%}$ FBA-noodles were significantly inhibited about $10{\%}$ in mitochondria and microsomes compared with control group. These results suggest that these FBA-noodles may play a desirable role in attenuating an oxygen radical formations and increasing a scavenger enzymes activity by some brown algae (Undaria pinnatifida) components.

  • This study was designed as a part of efforts to investigate the biochemical pollutant markers for diagnosis of marine pollutions by changes in oxygen radicals and their scavenger enzymes of the flounder (Paralichthys olivaceus) in the South Sea of Korea. Protein contents in brain and muscle of cultured flounder in the South Sea were remarkably lower ($19\~42\%$ and $25\~41\%$, respectively) than those of wild flounder in Pohang of the East Sea as a control group. Lipid Peroxide (LPO) levels in serum of cultured in the South Sea were considerably higher ($5\~33\%$) Elan those of wild flounder in Pohang. Hydroxyl radical formations in serum of cultured flounders in the South Sea were lower ($4\~25\%$) than those of wild flounders in Pohang. Superoxide dismutase (SOD) activities in serum of cultured flounders in South Sea were also significantly lower ($7\~31\%$) than those of wild flounders in Pohang. It is believed that significant decrease of protein contents in brain and muscle, and remarkable increases of LPO in serum of cultured flounders may be used as a biochemical pollutant markers for diagnosis of marine pollutions. These results suggest that near-coastal water as well as neritic water of the South Sea might be affected by pollutant input as well as the Yellow Sea of Korea.

Protein contents in brian and muscle of cultured flounder in Yellow Sea were remarkably lower(30-45% and 25-45%, respectively) than those of wild flounders in Pohang(control) of East Sea. Lipid peroxide(LPO) levels in serum of cultured and wild flounders in Yellow Sea were significanltly higher (30-80% and 125-145%, respectively)than those of wild flounder in Pohang. Hydroxide radical formations and superoxide dismutase(SOD) activities in serum of cultured flounders in Yellow Sea were significantly 15-30% and 15-35% lower than those of wild flounders in Pohang, but glutathione peroxidase (GSHPx) activities in brain of cultured flounders in Yellow Sea were significantly 15-25% higher than those of wild flounders in Pohang. It is believed that significantly decreases of protein contents in brain anad muscle, remakable increases of malondialdehyde(LPO) in serum and glutathione peroxidase (GSHPx)in brain of cultured flounders of Yellow Sea may be used as a biochemical pollutant markers for diagnosis of marine pollutions.

  • This study was designed as a part of efforts to investigate the biochemical pollutant markers for diagnosis of marine pollutions by changes in oxygen radicals and their scavenger enzymes of the mussel (Mytilus coruscus) in South Sea of Korea. Protein contents in muscle of cultured mussel in South Sea were remarkably lower (4-14%, respectively) than those of wild mussel in Pohang of East Sea. Superoxide radical activities in muscle of cultured in South Sea were significantly higher 82∼138% than those of wild mussel in Pohang. Hydroxyl radical formations in muscle of cultured mussels in South Sea were significantly 9∼25% higher than those of wild mussels in Pohang. Superoxide dismutase (SOD) activities in muscle of cultured mussels in South Sea were significantly 16∼28% lower than those of wild mussels in Pohang. It is believed that significantly decrease of protein contents in muscle, remarkable increases of superoxide radical and hydroxyl radical in muscle of cultured mussels of South Sea may be used as a biochemical pollutant markers for diagnosis of marine pollutions. These results suggest that near-coastal water as well as neritic water of the south sea might be affected by pollutant.

Effect of Nitric Oxide on Paraquat-Tolerance in Lettuce Leaves (상추잎의 Paraquat 내성에 미치는 Nitric oxide의 영향)

  • The protective effect of nitric oxide (NO) on the antioxidant system under paraquat(PQ) stress was investigated in leaves of 8-week-old lettuce (Lactuca sativa L.) plants. PQ stress caused a decrease of leaf growth including leaf length, width and weight. Application of NO donor, sodium nitroprusside (SNP), significantly alleviated PQ stress induced growth suppression. SNP permitted the survival of more green leaf tissue preventing chlorophyll content reduction and of higher quantum yield for photosystem II than in non-treated controls under PQ exposure, suggesting that NO has protective effect on chloroplast membrane in lettuce leaves. Flavonoids and anthocyanin were significantly accumulated in the leaves upon PQ exposure. However, the rapid increase of these compounds was alleviated in the SNP treated leaves. PQ treatment resulted in lipid peroxidation and induced accumulation of hydrogen peroxide ($H_2O_2$) in the leaves, while SNP prevented PQ induced increase in malondialdehyde (MDA) and $H_2O_2$. These results demonstrate that SNP serves as an antioxidant agent able to scavenge $H_2O_2$ to protect plant cells from oxidative damage. The activities of two antioxidant enzymes that scavenge reactive oxygen species, superoxide dismutase (SOD) and catalase (CAT) in lettuce leaves in the presence of NO donor under PQ stress were higher than those under PQ stress alone. Application of 2-(4-carboxyphenyl)-4, 4, 5, 5-tetramethylimidazoline-1-oxyl-3-oxide (PTIO), a specific NO scavenger, to the lettuce leaves arrested SNP mediated protective effect on leaf growth, photosynthetic pigment and antioxidant systems. However, PTIO had little effect on lettuce leaves under PQ stress compared with that of PQ stress alone. The obtained data suggest that the damage caused by PQ stress is in part due to increased generation of active oxygen by maintaining increased antioxidant enzyme activities and SNP protects plants from oxidative stress. From these results it is suggested that NO might act as a signal in activating active oxygen scavenging system that protects plants from oxidative damage induced by PQ stress and thus confer PQ tolerance.

  • $\alpha$-Phenyl-N-t-butylnitrone (PBN) is one of the most widely used spin-trapping compounds for investigating the existence of free radicals in biological systems. Recently, there has been considerable interest in the antioxidant nature of PBN on degenerative diseases, presumably related to oxidative stress. In the present study, the protective effect of PBN on the HepG2 cell line under oxidative stress was investigated. When the HepG2 cells were exposed to oxidant, such as hydrogen peroxide, menadione, or ethanol, the protective role of PBN was manifested as a reduction in trypan blue uptake and a decrease in the endogenous production of oxidants, as measured by the oxidation of 2',7'-dichlorodihydrofluorescin. The modulation of activity of major antioxidant enzymes, such as superoxide dismutase and catalase, was not significantly different either in the presence or in the absence of PBN. This indicates that PBN acts as a direct scavenger of reactive oxygen species.

Nitric Oxide Plays an Important Role in β-Aminobutyric Acid-Induced Resistance to Botrytis cinerea in Tomato Plants

  • β-Aminobutyric acid (BABA) has consistently been reported to enhance plant immunity. However, the specific mechanisms and downstream components that mediate this resistance are not yet agreed upon. Nitric oxide (NO) is an important signal molecule involved in a diverse range of physiological processes, and whether NO is involved in BABA-induced resistance is interesting. In this study, treatment with BABA significantly increased NO accumulation and reduced the sensitivity to Botrytis cinerea in tomato plants. BABA treatment reduced physical signs of infection and increased both the transcription of key defense marker genes and the activity of defensive enzymes. Interestingly, compared to treatment with BABA alone, treatment with BABA plus cPTIO (NO specific scavenger) not only significantly reduced NO accumulation, but also increased disease incidence and lesion area. These results suggest that NO accumulation plays an important role in BABA-induced resistance against B. cinerea in tomato plants.

Preparation of Egg White Liquid Hydrolysate (ELH) and Its Radical-Scavenging Activity

  • In the present study, an optimum protease was selected to hydrolyze the egg white liquid protein for the antioxidant peptides. Alcalase treatment yielded the highest amount of ${\alpha}$-amino groups (15.27 mg/mL), while the control (no enzymatic hydrolysis) showed the lowest amount of ${\alpha}$-amino groups (1.53 mg/mL). Alcalase also gave the highest degree of hydrolysis (DH) value (43.2%) and was more efficient for egg white liquid hydrolysis than the other enzymes. The Alcalase hydrolysate had the highest radical-scavenging activity (82.5%) at a concentration of 5.0 mg/mL. The conditions for enzymatic hydrolysis of egg white liquid with Alcalase were selected as substrate : water ratio of 2:1. Five percent Alacalse treatment did not show significant (P>0.05) increases of DH and ${\alpha}$-amino nitrogen content after 24 hhydrolysis. Thirty two hour-hydrolysis with 5% Alcalase is sufficient to make antioxidative egg white liquid hydrolysate from egg white liquid. DPPH and ABTS radical-scavenging activities were significantly (P<0.05) higher after enzymatic digestion. These results suggest that active peptides released from egg-white protein are effective radical-scavengers. Thus, this approach may be useful for the preparation of potent antioxidant products.

Effect of Pyroligneous Liquor on Oxygen Radicals and Their Scavenger Enzymes in Liver of CD Rats (흰쥐 간조직의 활성산소 및 제거효소에 미치는 목초액의 영향)

  • This study was designed to investigate the effects of pyroligneous liquor on oxygen radicals and their scavenger enzymes in the liver of Cri/Bgi CD rats (7 rats per group). Male rats were fed a basic diet prepared in our Lab., PL-0 (Control), PL-1, PL-25, PL-50 and PL-75 groups were Prepared to be 0%, 1%, 25%, 50% and 75%with distilled water using pyroligneous liquor (35% of Choa Co. Ltd.), and were administrated orally for 8 weeks. Superoxide radical contents in liver mitochondria and microsomes were significantly decreased to 12-14%, 11-15%, respectively, in these PL-25 and PL-50groups compared with the control group. Hydroxyl radical content in mitochondria and microsomes were markedly decreased to 12-20% and 17%, respectively, in these PL-25 and PL-50% groups compared with the control group. Hydrogen peroxide content in mitochondria and microsomes were significantly decreased about 15-12% and 22-20% in liver of PL-25 and PL-50 groups compared with the control group. Mn-SOD and Cu/Zn-SOD activities in liver of PL-25 and PL-50 groups were remarkably increased to 15-25%, 11-16%, respectively, compared with the control group. GPx activities in mitochondria and microsomes were significantly increased in the liver of PL-25 and PL-50 groups compared with the control group. CAT activities in mitochondria and cytosol were significantly increased to 12-14%, 15-27%, respectively, in the liver of PL-25 and PL-50 groups compared with the control group. These results suggest that long term administration orally of 25 and 50% pyroligneous liquor may effectively inhibit the formation of oxygen free radicals, and also scavenger enzyme activities significantly increase through the administration orally.

Protective Effect of Glycoprotein Isolated from Cudrania tricuspidata on Liver in $CCl_4$-treated A/J Mice (생쥐에 있어서 꾸지뽕 당단백질의 간보호 효과)

  • This study aimed to determine whether or not glycoprotein isolated from Cudrania tricuspidata Bureau fruit(CTB glycoprotein) exerts a hepatoprotective effect on liver injury induced by the administration of carbon tetrachloride($CCl_4$, 1.0mL/kg) to A/J mice. Following the administration of CTB glycoprotein(0-20mg/kg), the activities of antioxidant enzymes (superoxide dismutase(SOD), catalase(CAT), and glutathione peroxidase(GPx)), and the quantities of measured thiobarbituric acid reactive substances(TBARS), lactate dehydrogenase(LDH), and nitric oxide(NO) were evaluated from the murine liver tissues and plasma. Additionally, the activity of nuclear factor-kappa B(NF-${\kappa}B$) was assessed after pretreatment with $CCl_4$. When the mice were treated with $CCl_4$ alone, the activities of antioxidative enzymes reduced but amounts of TBARS, LDH, and NO increased. However, the results of treatment with CTB glycoprotein(10 and 20 mg/kg) revealed significantly increased activities of antioxidant enzymes(SOD, CAT, and GPx), as compared with $CCl_4$ alone. On the other hand, the result showed significant diminutions of the quantities of TBARS, LDH, and NO after treatment with CTB glycoprotein(10 and 20 mg/kg), as compared to $CCl_4$ alone. The activity of NF-${\kappa}B$ also declined after pretreatment with CTB glycoprotein, as compared with $CCl_4$ treatment alone. Thus, it is suggested that the CTB glycoprotein exerts a protective effect against $CCl_4$-induced liver injury in A/J mice.
