The effects of culture conditions on succinic acid production and its possible scale-up have been studied. Mannheimia succiniciproducens LPK7, engineered for enhanced production of succinic acid and reduced by-product secretion, was used for the experiments. Mannheimia succiniciproducens LPK7 is a knock-out strain of wild type deficient in the ldhA, pflB, and pta-ackA genes, and is derived from Mannheimia succiniciproducens MBEL55E. Process optimization of factors including optimal temperature, pH, carbon source, and nitrogen source was performed to enhance the production of succinic acid in flasks. To observe scale-up effects, batch fermentation was carried out at various working volumes. At a working volume of 7.0 l, the final succinic acid concentration and yield were 15.4g/l and 0.86g/g. This result shows similar amount of succinic acid obtained in lab-scale fermentation, and it is possible to scale up to larger fermentors without major problems.

Mulberry leaves containing 1-deoxynojirimycin (DNJ) known to be a strong inhibitory effect for ${\alpha}-glucosidase$. Thus, DNJ has been recognized as a potentially important source for prevent or treat hyperglycemia. More effective method for the DNJ high-production is needed because DNJ content of natural mulberry leaf are as low as 0.1%. Many researchers have studied for the DNJ high-production in mulberry leaves such as the harvest season, fermentation using microorganisms, optimal culture conditions, and optimal extraction conditions. In order to provide for useful data that is anticipated at the level of industrial scale, we investigated ${\alpha}-glucosidase$ inhibitory activity, pH value and DNJ content in large-scale based on the optimal culture conditions for mulberry leaf fermentation of small-scale in our previous study. The ${\alpha}-glucosidase$ inhibitory activity, pH value, and DNJ content in this study were measured from the mulberry leaf fermentation broth for 7 days. During mulberry leaf fermentation, the ${\alpha}-glucosidase$ inhibitory activity and DNJ content was increased until 2 to 4 days, but after 4 day was decreased. The pH value showed a decreasing trend up to 2 day, and little changes in 2 to 4 days. However, the pH was started to increase after 4 days.

Scale-up of hirudin production from Saccharomyces cerevisiae from bench-scale to pilot-scale was carried out based on constant volumetric oxygen transfer coefficient (K$\sub$L/a). Fed-batch mode of cultivation using step-wise feeding strategy of galactose was employed for the production of hirudin in a 30-L and a 300-L pilot-scale fermentor. The final hirudin concentrations were achieved 390 mg/L and 286.1 mg/L, and the volumetric productivities were 80.4% and 90.7% with the 30-L and 300-L fermentors, respectively, compared to the productivity of the 5-L bench-scale fermentor.

This study is to establish optimizing condition of alcohol fermentation in vinegar production with aronia, to confirm whether this can be industrially used, and to compare and analyze a change in anti-oxidative activity and quality characteristic according to alcohol fermentation of aronia. The optimized conditions for alcohol fermentation were as follows: Saccharomyces cerevisiae 5645 of yeast strain, a 5% inoculum size, aronia juice with a brix value of 14, and a glucose content of 7%. As a result to conduct scale up with optimizing conditions of alcohol fermentation of aronia, 8 days (192 hrs) of total alcohol fermentation time and 7.4% of the final alcohol content. The harvest volume accounted for approximately 90.2% with a loss of about 2.8%. As a result of antioxidant test, anti-oxidative activity of alcohol fermented liquor is lower than anti-oxidative activity of aronia extract, because of the decrease of antioxidant by oxidation of the fermentation process. An efficient and simple fermentation process was developed for the production of ${\gamma}$-aminobutyric acid (GABA) by Lactobacillus sakei B2-16. When the L. sakei B2-16 was cultivated in the rice bran extracts medium containing 4% sucrose, 1% yeast extract, and 12% monosodium glutamate, the maximum GABA concentration reached 660.0 mM with 100% conversion yield, showing the 2.4- fold higher GABA concentration compared with the modified MRS medium without the rice bran extracts. The GABA production was scaled-up from a laboratory scale (5 l) to a pilot (300 l) and a plant (5,000 l) scale to investigate the application possibility of GABA production to industrial fields. The production yields at the pilot and plant scales were similar to the laboratory scale using rice bran extracts medium, which could be effective for the low-cost production of GABA.
In order to develop economic processes for ethanol production from starch, a simultaneous saccharification and fermentation(SSF) process using Zymomonas mobilis and amyloglucosidase (AMG) was studied in semibatch modes using cell recycle. The cell recycle was carried out by adopting two different methods; microfiltration and settling. The cell recycle using microfiltration revealed higher productivity(5.4 g/l/h) than that using a settler(4.3 g/l/h). Taking the large-scale ethanol fermentation into account, the semibatch process using microfiltration system appeared most promising among others with respect to ethanol productivity, feasibility of scale-up and simplification of operation.
The effects of pH, temperature and oxygen transfer rate(OTR) on L-Ieucine fermentation were investigated employing Corynebacterium glutamicum CH 1516 in 71 fermentor. The optimum pH, temperature and OTR were determined to be 7.0, $30^{\circ}C$ and 0.21 kmole $O_2$/$m^3{\cdot}hr$, respectively. For the values of OTR lower than 0.19 kmole $O_2$/$m^3{\cdot}hr$ a significant amount of lactic acid was accumulated, while the packed cell volume(PCV) was rapidly increased at higher OTR values above 0.23 kmole $O_2$/$m^3{\cdot}hr$ and glutamic acid was produced to some extent. Scale-up studies for L-Ieucine fermentation which was carried out in 12001 pilot scale fermentor reaffirmed the results of 71 fermentation. The optimum redox potential value for L-Ieucine production was found to be -150 to -170 mY.
Distillation condensate generated from downstream processing of microbial alcohol fermentation imposes a serious burden to biological wastewater treatment or anaerobic digestion due to its high contents of SS (suspended solids) and TN (total nitrogen), A pilot scale microfiltration of the stillage condensate with a stainless steel SCEPTER membrane of 0.1 ${\mu}$m pore size was carried out to remove SS which was mostly composed of microbial cell residue. A stable permeate flux was achieved when the decanter effluent containing 0.7% of SS was filtered under the conditions of X10 VCR (volume concentration ratio), 2.5 bar of TMP (transmembrane pressure), and 60$^{\circ}C$. When stillage condensate with 2.6% SS was treated directly with microfiltration, VCR below X3 was recommended for a long duration of filtration. The permeate and retentate obtained from microfiltration were recycled to make-up medium of fermentation. Adding permeate or retentate up to 30% of fermentation volume showed no distinguished undesirable influence during the course of alcohol fermentation. Although only slight improvements in the final amount of CO$_2$ evolution and alcohol content were observed, fermentation rate increased so that the required time to reach 450 L/ton of CO$_2$ evolution was shortened to 72% of that with normal media.
The scale-up criterion of constant oxygen mass transfer coefficient ($k_La$) was applied for the production of itaconic acid (IA) in a 50 L pilot-scale fermentor by the fungal cells of Aspergillus terreus. Various operating conditions were examined to collect as many $k_La$ data as possible by adjusting the stirring speed and aeration rate in both 5 L and 50 L fermentor systems. In the fermentations performed with the 5 L fermentor, the highest IA production was obtained under the operating conditions of 200 rpm and 1.5 vvm. Accordingly, we intended to find out parallel agitation and aeration rates in the 50 L fermentor system, under which the $k_La$ value measured was almost identical to that ($0.02sec^{-1}$) of the 5 L system. The conditions of 180 rpm and 0.5 vvm in the 50 L system turned out to be optimal for providing almost the same volumetric amount of dissolved oxygen (DO) into the fermentor, without causing shear damage to the producing cells due to excessive agitation. Practically identical fermentation physiologies were observed in both fermentations performed under those respective operating conditions, as demonstrated by nearly the same values of volumetric ($Q_p$) and specific ($q_p$) IA production rates, IA production yield ($Y_{p/s}$), and specific growth rate (${\mu}$). Specifically, the negligible difference of the specific growth rate (${\mu}$) between the two cultures (i.e., $0.029h^{-1}$ vs. $0.031h^{-1}$) was notable, considering the fact that ${\mu}$ normally has a significant influence on $q_p$ in the biosynthesis of secondary metabolites such as itaconic acid.

This study was performed to investigate the application of dark $H_2$ fermentation to two-stage bioprocesses for organic waste treatment and energy production. We reviewed information about the two-stage bioprocesses combining dark $H_2$ fermentation with $CH_4$ fermentation, photo $H_2$ fermentation, microbial fuel cells (MFCs), or microbial electrolysis cells (MECs) by using academic information databases and university libraries. Dark fermentative bacteria use organic waste as the sole source of electrons and energy, converting it into $H_2$. The reactions related to dark $H_2$ fermentation are rapid and do not require sunlight, making them useful for treating organic waste. However, the degradation is not complete and organic acids remain. Thus, dark $H_2$ fermentation should be combined with a post-treatment process, such as $CH_4$ fermentation, photo $H_2$ fermentation, MFCs, or MECs. So far, dark $H_2$ fermentation followed by $CH_4$ fermentation is a promising two-stage bioprocess among them. However, if the problems of manufacturing expenses, operational cost, scale-up, and practical applications will be solved, the two-stage bioprocesses combining dark $H_2$ fermentation with photo $H_2$ fermentation, MFCs, or MECs have also infinite potential in organic waste treatment and energy production. This paper demonstrated the feasibility of two-stage bioprocesses combining dark $H_2$ fermentation as a novel system for organic waste treatment and energy production.
