볼록총채벌레 자동판정을 위한 후보영역 검출 (Detection of Candidate Areas for Automatic Identification of Scirtothrips Dorsalis)

  • 볼록총채벌레는 최근 감귤원 해충 피해의 주요 해충으로 인식되어 주기적인 예찰이 이루어지고 있으나 성충의 크기가 0.8mm 정도로 작아 육안 식별에 어려움이 있다. 본 논문에서는 예찰 트랩에 포집된 볼록총채벌레를 자동으로 판별하기 위한 후보 영역 검출 방법을 제안하였다. 본 논문에서 사용한 방법은 히스토그램 기반의 템플릿 매칭으로 그레이 이미지와 그레디언트 이미지를 합성한 이미지를 사용하였다. 50 배율의 광학 현미경으로 영상을 획득 하였고, 제안한 방법의 객관적인 성능 판별을 위해 기존 방법[8]과 노이즈 제거 이미지를 이용한 히스토그램 기반 템플릿 매칭방법 그리고 그레디언트 이미지를 이용한 히스토그램 기반 템플릿 매칭 방법들과 비교 실험을 하였다. 실험결과 본 논문에서 제안한 방법이 기존 전처리[8] 방법 보다 약 14.42% 향상된 성능을 보였고, 노이즈 제거 이미지를 이용한 방법보다 41.63%, 그레디언트 이미지를 이용한 방법보다 21.17% 높은 성능을 보였다.

A Bio-fluidic Device for Adaptive Sample Pretreatment and Its Application to Measurements of Escherichia coli Concentrations

  • In this paper, we describe a bio-fluidic device for adaptive sample pretreatment, in order to optimize the conditions under which absorbance assays can be conducted. This device can be successfully applied to the measurement of Escherichia coli (E. coli) concentrations using adaptive dilution, with which the dilution ratio can be adjusted during the dilution. Although many attempts have been previously made to miniaturize complex biochemical analyses at the chip scale, very few sample pretreatment processes have actually been miniaturized or automated at this point. Due to the lack of currently available on-chip pretreatments, analytical instruments tend to suffer from a limited range of analysis. This occasionally hinders the direct and quantitative analysis of specific analyses obtained from real samples. In order to overcome these issues, we exploit two novel strategies: dilution with a programmable ratio, and to-and-fro mixing. The bio-fluidic device consists of a rectangular chamber constructed of poly(dimethylsiloxane) (PDMS). This chamber has four openings, an inlet, an outlet, an air control, and an air vent. Each of the dilution cycles is comprised of four steps: detection, liquid drain, buffer injection, and to-and-fro mixing. When using adaptive sample pretreatment, the range in which E. coli concentrations can be measured is broadened, to an optical density (O.D.) range of $0.3{\sim}30$. This device may prove useful in the on-line monitoring of cell concentrations, in both fermenter and aqueous environments.

Identification of 1,531 cSNPs from Full-length Enriched cDNA Libraries of the Korean Native Pig Using in Silico Analysis

  • Sequences from the clones of full-length enriched cDNA libraries serve as valuable resources for functional genomics related studies, genome annotation and SNP discovery. We analyzed 7,392 high-quality chromatograms (Phred value ${\geq}$30) obtained from sequencing the 5' ends of clones derived from full-length enriched cDNA libraries of Korean native pigs including brainstem, liver, cerebellum, neocortex and spleen libraries. In addition, 50,000 EST sequence trace files obtained from GenBank were combined with our sequences to identify cSNPs in silico. The process generated 11,324 contigs, of which 2,895 contigs contained at least one SNP and among them 610 contigs had a minimum of one sequence from Korean native pigs. Of 610 contigs, we randomly selected 262 contigs and performed in silico analysis for the identification of cSNPs. From the results, we identified 1,531 putative coding single nucleotide polymorphisms (cSNPs) and the SNP detection frequency was one SNP per 465 bp. A large-scale sequencing result of clones from full-length enriched cDNA libraries and identified cSNPs will serve as a useful resource to functional genomics related projects such as a pig HapMap project in the near future.

시료용액의 특성에 따른 고정화된 단일벽 탄소나노튜브의 전기적 거동 (Electrical Property of Immobilized SWNTs Bundle as Bridge between Electrodes in Nanobiosensor Depending on Solvent Characteristics)

  • 현재 세계적인 이슈가 되고 있는 나노과학과 기술은 탄소나노튜브(CNTs)를 기반으로 한 바이오센서 성능 향상에 주력하고 있다. 다양한 기능성을 가진 CNTs는 높은 안정성과 바이오 수용체와 같은 생체물질과의 높은 적합성으로 이를 이용한 바이오 전극 기술에 힘입어 의학, 식품 및 환경에서 이슈가 되는 물질들을 검출하기 위한 산업적 응용 연구가 주목받고 있다. 본 연구에서는 이러한 CNTs를 이용한 전기화학적 바이오센서에 있어서 시료가 액체 상태로 검출이 예상되는데 그 시료의 화학적 특성에 따른 금 전극 사이에 고정화된 CNTs의 전자전달현상을 조사하였다. 그 결과, 시료가 극성인 경우와 무극성인 경우 고정화된 CNTs의 전자전달 현상이 다르게 나타났으며, 극성의 세기가 증가할수록 전자의 이동에 방해를 받는 것으로 확인되었다. 이는 CNTs의 양끝에 존재하는 극성 작용기와의 상호작용에 의한 것으로서 센서 디바이스 전체를 시료 용액에 침투시켜 전자이동을 관찰한 결과와 달리 안정적으로 저항값을 나타내는 것으로 확인되었다. 향후 민감도가 높은 CNTs 기반 나노바이오센서 개발 시 시료의 효과적인 전처리 공정에서 이러한 용매의 극성을 고려한 최적화 연구가 필요하다.

Detecting Incipient Caries Using Front-illuminated Infrared Light Scattering Imaging

  • A new method for early caries diagnosis was proposed and tested through a home-made optical examination system that used quantitative light fluorescence (QLF) and digital imaging fiber optic transillumination (FOTI) (DIFOTI), with light sources across a wide spectral range, from 350 nm to 1,000 nm. The front-illuminated infrared light scattering image (FIR) showed similar diagnostic abilities to that of DIFOTI. The FIR method was invented based on the observation that caries lesions lose the high transmittance and low scattering properties of benign enamel tissue. There are various methods for the early diagnosis of caries, such as visual examination, exploration, X-ray radiography, QLF, FOTI, and infrared fluorescence (diagnodent). Among them, methods based on optical properties are regarded as having the most potential. A comparative study was performed between the FOTI, QLF, diagnodent, optical coherence tomography, and FIR scattering image methods, using 20 extracted teeth samples with early caries. A scale of lesion measurement based on optical image contrast was proposed. The statistical analysis showed a significant correlation between the DIFOTI and FIR methods (r = 0.35, p < 0.05). However, the QLF and diagnodent methods showed little association with FIR images, as they have different detection principles as compared with FIR. Tomographic images obtained by OCT, using 1,330 nm super luminescent LED as a gold standard of tooth structure, verified that the FOTI and FIR results correctly represented the lack of homogeneity in dental tissue. The newly proposed FIR method attained similar diagnostic results to those of FOTI, but with an easier approach.

PZT 소자의 정압전 응답을 이용한 보 구조물의 모드 변형에너지기반 손상 모니터링 (Modal Strain Energy-based Damage Monitoring in Beam Structures using PZT's Direct Piezoelectric Response)

  • 본 연구에서는 PZT 소자의 정압전 효과에 의한 동적 응답신호를 이용하는 보 구조물 손상 모니터링 기법을 제안하였다. 특히, 모드 변형에너지기반 보 구조물 손상 모니터링에 PZT 정압전 응답신호를 입력자료로 활용하는 방안에 대한 연구에 주안점이 있다. 먼저, PZT 소자의 정압전 효과 및 동적 변형률 응답의 이론적 배경을 요약하였다. 다음으로, 모드 변형에너지기반 보 구조물 손상위치 모니터링 기법을 제시하였다. 제시된 기법의 적합성을 검증하기 위해, 캔틸레버 보 모형을 대상으로 강제진동 실험을 수행하였으며, 세 종류의 센서(가속도계, PZT 센서, 변형률계)를 통해 동적 응답신호가 계측되었다. 손상 전후에 계측된 이들 진동신호들을 사용하여 모드 변형에너지기반의 손상위치 모니터링이 수행되었다.

콘크리트 배면공동 탐지를 위한 GPR, IE 및 IR기법의 복합 적용 (Integrated Application of GPR, IE and IR Methods to Detection of the Rear Cavity of Concrete)

  • 콘크리트의 배면에 위치하는 공동을 탐지하기 위해 실제 구조물과 같은 스케일의 모형을 제작하여 비파괴 검사로 널리 사용되는 GPR (ground penetrating radar)탐사와 충격반향기법(IE; impact echo), 충격응답기법(IR; impulse response)을 복합적으로 적용하였다. 콘크리트 모형은 철근 및 공동의 유무에 따라 다양한 반응을 관찰할 수 있도록 설계 하였다. GPR 탐사결과 공동의 유무에 따라 반응의 차이가 나타났지만 정확하게 공동의 존재를 파악하기 어려웠으며, 주로 건전도 측정을 위해 지반조사에 활용되었던 IE 및 IR 기법은 공동의 유무를 구분할 수 있는 반응을 보여주었다. 각 탐사의 적용 결과 철근의 존재에 따라 각 방법별로 배면의 공동을 탐지하는데 반응의 변화가 나타나긴 했지만, 세 가지 방법을 복합적으로 적용하였을 때 단일 탐사를 이용한 결과보다 정확하게 배면 공동의 위치를 파악할 수 있었다.

DMMP 검출용 금속산화물을 첨가한 $SnO_2$ 가스센서 제조 (Fabrication of $SnO_2$ Gas Sensor added by Metal Oxide for DMMP)

  • $SnO_2$ gas sensor for the detection DMMP, simulant of nerve gas was fabricated and its characteristics were examined. Sensing materials were $SnO_2$ added by TEX>$\alpha$-$Al_{2}O_{3}$ with 0∼20wt.% and $In_{2}O_{3}$ with 0∼3wt.% and were physically mixed each material. They were deposited by screen printing method on alumina substrate. The sensor was consisted of sensing electrode with interdigit(IDT) type in front and a heater in back side. Its dimension was 7$\times$10$\times$0.6$\textrm{mm}^2$. Crystallite size 8t phase identification, specific surface area and morphology of fabricated $SnO_2$ powders were analyzed by X-ray diffraction(XRD), surface area analyzer(BET) and by a scanning electron microscope(SEM), respectively. Sensor was measured as flow type and sensor resistance change was monitored as real time using LabVIEW program. The best sensitivities were 75% at adding 4wt.% TEX>$\alpha$-$Al_{2}O_{3}$, operating temperature $300^{\circ}C$ and 87% at adding 2wt.% $In_{2}O_{3}$, operating temperature $350^{\circ}C$ to DMMP 0.5ppm. Response and recovery times were about 1 and 3 min., respectively. Repetition measurement was very good with $\pm$3% in full scale. As a result, operating temperature was lower TEX>$\alpha$-$Al_{2}O_{3}$ than $In_{2}O_{3}$, but sensitivity was higher $In_{2}O_{3}$ than $\alpha$-$Al_{2}O_{3}$.

Discrimination of Three Emotions using Parameters of Autonomic Nervous System Response

  • Objective: The aim of this study is to compare results of emotion recognition by several algorithms which classify three different emotional states(happiness, neutral, and surprise) using physiological features. Background: Recent emotion recognition studies have tried to detect human emotion by using physiological signals. It is important for emotion recognition to apply on human-computer interaction system for emotion detection. Method: 217 students participated in this experiment. While three kinds of emotional stimuli were presented to participants, ANS responses(EDA, SKT, ECG, RESP, and PPG) as physiological signals were measured in twice first one for 60 seconds as the baseline and 60 to 90 seconds during emotional states. The obtained signals from the session of the baseline and of the emotional states were equally analyzed for 30 seconds. Participants rated their own feelings to emotional stimuli on emotional assessment scale after presentation of emotional stimuli. The emotion classification was analyzed by Linear Discriminant Analysis(LDA, SPSS 15.0), Support Vector Machine (SVM), and Multilayer perceptron(MLP) using difference value which subtracts baseline from emotional state. Results: The emotional stimuli had 96% validity and 5.8 point efficiency on average. There were significant differences of ANS responses among three emotions by statistical analysis. The result of LDA showed that an accuracy of classification in three different emotions was 83.4%. And an accuracy of three emotions classification by SVM was 75.5% and 55.6% by MLP. Conclusion: This study confirmed that the three emotions can be better classified by LDA using various physiological features than SVM and MLP. Further study may need to get this result to get more stability and reliability, as comparing with the accuracy of emotions classification by using other algorithms. Application: This could help get better chances to recognize various human emotions by using physiological signals as well as be applied on human-computer interaction system for recognizing human emotions.

Effect of growth phase of cyanobacterium on release of intracellular geosmin from cells during microfiltration process

  • During low-pressure membrane treatments of cyanobacterial cells, including microfiltration (MF) and ultrafiltration (UF), there have reportedly been releases of intracellular compounds including cyanotoxins and compounds with an earthy-musty odor into the water, probably owing to cyanobacterial cell breakage retained on the membrane. However, to our knowledge, no information was reported regarding the effect of growth phase of cyanobacterial cells on the release of the intracellular compounds. In the present study, we used a geosmin-producing cyanobacterium, Anabaena smithii, to investigate the effect of the growth phase of the cyanobacterium on the release of intracellular geosmin during laboratory-scale MF experiments with the cells in either the logarithmic growth or stationary phase. Separate detection of damaged and intact cells revealed that the extent of cell breakage on the MF membrane was almost the same for logarithmic growth and stationary phase cells. However, whereas the geosmin concentration in the MF permeate increased after 3 h of filtration with cells in the logarithmic growth phase, it did not increase during filtration with cells in the stationary phase: the trend in the geosmin concentration in the MF permeate with time was much different between the logarithmic growth and stationary phases. Adsorption of geosmin to algogenic organic matter (AOM) retained on the MF membrane and/or pore blocking with the AOM were greater when the cells were in the stationary phase versus the logarithmic growth phase, the result being a decrease in the apparent release of intracellular geosmin from the stationary phase cells. In actual drinking water treatment plants employing membrane processes, more attention should be paid to the cyanobacterial cells in logarithmic growth phase than in stationary phase from a viewpoint of preventing the leakage of intracellular earthy-musty odor compounds to finished water.
