Distribution of Ectomycorrhizal Mushroom According to Altitude in NaeJangsan National Park (내장산국립공원의 고도에 따른 외생균근성 버섯 분포)

  • This study was conducted to investigate the diversity of ectomycorrhizal mushroom by surveying sites from June 2004 to October 2005. The obtained results from investigation were as follows. The total of 5 classes 16 orders 63 families 149 genera and 358 species including saprophytic and ectomycorrhizal fungi was investigated. A total of 17 families 36 genera 152 species (1,285ea.) of ectomycorrhizal mushroom was investigated. The mushrooms are classified into 9 families 27 genera and 136 species in Agaricales, 5 families 6 genera and 12 species in Aphyllophorales and 3 families 3 genera and 4 species in Gasteromycetes. Dorminant species were Russulaceae (35 species) followed by Boletaceae (34 species), Amanitaceae(22 species) and Cortinariaceae (21 species). The mushroom occurrence of octomycorrhizal fungi was closely related to climatic conditions such as high air temperature and lots of rainfall from July to September. The environment factors which have a favorable influence of mushroom occurrence were soil pH, available $P_{2}O_{5}$ of soil and rainfall and air temperature of climatic environment.

Antagonistic Effects of the Bacterium Alcaligenes sp. HC12 on Browning Disease Caused by Pseudomonas agarici (버섯 세균성회색무늬병균(Pseudomonas agarici)에 대한 Alcaligenes sp. HC12의 항균활성)

  • A gram-negative bacterium was isolated from spent substrates of Agaricus bisporus and showed significant antagonistic activity against Pseudomonas agarici. The bacterium was identified as Alcaligenes sp. based on cultural, biochemical, physiological characteristics and a 16S rRNA sequence analysis. The isolate is saprophytic, but not parasitic or pathogenic on cultivated mushroom, whereas it showed strong inhibitory effects against P. agarici cells in vitro. The control efficacy of Alcaligenes sp. HC12 against brown blotch of P. agarici was up to 63% on Agaricus bisporus. The suppressive bacterium may be useful for the development of biocontrol systems.

Antagonistic Effect of Bacillus safensis HC42 on Brown Blotch Mushroom Disease Caused by Pseudomonas agarici (버섯 세균성회색무늬병균 (Pseudomonas agarici) 에 대한 항균활성을 가지는 Bacillus safensis HC42)

  • A gram-positive bacterium was isolated from the spent substrate of Agaricus bisporus that showed a marked antagonistic activity against Pseudomonas agarici. It was identified as Bacillus safensis HC42 based on its cultural, biochemical, and physiological characteristics, and 16S rRNA sequence. B. safensis HC42 was saprophytic, but not parasitic or pathogenic, in cultivated mushrooms and showed strong inhibition of P. agarici in vitro. Moreover, it showed a control efficacy of 66 % against browning disease caused by P. agarici in Agaricus bisporus. Therefore, B. safensis HC42 may be useful in the future for the development of a biocontrol system.

Effect of Thinning on Environmental Factors and Wild Mushroom Fruting in Quercus mongolica Forest (신갈나무림에서 솎아베기가 임내환경과 자연버섯 발생에 미치는 영향)

  • In order to determine the effects of thinning on mushroom fruiting, microclimate and air temperature in Quercus mongolica forests, this study investigated changes in crown density, soil temperature, air temperature and throughfall in the natural Q. mongolica forest which was thinned by approximately 45% four years ago. The results showed that crown density was smaller than control by 6% from April to October, The soil temperature and air temperature in the thinned stands were significantly higher than those of the control by $1{\sim}2^{\circ}C$ until August and differences in air and soil temperature. The average daily temperature difference was higher than control by $0.2{\sim}0.7^{\circ}C$ until October. Throughfall from July to September in the thinned was approximately 135 mm higher than in the control. The maximum difference in throughfall per unit time was 3.5 mm/h. Soil moisture in the thinned site increased by approximately 5% compared with the control and reduced to the normal moisture after 4 days in both sites. 55 mushroom species were found in the thinned area between July and September. The thinned site contained 10 mycorrhizal mushrooms more than the control and 1 saprophytic mushroom species more than the control. Shannon-Wiener Index was 3.2, approximately 0.5 higher than the control. Tylopilus neofelleus, etc., occurred in the thinned site more quickly, but Aaricus subrutilescens, Clitocybe sp, occurred later. In the thinned site, the dominance of Tylopilus neofellelus and Armillaria sp. increased approximately 6% and 30% and yield about 1.5 times and 20 times, respectively. In conclusion, thinning in the Q. mongolica forest increased the soil and air temperature, soil moisture, throughfall and the diversity of mushroom species, and advanced the period of occurrence, and increased dominance and yield of some mycorrhizal mushrooms and Amillaria sp.

Clinical Characteristics of Pulmonary Aspergillosis (폐 국균증의 임상적 특성)

  • Background: Genus of Aspergilli are ubiquitous saprophytic molds in nature, but its change from a saprophytic fungus to a pathogenic organism has occurred since the use of various antibiotics. The fungus affects the chronically ill and debilitated population. Recently frequency of the fungal infection is increasing in Korea with abuse of antibiotics and glucocorticoids. Method: We analyzed medical records of 52 patients with pulmonary aspergillosis seen at Hanyang University Hospital from 1980 to 1994. The results were as follows; Results: 1) Ages ranged between second to eighth decades with majority(50%) in the fourth to fifth decades. The male to female ratio was 1.1:1. 2) Hemoptysis and productive cough, the leading symptoms, occurred in 42.3% and 25% respectively. 3) On chest X-ray fingings, the characteristic "fungus ball" pattern were observed in 53.8% of the 52cases. 4) Sputum culture for aspergilli were positive in 21.6% of the cases. We performed fine needle aspiration in 22 patients and the diagnostic yield was 100%. 5) Thirty-six patients had history of treatment with antituberculous drugs under diagnosis of pulmonary tuberculosis for an average of 27.3 months. But sputum analysis for acid-fast bacilli were positive in 5.6%(2cases of 36cases), and postoperative pathologic findings showed that 38.9%(12 cases of 28cases) were combined with tuberculosis. 6) Right upper and left upper lobes were predominantly involved(34.6% and 19.2% respectively) and lobectomies were performed in 21 cases. 7) Underlying diseases were present in 47 cases and 48.9% of them were pulmonary tuberculosis. Conclusion: These results showed that pulmonary aspergillosis usually develops in patients with open cavitary pulmonary tuberculosis. And we must consider the possibility of pulmonary aspergillosis in a patient with hemoptysis and cavitary lung lesion.

  • A Gram-negative bacterium was isolated from mushroom media that markedly reduces the level of extracellular toxins (i.e., tolaasins) produced by Pseudomonas tolaasii, the most destructive pathogen of cultivated mushrooms. The HC1 strain was selected as detoxifying tolaasin by bioassay on potato and it was identified Pseudomonas sp. by the cultural, morphological and physiological characteristics, and analysis of the 16S rRNA. The isolated bacterium is saprophytic but not parasitic nor pathogenic to cultivation mushroom. The isolated bacterium for P. tolaasii cell, was sufficient for detoxification in vitro. Inoculation of the isolated bacterium prevents the development of bacterial disease in Pleurotus ostreatus, Flammunia velutipes and Agaricus bisporus. Control efficacy of brown blotch of strain HC1 treatment was 69, 68 and 55% on Agaricus bisporus, Flammulina velutipes and Pleurotus ostreatus, respectively. The suppressive bacterium may be useful in future for the development of biocontrol system and the construction of genetically modified edible fungi resistant to the disease caused by P. tolaasii.

Diversity and Occurrence Frequency of Ectomycorrhizal Fruiting Bodies by Planting Sites (식재지별 외생균근성 버섯의 다양성 및 발생 빈도 비교)

  • This study was conducted to investigate the diversity and occurrence frequency of ectomycorrhizal fruit bodies by planting sites from June 2000 to October 2001. A total of 3 classes 3 subclasses 8 orders 22 families 41 genera and 72 species (including two varieties) including saprophytic and ectomycorrhizal fungi was investigated. The mushrooms are classified into 9 families 21 genera and 48 species in Agaricales, 5 families 11 genera and 13 species in Aphllophorales, 3 families 3 genera and 4 species in Heterobasidiomycetes and 5 families 6 genus and 7 species in Gasteromycetdae. A total of 7 families 11 genera 30 species (2,451 ea.) of ectomycorrhizal mushroom was investigated. The occurrence frequency of mushrooms was 1,225, 179 and 130 times for Laccaria vinaceoavellanea, Amanita longistriata and Laccaria amethystea, respectively. The mushroom occurrence of ectomycorrhizal fungi was closely related to climatic conditions such as high air temperature, relative humidity and lots of rainfall from July to August. Diversity and distribution of ectomycorrhizal fungi by plots were very different because of variable local environments and different host plants in experimental plots. Laccaria vinaceoavellanea has showed very low host range of plant specificity because of mushroom occurrence in only Quercus sp. and Amanita longistriata, Russula bella and Inocybe sp. have showed wide host range of plant specificity because of mushroom occurrence in coniferous and broadleaved trees. The environment which has a favorable influence of mushroom occurrence was soil pH, organic matter and T/N ratio of soil enviromental and humidity of climatic environment.

Fungi Associated with Soybean Seed, their Pathogenicity and Seed Treatment (콩 종자(種子)에서 분리(分離)한 사상균(絲狀菌), 그 병원성(病原性) 및 종자(種子) 소독(消毒)에 관한 연구(硏究))

  • Alternaria tenuis, Arthrobotrytis sp., Aspergillus spp., Cephalosporium sp., Cladosporium sp., Cylindrocarpon sp., Fusarium equiseti, F. moniliforme, F. semitectum, F. solani, Penicillium spp., Rhizopus sp. were saprophytic fungi and Cercospora kikuchii, Colletotrichum truncatum, Diaporthe phaseolorum var. sojae and Fusarium oxysporum were pathogenic fungi detected from 14 seed samples of soybean. Initial symptoms caused by C. kikuchii, C. truncatum, D. phaseolorum sojae and F. oxysporum on seedlings from naturally infected seed by the test tube agar method have been described and discussed. Soybean seeds infected with C. truncatum, D. phaseolorum sojae and F. oxysporum reduced germination of seeds and have influenced on the growth of soybean seedling caused by C. kikuchii. Surface-sterilized soybean seedlings became diseased in the test tube agar artificially inoculated with C. kikuchii, C. truncatum and D. phaseolorum sojae isolated from naturally infected soybean seeds. F. oxysporum showed very weak pathogenicity. Seed disinfectants of Benlate-T, Homai, Tecto and Sisthane have effective to C. kikuchii, C. truncatum, D. phaseolorum sojae and F. oxysporum. Arasan, Captan, Busan-30 and Mercron were inferior to C. kikuchii but effective against others. Seed disinfectants treated in this experiment have increased seed germination campared with non-treatment.

Effect of Different Abiotic Factors on Chemotaxis of Bacteria Towards Fungal Propagules (종류가 다른 무생물적 요인이 진균류에 미치는 세균의 주화성)

  • Chemotactic responses of five motile saprophytic and one phytopathogenic bacteria e.g. Agrobacterium radiobacter, Bacillus subtilis, B. polymyxa, Pseudomonas aeruginosa, P. fluorescens and Xanthomonas malvacearum towards exudate of Cochliobolus sativus conidia, Fusarium of oxysporum f. sp. ciceri chlamydospores, Macrophomina phaseolina sclerotia and Phytophthora drechsleri f. sp. cajani oospores were determined in vitro at different abiotic conditions. In general, a positive correlation (r=0.76 to 0.89; P=0.05) was observed between concentration of fungal exudates and attraction of bacterial cells. Similarly, a significant (P=0.05; r=+0.82 to 0.95) positive correlation was noticed between chemotactic response and incubation period. The chemotactic response of bacteria was greatly influenced by temperature and pH of the test fungal exudate. The optimum temperature for maximum chemotaxis was $25^{\circ}C$ for A. radiobacter, $30^{\circ}C$ for B. polymyxa, P. aerugionosa, P. fluorescens and X. malvacearum and $35^{\circ}C$ for B. subtilis. Fungal exudates maintained at pH 7 attracted maximum number of bacteria. The response of bacterial cells to exudates at pH 3 and 11 was not significantly (P=0.05) different than that to the buffer (control). Chemotaxis of bacteria was observed towards attractants (fungal propagules and their exudates) when they were kept apart and bridged with the capillaries filled with non-attractant (buffer) or attractant (exudate).

  • Present experiments were conducted to determine the possibility of cultivation of tropical and subtropical saprophytic fungus, Volvariella volvacea (Bull. ex Fr.) Sing. under Korean climatic conditions and the-possibility of applying the method of indoor cultivation which was modified from the cultivation method of the common mushroom, Agaricus bisporus (Lange) Sing.. The results obtained were as follows: 1. In conventional outdoor cultivation, the adequate cultivation period of this fungus was from around the end of June to the end of August. 2. When rice straws submerged in water for 5 days were peak-heated for 6 days in a mush room house, the yield of the fungus was higher than that obtained from the rice straws peak-heated immediately after outdoor composting. 3. The highest yield, 20.49kg per 100kg rice straws, was obtained from the water-soaked and peak-heated rice straws which were cased with a 2 cm layer of loam at 7 days after spawning.
