홍삼성분이 섬유아세포의 콜라겐 생합성과 MMP-1 활성에 미치는 영향 (Effect of Korean red ginseng on collagen biosynthesis and MMP-I activity in human dermal fibroblast)

  • 본 실험은 홍삼을 이용하여 주름 예방에 효과가 있는 건강 기능 식품을 개발하기 위한 기초 자료로 활용하고자 홍삼이 사람의 섬유아세포에 대한 콜라겐 생합성 정도, 콜라겐 분해효소 활성, 세포 증식, 항산화 활성에 미치는 영향을 조사하였다. 조사포닌은 홍삼물엑기스 (WE), 에탄올 엑기스 (EE), 에탄올엑기스를 88% 주정으로 용해시켜 정제한 엑기스 (PE) 에서 각각 72 mg/g, 107 mg/g, 220 mg/g으로 함유되어 있었다. 홍삼시료는 1 ${\mu}g/ml$, 5 ${\mu}g/ml$, 10 ${\mu}g/ml$의 농도로 첨가하여 섬유아세포를 배양하였다 . 콜라겐 생합성 정도는 대조군이 1.86 ${\mu}g/ml$이었고 Retinoic acid의 경우 0.03 ${\mu}g/ml$의 농도로 처리했을 때 2.85 ${\mu}g/ml$이었다 . 홍삼의 첨가농도 10 ${\mu}g/ml$을 기준으로 할 때 물엑기스에서는 대조군과 유사하였고, EE (ethanol extract)에서는 2.05 ${\mu}g/ml$, PE (purified ethanol extract)에서는 2.58 ${\mu}g/ml$, ginsenoside $Rg_1$은 2.01 ${\mu}g/ml$이었으며 $Rb_1$은 3.07 ${\mu}g/ml$의 콜라겐을 생성하여 대조군에 비하여 165% 증가하였으며 retinoic acid보다 더 높았다. MMP-1 활성에서는 10 ${\mu}g/ml$의 농도로 배양했을 때 EE, PE, $Rg_1$, $Rb_1$에서 대조군의 92%, 94%, 91%, 48%을 나타내어 $Rb_1$이 저해효과가 뚜렷하였다. MTT assay에서 모든 시료는 대조군에 비하여 84%${\sim}$96%의 수준으로 세포 생존율을 나타내어 의미있는 세포 독성을 나타내지 않았다. 항산화 정도를 나타내는 SOD 활성은 높은 수준은 아니었으나 Vit.C에 비하여 28${\sim}$79%의 활성을 나타내었다. 이상의 실험 결과를 종합해 볼 때 홍삼 시료는 사람의 섬유아세포에 대하여 콜라겐 생합성을 증가시키고 MMP-I 활성을 억제하였으며 항산화 효과를 나타내어 피부 주름 생성을 억제하는 효과가 있는 것으로 확인되었고, 홍삼의 사포닌 성분 중에서는 $ginsenoside-Rg_1$에 비하여 $ginsenoside-Rb_1$이 더 효과적이었다.

알칼리수를 이용한 홍삼 추출물의 품질특성 (Quality Characteristics of Red Ginseng Extracts Prepared Using Alkaline Water)

  • 본 연구는 알칼리수를 이용한 홍삼추출물의 품질특성을 조사하였다. 알칼리수의 미네랄 함량은 칼슘이 $61.73{\sim}180.63\;mg%$로 가장 높았으며, 이온화 무기염류의 첨가량이 높을수록 알칼리도는 증가하는 것으로 나타났다. AKRGE 3, 6 및 9의 pH는 $4.9{\sim}5.0$, 당도 $3.03{\sim}3.43^{\circ}Brix$, 가용성 고형분 $2.33{\sim}2.60%$, 갈색도 3.0으로 나타났으며 RGE와 큰 차이는 없었다. AKRGE 6과 AKRGE 9의 칼슘함량은 107.80 mg% 및 171.81 mg%로, RGE와 시판 홍삼 파우치 제품 5종에 비해서 약 $18{\sim}29$배 높게, 알칼리도는 약 5배$\sim$11배 이상 증가한 것으로 나타났다. 조사포닌 함량은 AKRGE가 RGE에 비해서 조금 낮게 나타났으나 ginsenoside 총량에는 큰 차이가 없었다. 알칼리성 홍삼추출물의 DPPH free radical 소거활성은 $12{\sim}13%$, superoxide radical 소거활성은 약 20%로 RGE와 큰 차이는 없었다. 관능검사 결과 AKRGE 3과 6은 RGE보다 쓴맛은 감소하고, 전반적인 기호도에서 더 높은 수치를 나타내었다.

압출성형 수삼의 추출수율과 추출물의 저온압출 구형과립화 (Extraction Yield of Extruded Ginseng and Granulation of Its Extracts by Cold Extrusion-Spheronization)

  • 압출성형공법을 이용한 수삼의 전처리를 통한 추출수율의 향상과 최적조건에서 열수 추출한 인삼추출액의 첨가량을 달리하여 저온압출공정으로 제조한 구형과립의 입자분포, 마모율, 용적밀도, 진동밀도, 압축지수와 용출패턴을 결정하였다. 열수추출수율에서 수삼을 압출성형할 때 수분함량이 가장 유의적으로 영향을 미쳤다. 수분함량의 감소와 함께 스크류 회전속도가 증가할수록 압출성형 수삼의 추출수율이 크게 증가하였다. 추출수율이 최대가 되는 압출성형 공정변수는 수분함량 15%, 스크류 회전속도 250 rpm, 사출구 부위온도 13$0^{\circ}C$였다. 동일한 추출조건에서 15개 압출성형 수삼 시료의 평균 추출수율(43.5%)은 홍삼(38.3%)과 백삼(29.0%) 추출수율과 비교하여 크게 향상되었다. 저온압출공정으로 압출성형 수삼농축액(고형분함량 59.2%)의 농도를 5, 20, 30, 40, 50, 60%로 각각 증류수에 희석한 용액 200 mL과 200 g 미세결정 셀룰로오즈를 이용하여 과립화시킬 수 있는 압출성형 수삼추출액의 최대 농도는 60%였으며 추출액의 증가와 함께 입자수율의 증가, 마모율의 감소, 압출지수의 감소 등으로 입자의 물리적 특성은 구형과립을 젤라틴 캡슐에 포장하여 제품화할 수 있었다. 또한 압출성형 수삼 추출액을 포함한 구형과립으로부터 농축액 성분이 10분 이내에 쉽게 용출되므로 구형과립은 젤라틴 캡슐에 적합하다는 결론을 내릴 수가 있었다.

홍삼 사포닌의 항불안 효과 (Anxiolytic Effect of Ginseng Total Saponin)

  • 홍삼사포닌분획의 항불안작용의 양상을 검색하고자 사포닌분획을 사용하여 실험 동물에서 항불안작용을 검색하고 이들의 작용을 benzodiazepine의 대표적 약물인 diazepam의 효과와 비교하여 보았다. 실험동물에서 항불안효과의 검정을 위하여 생쥐에게 각각 상이 한 단일성분 함량을 지닌 여러분획들을 투여 후 elevated plus maze법을 사용하여 항불안효과를 비교, 관찰하였다. 실험결과 홍삼 ginsenoside Rg1, Rf 및 Re 등의 함량이 보다 높은 TSI 분획에서 항불안 효과를 관찰하였다. 중추신경계에 작용하여 진정효과를 나타내는 약물 중 특히 항불안제의 작용에 관여하는 신경전달물질 수용체로 현재까지 가장 주목을 받고 있는 것은 GABA A-benzodiazepine 수용체-chloride통로 복합체에 있는 benzodiazepine 수용체이다. 본 실험결과 관찰한 홍삼 사포닌의 항불안작용의 기전 규명을 위한 접근방법으로 홍삼 사포닌의 benzodiazepine 수용체에 대한 수용체 결합력을 관찰하고자, 흰쥐의 대뇌 피질을 검체로 하여 benzodiazepine 수용체에 대한 [$^3H$] Ro15-1788 결합 실험을 실시하였다. 홍삼 사포닌 분획들은 benzodiazpine 수용체에 결합하는 반응성을 보였으며, ginsenoside Rb, Re 및 Rd등의 함량이 높은 TSIV 분획이 가장 높은 결합력을 나타내었는데, 이 분획에서는 항불안 작용을 관찰할 수 없었다. 이상의 결과에서 볼 때 홍삼사포닌은 항불안작용을 나타내었으며, 이 항불안작용과 benzodiazepine 수용체에 대한 결합력과의 연관성은 관찰되지 않았다.

Traditional quality measures were elucidated in accordance with modern analytical chemistry resulting in the importance of peptides in the centrat part rather than ginsenosides For large root production growth promoting rootzone micrcorganisms (PGPRM) were isolated and active compounds were identified. Field test on PGPRM was on going. Varictus methods formality improvement through cultivation were developed. Management research of ginseng production was rare Extension was active throuch official and private organizations and through workshop for the extension specialists, and direct lectures to grower's. Extension services made the researcher to understand the existing problems at grower's fields. Research environment for ginseng production was in prime time only for three years when Korea Ginseng Research Institute was established then gradually aggravated.

인삼의 항암작용에 대한 한의학 관련 논문 분석 (Oriental Medicine papers review on Anticancer Effect of Ginseng)

  • Backgrounds: Multidisciplinary approaches including surgery, chemotherapy, and radiation therapy are currently being performed to target various cancers in Western Medicine. However, some cancers still remain difficult to battle, which has long attracted many scientists for the discovery of new agents to fight cancers. Ginseng is one of the herbs used in Oriental Medicine including Korea, China and Japan. We have further investigated ginseng for its anticancer effect. Objective: This is a comprehensive review summary of anticancer effect of ginseng and ginsenoids as a possible agent for future cancer treatment. Methods: Data were retrieved from two web sites; www.pubmed.com and www.riss.kr, and authorized texts concerning anticancer effects of ginseng. From collected data, information on anticancer effect of ginseng was thoroughly sorted, restructured, then assessed. Results: Panax Ginseng C.A. Meyer belongs to Araliaceae Panax family, a perennial prairie plant with its root known as Ginseng Radix. Ginseng induces anticancer effect through cell cycle arrest, acceleration of apoptosis, anti-angiogenesis, and suppression of metastasis. Anticancer effect of ginseng may be due to single compound or multi-compound actions. Many studies report involvement of immune mechanisms of cytokines, Natural Killer (NK) cells, macrophages and some antibodies in enhancing anticancer effect of ginseng. In near future, possibility of applying these mechanisms into clinical trials is convinced. There were some important findings on saponin in ginsenoids in reviewing for this article; First, eradication of metastatic tumors were influenced by macrophage activation. Second, suppression of malignant melanoma cell metastasis to lung were induced by macrophage and NK cell activation in spleen with red ginseng acidic polysaccharide (RGAP). Third, final metabolites of M1, M4 had exerted anticancer effect of ginseng. Conclusion: Unknown anticancer mechanisms of ginseng have been studied for many years up until now. Ginseng is comprised of multiple bio-chemical compounds that create complex pharmaceutical interactions. Therefore, for its proper usage and safe prescription, studies on different types of ginseng and patients' susceptibility to ginseng according to their constitution and stages of the disease should be further pursued. More efforts are needed to understand the anticancer mechanisms of ginseng as well.

반복 스트레스에 의한 흰쥐 해마조직내 신경전구세포의 생성과 brain-derived neurotrophic factor (BDNF) mRNA 발현 변동에 미치는 고려홍삼 사포닌의 반복 투여 효과 (Effects of Korea Red Ginseng Total Saponin on Repeated Unpredictable Stress-induced Changes of Proliferation of Neural Progenitor Cells and BDNF mRNA Expression in Adult Rat Hippocampus)

  • 본 연구 결과를 통하여 홍삼 성분인 고려홍삼 사포닌을 반복 투여시 흰쥐 해마 SGZ 부위의 신경전구세포 생성이 유의하게 증가되었으며, 이와 같은 경향은 반복 스트레스에 노출되어도 유지되었다. 또한 스트레스를 가하지 않은 흰쥐에서 고려홍삼 사포닌 반복 투여시 해마 CA3와 CA1 부위에서 BDNF mRNA의 발현이 증가되었으나, 반복 스트레스를 가한 흰쥐의 CA3와 CA1부위에서 BDNF mRNA의 감소를 차단하지는 못하였다. 따라서 고려홍삼 사포닌 반복 처치에 의한 해마 신경전구세포의 생성에 BDNF 보다는 다른 요인이 관여할 가능성이 클 것으로 추정된다.

고온 열처리가 홍삼물추출물의 이화학적 특성에 미치는 영향 (Effects of High Temperature Heating on the Some Physicochemical Properties of Korean Red Ginseng (Panax ginseng C.A. Meyer) Water Extract)

  • This study was carried out to investigate the some physicochemical properties of Korean red ginseng (Panax ginseng C.A. Meyer) water extract (RGWE) after heated with high temperatures above $100^{\circ}C$ for 2 hours. RGWEs were heated at 100, 110 and $120^{\circ}C$ for 2 hours by using autoclave. After RGWEs were heated at high temperature for 2 hours without not adjustment of pH, the changes of saponin, free sugars, mineral and color in the RGWEs were investigated. Total ginsenoside content in control was 1.99%, while those of RGWE were 1.65, 1.49 and 1.29% when treated at 100, 110 and $120^{\circ}C$, respectively. The contents of total ginsenoside showed decreased tendency as heating temperatures were increased. The ginsenoside-$Rh_{2}$ and $-Rg_{3}$, which have been reported as very stable red ginseng ginsenosides, showed relatively strong spots on TLC when RGWEs were heated at 110 and $120^{\circ}C$. In case of free sugars in RGWEs, fructose, glucose and maltose showed high contents when compared with control, while Fe, Ca and Mg ions showed very low contents. Value of L in RGWE treated with high temperature was almost the same with control, while values of a and b were increased. Values of a were increased from -0.86 of control to +0.04, +0.05 and +1.14 when treated with 100, 110 and $120^{\circ}C$, respectively. Values of b also were increased from 27.68 of control to 33.61, 33.61 and 37.42 when treated with 100, 110 and $120^{\circ}C$, respectively. Values of total color in RGWEs treated with high temperatures, E, were finally increased by values of a and b.

Correlation between the Content and Pharmacokinetics of Ginsenosides from Four Different Preparation of Panax Ginseng C.A. Meyer in Rats

  • We aimed to compare the content of ginsenosides and the pharmacokinetics after the oral administration of four different ginseng products at a dose of 1 g/kg in rats. The four different ginseng products were fresh ginseng extract, red ginseng extract, white ginseng extract, and saponin enriched white ginseng extract prepared from the radix of Panax ginseng C.A. Meyer. The ginsenoside concentrations in the ginseng product and the rat plasma samples were determined using a liquid chromatography-tandem mass spectrometry (LC-MS/MS). Eight or nine ginsenosides of the 15 tested ginsenosides were detected; however, the content and total ginsenosides varied depending on the preparation method. Moreover, the content of triglycosylated ginsenosides was higher than that of diglycosylated ginsenosides, and deglycosylated ginsenosides were not present in any preparation. After the single oral administrations of four different ginseng products in rats, only four ginsenosides, such as 20(S)-ginsenosides Rb1 (GRb1), GRb2, GRc, and GRd, were detected in the rat plasma samples among the 15 ginsenosides tested. The plasma concentrations of GRb1, GRb2, GRc, and GRd were different depends on the preparation method but pharmacokinetic features of the four ginseng products were similar. In conclusion, a good correlation between the area under the concentration curve and the content of GRb1, GRb2, and GRc, but not GRd, in the ginseng products was identified and it might be the result of their higher content and intestinal biotransformation of the ginseng product.

백삼 및 백미삼 추출물의 초단파 및 식초 처리에 의한 인삼 사포닌 성분 변화 (The Change of Ginsenoside Composition in White Ginseng and Fine White Ginseng Extract by the Microwave and Vinegar Process)

  • The purpose of this study is to develop a new preparation process of ginseng extracts having high concentrations of ginsenoside $Rg_3$, $Rg_5$ and $Rk_1$, a special component of Red ginseng. Chemical transformation from ginseng saponin glycosides to prosapogenin was analyzed by the HPLC. Extracts of White ginseng (Panax ginseng) and Fine White ginseng were processed under several treatment conditions including microwave and vinegar (about 14% acidity) treatments. Results of those treatments showed that the quantity of ginsenoside $Rg_3$ increased by over 0.6% at 4 minutes of pH 2~4 vinegar and microwave treatments. The results of processing with MWG-4 indicate that the Microwave and vinegar processed white ginseng extracts (about 14% acidity) that had gone through 4-minute treatments were found to contain the largest amount of ginsenoside $Rg_3$ (0.626%), $Rg_5$ (0.514%) and $Rk_1$ (0.220%). Results of treatments with MFWG-5 showed that the Fine White ginseng extracts that had been processed with microwave and vinegar (about 14% acidity) for 5 minutes were found to contain the largest amount of ginsenoside $Rg_3$ (4.484%), $Rg_5$ (3.192%) and $Rk_1$ (1.684%). It is thought that such results provide basic information in preparing White ginseng and Fine White ginseng extracts with functionality enhanced.
